| 7 | Cholesterol-tuned liposomal membrane rigidity directs tumor penetration and anti-tumor effect显示文摘Recently, liposomes have been widely used in cancer therapeutics, but their anti-tumor effects are suboptimal due to limited tumor penetration. To solve this problem, researchers have made significant efforts to optimize liposomal diameters and potentials, but little attention has been paid to liposomal membrane rigidity. Herein, we sought to demonstrate the effects of cholesterol-tuned liposomal membrane rigidity on tumor penetration and anti-tumor effects. In this study, liposomes composed of hydrogenated soybean phospholipids(HSPC), 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy(polyethylene glycol)-2000](DSPE-PEG2000) and different concentrations of cholesterol were prepared. It was revealed that liposomal membrane rigidity decreased with the addition of cholesterol. Moderate cholesterol content conferred excellent diffusivity to liposomes in simulated diffusion medium, while excessive cholesterol limited the diffusion process. We concluded that the differences of the diffusion rates likely stemmed from the alterations in liposomal membrane rigidity, with moderate rigidity leading to improved diffusion. Next, the in vitro tumor penetration and the in vivo anti-tumor effects were analyzed. The results showed that liposomes with moderate rigidity gained excellent tumor penetration and enhanced anti-tumor effects. | 12 | Chitosan oligosaccharide modified liposomes enhance lung cancer delivery of paclitaxel显示文摘Lung cancer is one of the leading causes of cancer-related death worldwide.Various therapeutic failed in the effective treatment of the lung cancer due to their limited accumulation and exposure in tumors.In order to promote the chemotherapeutics delivery to lung tumor,we introduced chitosan oligosaccharide(CSO)modification on the liposomes.CSO conjugated Pluronic P123 polymers with different CSO grafting amounts,called as CP50 and CP20,were synthesized and used to prepare CSO modified liposomes(CP50-LSs and CP20-LSs).CP50-LSs and CP20-LSs displayed significantly enhanced cellular uptake in A549 cells in vitro as well as superior tumor accumulation in vivo compared with non-CSO modified liposomes(P-LSs).This phenomenon was related to the increased affinity between CSO modified liposomes and tumor cells following massive adsorption of collagen,which was highly expressed in lung tumors.In the A549 tumor-bearing mouse model,intravenous injection of paclitaxel(PTX)-loaded CP50-LSs every 3 days for 21 days resulted in optimal antitumor therapeutic performance with an inhibition rate of 86.4%.These results reveal that CSO modification provides promising applicability for nanomedicine design in the lung cancer treatment. | Yun-qiu Miao Ming-shu Chen Xin Zhou Lin-miao Guo Jing-jing Zhu Rui Wang Xin-xin Zhang Yong Gan | 2021 | Acta Pharmacologica Sinica2021,42,10: | 2 |
| 13 | Tree ring based drought variability in Northwest Tajikistan since 1895 AD显示文摘Determining the mechanisms controlling the changes of wet and dry conditions will improve our understanding of climate change over the past hundred years,which is of great significance to the study of climate and environmental changes in the arid regions of Central Asia.Forest trees are ecologically significant in the local environment,and therefore the tree ring analysis can provide a clear record of regional historical climate.This study analyzed the correlation between the tree ring width chronology of Juniperus turkestanica Komarov and the standardized precipitation evapotranspiration index(SPEI)in Northwest Tajikistan,based on 56 tree ring samples collected from Shahristan in the Pamir region.Climate data including precipitation,temperature and the SPEI were downloaded from the Climate Research Unit(CRU)TS 4.00.The COFECHA program was used for cross-dating,and the ARSTAN program was used to remove the growth trend of the tree itself and the influence of non-climatic factors on the growth of the trees.A significant correlation was found between the radial growth of J.turkestanica trees and the monthly mean SPEI of February–April.The monthly mean SPEI sequence of February–April during the period of 1895–2016 was reconstructed,and the reconstruction equation explained 42.5%of the variance.During the past 122 a(1895–2016),the study area has experienced three wetter periods(precipitation above average):1901–1919,1945–1983 and 1995–2010,and four drier periods(precipitation below average):1895–1900,1920–1944,1984–1994 and 2011–2016.The spatial correlation analysis revealed that the monthly mean SPEI reconstruction sequence of February–April could be used to characterize the large-scale dry-wet variations in Northwest Tajikistan during the period of 1895–2016.This study could provide comparative data for validating the projections of climate models and scientific basis for managing water resources in Tajikistan in the context of climate change. | YANG Meilin YU Yang ZHANG Haiyan WANG Qian GAN Miao YU Ruide | 2020 | Journal of Arid Land2020,12,3: | 2 |

| 18 | Trapping precursor-level functionalities in hierarchically porous carbons prepared by a pre-stabilization route for superior supercapacitors显示文摘Versatile module design of precursor networks enables flexible functionalization of nano-carbon electrode materials to meet the adaptable energy-storage demand. Functionalized heterogeneous networks are more likely to decompose by swift temperature programming together with predesign module removal, so high functionality/network transfer from precursor to carbon is still a work in progress. A pre-stabilization route is proposed here to enhance the network strength at early pyrolysis and pin up precursor-level functionalities on the final carbon. Such strategy successfully fixes more electroactive N(4.28-8.86 wt%) into the resultant carbon microspheres compared with non-pretreated carbon(2.89wt%), as well as achieves broad ion-accessible platforms of 1575-2269 m^(2)/g with preset structural superiorities. As a result, a typical acidic device reveals an outstanding specific capacitance of 383 F/g at 10 mV/s. Taking advantage of a novel LiNO_(3)-PAM polymer electrolyte, the upgraded symmetric device displays the maximum specific capacitance of 229 F/g, along with a boosted energy density of 41.1 Wh/kg at 643.4 W/kg. | 20 | From interpenetrating polymer networks to hierarchical porous carbons for advanced supercapacitor electrodes显示文摘Hierarchical porous carbons (HPCs) are obtained via in-situ activation of interpenetrating polymer networks (IPNs) obtained from simultaneous polymerization of resorcinol/formaldehyde (R/F) and polyacrylamide (PAM). The hierarchically micro-, meso-and macroporous structure of as-prepared HPCs is attributed to the synergistic pore-forming effect of PAM and KOH, including PAM decomposition, KOH chemical activation, and a foaming process of potassium polyacrylate formed by partial hydrolysis of PAM in KOH aqueous solution. The typical HPC electrode with the highest surface area (2544 m2/g) shows a high specific capacitance of 261 F/g at 1.0 A/g and a superior rate capability of 216 F/g at 20 A/g in alkaline electrolyte. Moreover, the electrode maintains the capacitance retention of 90.8% after 10000 chargingdischarging cycles at 1.0 A/g, exhibiting long cycling life.
