| 3 | Regulation of Hepatitis C Virus Replication and Gene Expression by the MAPK-ERK Pathway显示文摘The mitogen activated protein kinases-extracellular signal regulated kinases (MAPK-ERK) pathway is involved in regulation of multiple cellular processes including the cell cycle. In the present study using a Huh7 cell line Con1 with an HCV replicon, we have shown that the MAPK-ERK pathway plays a significant role in the modulation of HCV replication and protein expression and might influence IFN-α signalling. Epithelial growth factor (EGF) was able to stimulate ERK activation and decreased HCV RNA load while a MAPK-ERK pathway inhibitor U0126 led to an elevated HCV RNA load and higher NS5A protein amounts in Con1 cells. It could be further demonstrated that the inhibition of the MAPK-ERK pathway facilitated the translation directed by the HCV internal ribosome entry site. Consistently, a U0126 treatment enhanced activity of the HCV reporter replicon in transient transfection assays. Thus, the MAPK-ERK pathway plays an important role in the regulation of HCV gene expression and replication. In addition, cyclin-dependent kinases (CDKs) downstream of ERK may also be involved in the modulation of HCV replication since roscovitine, an inhibitor of CDKs had a similar effect to that of U0126. Modulation of the cell cycle progression by cell cycle inhibitor or RNAi resulted consistently in changes of HCV RNA levels. Further, the replication of HCV replicon in Con1 cells was inhibited by IFN-α. The inhibitory effect of IFN-α could be partly reversed by pre-incubation of Con-1 cells with inhibitors of the MAPK-ERK pathway and CDKs. | 4 | Disruption of crosstalk between LX-2 and liver cancer stem-like cells from MHCC97H cells by DFOG via inhibiting FOXM1显示文摘Hepatic stellate cell(HSC)line LX-2 is activated by liver cancer stem-like cells(LCSLCs)and produces various cytokines that make up most of the hepatocellular carcinoma(HCC)microenvironment.The new genistein derivative,7-difluoromethoxyl-5,4-dirboctylgenistein(DFOG),shows anticancer effects in multiple malignancies by controlling forkhead box M1(FOXM1).In this study,we aimed to assess whether DFOG disrupts the crosstalk between human HSC LX-2 cells and LCSLCs.Distinct gen erations of MHCC97H-derived spheres were obtained with the sec ond generation considered as LCSLCs which displayed enhanced self-renewal ability and elevated expression levels of CD133,CD44,and EpCAM proteins,as well as tumorigenicity,as revealed by colony formation assay in vitro and tumorigenicity assay in vivo.LX-2 and MHCC97H cells were co-cultured with/without DFOG(1,5,and 10 pM,respectively)using the transwell system.FOXM1 overexpression and/or knockdown were employed for mechanistic investigations.Our results suggested that Co-CM promoted LX-2 cell transformation into liver cancer-associated HSCs.Meanwhile,FOXM1 was up-regulated and the level of hepatocyte growth factor(HGF)was in creased in LX-2 cells and in the super nata nt after Co-CM stimulati on.Sphere and colony formation abilities in MHCC97H cells,and protein levels of CD133r CD44,and EpCAM,were also markedly elevated.DFOG dose-dependently inhibited the above effects,similar to FOXM1 knockdown in LX-2 cells.FOXM1 overexpression reversed the inhibitory effects of DFOG or FOXM1 knockdown or both on LX-2 cell activation and LCSLC feature induction in MHCC97H cells by LCSLC/LX-2 co-culture.This study demonstrated that DFOG disrupts the crosstalk between HSCs and LCSLCs to suppress LCSLC features via dowrrregulating FOXM1 expression and reducing HGF secretion in HSCs. | A Chen Chang Xu Yimin Luo Lihua Liu Kun Song Guangqi Deng Mengjie Yang Jianguo Cao Liming Yuan Xiang Li | 2019 | Acta Biochimica et Biophysica Sinica2019,51,12: | 2 |

| 9 | Influence of positive end-expiratory pressure upregulation on the right ventricle in critical patients with acute respiratory distress syndrome:an observational cohort study显示文摘Background This study aimed to investigate the influence of positive end-expiratory pressure(PEEP)on the right ventricle(RV)of mechanical ventilation-assisted patients through echocardiography.Methods Seventy-six patients assisted with mechanical ventilation were enrolled in this study.Positive end-expiratory pressure was upregulated by 4 cm H_(2)O to treat acute respiratory distress syndrome,wherein echocardiography was performed before and after this process.Hemodynamic data were also recorded.All variables were compared before and after PEEP upregulation.The effect of PEEP was also evaluated in patients with and without decreased static lung compliance(SLC).Results Positive end-expiratory pressure upregulation significantly affected the RV function.Remarkable differences were observed in the following:Tei index(P=0.027),pulmonary artery pressure(P=0.039),tricuspid annular plane systolic excursion(P=0.014),early wave/atrial wave(P=0.002),diaphragm excursion(P<0.001),inferior vena cava collapsing index(P<0.001),and SLC(P<0.001).There were no significant changes in heart rate,respiratory rate,central venous pressure,mean arterial pressure,and base excess(P>0.05).Furthermore,the cardiac output of the RV was not significantly affected.In patients with decreased SLC(n=41),there were more significant changes in diaphragm excursion(P<0.001),inferior vena cava collapse index(P=0.025),pulmonary artery pressure(P<0.001),and tricuspid annular plane systolic excursion(P=0.007)than in those without decreased SLC(n=35).Conclusion Positive end-expiratory pressure upregulation significantly affected the RV function of critically ill patients with acute respiratory distress syndrome,especially in those with decreased SLC. | Hui Liu Mengjie Song Li Wang Jianguo Xiao Feihu Zhou | 2023 | Emergency and Critical Care Medicine2023,3,3: | 0 |

| 11 | Proteome comparisons reveal influence of different dietary proteins on the development of rat jejunum显示文摘This study compared proteome profiles and morphological changes of rat jejunum in response to different dietary proteins. Fifty male Sprague-Dawley rats were fed with casein(control), and isolated beef, pork, fish and chicken proteins for 14 days. Proteome analysis,histological observation and PEPT1 quantification of the jejunum were performed. The results indicated that rats fed with chicken proteins had higher PEPT1 mRNA and protein levels(P < 0.05) but lower villus height and ratio of villus height to crypt depth(V/C ratio, P < 0.05) than those fed with casein and pork protein. Label-free LC-MS/MS indicated that, as compared to casein, intake of chicken protein can regulate oligopeptide transport mainly by upregulating PEPT1 protein expression and reducing dipeptidyl-peptidase activity related to biological oxidation, and can reduce oligopeptide absorption capacity by regulating Hippo signaling pathway. | 13 | Effects of phosphate on the transport of graphene oxide nanoparticles in saturated clean and iron oxide-coated sand columns显示文摘In this study, transport behaviors of graphene oxide(GO) in saturated uncoated(i.e., clean sand) and goethite-coated sand porous media were examined as a function of the phosphate. We found that phosphate enhanced the transport of GO over a wide range of solution chemistry(i.e., pH 5.0–9.0 and the presence of 10 mmol/L Na^(+) or 0.5 mmol/L Ca^(2+)). The results were mainly ascribed to the increase of electrostatic repulsion between nanoparticles and porous media. Meanwhile, deposition site competition induced by the retained phosphate was another important mechanism leading to promote GO transport. Interestingly, when the phosphate concentration increased from 0.1 to 1.0 mmol/L, the transportenhancement effect of phosphate in goethite-coated sand was to a much larger extent than that in clean sand. The observations were primarily related to the difference in the total mass of retained phosphate between the iron oxide-coated sand and clean sand columns, which resulted in different degrees of the electrostatic repulsion and competitive effect of phosphate. When the background solution contained 0.5 mmol/L Ca^(2+), phosphate could be bind to sand/goethite-coated sand surface by cation bridging;and consequently, promoted competition between phosphate and nanoparticles for deposition sites, which was an important mechanism for the enhanced effect of phosphate. Moreover, the DLVO theory was applicable to describe GO transport behaviors in porous media in the absence or presence of phosphate. | 19 | A modeling study of different kinds of sessile droplets on the horizontal surface with surface wettability and gravity effects considered显示文摘Droplet phase change is important for energy storage and saving technology.The initial profile of the droplet is extremely important for its vaporization or solidification on a horizontal surface.To understand the effect of liquid physical properties on droplet profile,a theoretical model was developed in this study,based on the Young-Laplace equation with gravity effect specially considered.After the model was experimentally validated by comparing the geometric shape of water droplets,it was further used for predicting droplet shapes of other materials,and thus analyzing the influence of different physical properties,such as temperature,pressure,surface wettability,etc.Results show that the results of this model agree well with the experimental data.The maximum and average deviations are less than 4.5%and 1.5%,respectively.For all kinds of droplets on the fixed surfaces,when the temperature increases,the droplet contact radius increases and height decreases.The droplets of nitro-gen and carbon dioxide are more sensitive to temperature than ethanol and ethylene glycol droplets.For 20��L droplets on the surface of contact angle 150°,when the temperature changes from 273.15 K to 293.15 K,the droplet contact radiuses increase by 30.6%for carbon dioxide,1.2%for ethanol and 0.67%for ethylene glycol,and the droplet heights decrease by 42.9%,2.5%,1.1%,respectively.Results of this study are meaningful for predicting the phase change process of droplets on the horizontal surface by controlling their initial profiles. | DANG Qun SONG Mengjie LUO Xiaoyan QU Minglu WANG Xiaotao | 2022 | Energy Storage and Saving2022,1,1: | 0 |
