1Expression of vascular endothelial growth factor and its role in oncogenesis of human gastric carcinoma显示文摘AIM To establish the role of vascular endothelial growth factor (VEGF) in the oncogenesisof human gastric carcinoma more directly.METHODS The expression of VEGF and its receptor kinase-domain insert containing receptor (KDR) in human gastric cancer tissue were observed by immunohistochemical staining. VEGF levels were manipulated in human gastric cancer cell using eukaryotic expression constructs designed to express the complete VEGF165 complimentary DNA in either the sense or antisense orientation. The biological changes of the cells were observed in which VEGF was up-regulated or downregulated.RESULTS VEGF-positive rate was 50%, and VEGF was mainly localized in the cytoplasm and membrane of the tumor cells, while KDR was mainly located in the membrane of vascular endothelial cells in gastric cancer tissues and peri-cancerous tissue. In 2 cases of 50 specimens, the gastric cancer cells expressed KDR,localized in both the cytoplasm and membrane.Introduction of VEGF165 antisense into human gastric cancer cells ( SGC-7901, immunofluorescence intensity,31.6%)) resulted in a significant reduction in VEGFspecific messenger RNA and total and cell surface VEGF protein ( immunofluorescence intensity, 8.9%)(P<0.05). Conversely, stable integration of VEGF165 in the sense orientation resulted in an increase in cellular and cell surface VEGF (immunofluorescence intensity,75.4%) (P<0.05). 2Real world effectiveness of PCSK-9 inhibitors combined with statins versus statins-based therapy among patients with very high risk of atherosclerotic cardiovascular disease in China(RWE-PCSK study)显示文摘BACKGROUND The efficacy and safety of proprotein convertase subtilisin/kexin type 9(PCSK-9) inhibitors were confirmed by several clinical trials, but its effectiveness in routine clinical practice in China has not been evaluated. This study aims to describe the real world effectiveness of PCSK-9 inhibitors combined with statins compared with statins-based therapy among patients with very high risk of atherosclerotic cardiovascular disease(ASCVD).METHODS This is a multi-center observational study, enrolled patients from 32 hospitals who underwent percutaneous coronary intervention(PCI) from January to June in 2019. There are 453 patients treated with PCSK-9 inhibitors combined with statins in PCSK-9 inhibitor group and 2,610 patients treated with statins-based lipid lowering therapies in statins-based group. The lipid control rate and incidence of major adverse cardiovascular events(MACE) over six months were compared between two groups.A propensity score-matched(PSM) analysis was used to balance two groups on confounding factors. Survival analysis using Kaplan-Meier methods was applied for MACE.RESULTS In a total of 3,063 patients, 89.91% of patients had received moderate or high-intensity statins-based therapy before PCI, but only 9.47% of patients had low-density lipoprotein cholesterol(LDL-C) levels below 1.4 mmol/L at baseline. In the PSM selected patients, LDL-C level was reduced by 42.57% in PCSK-9 inhibitor group and 30.81%(P < 0.001) in statins-based group after six months. The proportion of LDL-C ≤ 1.0 mmol/L increased from 5.29% to 29.26% in PCSK-9 inhibitor group and 0.23% to 6.11% in statins-based group, and the proportion of LDL-C ≤ 1.4 mmol/L increased from 10.36% to 47.69% in PCSK-9 inhibitor group and 2.99% to 18.43% in statins-based group(P < 0.001 for both). There was no significant difference between PCSK-9 inhibitor and statins-based treatment in reducing the risk of MACE(hazard ratio = 2.52, 95% CI: 0.49-12.97, P = 0.250).CONCLUSIONS In the real world, PCSK-9 inhibitors combined with statins could significantly reduce LDL-C levels among patients with very high risk of ASCVD in China. 3Analysis of Gene Expression Pattern of Lumbar Intervertebral Disc Degeneration in Human显示文摘ObjectiveTo investigate the gene expression changes in normal and degeneration lumbar intervertebral disc in humans, providing information for clinical. MethodsThe PCR products of 4096 human genes were spotted onto a kind of chemical-material-coated-glass slides. The total RNAs were isolated from the tissues. Both the mRNAs from the degeneration and normal lumbar intervertebral disc in humans were reversely transcribed to the cDNAs, which used as the hybridization probes with the incorporations of fluorescent dUTP. The mixed probes were then hybridized to the cDNA microarray. After high-stringent washing, the cDNA microarray was scanned for the fluorescent signals and analyzed with computer image analysis. ResultsAmong the 4096 targets, there were 706 genes whose expression levels differed between the degeneration and normal lumbar intervertebral disc in all cases, comprising 298 up-regulated and 358 down-regulated ones. ConclusionDNA microarray technology is an effective technique in screening for differently expressed genes between the degeneration and normal lumbar intervertebral disc. 7A survey of the development of quadruped robots: Joint configuration, dynamic locomotion control method and mobile manipulation approach显示文摘Some quadruped robots developed recently show better dynamic performance and environmental adaptability than ever, and have been preliminarily applied in the field of emergency disposal, military reconnaissance and infrastructure construction. The development route, mechanisms design, control methods and mobile manipulating approaches of the quadruped robots are surveyed in this article. Firstly, the development route of the quadruped robot is combed, as the references of the forecast of the future work on quadruped robots. Then the bionic structure and the motion control method of the quadruped robot is summarized, the advantages and disadvantages are analyzed in aspects of gait switching, terrain adaption and disturbance resistance. Subsequently, aiming at the mobile manipulation of the quadruped robot, the representative leg-arm collaborative robots and the multi-task-oriented Whole-body Control (WBC) methods are introduced. 8Gaseous emissions from compressed natural gas buses in urban road and highway tests in China显示文摘The natural gas vehicle market is rapidly developing throughout the world, and the majority of such vehicles operate on compressed natural gas(CNG). However, most studies on the emission characteristics of CNG vehicles rely on laboratory chassis dynamometer measurements, which do not accurately represent actual road driving conditions. To further investigate the emission characteristics of CNG vehicles, two CNG city buses and two CNG coaches were tested on public urban roads and highway sections. Our results show that when speeds of 0–10 km/hr were increased to 10–20 km/hr, the CO_2, CO, nitrogen oxide(NO_x), and total hydrocarbon(THC) emission factors decreased by(71.6 ± 4.3)%,(65.6 ± 9.5)%,(64.9 ± 9.2)% and(67.8 ± 0.3)%, respectively. In this study, The Beijing city buses with stricter emission standards(Euro Ⅳ) did not have lower emission factors than the Chongqing coaches with Euro Ⅱ emission standards. Both the higher emission factors at 0–10 km/hr speeds and the higher percentage of driving in the low-speed regime during the entire road cycle may have contributed to the higher CO_2 and CO emission factors of these city buses. 10Elevating kinetics of passivated Fe anodes with NH_(4)Cl regulator:Toward low-cost,long-cyclic and green cathode-free Fe-ion aqueous batteries显示文摘The environment benignity and battery cost are major concerns for grid-scale energy storage applications.The emerging dendrite-free Fe-ion aqueous batteries are promising due to the rich natural abundance,low cost and non-toxicity for Fe resources.However,serious passivation reactions on Fe anodes and poor long-term cyclability for matched cathodes still stand in the way for their practical usage.To settle above constraints,we herein use NH_(4)Cl as the electrolyte regulator to elevate the reaction kinetics of passivated Fe anodes,and also propose a special cathode-free design to prolong the cells lifetime over 1,000 cycles.The added NH_(4)Cl can erode/break inert passivation layers and strengthen the ion conductivity of electrolytes,facilitating the reversible Fe plating/stripping and Fe^(2+)shuttling.The highly puffed nano carbon foams function as current collectors and actives anchoring hosts,enabling expedite Fe^(2+)adsorption/desorption,FeII/FeIII redox conversions and FeIII deposition.The configured rocking-chair Fe-ion cells have good environmental benignity and decent energy-storage behaviors,including high reactivity/reversibility,outstanding cyclic stability and far enhanced operation longevity.Such economical,long-cyclic and green cathode-free Fe-ion batteries may hold great potential in near-future energy-storage power stations.Shibo Chai Jianhui Zhu Jian Jiang Chang Ming Li 2022Nano Research2022,15,4:1
11Multivariate Rational Response Surface Approximation of Nodal Displacements of Truss Structures显示文摘Polynomial-basis response surface method has some shortcomings for truss structures in structural optimization,concluding the low fitting accuracy and the great computational effort. Based on the theory of approximation, a response surface method based on Multivariate Rational Function basis(MRRSM) is proposed. In order to further reduce the computational workload of MRRSM, focusing on the law between the cross-sectional area and the nodal displacements of truss structure, a conjecture that the determinant of the stiffness matrix and the corresponding elements of adjoint matrix involved in displacement determination are polynomials with the same order as their respective matrices, each term of which is the product of cross-sectional areas, is proposed. The conjecture is proved theoretically for statically determinate truss structure, and is shown corrected by a large number of statically indeterminate truss structures. The theoretical analysis and a large number of numerical examples show that MRRSM has a high fitting accuracy and less computational effort. 16Structural Basis of Glycan Recognition in Globally Predominant Human P[8]Rotavirus显示文摘Rotavirus(RV)causes acute gastroenteritis in infants and children worldwide.Recent studies showed that glycans such as histo-blood group antigens(HBGAs)function as cell attachment factors affecting RV host susceptibility and prevalence.P[8]is the predominant RV genotype in humans,but the structural basis of how P[8]RVs interact with glycan ligands remains elusive.In this study,we characterized the interactions between P[8]VP8~*s and glycans which showed that VP8~*,the RV glycan binding domain,recognized both mucin core 2 and H type 1 antigens according to the ELISA-based oligosaccharide binding assays.Importantly,we determined the structural basis of P[8]RV-glycans interaction from the crystal structures of a Rotateq P[8]VP8~*in complex with core 2 and H type 1 glycans at 1.82.3?,respectively,revealing a common binding pocket and similar binding mode.Structural and sequence analysis demonstrated that the glycan binding site is conserved among RVs in the P[Ⅱ]genogroup,while genotype-specific amino acid variations determined different glycan binding preference.Our data elucidated the detailed structural basis of the interactions between human P[8]RVs and different host glycan factors,shedding light on RV infection,epidemiology,and development of anti-viral agents.Xiaoman Sun Lei Dang Dandi Li Jianxun Qi Mengxuan Wang Wengang Chai Qing Zhang Hong Wang Ruixia Bai Ming Tan Zhaojun Duan 2020Virologica Sinica2020,35,2:1
