1MiR-30a Positively Regulates the Inflammatory Response of Microglia in Experimental Autoimmune Encephalomyelitis显示文摘Multiple sclerosis(MS) is a classical inflammatory demyelinating disease of the central nervous system(CNS). Microglia are the main resident immune cells in the CNS and are closely associated with the pathogenesis of MS.In the present study, we found that mi R-30 a was highly expressed in jellyfish-like microglia in chronic active lesions of MS patients, as well as in the microglia of mice with experimental autoimmune encephalomyelitis(EAE) at the chronic phase. In vitro, the conditioned supernatant of mouse microglia overexpressing miR-30 a promoted the apoptosis of oligodendrocyte precursor cells(OPCs), and inhibited OPC differentiation. In vivo, overexpressing miR-30 a in transplanted microglia exacerbated the progression of EAE.Overexpression and knock-down experiments in primary cultured mouse microglia showed that mi R-30 a increased the expression of IL-1 b and i NOS, which are pro-inflammatory, while inhibiting the expression of Ym-1 and CD206.Mechanistically, mi R-30 a inhibited the expression of Ppargc1 b, which is the co-activator of peroxisome proliferator-activated receptor gamma, resulting in pro-inflammatory effects. 2Clinicopathological significance of p53 and mdm2 protein expression in human pancreatic cancer显示文摘AIM: To study the clinicopathological significance of p53 and mdm2 protein expression in human pancreatic cancer. METHODS: To investigate the expression of p53 and mdm2 in pancreatic cancer by immunohistochemistry, and the relationships between the p53 and mdm2 protein expression and clinicopathological parameters in pancreatic cancer.RESULTS: The positive expression of p53 protein was found in 40 of 59 patients (67.8%) and that of mdm2 protein in 17 of 59 patients (28.8%). No obvious relationships were found between p53 as well as mdm2 expression and sex, tumor site, TNM staging and histological differentiation. p53 expression was increased in patients younger than 65 years old, while mdm2 had no relationship with age. The survival time of the patients with the positive expression of p53 and mdm2 proteins was obviously shorter than the other groups. CONCLUSION: Both p53 and mdm2 presented relatively high expression in human pancreatic cancer. 3Enhanced Poly(ethylene terephthalate)Hydrolase Activity by Protein Engineering显示文摘Poly(ethylene terephthalate)hydrolase(PETase)from Ideonella sakaiensis exhibits a strong ability to degrade poly(ethylene terephthalate)(PET)at room temperature,and is thus regarded as a potential tool to solve the issue of polyester plastic pollution.Therefore,we explored the interaction between PETase and the substrate(a dimer of the PET monomer ethylene terephthalate,2PET),using a model of PETase and its substrate.In this study,we focused on six key residues around the substrate-binding groove in order to create novel high-efficiency PETase mutants through protein engineering.These PETase mutants were designed and tested.The enzymatic activities of the R61A,L88F,and I179F mutants,which were obtained with a rapid cell-free screening system,exhibited 1.4 fold,2.1 fold,and 2.5 fold increases,respectively,in comparison with wild-type PETase.The I179F mutant showed the highest activity,with the degradation rate of a PET film reaching 22.5 mg perμmol·L^-1 PETase per day.Thus,this study has created enhanced artificial PETase enzymes through the rational protein engineering of key hydrophobic sites,and has further illustrated the potential of biodegradable plastics.Yuan Ma Mingdong Yao Bingzhi Li Mingzhu Ding Bo He Si Chen Xiao Zhou Yingjin Yuan 2018Engineering2018,4,6:10
4Effect of Sanhuangwuji powder,anti-rheumatic drugs,and ginger-partitioned acupoint stimulation on the treatment of rheumatoid arthritis with peptic ulcer:a randomized controlled study显示文摘OBjECTIVE:To observe the efficacy and safety of oral Sanhuangwuji powder,anti-rheumatic drugs(ARDs),and ginger-partitioned acupoint stimulation at Zusanli(ST 36) on the treatment of rheumatoid arthritis(RA) complicated by peptic ulcer.METHODS:This prospective randomized controlled study included 180 eligible inpatients and outpatients randomly assigned to an ARD treatment(n = 60),ginger-partitioned stimulation(n =60),or combination treatment(n = 60).Patients assigned to the ARD group were given oral celecoxib,methotrexate,and esomeprazole.Patients assigned to theginger-partitioned stimulation group were given ginger-partitioned acupoint stimulation at Zusanli(ST 36) in addition to the ARDs.Patients in the combination treatment group were given oral Sanhuangwuji powder,ginger-partitioned acupoint stimulation at Zusanli(ST 36),and ARDs.All patients were followed up for 2 months to evaluate clinical effects and safety.The study was registered in the World Health Organization database at the General Hospital of Chengdu Military Area Command Chinese People's Liberation Army(ChiCTR-TCC12002824).RESULTS:The combination treatment group had significantly greater improvements in RA symptoms,laboratory outcomes,and gastrointestinal symptom scores,compared with the other groups(P < 0.05).The peptic ulcer healing rate in the combination treatment group was significantly greater than that in the ARD treatment group(x^2 = 16.875,P < 0.05) and the ginger-partitioned stimulation group(x^2 = 6.171,P<0.05).CONCLUSION:Combination treatment with ginger-partitioned acupoint stimulation at Zusanli(ST36),oral Sanhuangwuji powder,and ARDs had a better clinical effect for RA with complicated peptic ulcer,compared with ARD treatmentalone or in combination with ginger-partitioned acupoint stimulation.Liu Defang Guo Mingyang Hu Yonghe Liu Taihua Yan Jiao Luo Yong Yun Mingdong Yang Min Zhang Jun Guo Linglin 2015Journal of Traditional Chinese Medicine2015,35,3:9
5Engineering the Biosynthesis of Caffeic Acid in Saccharomyces cerevisiae with Heterologous Enzyme Combinations显示文摘Engineering the biosynthesis of plant-derived natural products in microbes presents several challenges, especially when the expression and activation of the plant cytochrome P450 enzyme is required. By recruiting two enzymes—HpaB and HpaC—from several bacteria, we constructed functional 4- hydroxyphenylacetate 3-hydroxylase (4HPA3H) in Saccharomyces cerevisiae to take on a role similar to that of the plant-derived cytochrome P450 enzyme and produce caffeic acid. Along with a common tyrosine ammonia lyase (TAL), the different combinations of HpaB and HpaC presented varied capabilities in producing the target product, caffeic acid, from the substrate, L-tyrosine. The highest production of caffeic acid was obtained with the enzyme combination of HpaB from Pseudomonas aeruginosa and HpaC from Salmonella enterica, which yielded up to (289.4 ± 4.6) mg-L1 in shake-flask cultivation. The compatibility of heterologous enzymes within a yeast chassis was effectively improved, as the caffeic acid production was increased by 40 times from the initial yield. Six key amino acid residues around the flavin adenine dinucleotide (FAD) binding domain in HpaB from Pseudomonas aeruginosa were differentiate from those other HpaBs, and might play critical roles in affecting enzyme activity. 6Clinicopathological significance of Bcl-2 and Bax protein expression in human pancreatic cancer显示文摘AIM: To assess the clinicopathological significance of the expression of the apoptosis-inhibitory Bcl-2 protein (pBcl-2) and the apoptosis-promoting Bax protein (pBax) in human invasive ductal carcinomas (IDCs) of the pancreas. METHODS: Fifty-nine surgical specimens of IDCs of the pancreas were stained immunohistochemically to detectpBcl-2 and pBax expressions whose correlation to tumor classification, staging, and prognosis was analyzed by univariate and multivariate analyses. RESULTS: The expression of pBcl-2 and pBax was detected in 21 of 59 (35.6%) and in 29 of 59 (49.2%) patients with IDCs of the pancreas, respectively. Neither pBcl-2 nor pBax alone was correlated to TNM staging and differentiation degree of IDCs of the pancreas according to univariate analysis. By Mantel-Cox test, the median survival time after surgery for pBcl-2(+) and pBcl-2(-) groups were 14.3 and 7.3 mo, respectively (χ2= 9.357, P = 0.002) and that for pBax(+) and pBax(-) groups were 12.9 and 10.2 mo, respectively (χ2= 0.285, P>0.05).Contingency coefficient between pBd-2 and pBax expression was 0.298, indicating that there was correlation between them (χ2= 5.74, P<0.05). The median survival time after surgery for pBd-2(+)pBax(+) and pBcl-2(+)pBax(-) groups were 14.3 and 14.1 mo, respectively, and that for pBcl-2 (-)pBax(+) and pBcl-2(-)pBax(-) groups were 5.9 and 9.9 mo, respectively. There was a significant difference between pBcl-2(+)pBax(+) and pBcl-2(-)pBax(+) (χ2 = 5.06,P<0.05), such was the case for pBcl-2(+)pBax(+) andpBcl-2(-)pBax(-) (χ2= 7.18, P<0.01). Cox proportional hazards model for multivariate analysis was applied, indicating that pBcl-2, TNM staging, age and pBax were high risk factors of post-surgical survival time. 7Services and Key Technologies of the Internet of Things显示文摘This article introduces the services and development of the Internet of Things,and analyzes the driving forces and obstacles behind such development. Looking at application types and the different development stages of the Internet of Things,this article categorizes its services into four types:identity related services,information aggregation services,collaborative-aware services,and ubiquitous services. For the first two types of services,applications and system framework are discussed; for the last two types,development trends are discussed. Services provided by the Internet of Things will gradually be integrated into human life and society; with the development of the Internet of Things,applications will evolve from relatively simple identity-related and information aggregation-related applications,to collaboratively-aware,and finally ubiquitous applications. 11Microbionic and peristaltic robots in a pipe显示文摘A microbionic and peristaltic robot that simulates the motion of an earthworm to move within a micropipe is proposed. The robot consists of three flexible units. Each unit is composed of two plates connected with three shape memory alloys (SMA) 120° apart, the rubber gasbag around the SMA wires inflated with air inside. Each unit corresponds to a segment of an earthworm, and the SMA and rubber gasbag have the same functions as the cricoid and longitudinal muscles of the earthworm. 12Growing vertical aligned mesoporous silica thin film on nanoporous substrate for enhanced degradation,drug delivery and bioactivity显示文摘Mesoporous silica thin film has been widely used in various fields,particularly the medical implant coating for drug delivery.However,some drawbacks remain with the films produced by traditional method(evaporation-induced self-assembly,EISA),such as the poor permeability caused by their horizontal aligned mesochannels.In this study,the vertical aligned mesoporous silica thin film(VMSTF)is uniformly grown alongside the walls of titania nanotubes array via a biphase stratification growth method,resulting in a hierarchical two-layered nanotubular structure.Due to the exposure of opened mesopores,VMSTF exhibits more appealing performances,including rapid degradation,efficient small-molecular drug(dexamethasone)loading and release,enhanced early adhesion and osteogenic differentiation of MC3T3-E1 cells.This is the first time successfully depositing VMSTF on nanoporous substrate and our findings suggest that the VMSTF may be a promising candidate for bone implant surface coating to obtain bioactive performances.Zhe Li Yide He Lasse Hyldgaard Klausen Ning Yan Jing Liu Fanghao Chen Wen Song Mingdong Dong Yumei Zhang 2021Bioactive Materials2021,6,5:4
13WEB SERVICE SELECTION ALGORITHM BASED ON PRINCIPAL COMPONENT ANALYSIS显示文摘Existing Web service selection approaches usually assume that preferences of users have been provided in a quantitative form by users. However, due to the subjectivity and vagueness of preferences, it may be impractical for users to specify quantitative and exact preferences. Moreover, due to that Quality of Service (QoS) attributes are often interrelated, existing Web service selection approaches which employ weighted summation of QoS attribute values to compute the overall QoS of Web services may produce inaccurate results, since they do not take correlations among QoS attributes into account. To resolve these problems, a Web service selection framework considering user's preference priority is proposed, which incorporates a searching mechanism with QoS range setting to identify services satisfying the user's QoS constraints. With the identified service candidates, based on the idea of Principal Component Analysis (PCA), an algorithm of Web service selection named PCA-WSS (Web Service Selection based on PCA) is proposed, which can eliminate the correlations among QoS attributes and compute the overall QoS of Web services accurately. After computing the overall QoS for each service, the algorithm ranks the Web service candidates based on their overall QoS and recommends services with top QoS values to users. 14Effect of Sanhuangyilong decoction plus methotrexate on tumor necrosis factor alpha and interferon gamma in serum and synovial fluid in rheumatoid arthritis patients with symptom pattern of damp heat obstruction显示文摘OBJECTIVE: To investigate the effect of Sanhuangyilong decoction plus methotrexate(MTX) on Interferon gamma(IFN-γ) and tumor necrosis factor alpha(TNF-α) in the serum and synovial fluid of rheumatoid arthritis(RA) patients with damp-heat-obstruction symptom pattern, Sanhuangyilong decoction and the role of TNF-α and IFN-γ in the development of RA.METHODS: RA inpatients with damp-heat-obstruction symptom pattern(partly with knee joint effusion) were selected as the research subjects. Before the treatment, healthy subjects and osteoarthritis(OA) patients with knee joint effusion were assigned to the serum control group and the synovial fluid control group, respectively; during the treatment, RA patients with damp-heat-obstruction symptom pattern were divided into two groups:one is combined group that was administered Sanhuangyilong decoction plus MTX; the other group was MTX group that received MTX only. The expression levels of TNF-α and IFN-γ in the serum and synovial fluid were measured with enzyme-linked immunosorbent assay(ELISA) before and after the treatment, and the peripheral blood levels of erythrocyte sedimentation rate(ESR), C-reactive protein(CRP) and disease activity score in 28 joints(DAS28)were determined.RESULTS: Before treatment, the serum levels of TNF-α and IFN-γ in the RA patients with dampheat-obstruction symptom pattern were higher than those in healthy control group(P < 0.05).The expression levels of TNF-α and IFN-γ in the synovial fluid of the RA patients were higher than those in the serum of the RA patients(P < 0.05). The expression levels of TNF-α and IFN-γ in the synovial fluid of the RA patients were higher than those of the sy-novial fluid of the osteoarthritis patients(P < 0.05).The expression of TNF-α and IFN-γ in the serum and synovial fluid of the RA patients had no correlation with the inflammatory activity index ESR, CRP,or DAS28(P > 0.05). After 2 weeks of treatment, the expression level of TNF-α and IFN-γ in the combined group had increased, although the difference was not statistically significant(P > 0.05); in contrast, ESR, CRP, and DAS28 decreased, and the difference was statistically significant(P < 0.01). After 4 weeks of therapy, TNF-alpha and IFN-γ, ESR,CRP, and DAS28 in the combined group decreased compared with the before-treatment levels(P <0.01). After 2 w of treatment, the differences in the TNF-α and IFN-γ expression levels in the combined group were not statistically significant(P > 0.05)compared with that in the MTX group, although there were statistically significant differences in the ESR, CRP, and DAS28(P < 0.05). After 4 weeks of treatment, differences in TNF-α, IFN-γ, ESR, CRP, and DAS28 in the combined group compared with MTX group were statistically significant(P < 0.01).CONCLUSION: TNF-α and IFN-γ might be involved in the development of RA. 15Ultrathin Ti_(3)C_(2)T_(x)(MXene) Nanosheet-Wrapped NiSe_(2) Octahedral Crystal for Enhanced Supercapacitor Performance and Synergetic Electrocatalytic Water Splitting显示文摘Metal selenides,such as NiSe2,have exhibited great potentials as multifunctional materials for energy storage and conversation.However,the utilization of pure NiSe2 as electrode materials is limited by its poor cycling stability,low electrical conductivity,and insufficient electrochemically active sites.To remedy these defects,herein,a novel NiSe2/Ti3C2Tx hybrid with strong interfacial interaction and electrical properties is fabricated,by wrapping NiSe2 octahedral crystal with ultrathin Ti3C2Tx MXene nanosheet.The NiSe2/Ti3C2Tx hybrid exhibits excellent electrochemical performance,with a high specific capacitance of 531.2 Fg-1 at1 A g-1 for supercapacitor,low overpotential of 200 mV at 10 mA g-1,and small Tafel slope of 37.7 mV dec-1 for hydrogen evolution reaction(HER).Furthermore,greater cycling stabilities for NiSe2/Ti3C2Tx hybrid in both supercapacitor and HER have also been achieved.These significant improvements compared with unmodified NiSe2 should be owing to thestrong interfacial interaction between NiSe2 octahedral crystal and Ti3C2Tx MXene,which provides enhanced conductivity,fast charge transfer as well as abundant active sites,and highlight the promising potentials in combinations of MXene with metal selenides for multifunctional applications such as energy storage and conversion.Hanmei Jiang Zegao Wang Qian Yang Luxi Tan Lichun Dong Mingdong Dong 2019Nano-Micro Letters2019,11,2:4
16Formation Mechanism of Ground Fissures Originated from the Hanging Wall of Normal Fault:A Case in Fen-Wei Basin,China显示文摘This paper takes Fen-Wei Basin(FWB)as a case to study the ground fissures controlled by normal fault.Based on the field investigation,geophysical exploration,drilling,GNSS data and numerical calculation,the characteristics and mechanism of ground fissures originated from the hanging wall of normal faults are revealed.The results show that the distribution of ground fissures in the hanging wall and heading wall of the active faults is not uniform.Ground fissures are mostly distributed in the hanging wall of active faults and show a linear distribution on the surface,their strike is consistent with the fault,mainly characterized by vertical offset and horizontal tension.Ground fissures destroy the farmland and building foundation through which they pass and cause the rupture or displacement.In profile section,the ground fissure shows the characteristics of normal faults and dislocates the strata,and is connected with the underlying faults.Numerical analysis shows that the vertical displacement of normal fault activity in hanging wall is much larger than that in heading wall,which is the reason that tectonic ground fissures mainly originate from hanging wall.The range of dangerous area of ground fissures is controlled by the depth of fault,the strength of the ground fissures disaster is mainly controlled by the activity of fault.The formation of the ground fissures originated from the hanging wall of the fault experienced three stages:the main fault activity stage,the secondary fault activity stage and the fissure formation stage.Zhijie Jia Jianbing Peng Quanzhong Lu Jianwei Qiao Feiyong Wang Mingdong Zang Yang Liu Junyan Zhao Fengji Zhu 2022Journal of Earth Science2022,33,2:2
17Application of machine learning in predicting the rate-dependent compressive strength of rocks显示文摘Accurate prediction of compressive strength of rocks relies on the rate-dependent behaviors of rocks, and correlation among the geometrical, physical, and mechanical properties of rocks. However, these properties may not be easy to control in laboratory experiments, particularly in dynamic compression experiments. By training three machine learning models based on the support vector machine(SVM), backpropagation neural network(BPNN), and random forest(RF) algorithms, we isolated different input parameters, such as static compressive strength, P-wave velocity, specimen dimension, grain size, bulk density, and strain rate, to identify their importance in the strength prediction. Our results demonstrated that the RF algorithm shows a better performance than the other two algorithms. The strain rate is a key input parameter influencing the performance of these models, while the others(e.g. static compressive strength and P-wave velocity) are less important as their roles can be compensated by alternative parameters. The results also revealed that the effect of specimen dimension on the rock strength can be overshadowed at high strain rates, while the effect on the dynamic increase factor(i.e. the ratio of dynamic to static compressive strength) becomes significant. The dynamic increase factors for different specimen dimensions bifurcate when the strain rate reaches a relatively high value, a clue to improve our understanding of the transitional behaviors of rocks from low to high strain rates.Mingdong Wei Wenzhao Meng Feng Dai Wei Wu 2022Journal of Rock Mechanics and Geotechnical Engineering2022,14,5:2

20Effects of Antiadhesion preparation on free fibrinogen and fibrin degrading products in abdominal exudates of rabbits postoperatively显示文摘AIM: To observe effects of ACOL on fibrinogen (FIB), fibrin degrading products (FDP) and changes of FIB and FDP concentration in rabbits with intro-abdominal exudates during 7 d after major abdominal surgery.METHODS: Sixty New Zealand rabbits were randomly divided into 4 groups: ACOL group, the control group, DCT group and the normal group.
