| 1 | Schistosoma mansoni proteins attenuate gastrointestinal motility disturbances during experimental colitis in mice显示文摘AIM:To investigate the therapeutic effect of Schistosoma mansoni(S.mansoni) soluble worm proteins on gastrointestinal motility disturbances during experimental colitis in mice. METHODS:Colitis was induced by intrarectal injection of trinitrobenzene sulphate(TNBS) and 6 h later,mice were treated ip with S.mansoni proteins.Experiments were performed 5 d after TNBS injection.Inflammationwas quantified using validated inflammation parameters. Gastric emptying and geometric center were measured to assess in vivo gastrointestinal motility.Peristaltic activity of distal colonic segments was studied in vitro using a modified Trendelenburg set-up.Cytokine profiles of T-lymphocytes isolated from the colon were determined by real time reverse transcriptase-polymerase chain reaction. RESULTS:Intracolonic injection of TNBS caused severe colitis.Treatment with S.mansoni proteins significantly ameliorated colonic inflammation after 5 d.TNBS did not affect gastric emptying but significantly decreased the geometric center and impaired colonic peristaltic activity 5 d after the induction of colitis.Treatment with S.mansoni proteins ameliorated these in vivo and in vitro motility disturbances.In addition,TNBS injection caused a downregulation of effector T cell cytokines after 5 d,whereas a S.mansoni protein effect was no longer observed at this time point. | 2 | Weather Information Acquisition and Health Significance during Extreme Cold Weather in a Subtropical City:A Crosssectional Survey in Hong Kong显示文摘Health and disaster risk reduction are important and necessary components in building a smart city,especially when climate change may increase the frequency of extreme temperatures and the health risks of urban dwellers.However,limited knowledge is available about the best way to disseminate weather warnings and health protection information.This study explores the weather information acquisition patterns of the Hong Kong public and examines the sociodemographic predictors of these patterns to establish the potential public health implications of smart city development.A population-based,stratified crosssectional,random digit dialing telephone survey was conducted among the Cantonese-speaking population aged over 15 years in Hong Kong in early 2016.Analyses were conducted based on 1017 valid samples,with a response rate of 63.6%.Cold Weather Warnings were well disseminated in Hong Kong,with 95.7% of the respondents reporting awareness of the public warnings.Television and smartphone apps were the two most important channels for weather information acquisition.Age and education level are the main social-demographic variables associated with the current utilization and future preference of smartphone technology.Among those who were not using a preferredchannel to acquire weather information,61.3% considered switching to a smartphone app.Moreover,the patterns of individual health protection measures and self-reported health impacts were significantly different between smartphone app users and non-users.Weather information dissemination should be tailored to the sociodemographic characteristics of the users. | Emily Ying Yang Chan Zhe Huang Carman Ka Man Mark Chunlan Guo | 2017 | International Journal of Disaster Risk Science2017,8,2: | 2 |

| 20 | MicroRNA-17-3p suppresses NF-κB-mediated endothelial inflammation by targeting NIK and IKKβ binding protein显示文摘Nuclear factor kappa B(NF-κB)activation contributes to many vascular inflammatory diseases.The present study tested the hypothesis that microRNA-17-3p(miR-17-3p)suppresses the pro-inflammatory responses via NF-κB signaling in vascular endothelium.Human umbilical vein endothelial cells(HUVECs),transfected with or without miR-17-3p agomir/antagomir,were exposed to lipopolysaccharide(LPS),and the inflammatory responses were determined.The cellular target of miR-17-3p was examined with dual-luciferase reporter assay.Mice were treated with miR-17-3p agomir and the degree of LPS-induced inflammation was determined.In HUVECs,LPS caused upregulation of miR-17-3p.Overexpression of miR-17-3p in HUVECs inhibited NIK and IKKβ binding protein(NIBP)protein expression and suppressed LPS-induced phosphorylation of inhibitor of kappa Bα(IκBα)and NF-κB-p65.The reduced NF-κB activity was paralleled by decreased protein levels of NF-κB-target gene products including pro-inflammatory cytokine[interleukin 6],chemokines[interleukin 8 and monocyte chemoattractant protein-1]and adhesion molecules[vascular cell adhesion molecule-1,intercellular adhesion molecule-1 and E-selectin].Immunostaining revealed that overexpression of miR-17-3p reduced monocyte adhesion to LPS-stimulated endothelial cells.Inhibition of miR-17-3p with antagomir has the opposite effect on LPS-induced inflammatory responses in HUVECs.The anti-inflammatory effect of miR-17-3p was mimicked by NIBP knockdown.In mice treated with LPS,miR-17-3p expression was significantly increased.Systemic administration of miR-17-3p for 3 days suppressed LPS-induced NF-κB activation and monocyte adhesion to endothelium in lung tissues of the mice.In conclusion,miR-17-3p inhibits LPS-induced NF-κB activation in HUVECs by targeting NIBP.The findings therefore suggest that miR-17-3p is a potential therapeutic target/agent in the management of vascular inflammatory diseases. | Yin Cai Yu Zhang Hui Chen Xing-hui Sun Peng Zhang Lu Zhang Meng-yang Liao Fang Zhang Zheng-yuan Xia Ricky Ying-keung Man Mark WFeinberg Susan Wai-Sum Leung | 2021 | Acta Pharmacologica Sinica2021,42,12: | 1 |
