Architectural proteins for the formation and maintenance of the 3D genome显示文摘Eukaryotic genomes are densely packaged into hierarchical three-dimensional(3D) structures that contain information about gene regulation and many other biological processes. With the development of imaging and sequencing-based technologies, 3D genome studies have revealed that the high-order chromatin structure is composed of hierarchical levels, including chromosome territories, A/B compartments, topologically associated domains, and chromatin loops. However, how this chromatin architecture is formed and maintained is not completely clear. In this review, we introduce experimental methods to investigate the 3D genome, review major architectural proteins that regulate 3D chromatin organization in mammalian cells, such as CTCF(CCCTC-binding factor), cohesin, lamins, and transcription factors, and discuss relevant mechanisms such as phase separation. | Mengfan Li Jingbo Gan Yuao Sun Zihan Xu Junsheng Yang Yujie Sun Cheng Li | 2020 | Science China(Life Sciences)2020,63,6: | 7
Reconstruction methodology for rational secret sharing based on mechanism design显示文摘Rational secret sharing[1]is the intersection of game theory[2]and traditional secret sharing[3].In rational secret sharing, the players are selfish and always behave in accordance with profit maximization. What this implies, primarily,is that rational players prefer obtaining the secret rather than not obtaining it and, secondarily, that players prefer as few other players as possible obtaining the secret.Motivated by selfishness, rational players either keep silent or send incorrect shares during the reconstruction phase, thereby the | Hai LIU Xinghua LI Jianfeng MA Mengfan XU Jingjing GUO | 2017 | Science China(Information Sciences)2017,60,8: | 3
Metabolic regulation mechanism of Aconiti Radix Cocta extract in rats based on ^(1)H-NMR metabonomics显示文摘Objective:To establish a metabonomics research technique based on the combination of^(1)H-NMR and multivariate statistical analysis,so as to explore the metabolic regulation mechanism of Aconiti Radix Cocta extract(ARCE)in rat tissues and serum. Methods:SD rats were randomly divided into blank group, female group and male group.The^(1)H-NMR technique was used to collect the information of rat tissues and serum samples in each group. The principal component analysis(PCA),partial least squares discriminant analysis(PLS-DA)and other methods were used for data pattern recognition,so as to screen out potential differential metabolites and metabolic pathways, and then network analysis and KEGG database were used to analyze the relationship between metabolites, metabolic pathways and diseases. Results:The external features and^(1)H-NMR analysis showed that the sex of rats had no obvious effect on the drug action.A total of 15 potential differential metabolites and six metabolic pathways were screened out through data pattern recognition.Through network analysis and KEGG pathway analysis, three target diseases closely related to differential metabolites were found, and the metabolic pathway related to lung cancer was the central carbon metabolism of cancer. Conclusion:This study shows that Aconiti Radix Cocta(ARC)may regulate the energy metabolism of the body by influencing arginine synthesis,so as to play the roles of anti-inflammation,analgesia,anti-tumor and immune regulation. | Wenfang Jin Jianli Bi Sheng Xu Mengfan Rao Qi Wang Yan Yuan Baolei Fan | 2022 | Chinese Herbal Medicines2022,14,4: | 1
Stretchable polymer composites with ultrahigh piezoelectric performance显示文摘Flexible piezoelectric materials capable of withstanding large deformation play key roles in flexible electronics.Ferroelectric ceramics with a high piezoelectric coefficient are inherently brittle, whereas polar polymers exhibit a low piezoelectric coefficient. Here we report a highly stretchable/compressible piezoelectric composite composed of ferroelectric ceramic skeleton, elastomer matrix and relaxor ferroelectric-based hybrid at the ceramic/matrix interface as dielectric transition layers, exhibiting a giant piezoelectric coefficient of 250 picometers per volt, high electromechanical coupling factor keff of 65%,ultralow acoustic impedance of 3MRyl and high cyclic stability under 50%compression strain. The superior flexibility and piezoelectric properties are attributed to the electric polarization and mechanical load transfer paths formed by the ceramic skeleton, and dielectric mismatch mitigation between ceramic fillers and elastomer matrix by the dielectric transition layer. The synergistic fusion of ultrahigh piezoelectric properties and superior flexibility in these polymer composites is expected to drive emerging applications in flexible smart electronics. | Tongxiang Tang Zhonghui Shen Jian Wang Shiqi Xu Jiaxi Jiang Jiahui Chang Mengfan Guo Youjun Fan Yao Xiao Zhihao Dong Houbing Huang Xiaoyan Li Yihui Zhang Danyang Wang Long-Qing Chen Ke Wang Shujun Zhang Ce-Wen Nan Yang Shen | 2023 | National Science Review2023,10,8: | 1
Novel potential XOD inhibitory peptides derived from Trachinotus ovatus:Isolation, identification and structure-function analysis显示文摘This study aimed to isolate and identify the novel potential Xanthine oxidase(XOD)inhibitory peptides derived from Trachinotus ovatus hydrolysate(TOH)(hydrolysis by neutral protease),and investigate their structure-function relationship.Initially,TOH was separated to four fractions(F1–F4)by gel filtration chromatography and F4 was chosen with higher XOD inhibitory rate(85.16%±0.566,25 mg/mL)and lower molecular weight(95.73%,<1000 Da).F4 was further characterized by reversed-phase high performance liquid chromatography(RP-HPLC)coupled with Nano-HPLC-MS/MS and four peptides(FPAW,LLPW,WLLP and FHLP)were identified with IC50 value of 3.81±0.18 mM,4.17±0.12 mM,43.06±0.73 mM,and over 50.00 mM respectively. It can be concluded that Trp-containing peptides especially located in C-terminal could inhibit XOD effectively. Moreover, the XOD-inhibiting actions of these peptides involvedπ-πstacking(Phe914 and Phe1009 in XOD),hydrogen bonding via molecular docking.Lineweaver-Burk equation showed the inhibition type of FPAW was a mixed inhibition.Circular dichroism(CD)spectra indicated FPAW could combine and change the secondary structure of XOD.Therefore, two peptides of FPAW and LLPW might have potential to be used to alleviate hyperuricemia in the future. | Mengfan Hou Huan Xiang Xiao Hu Shengjun Chen Yanyan Wu Jiachao Xu Xianqing Yang | 2022 | Food Bioscience2022,47,3: | 1
Fully Printed High‑Performance n‑Type Metal Oxide Thin‑Film Transistors Utilizing Coffee‑Ring Effect显示文摘Metal oxide thin-films transistors(TFTs)produced from solution-based printing techniques can lead to large-area electronics with low cost. However, the performance of current printed devices is inferior to those from vacuum-based methods due to poor film uniformity induced by the“coffeering”effect. Here,we report a novel approach to print highperformance indium tin oxide(ITO)-based TFTs and logic inverters by taking advantage of such notorious effect.ITO has high electrical conductivity and is generally used as an electrode material. However,by reducing the film thickness down to nanometers scale, the carrier concentration of ITO can be effectively reduced to enable new applications as active channels in transistors. The ultrathin(~10-nm-thick)ITO film in the center of the coffee-ring worked as semiconducting channels, while the thick ITO ridges(>18-nm-thick)served as the contact electrodes. The fully inkjet-printed ITO TFTs exhibited a high saturation mobility of 34.9 cm2 V^(−1) s^(−1) and a low subthreshold swing of 105 mV dec^(−1).In addition, the devices exhibited excellent electrical stability under positive bias illumination stress(PBIS,ΔV_(th)=0.31 V)and negative bias illuminaiton stress(NBIS,ΔV_(th)=−0.29 V)after 10,000 s voltage bias tests.More remarkably, fully printed n-type metal–oxide–semiconductor(NMOS)inverter based on ITO TFTs exhibited an extremely high gain of 181 at a low-supply voltage of 3 V,promising for advanced electronics applications. | Kun Liang Dingwei Li Huihui Ren Momo Zhao Hong Wang Mengfan Ding Guangwei Xu Xiaolong Zhao Shibing Long Siyuan Zhu Pei Sheng Wenbin Li Xiao Lin Bowen Zhu | 2021 | Nano-Micro Letters2021,13,11: | 1
Breaking the responsivity-speed dilemma of a-GaO_(x) photodetector by alternating gate modulation显示文摘The gallium oxide(Ga_(2)O_(3))based photodetectors are usually confronted with the typical challenge of the responsivity-speed(RS)dilemma,namely high photocurrent induces a long decay time,as shown in Figure 1(a).The RS dilemma involves a trade-off issue, which is independent of photoelectric materials. | Zhongfang ZHANG Pengju TAN Xiaohu HOU Xiaolan MA Mengfan DING Shunjie YU Guangwei XU Xiaolong ZHAO Shibing LONG | 2023 | Science China(Information Sciences)2023,66,12: | 0
Nutrition in Alzheimer's disease:a review of an underappreciated pathophysiological mechanism显示文摘Alzheimer's disease(AD)is the leading cause of dementia in older individuals and is an escalating challenge to global public health.Pharmacy therapy of AD is one of the well-funded areas;however, little progress has been made due to the complex pathogenesis.Recent evidence has demonstrated that modifying risk factors and lifestyle may prevent or delay the incidence of AD by 40%,which suggests that the management should pivot from single pharmacotherapy toward a multipronged approach because AD is a complex and multifaceted disease.Recently, the gut-microbiota-brain axis has gained tremendous traction in the pathogenesis of AD through bidirectional communication with multiple neural, immune, and metabolic pathways, providing new insights into novel therapeutic strategies.Dietary nutrition is an important and profound environmental factor that influences the composition and function of the microbiota. The Nutrition for Dementia Prevention Working Group recently found that dietary nutrition can affect cognition in AD-related dementia directly or indirectly through complex interactions of behavioral, genetic, systemic, and brain factors. Thus, considering the multiple etiologies of AD,nutrition represents a multidimensional factor that has a profound effect on AD onset and development. However, mechanistically, the effect of nutrition on AD is uncertain;therefore, optimal strategies or the timing of nutritional intervention to prevent or treat AD has not been established. Thus, this review summarizes the current state of knowledge concerning nutritional disorders,AD patient and caregiver burden, and the roles of nutrition in the pathophysiology of AD.We aim to emphasize knowledge gaps to provide direction for future research and to establish optimal nutrition-based intervention strategies for AD. | Jiwei Jiang Hanping Shi Shirui Jiang Anxin Wang Xinying Zou Yanli Wang Wenyi Li Yuan Zhang Mengfan Sun Qiwei Ren Jun Xu | 2023 | Science China(Life Sciences)2023,66,10: | 0
Stratified Analysis of Survival Benefit for ABO-incompatible Deceased-donor Liver Transplantation:Multicenter Propensity Score-matched Study显示文摘Background and Aims:Liver transplantation(LT)using ABO-incompatible(ABOi)grafts can extend the donor pool to a certain extent and hence reduce the waiting time for transplantation. However, concerns of the impending prognosis associated with this option, especially for patients with liver failure and higher model for end-stage liver disease(MELD)scores, who tend to be more fragile during the waiting period before LT.Methods:Recipients undergoing LT for acute-onchronic liver failure or acute liver failure were retrospectively enrolled at four institutions.Overall survival was compared and a Cox regression analysis was performed.Propensity score matching was performed for further comparison.Patients were stratified by MELD score and cold ischemia time(CIT)to determine the subgroups with survival benefits. Results:Two hundred ten recipients who underwent ABOi LT and 1,829 who underwent ABO compatible(ABOc)LT were enrolled. The 5-year overall survival rate was significantly inferior in the ABOi group compared with the ABOc group after matching(50.6%vs.75.7%,p<0.05).For patients with MELD scores≤30,using ABOi grafts achieved a comparable overall survival rate as using ABOc grafts(p>0.05).Comparison of the survival rates revealed no statistically significant difference for patients with MELD scores≥40(p>0.05).For patients with MELD scores of 31-39,the overall survival rate was significantly inferior in the ABOi group compared with the ABOc group(p<0.001);however,the rate was increased when the liver graft CIT was<8 h.Conclusions:For recipients with MELD scores≤30,ABOi LT had a prognosis comparable to that of ABOc LT and can be regarded as a feasible option. For recipients with MELD scores≥40,ABOi should be adopted with caution in emergency cases. For recipients with MELD scores of 31-39,the ABOi LT prognosis was worse. However, those patients benefited from receiving ABOi grafts with a CIT of<8 h. | Mengfan Yang Xuyong Wei Abdul Rehman Khan Qiang Wei Rui Wang Binhua Pan Kun Wang Zhisheng Zhou Di Lu Beini Cen Shuijun Zhang Wenzhi Guo Shusen Zheng Yang Yang Xiao Xu | 2023 | Journal of Clinical and Translational Hepatology2023,11,4: | 0
Systematic Review and Meta-analysis:Immunosuppressive Agents for the Treatment of Primary Biliary Cholangitis显示文摘Background and aim:There are still no clinically satisfactory therapy for PBC.This study was performed to assess the safety and efficacy of IAs for the therapy of PBC.Methods:Relevant studies were identified and selected by searching PubMed,Web of Science and Cochrane Library databases. The primary outcome was defined as the need for mortality or liver transplantation.Adverse effects and liver biochemical variables were a secondary outcome. Results:Nine randomized controlled trials, involving six different treatment regimens with a total of 996 patients, were included in the analysis. On meta-analysis,IAs was not associated with a reduction in risk of mortality or liver transplantation(risk ratio[RR]:0.92,95% confidence interval[CI]:0.69-1.22,P=0.57,P=0%),and have resulted in more adverse effects(RR:1.44,95%CI:1.08-1.92,P=0.01,P=19%).Subgroup analysis showed that IAs monotherapy caused adverse effects such as diarrthea, abdominal pain, and renal insufficiency(RR:1.36,95% CI:1.01-1.82,P=0.04,P=48%).IAs therapy did not prominently improve markers of liver function except for alkaline phosphatases(weighted mean difference[WMD]:-0.38,95% CI:-0.62 to-0.14,P=0.002).Conclusions:IAs cannot reduce the risk of mortality or liver transplantation, whether in IAs monotherapy or combination therapy, and even be associated with more adverse effects. | Mengfan Du Bing Xu Ru Xiang Qian Li Yang Chen Yu Fan | 2021 | Journal of Clinical and Nursing Research2021,5,4: | 0
