2Chronic consumption of thermally processed palm oil or canola oil modified gut microflora of rats显示文摘Dietary oils have critical influences on human health,and thermally cooking or frying modify the components and nutritional functions of oils.Palm oil was the most widely used oil in food processing industry,but its health effects remain debatable.In the current study,we aimed to compare the effects of thermally oxidized palm oil and canola oil on gut microbiota.Palm oil or canola oil were heated at 180°C for 10 h to prepare high-fat diets.Rats were fed high-fat diets for 3 months,and hematological properties,gut microflora composition and intestinal gene expression were examined.The results indicated that heated canola oil consumption elevated plasma total cholesterol and LDL-c levels compared with unheated canola oil,but heated palm oil do not had these effects;and consumption of heated palm oil significantly elevated the relative abundance of Lactobacillucs and Roseburia in gut,compared with non-heated palm oil or two canola oil groups.Moreover,intestinal expression of IL-22 was increased in heated palm oil fed animal,though ZO-1 and GPR41 were reduced.In conclusion,heating process may enhance the effects of palm oil on proliferation of probiotics Lactobacillucs,and weaken the effects of canola oil on cholesterol transport and metabolism.Mengcheng Ruan Yiran Bu Fangjie Wu Shijie Zhang Rulong Chen Na Li Zhiguo Liu Hualin Wan 2021Food Science and Human Wellness2021,10,1:1
3Synergistic Balance of Strength and Corrosion Resistance in Al-Mg-Er Alloys显示文摘High-strength,corrosion-resistant,and lightweight Al-Mg alloys perform an important function in harsh coastal service environments.Corrosion resistance is generally inversely correlated with strength;hence,it is difficult to simultaneously optimize both.In this study,a low-magnesium Er-containing Al-based alloy that is stronger and more corrosion resistant than Al-based alloys have been reported.The alloy contains Er,and the precipitation of Al3 Er within a face-centered cubic matrix is obtained by a series of smelting-casting,heat treatment,and rolling processes.It is presumed that the strengthening phase of Al3 Er pinning dislocations improves alloy strength.It also increases the recrystallization temperature of cold-rolled matrix and induces the distribution of small-angle grain boundaries,thus allowing the alloy to achieve excellent environmental corrosion resistance.As a result,strength and corrosion resistance are simultaneously improved.Haifei Zhou Zhouhai Qian Mengcheng Zhou Xuebing Liu Yong Li Xinfang Zhang 2020Acta Metallurgica Sinica(English Letters)2020,33,5:1
4PSPC1 regulates CHK1 phosphorylation through phase separation and participates in mouse oocyte maturation显示文摘Liquid–liquid phase separation (LLPS) underlies the formation of membraneless compartments in mammal cells. However, there are few reports that focus on the correlation of mouse oocyte maturation with LLPS. Previous studies have reported that paraspeckle component 1 (PSPC1) is related to the occurrence and development of tumors, but whether PSPC1 functions in mouse oocyte maturation is still unclear. Sequence analysis of PSPC1 protein showed that it contains a prion-like domain (PrLD) that is required for phase separation of proteins. In this study, we found that PSPC1 could undergo phase separation. Moreover, the loss of PrLD domain of PSPC1 could greatly weaken its phase separation ability. The immunofluorescence assays showed that PSPC1 is present in mouse oocytes in the germinal vesicle (GV) stage. Knockdown of PSPC1 significantly impeded the maturation of mouse oocytes in vitro. CHK1 has been reported to play important roles in the GV stage of mouse oocytes. Co-IP experiment revealed that PSPC1 could interact with phosphatase serine/threonine-protein phosphatase 5 (PPP5C), which regulates CHK1 phosphorylation. Western blot analysis revealed that PSPC1 could regulate the phosphorylation of CHK1 through PPP5C;however, PSPC1 without PrLD domain was inactive, suggesting that the lack of phase separation ability led to the abnormal function of PSPC1 in regulating CHK1 phosphorylation. Thus, we conclude that PSPC1 may undergo phase separation to regulate the phosphorylation level of CHK1 via PPP5C and participate in mouse oocyte maturation. 5Per1/Per2 double knockout transcriptome analysis reveals circadian regulation of hepatic lipid metabolism显示文摘Scope: Circadian disorder and high-fat diet(HFD)can disturb lipid metabolism homeostasis and may promote the development of various metabolic diseases. The relationship between them is of great concern. This study aimed to explore the effects of Per1/Per2 double knockout(DKO)on hepatic lipid metabolism in mice under HFD and HFD with docosahexaenoic acid(DHA)substitution. Methods and results: Both wild type(WT)and DKO male C57BL/6 mice were fed with normal chow diet(CON), HFD, or HFD with DHA substitution(AO)for 15 weeks. At the end of the experiment, mice were sacrificed at zeitgeber time(ZT)0(7:00 am)or ZT12(7:00 pm). Pathological indicators were determined using histological and biochemical methods. Hepatic transcriptome sequencing analysis showed that DKO mice exhibited multiple dysfunctions in diurnal rhythm, drug metabolism, cell cycle, cancer pathways, and lipid metabolism. HFD had greater effects on fatty acid oxidation and cholesterol synthesis and metabolism in Per1-/-Per2-/-mice, which was improved by DHA substitution. Conclusions: Per1/Per2 played an important role in the circadian regulation of hepatic lipid metabolism, and DKO mice were more sensitive to HFD. 6Transport signatures of the topological surface state induced by the size effect in superconductor β-PdBi_(2)显示文摘The superconductivity and nontrivial topological electronic state are key hallmarks of topological superconductors.Here,we focus on the transport signals of possible topological surface state in the topological superconductor candidate β-PdBi_(2) nanoflake with a thickness of 21 nm.The resistance demonstrates a semiconductor-metal transition followed by an upturned behavior as the temperature decreases.A large and unsaturated longitudinal magnetoresistance(MR),accompanied by distinct Shubnikov-de Hass oscillation in Hall resistance,is observed.An analysis of Hall resistance reveals that the carriers present the characteristics of relativistic particles with small effective mass and extremely high mobility.The angle-dependent quantum oscillations demonstrate a two-dimensional Fermi surface topology.A giant anisotropic MR as large as 98%is detected when rotating the magnetic field.These results provide the possible transport signals of a nontrivial topological electronic state,establishing a further understanding of the topological properties of the low-dimensional topological superconductor candidate β-PdBi_(2).Ankang Zhu Zheng Chen Yuyan Han Mengcheng Zhu Huanhuan Wang Minglong Han Liang Li Xue Liu Guolin Zheng Xiangde Zhu Wenshuai Gao Mingliang Tian 2023Science China(Physics,Mechanics & Astronomy)2023,66,7:0

9Effect of soil surface roughness on emergence rate and yield of mechanized direct-seeded rapeseed based on 3D laser scanning显示文摘The quality of seedbed after sowing such as soil surface roughness is one of the key factors affecting the seedling emergence of rapeseed,which ultimately affected crop yield.However,the effect of soil surface roughness on seedling emergence and yield of rapeseed is still unclear.In this study,field experiments at the experimental site of Jianli and Shayang were carried out.Three treatments were designed:relative slow(M1),medium(M2),and fast(M3)forward speed of the unit.Soil surface roughness measured by a 3D laser scanner,seed quantity of the seeder,emergence rate and yield of rapeseed were determined to investigate the soil surface roughness effect on emergence rate and yield of rapeseed.The results showed that as the forward speed of the unit increased,the compartment surface became rougher.Compared with the M1 and M2 treatments,soil surface roughness under the M3 treatment increased by 36.5%and 9.8%,respectively.The actual seed quantity of the seeder under different treatments ranged from 3806.56 to 4158.18 g/hm2.The average error rate of the actual and theoretical seed quantity was less than 5%,which met the operational quality requirements for seeding rapeseed crops.As the forward speed of the unit increased,the actual seed quantity of the seeder gradually increased while the emergence rate and yield of rapeseed decreased.The seed quantity under the M3 treatment increased by 6.9%and 4.7%,while the emergence rate of rapeseed decreased by 3.3%and 2.0%,and the yield decreased by 23.2%and 13.1%,compared with the M1 and M2 treatments,respectively.Correlation analysis indicated that emergence rate and yield of rapeseed were negatively influenced by soil surface roughness.Considering rapeseed emergence rate,seed yield,and economic benefits,the M1 treatment was recommended.But considering the factor that the M1 treatment may reduce the unit operation efficiency,and thus resulting in lower cost of production,M2 could be recommended in actual farming.The results of this study laid a theoretical foundation for analyzing the relationship between the seedbed surface quality and seedling emergence and yield.Hui Chen Liping Gao Mengcheng Li Yitao Liao Qingxi Liao 2023International Journal of Agricultural and Biological Engineering2023,16,3:0
10Nuclear localization of human MEIOB requires its NLS in the OB domain and interaction with SPATA22显示文摘MEIOB is a vital protein in meiotic homologous recombination and plays an indispensable role in human gametogenesis.In mammals,MEIOB and its partner SPATA22 form a heterodimer,ensuring their effective localization on single-strand DNA(ssDNA)and proper synapsis processes.Mutations in human MEIOB(hMEIOB)cause human infertility attributed to the failure of its interaction with human SPATA22(hSPATA22)and ssDNA binding.However,the detailed mechanism is still unclear.In our study,truncated or full-length hMEIOB and hSPATA22 are traced by fused expression with fluorescent proteins(i.e.,copGFP or mCherry),and the live cell imaging system is used to observe the expression and localization of the proteins.When transfected alone,hMEIOB accumulates in the cytoplasm.Interestingly,a covered NLS in the OB domain of hMEIOB is identified,which can be exposed by hSPATA22 and is necessary for the nuclear localization of hMEIOB.When hSPATA22 loses its hMEIOB interacting domain or NLS,the nuclear localization of hMEIOB is aborted.Collectively,our results prove that the NLS in the OB domain of hMEIOB and interaction with hSPATA22 are required for hMEIOB nuclear localization.Yating Xu Zhen Chen Panfeng Wu Wei Qu Hanqing Shi Muyang Cheng Yumin Xu Tingyu Jin Cong Liu Cong Liu Yi Li Mengcheng Luo 2023Acta Biochimica et Biophysica Sinica2023,55,1:0
