| 1 | The chromosome-level quality genome provides insights into the evolution of the biosynthesis genes for aroma compounds of Osmanthus fragrans显示文摘Sweet osmanthus(Osmanthus fragrans)is a very popular ornamental tree species throughout Southeast Asia and USA particularly for its extremely fragrant aroma.We constructed a chromosome-level reference genome of O.fragrans to assist in studies of the evolution,genetic diversity,and molecular mechanism of aroma development.A total of over 118 Gb of polished reads was produced from HiSeq(45.1 Gb)and PacBio Sequel(73.35 Gb),giving 100×depth coverage for long reads.The combination of Illumina-short reads,PacBio-long reads,and Hi-C data produced the final chromosome quality genome of O.fragrans with a genome size of 727 Mb and a heterozygosity of 1.45%.The genome was annotated using de novo and homology comparison and further refined with transcriptome data.The genome of O.fragrans was predicted to have 45,542 genes,of which 95.68%were functionally annotated.Genome annotation found 49.35%as the repetitive sequences,with long terminal repeats(LTR)being the richest(28.94%).Genome evolution analysis indicated the evidence of whole-genome duplication 15 million years ago,which contributed to the current content of 45,242 genes.Metabolic analysis revealed that linalool,a monoterpene is the main aroma compound.Based on the genome and transcriptome,we further demonstrated the direct connection between terpene synthases(TPSs)and the rich aromatic molecules in O.fragrans.We identified three new flower-specific TPS genes,of which the expression coincided with the production of linalool.Our results suggest that the high number of TPS genes and the flower tissue-and stage-specific TPS genes expressions might drive the strong unique aroma production of O.fragrans. | Xiulian Yang Yuanzheng Yue Haiyan Li Wenjie Ding Gongwei Chen Tingting Shi Junhao Chen Min S.Park Fei Chen Lianggui Wang | 2018 | Horticulture Research2018,5,1: | 16 |
| 2 | Mouse macrophage specific knockout of SIRT1 influences macrophage polarization and promotes angiotensin Ⅱ-induced abdominal aortic aneurysm formation显示文摘Abdominal aortic aneurysm(AAA) is a vascular degenerative disease. Macrophage polarization and the balance between classically activated macrophages(M1) and alternatively activated macrophages(M2)are crucial for AAA pathogenesis. The present study aims to investigate the roles of macrophage SIRT1 in AAA formation and macrophage polarization. We found that in mouse peritoneal macrophages, SIRT1 expression was decreased after M1 stimulation, but was enhanced after M2 stimulation. Results from SIRT1^(flox/flox) mice and macrophage specific SIRT1 knockout mice with treatment of angiotensin II(Ang II)for 4 weeks showed that macrophage specific deficiency of SIRT1 increased the incidence of AAA and exacerbated the severity, including more severe aneurysm types, enlarged diameter of the aneurysm and increased degradation of elastin. In mouse aortas, SIRT1 deficiency increased the proinflammatory M1 molecule inducible nitric oxide synthase(i NOS), and decreased M2 molecules such as arginase 1(Arg1) and mannose receptor(MR). Furthermore, in peritoneal macrophages, SIRT1 deficiency increased the expression of M1 inflammatory molecules, but decreased the expression of M2 molecules. Overexpression of SIRT1 had the opposite effects. | 3 | Anlotinib has good efficacy and low toxicity: a phase Ⅱ study of anlotinib in pre-treated HER-2 negative metastatic breast cancer显示文摘Objective:Anlotinib is a novel tyrosine kinase inhibitor blocking angiogenesis.This study was performed to assess the efficacy and safety of anlotinib in patients with metastatic breast cancer.Methods:Patients with HER2-negative breast cancer,who were pre-treated with anthracycline or taxanes in a neoadjuvant,adjuvant,or metastatic setting,and had treatment failure after at least one prior chemotherapy regimen in the metastatic setting were enrolled.Anlotinib was administered at 12 mg daily for 14 days in a 21-day cycle until disease progression or unacceptable toxicity occurred.Simultaneously,5–10 m L of venous blood was collected to perform circulating tumor DNA(ct DNA)testing every 2 treatment cycles.The primary endpoint was the objective response rate(ORR).Secondary endpoints included the disease control rate(DCR),progression-free survival(PFS),overall survival,safety,and biomarkers.Results:Twenty-six eligible patients were enrolled,with a median age of 56(30–75)years.The median follow-up time was 10.5 months.The ORR was 15.4%,the DCR was 80.8%,and the median PFS was 5.22 months(95%confidence interval 2.86–6.24).Fourteen(53.8%)patients survived for more than 10 months.The changes in the detectable ct DNA variant allele frequency were consistent with the tumor response.The most common treatment-related adverse events were hypertension(57.7%),thyroidstimulating hormone elevation(34.6%),and hand-foot syndrome(23.1%).Conclusion:Anlotinib showed objective efficacy with tolerable toxicity in heavily pre-treated,metastatic HER2-negative breast cancer.The dynamic changes in the ct DNA variant allele fraction may be predictive of the tumor response. | Nanlin Hu Yiran Si Jian Yue Tingting Sun Xue Wang Zhuqing Jia Songlin Gao Qiao Li Yang Shao Jiayu Wang Yang Luo Fei Ma Binghe Xu Peng Yuan | 2021 | Cancer Biology & Medicine2021,18,3: | 11 |

WSNs are applied in almos every aspect of smart grid, including power generation, power transmission, power distribution, power utilization and power dispatch, and the data query processing of‘WSNs in power grid' become an hotspo issue due to the amount of data of power grid is very large and the requirement of response time is very high. To meet the demands top-k query processing is a good choice which performs the cooperative query by aggregating the database objects' degree of match for each different query predicate and returning the best k matching objects. In this paper, a framework that can effectively apply top-k query to wireless sensor network in smart grid is proposed, which is based on the cluster-topology sensor network. In the new method, local indices are used to optimize the necessary query routing and process intermediate results inside the cluster to cu down the data traffic, and the hierarchical join query is executed based on the local resultsBesides, top-k query results are verified by the clean-up process, and two schemes are taken to deal with the problem of node's dynamicity, which further reduce communication cost. | 10 | Gaseous emissions from compressed natural gas buses in urban road and highway tests in China显示文摘The natural gas vehicle market is rapidly developing throughout the world, and the majority of such vehicles operate on compressed natural gas(CNG). However, most studies on the emission characteristics of CNG vehicles rely on laboratory chassis dynamometer measurements, which do not accurately represent actual road driving conditions. To further investigate the emission characteristics of CNG vehicles, two CNG city buses and two CNG coaches were tested on public urban roads and highway sections. Our results show that when speeds of 0–10 km/hr were increased to 10–20 km/hr, the CO_2, CO, nitrogen oxide(NO_x), and total hydrocarbon(THC) emission factors decreased by(71.6 ± 4.3)%,(65.6 ± 9.5)%,(64.9 ± 9.2)% and(67.8 ± 0.3)%, respectively. In this study, The Beijing city buses with stricter emission standards(Euro Ⅳ) did not have lower emission factors than the Chongqing coaches with Euro Ⅱ emission standards. Both the higher emission factors at 0–10 km/hr speeds and the higher percentage of driving in the low-speed regime during the entire road cycle may have contributed to the higher CO_2 and CO emission factors of these city buses. | 12 | Genomic Epidemiology of Carbapenemaseproducing Klebsiella pneumoniae in China显示文摘The rapid spread of carbapenemase-producing Klebsiella pneumoniae(cpKP)poses serious threats to public health;however,the underlying genetic basis for its dissemination is still unknown.We conducted a comprehensive genomic epidemiology analysis on 420 cpKP isolates collected from 70 hospitals in 24 provinces/autonomous regions/municipalities of China during 2009–2017 by short-/long-read sequencing.The results showed that most cpKP isolates were categorized into clonal group 258(CG258),in which ST11 was the dominant clone.Phylogenetic analysis revealed three major clades including the top one of Clade 3 for CG258 cpKP isolates.Additionally,carbapenemase gene analysis indicated that blaKPC was dominant in the cpKP isolates,and most blaKPC genes were located in five major incompatibility(Inc)groups of blaKPC-harboring plasmids.Importantly,three advantageous combinations of host–blaKPC-carrying plasmid(Clade 3.1+3.2–IncFIIpHN7A8,Clade 3.1+3.2–IncFIIpHN7A8:IncR,and Clade 3.3–IncFIIpHN7A8:IncpA1763-KPC)were identified to confer cpKP isolates the advantages in both genotypes(strong correlation/coevolution)and phenotypes(resistance/growth/competition)to facilitate the nationwide spread of ST11/CG258 cpKP.Intriguingly,Bayesian skyline analysis illustrated that the three advantageous combinations might be directly associated with the strong population expansion during 2007–2008 and subsequent maintenance of the population of ST11/CG258 cpKP after 2008.We then examined drug resistance profiles of these cpKP isolates and proposed combination treatment regimens for CG258/non-CG258 cpKP infections.Thus,the findings of our systematical analysis shed light on the molecular epidemiology and genetic basis for the dissemination of ST11/CG258 cpKP in China,and much emphasis should be given to the close monitoring of advantageous cpKP–plasmid combinations. | Cuidan Li Xiaoyuan Jiang Tingting Yang Yingjiao Ju Zhe Yin Liya Yue Guannan Ma Xuebing Wang Ying Jing Xinhua Luo Shuangshuang Li Xue Yang Fei Chen Dongsheng Zhou | 2022 | Genomics, Proteomics & Bioinformatics2022,20,6: | 1 |

| 15 | SEC1A and SEC6 synergistically regulate pollen tube polar growth显示文摘Pollen tube polar growth is a key physiological activity for angiosperms to complete double fertilization, which is highly dependent on the transport of polar substances mediated by secretory vesicles.The exocyst and Sec1/Munc18(SM) proteins are involved in the regulation of the tethering and fusion of vesicles and plasma membranes, but the molecular mechanism by which they regulate pollen tube polar growth is still unclear. In this study, we found that loss of function of SEC1A, a member of the SM protein family in Arabidopsis thaliana, resulted in reducing pollen tube growth and a significant increase in pollen tube width. SEC1A was diffusely distributed in the pollen tube cytoplasm, and was more concentrated at the tip of the pollen tube. Through coimmunoprecipitation-mass spectrometry screening,protein interaction analysis and in vivo microscopy,we found that SEC1A interacted with the exocyst subunit SEC6, and they mutually affected the distribution and secretion rate at the tip of the pollen tube. Meanwhile, the functional loss of SEC1A and SEC6 significantly affected the distribution of the SNARE(soluble N-ethylmaleimide-sensitive factor attachment protein receptor) complex member SYP125 at the tip of the pollen tube, and led to the disorder of pollen tube cell wall components. Genetic analysis revealed that the pollen tube-related phenotype of the sec1a sec6 double mutant was significantly enhanced compared with their respective single mutants. | 17 | Potential cuckoo hosts have similar egg rejection rates to parasitized host species显示文摘Background:Thrush species are rarely parasitized by cuckoos,but many have a strong egg recognition ability.To date,there is a limited understanding of the relationship between host egg rejection and cuckoo parasitism rate.Methods:By using egg experiments in the field,we compared egg rejection between two non‑parasitized potential host species and two parasitized hosts of cuckoos in the same region.Results:The White‑bellied Redstart(Luscinia phoenicuroides),a host of the Common Cuckoo(Cuculus canorus),rejected 66.6%of blue model eggs;the Elliot’s Laughingthrush(Trochalopteron elliotii),a host of the Large Hawk Cuckoo(Hierococcyx sparverioides),rejected 25%of blue model eggs and 46.1%of white model eggs;and the Chestnut Thrush(Turdus rubrocanus)and the Chinese Thrush(T.mupinensis),in which cuckoo parasitism has not been recorded,rejected 41.1 and 83.3%of blue model eggs,respectively.There were no significant differences in the egg rejection among them,although the Chinese Thrush showed the highest rate of egg rejection.Conclusions:This study indicates that the egg recognition ability of cuckoo hosts has no correlation with the actual parasitism rate of cuckoos.We suggest that the egg recognition ability of the two potential host species may have been retained from a parasitic history with the cuckoo,while the two common host species have developed their egg rejection abilities due to current parasitism pressure.In addition,our study highlights the importance of the multicuckoo parasite system for better understanding the selection pressure of parasitism on the evolution of host egg recognition abilities. | Tingting Yi Yue‑Hua Sun Wei Liang | 2020 | Avian Research2020,11,3: | 1 |

| 19 | Flexible and flame-retarding phosphorylated MXene/polypropylene composites for efficient electromagnetic interference shielding显示文摘Flame-retardant composites with high electromagnetic interference(EMI)shielding performance are desirable for electronic device packaging.Despite great potential of MXene for high EMI,it still remains a great challenge to develop high-performance flame-retardant polymer/MXene composites with excellent EMI shielding effectiveness because of the poor oxidative stability of MXene.Herein,phosphorylated MXene/polypropylene(PP)composites are prepared by coating phosphorylated MXene on PP fabric followed by spraying polyethylenimine(PEI)and hot-pressing.The phosphorylated MXene proves to be more durable against oxidation than pure MXene due to the protection effect of polyphosphates.Upon hot-pressing,melted PP fibers are fused together at their contact points and thus as-prepared composites are bi-continuous with two interpenetrating phases.The composites show significantly improved thermal stability and flame retardancy relative to pure PP,with a low total heat release(THR)of 3.7 kJ/g and a heat release rate(HRR)of 50.0 W/g,which are reduced by 78%and 87%,respectively.In addition,the composites exhibit a high electrical conductivity of~36,700 S/m and an EMI shielding performance of~90 d B over the whole frequency range of 8–12 GHz with a thickness of~400μm.The as-developed PP/MXene composites hold great promise for reliable protection of next-generation electronic devices working in complex environments. | Tingting Tang Shanchi Wang Yue Jiang Zhiguang Xu Yu Chen Tianshu Peng Fawad Khan Jiabing Feng Pingan Song Yan Zhao | 2022 | Journal of Materials Science & Technology2022,,16: | 1 |
| 20 | Transcriptome shock in an interspecific F1 triploid hybrid of Oryza revealed by RNA sequencing显示文摘Interspecific hybridization is a driving force in evolution and speciation of higher plants. Interspecific hybridization often induces immediate and saltational changes in gene expression,a phenomenon collectively termed 'transcriptome shock'. Although transcriptome shock has been reported in various plant and animal taxa,the extent and pattern of shock-induced expression changes are often highly idiosyncratic,and hence entails additional investigations. Here,we produced a set of interspecific F_1 triploid hybrid plants between Oryza sativa,ssp. japonica(2n=2x=24,genome AA) and the tetraploid form of O. punctata(2n=4x=48,genome,BBCC),and conducted RNA-seq transcriptome profiling of the hybrids and their exact parental plants. We analyzed both homeolog expression bias and overall gene expression level difference in the hybrids relative to the in silico 'hybrids'(parental mixtures). We found that approximately 16%(2,541) of the 16,112 expressed genes in leaf tissue of the F_1 hybrids showed nonadditive expression, which were specifically enriched in photosynthesis-related pathways. Interestingly,changes in the maternal homeolog expression, including non-stochastic silencing, were the major causes for altered homeolog expression partitioning in the F_1 hybrids.
