| 1 | Historical trends of PCBs and PBDEs as reconstructed in a lake sediment from southern Tibetan Plateau显示文摘High-altitude lake sediment can be used as a natural archive to reconstruct the history of pollutants. In this work, the temporal distribution of polychlorinated biphenyls(PCBs) and polybrominated diphenyl ethers(PBDEs) were determined using a high-resolution gas chromatography coupled with high-resolution mass spectrometer(HRGC/HRMS) in an alpine lake sediment core collected from the southern Tibetan Plateau(TP) to examine whether the expected decreasing trends due to the implementation of the international Conventions could be observed. The concentrations of PCBs and PBDEs in the sediment core were in the range of 11.8–142 pg/g dw and ND-457 pg/g dw, and their fluxes were in the range of 2.51–31.7 ng/(m^2 ·yr) and ND-43.2 ng/(m^2 ·yr), respectively. The prevalence of low-chlorinated(tri-CB) PCBs and low-brominated(tri-to tetra-) PBDEs in most sections of the sediment profiles was observed, suggesting that the light molecular weight PCBs and PBDEs have most likely reached lake sediments by long-range atmospheric transport from distant sources. Despite the restrictions on their applications, the sediment records for the concentrations and fluxes showed no corresponding decreasing trend with restrictions for PCBs, which suggested that these POPs(e.g., PCBs) were still emitted to the environment owing to the influence of primary or secondary emissions. | 2 | HGF percutaneous endocardial injection induces cardiomyocyte proliferation and rescues cardiac function in pigs显示文摘Objective:To investigate the effect of cardiomyocyte proliferation induced by human hepatocyte growth factor(HGF)in pigs with chronic myocardial infarction(CMI).Methods:A steerable,deflectable 7F catheter incorporating a 27-guage needle was advanced percutaneously to the left ventricular myocardium of 18 pigs with CMI.Pigs were randomized(1:1:1)to receive adenoviral vector HGF(total dose,1×1010 genome copies),which was administered as five injections into the infarcted myocardium(total,1.0 mL),or saline,or Ad-null(control groups).Injections were guided by Ensite NavX left ventricular electroanatomical mapping.HGF and cyclin proteins were detected by western blot and immunoprecipitation analysis.Histological and immunohistochemical analysis determined proliferating cardiomyocytes.Myocardial perfusion and cardiac function were estimated by Gated-Single Photon Emission Computed Tomography(G-SPECT).Results:Western blot analyses showed that HGF were predominantly expressed in the infarct core and border in the myocardium of the infarcted heart.G-SPECT analysis indicated that the HGF group had better cardiac function and myocardial perfusion four weeks after the injection of Ad-HGF than before the injection of Ad-HGF.After treatment there were more proliferating cardiomyocytes in the HGF group compared to either of the control groups.Furthermore,the HGF group myocardial samples expressed higher levels of p-Akt,cyclin A,cyclin E,cyclin D1,cdk2,cdk4 than those in the control groups.Conclusion:The over-expression of HGF activates pro-survival pathways,induces cardiomyocyte proliferation,and improves the perfusion and function of the porcine CMI heart. | Zhengxian Tao Bo Chen Yingming Zhao Hongwu Chen Liansheng Wang Yonghong Yong Kejiang Cao Qifeng Yu Danian Ke` Hua Wang Zuze Wu Zhijian Yang | 2010 | The Journal of Biomedical Research2010,24,3: | 3 |
| 3 | Tuning filament composition and microstructure of 3D-printed bioceramic scaffolds facilitate bone defect regeneration and repair显示文摘It is still a challenge to optimize the component distribution and microporous structures in scaffolds for tailoring biodegradation(ion releasing)and enhancing bone defect repair within an expected time stage.Herein,the core–shell-typed nonstoichiometric wollastonite(4%and 10%Mg-doping calcium silicate;CSiMg4,CSiMg10)macroporous scaffolds with microporous shells(adding~μ10 μm PS microspheres into shell-layer slurry)were fabricated via 3D printing.The initial mechanical properties and bio-dissolution(ion releasing)in vitro,and osteogenic capacity in vivo of the bioceramic scaffolds were evaluated systematically.It was shown that endowing high-density micropores in the sparingly dissolvable CSiMg10 or dissolvable CSiMg4 shell layer inevitably led to nearly 30%reduction of compressive strength,but such micropores could readily tune the ion release behaviour of the scaffolds(CSiMg4@CSiMg10 vs.CSiMg4@CSiMg10-p;CSiMg10@CSiMg4 vs.CSiMg10@CSiMg4-p).Based on the in rabbit femoral bone defect repair model,the 3D μCT reconstruction and histological observation demonstrated that the CSiMg4@CSiMg10-p scaffolds displayed markedly higher osteogenic capability than the other scaffolds after 12weeks of implantation.It demonstrated that core–shell bioceramic 3D printing technique can be developed to fabricate single-phase or biphasic bioactive ceramic scaffolds with accurately tailored filament biodegradation for promoting bone defect regeneration and repair in some specific pathological conditions. | Yi Chen Jiaping Huang Jiamei Liu Yingming Wei Xianyan Yang Lihong Lei Lili Chen Yanmin Wu Zhongru Gou | 2021 | Regenerative Biomaterials2021,8,2: | 3 |
| 4 | Estimationof genetic parameters for juvenile growth performance traits in oliveflounder(Paralichthys olivaceus)显示文摘The aim of this study was to evaluate genetic parameters of growth traits for improving selective breeding in olive flounder(Paralichthys olivaceus).A total of 82 full-sib families from 2 year-classes with complete pedigree information were used to estimate heritabilities and genetic/phenotypic correlations for 4 growth performance traits(body weight BW,total length TL,body height BH and condition factor K)based on uni-trait and multi-trait linear mixed models,respectively.Data were analyzed by the restricted maximum likelihood(REML)method.The heritabilities of BW,TL and BH were 0.81,0.70 and 0.61,which are relatively high,however,heritability for K is low(0.11).For all growth traits,the common full-sib effects(c^(2))were at a low to moderate level(0.14–0.20);the maternal effects(m^(2))of the traits(except K)were also detected(0.15–0.19).The phenotypic and genetic correlations among BW,TL and BH were highly positive(0.70–0.91),however,the correlations between K and the other 3 traits was low or negligible(−0.22–0.26).According to our results,these traits(except K)could be selected and enhanced simultaneously.In addition,direct selection based on TL or BH,which is an easily measurable trait,will be more favorable than selection on BW.Moreover,our results indicated that growth parameters estimated in the juvenile stage could be used as a predictor of growth performance at later growth stages. | Yangzhen Li Bo Zhang Yingming Yang Songlin Chen | 2019 | Aquaculture and Fisheries2019,4,2: | 3 |
| 5 | The vicK gene of Streptococcus mutans mediates its cariogenicity via exopolysaccharides metabolism显示文摘Streptococcus mutans(S.mutans)is generally regarded as a major contributor to dental caries because of its ability to synthesize extracellular polysaccharides(EPS)that aid in the formation of plaque biofilm.The VicRKX system of S.mutans plays an important role in biofilm formation.The aim of this study was to investigate the effects of vicK gene on specific characteristics of EPS in S.mutans biofilm.We constructed single-species biofilms formed by different mutants of vicK gene.Production and distribution of EPS were detected through atomic force microscopy,scanning electron microscopy and confocal laser scanning microscopy.Microcosmic structures of EPS were analyzed by gel permeation chromatography and gas chromatography-mass spectrometry.Cariogenicity of the vicK mutant was assessed in a specific pathogen-free rat model.Transcriptional levels of cariogenicityassociated genes were confirmed by quantitative real-time polymerase chain reaction.The results showed that deletion of vicK gene suppressed biofilm formation as well as EPS production,and EPS were synthesized mostly around the cells.Molecular weight and monosaccharide components underwent evident alterations.Biofilms formed in vivo were sparse and contributed a decreased degree of caries.Moreover,expressional levels of genes related to EPS synthesis were down-regulated,except for gtfB.Our report demonstrates that vicK gene enhances biofilm formation and subsequent caries development.And this may due to its regulations on EPS metabolism,like synthesis or microcosmic features of EPS.This study suggests that vicK gene and EPS can be considered as promising targets to modulate dental caries. | Yalan Deng Yingming Yang Bin Zhang Hong Chen Yangyu Lu Shirui Ren Lei Lei Tao Hu | 2021 | International Journal of Oral Science2021,13,4: | 2 |

| 18 | Functional analysis of the promoter of the dmrt1 gene in Chinese tongue sole,Cynoglossus semilaevis显示文摘The dmrt1 (doublesex and mab-3 related transcription factor 1) gene is considered to play an essential role in testis differentiation and development among metazoan species. As an economically important marine fish in China, Chinese tongue sole (Cynoglossus semilaevis) possess a ZW/ZZ sex-determining system, and the females usually grow two to four times bigger than males. Previous studies have confirmed that Z-linked dmrt1 gene is the male determining gene in C. semilaevis and TALENs-mediated knocking-out individuals showed increased growth phenotype. In order to investigate the function of dmrt1 regulatory regions and its potential application, we have cloned the promoter of C. semilaevis dmrt1. The acquired sequence consisted of a 5'-upstream sequence and a partial exon1;functional analysis showed that there were two positive regulatory regions in the promoter, as well as one negative region and one neutral region. These positive regions were then combined to obtain several newly constructed promoters. Both in vitro and in vivo analysis confirmed one of them (dmrt1-Δ3+Δ4) showed elevated promoting activity compared to the full-length promoter and could drive exogenous gene expression in tongue sole gonads.
