| 1 | Postconditioning of sevoflurane and propofol is associated with mitochondrial permeability transition pore显示文摘Background: Sevoflurane and propofol are effective cardioprotective anaesthetic agents, though the cardioprotection of propofol has not been shown in humans. Their roles and underlying mechanisms in anesthetic postconditioning are unclear. Mitochondrial permeability transition pore (MPTP) opening is a major cause of ischemia-reperfusion injury. Here we investigated sevoflurane- and propofol-induced postconditioning and their relationship with MPTP. Methods: Isolated perfused rat hearts were exposed to 40 min of ischemia followed by 1 h of reperfusion. During the first 15 min of reperfusion, hearts were treated with either control buffer (CTRL group) or buffer containing 20 μmol/L atractyloside (ATR group), 3% (v/v) sevoflurane (SPC group), 50 μmol/L propofol (PPC group), or the combination of atractyloside with respective anesthetics (SPC+ATR and PPC+ATR groups). Infarct size was determined by dividing the total necrotic area of the left ventricle by the total left ventricular slice area (percent necrotic area). Results: Hearts treated with sevoflurane or propofol showed significantly better recovery of coronary flow, end-diastolic pressures, left ventricular developed pressure and derivatives compared with controls. Sevoflurane resulted in more protective alteration of hemodynamics at most time point of reperfusion than propofol. These improvements were paralleled with the reduction of lactate dehydrogenase release and the decrease of infarct size (SPC vs CTRL: (17.48±2.70)% vs (48.47±6.03)%, P<0.05; PPC vs CTRL: (35.60±2.10)% vs (48.47±6.03)%, P<0.05). SPC group had less infarct size than PPC group (SPC vs PPC: (17.48±2.70)% vs (35.60±2.10)%, P<0.05). Atractyloside coadministration attenuated or completely blocked the cardioprotective effect of postconditioning of sevoflurane and propofol. Conclusion: Postconditioning of sevoflurane and propofol has cardio-protective effect against ischemia-reperfusion injury of heart, which is associated with inhibition of MPTP opening. | 2 | Sevoflurane postconditioning reduces myocardial reperfusion injury in rat isolated hearts via activation of PI3K/Akt signaling and modulation of Bcl-2 family proteins显示文摘Sevoflurane postconditioning reduces myocardial infarct size.The objective of this study was to examine the role of the phosphatidylinositol-3-kinase(PI3K)/Akt pathway in anesthetic postconditioning and to determine whether PI3K/Akt signaling modulates the expression of pro-and antiapoptotic proteins in sevoflurane postconditioning.Isolated and perfused rat hearts were prepared first,and then randomly assigned to the following groups:Sham-operation(Sham),ischemia/reperfusion(Con),sevoflurane postconditioning(SPC),Sham plus 100 nmol/L wortmannin(Sham+Wort),Con+Wort,SPC+Wort,and Con+dimethylsulphoxide(DMSO).Sevoflurane postconditioning was induced by administration of sevoflurane(2.5%,v/v) for 10 min from the onset of reperfusion.Left ventricular developed pressure(LVDP),left ventricular end-diastolic pressure(LVEDP),maximum increase in rate of LVDP(+dP/dt),maximum decrease in rate of LVDP(?dP/dt),heart rate(HR),and coronary flow(CF) were measured at baseline,R30 min(30 min of reperfusion),R60 min,R90 min,and R120 min.Creatine kinase(CK) and lactate dehydrogenase(LDH) were measured after 5 min and 10 min reperfusion.Infarct size was determined by triphenyltetrazolium chloride staining at the end of reperfusion.Total Akt and phosphorylated Akt(phospho-Akt),Bax,Bcl-2,Bad,and phospho-Bad were determined by Western blot analysis.Analysis of variance(ANOVA) and Student-Newman-Keuls' test were used to investigate the significance of differences between groups.The LVDP,±dP/dt,and CF were higher and LVEDP was lower in the SPC group than in the Con group at all points of reperfusion(P<0.05).The SPC group had significantly reduced CK and LDH release and decreased infarct size compared with the Con group [(22.9±8)% vs.(42.4±9.4)%,respectively;P<0.05].The SPC group also had increased the expression of phospho-Akt,Bcl-2,and phospho-Bad,and decreased the expression of Bax.Wortmannin abolished the cardioprotection of sevoflurane postconditioning.Sevoflurane postconditioning may protect the isolated rat heart.Activation of PI3K and modulation of the expression of pro-and antiapoptotic proteins may play an important role in sevoflurane-induced myocardial protection. | Li-na YU Jing YU Feng-jiang ZHANG Mei-juan YANG Ting-ting DING Jun-kuan WANG Wei HE Tao FANG Gang CHEN Min YAN | 2010 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2010,11,9: | 36 |

| 4 | Expressions of inducible nitric oxide synthase and matrix metalloproteinase-9 and their effects on angiogenesis and progression of hepatocellular carcinoma显示文摘AIM: To determine the expressions of inducible nitric oxide synthase (iNOS) and matrix metalloproteinase-9 (MMP-9)in hepatocellular carcinoma (HCC) and to investigate the relationship between iNOS and MMP-9 expression and their effects on angiogenesis and progression of HCC.METHODS: Tn this study, we examined iNOS, MMP-9,and CD34 expression in specimens surgically removed from 32 HCC patients and 7 normal liver tissues by immunohistochemical staining. Meanwhile, microvessel density (MVD) was determined as a marker of angiogenesis by counting CD34-positive cells.RESULTS: The positive rates of iNOS and MMP-9expression were 71.88% (23/32) and 78.13% (25/32) in HCC. MMP-g expression was significantly correlated with tumor size, capsule status, TNM stage, and risk of HCC recurrence (P= 0.032, P= 0.033, P= 0.007, and P= 0.001,respectively). There was also a significant relationship between iNOS expression and capsule status and risk of HCC recurrence (P = 0.04g and P = 0.004, respectively),but no correlation between iNOS expression and tumor size and TNM stage. There was a positive association between MVD and TNM stage and risk of HCC recurrence (P = 0.037 and P = 0.000, respectively). The count of MVD was significantly different in different iNOS and MMP-9 immunoreactivity groups (F= 17.713 and 17.097, P = 0.000 and P = 0.000, respectively). The examination of Spearman's rank correlation coefficient showed that there was a significant positive correlation between MVD and iNOS,MMP-9 immunoreactivity (r= 0.754 and 0.751, P= 0.000 and P=0.000, respectively). There was also a significant association between MMP-9 and iNOS expression in HCC (P = 0.010).CONCLUSION: Nitric oxide (NO) produced by iNOS could modulate MMP-9 production and therefore contribute to tumor cell angiogenesis and invasion and metastasis in HCC. The strong expression of iNOS and MMP-9 in HCC may be helpful in evaluating the recurrence of HCC,predicting poor prognosis. | 10 | Effect of perioperative autologous versus allogeneic blood transfusion on the immune system in gastric cancer patients显示文摘Background: Allogeneic blood transfusion-induced immunomodulation (TRIM) and its adverse effect on the prognosis of patients treated surgically for cancer remain complex and controversial. However, the potential risk associated with allogeneic blood transfusion has heightened interest in the use of autologous blood transfusion. In the present study, the serum concentrations of neopterin, interferon-gamma (IFN-γ), T lymphocyte subsets (CD3+, CD4+, CD8+, CD4+/CD8+) and a possible association between these variables were investigated. The purpose was to further evaluate the effect of autologous versus allogeneic blood transfusion on immunological status in patients undergoing surgery for gastric cancer. Methods: Sixty ASA I~II (American Society of Anesthesiologists) patients undergoing elective radical resection for stomach cancer were randomly allocated to receive either allogeneic blood transfusion (n=30) or autologous blood transfusion (n=30). Serum concentrations of the neopterin, IFN-γ and T lymphocyte subsets in the recipients were measured before induction of anesthesia, after operation, and on the 5th postoperative day. Results: Both two groups, serum neopterin, IFN-γ, percentages of T-cell subsets (CD3+, CD4+), and CD4+/CD8+ ratio had significantly decreased after operation, but decreased more significantly in group H (receiving allogeneic blood transfusion) than those in group A (receiving autologous whole blood transfusion) (P<0.05). On the 5th postoperative day, serum neopterin, IFN-γ, CD3+, CD4+ T-cells, and CD4+/CD8+ ratio returned to the baseline values in group A. In contrast, the above remain decreasing in group H, where there were no significant relations between serum neopterin and IFN-γ. Conclusion: Perioperative surgical trauma and stress have an immunosuppressive impact on gastric cancer patients. Allogeneic blood transfusion exacerbates the impaired immune response. | 11 | Effects of sevoflurane preconditioning and postconditioning on rat myocardial stunning in ischemic reperfusion injury显示文摘Ischemic preconditioning and postconditioning distinctly attenuate ventricular arrhythmia after ischemia without affecting the severity of myocardial stunning. Therefore,we report the effects of sevoflurane preconditioning and postconditioning on stunned myocardium in isolated rat hearts. Isolated rat hearts were underwent 20 min of global ischemia and 40 min of reperfusion. After an equilibration period (20 min),the hearts in the preconditioning group were exposed to sevoflurane for 5 min and next washout for 5 min before ischemia. Hearts in the sevoflurane postconditioning group underwent equilibration and ischemia,followed immediately by sevoflurane exposure for the first 5 min of reperfusion. The control group received no treatment before and after ischemia. Left ventricular pressure,heart rate,coronary flow,electrocardiogram,and tissue histology were measured as variables of ventricular function and cellular injury,respectively. There was no significant difference in the duration of reperfusion ventricular ar-rhythmias between control and sevoflurane preconditioning group (P=0.195). The duration of reperfusion ventricular arrhythmias in the sevoflurane postconditioning group was significantly shorter than that in the other two groups (P<0.05). ±(dP/dt)max in the sevoflurane preconditioning group at 5,10,15,20,and 30 min after reperfusion was sig-nificantly higher than that in the control group (P<0.05),and there were no significant differences at 40 min after reperfusion among the three groups (P>0.05). As expected,for a 20-min general ischemia,infarct size in heart slices determined by 2,3,5-triphenyltetrazolium chloride staining among the groups was not obvious. Sevoflurane postcon-ditioning reduces reperfusion arrhythmias without affecting the severity of myocardial stunning. In contrast,sevoflu-rane preconditioning has no beneficial effects on reperfusion arrhythmias,but it is in favor of improving ventricular function and recovering myocardial stunning. | 13 | Evaluating effective swath width and droplet distribution of aerial spraying systems on M-18B and Thrush 510G airplanes显示文摘Aerial spraying plays an important role in promoting agricultural production and protecting the biological environment due to its flexibility,high effectiveness,and large operational area per unit of time.In order to evaluate the performance parameters of the spraying systems on two fixed wing airplanes M-18B and Thrush 510G,the effective swath width and uniformity of droplet deposition under headwind flight were tested while the planes operated at the altitudes of 5 m and 4 m.The results showed that although wind velocities varied from 0.9 m/s to 4.6 m/s,and the directions of the atomizer switched upward and downward in eight flights,the effective swath widths were kept approximately at 27 m and 15 m for the M-18B and Thrush 510G,respectively,and the latter was more stable.In addition,through analyzing the coefficients of variation(CVs)of droplet distribution,it was found that the CVs of the M-18B were 39.57%,33.54%,47.95%,and 59.04% at wind velocities of 0.9,1.1,1.4 and 4.6 m/s,respectively,gradually enhancing with the increasing of wind speed;the CVs of Thrush 510G were 79.12%,46.19%,14.90%,and 48.69% at wind velocities of 1.3,2.3,3.0 and 3.4 m/s,respectively,which displayed the irregularity maybe due to change of instantaneous wind direction.Moreover,in terms of the CVs and features of droplet distribution uniformity for both airplanes in the spray swath,choosing smaller CV(20%-45%)as the standard of estimation,it was found that the Thrush 510G had a better uniform droplet distribution than the M-18B.The results provide a research foundation for promoting the development of aerial spraying in China. | Zhang Dongyan Chen Liping Zhang Ruirui Xu Gang Lan Yubin Wesley Clint Hoffmann Wang Xiu Xu Min | 2015 | International Journal of Agricultural and Biological Engineering2015,8,2: | 18 |

Qingfei Xiaoyan Wan(QF), a traditional Chinese medicine(TCM)formula, has been used clinically in China for the treatment of upper respiratory tract infections, acute or chronic bronchitis and pulmonary infection. In this study, the effects of QF on Pseudomonas aeruginosa–induced acute pneumonia in mice were evaluated. The mechanisms by which four typical antiinflammatory ingredients from QF, arctigenin(ATG), cholic acid(CLA), chlorogenic acid(CGA) and sinapic acid(SPA), regulate anti-inflammatory signaling pathways and related targets were investigated using molecular biology and molecular docking techniques. The results showed that pretreatment with QF significantly inhibits the release of cytokines(TNF-α and IL-6) and chemokines(IL-8 and RANTES),reduces leukocytes recruitment into inflamed tissues and ameliorates pulmonary edema and necrosis. In addition, ATG was identified as the primary anti-inflammatory agent with action on the PI3K/AKT and Ras/MAPK pathways. CLA and CGA enhanced the actions of ATG and exhibited synergistic NF-κB inactivation effects possibly via the Ras/MAPK signaling pathway. Moreover, CLA is speculated to target FGFR and MEK firstly. | 17 | Interleukin-8, a promising predictor for prognosis of pancreatic cancer显示文摘AIM: To investigate the value of interleukin-8 (IL-8), a pro-inflammatory chemokine, in predicting the prognosis of pancreatic cancer. METHODS: Expression of IL-8 and its receptor CXCR1 was assessed by immunohistochemistry in pancreatic cancer and chronic pancreatitis samples. Enzyme-linked immunosorbent assay was used to detect the serum IL-8 levels in pancreatic cancer patients. Human pancreatic cancer tissues were heterotopically transplanted to the immune-deficiency mice to evaluate the effect of serum IL-8 on the tumorigenesis of the cancer samples.RESULTS: IL-8 and CXCR1 proteins were both overexpressed in pancreatic adenocarcinoma samples (55.6% and 65.4%, respectively) compared with the matched para-cancer tissues (25.9% and 12.3%, P < 0.01), or chronic pancreatitis (0% and 25%, P < 0.05). Serum IL-8 levels in pancreatic cancer patients (271.1 ± 187.7 ng/mL) were higher than in other digestive system tumors, such as gastric cancer (41.77 ± 9.11 ng/mL, P = 0.025), colorectal carcinoma (78.72 ± 80.60 ng/mL, P = 0.032) and hepatocellular carcinoma (59.60 ± 19.80 ng/mL, P = 0.016). In vivo tumorigenesis analysis further proved that tumor tissues from patients with higher serum IL-8 levels grew faster than those with lower IL-8 levels. | 18 | Effects of warming and clipping on plant and soil properties of an alpine meadow in the Qinghai-Tibetan Plateau, China显示文摘Climate warming and livestock grazing are known to have great influences on alpine ecosystems like those of the Qinghai-Tibetan Plateau(QTP) in China. However, it is lacking of studies on the effects of warming and grazing on plant and soil properties in these alpine ecosystems. In this study, we reported the related research from manipulative experiment in 2010–2012 in the QTP. The aim of this study was to investigate the individual and combined effects of warming and clipping on plant and soil properties in the alpine meadow ecosystem. Infrared radiators were used to simulate climate warming starting in July 2010, while clipping was performed once in October 2011 to simulate the local livestock grazing. The experiment was designed as a randomized block consisting of five replications and four treatments: control(CK), warming(W), clipping(C) and warming+clipping combination(WC). The plant and soil properties were investigated in the growing season of the alpine meadow in 2012. The results showed that W and WC treatments significantly decreased relative humidity at 20-cm height above ground as well as significantly increases air temperature at the same height, surface temperature, and soil temperature at the depth of 0–30 cm. However, the C treatment did not significantly decrease soil moisture and soil temperature at the depth of 0–60 cm. Relative to CK, vegetation height and species number increased significantly in W and WC treatment, respectively, while vegetation aboveground biomass decreased significantly in C treatment in the early growing season. However, vegetation cover, species diversity, belowground biomass and soil properties at the depth of 0–30 cm did not differ significantly in W, C and WC treatments. Soil moisture increased at the depth of 40–100 cm in W and WC treatments, while belowground biomass, soil activated carbon, organic carbon and total nitrogen increased in the 30–50 cm soil layer in W, C and WC treatments. Although the initial responses of plant and soil properties to experimental warming and clipping were slow and weak, the drought induced by the downward shift of soil moisture in the upper soil layers may induce plant belowground biomass to transfer to the deeper soil layers. This movement would modify the distributions of soil activated carbon, organic carbon and total nitrogen. | 19 | The Predictive Value of Baseline HBs Ag Level and Early Response for HBs Ag Loss in Patients with HBe Ag-positive Chronic Hepatitis B during Pegylated Interferon Alpha-2a Treatment显示文摘Objective To explore the predictive value of baseline HBsAg level and early response for HBsAg loss in patients with HBeAg-positive chronic hepatitis B during pegylated interferon alpha-2a treatment. Methods A total of 121 patients with HBeAg-positive chronic hepatitis B who achieved HBsAg loss were enrolled; all patients were treated with PEG-IFNα-2a 180 μg/week. Serum HBV DNA and serological indicators(HBsAg, anti-HBs, HBeAg, and anti-HBe) were determined before and every 3 months during treatment. Results The median treatment time for HBsAg loss was 84 weeks(7-273 weeks), and 74.38%(90 cases) of the patients needed extended treatment(> 48 weeks). The correlation between baseline HBsAg levels and the treatment time of HBsAg loss was significant(B = 14.465, t = 2.342, P = 0.021). Baseline HBsAg levels together with the decline range of HBsAg at 24 weeks significantly correlated with the treatment time of HBsAg loss(B = 29.862, t = 4.890, P = 0.000 and B = 27.993, t = 27.993, P = 0.005). Conclusion Baseline HBsAg levels and extended therapy are critical steps toward HBsAg loss.
