| 1 | Altered trends in carbon uptake in China’s terrestrial ecosystems under the enhanced summer monsoon and warming hiatus显示文摘The carbon budgets in terrestrial ecosystems in China are strongly coupled with climate changes.Over the past decade,China has experienced dramatic climate changes characterized by enhanced summer monsoon and decelerated warming.However,the changes in the trends of terrestrial net ecosystem production(NEP)in China under climate changes are not well documented.Here,we used three ecosystem models to simulate the spatiotemporal variations in China's NEP during 1982–2010 and quantify the contribution of the strengthened summer monsoon and warming hiatus to the NEP variations in four distinct climatic regions of the country.Our results revealed a decadal-scale shift in NEP from a downtrend of–5.95 Tg C/yr^2(reduced sink)during 1982–2000 to an uptrend of 14.22 Tg C/yr^2(enhanced sink)during 2000–10.This shift was essentially induced by the strengthened summer monsoon,which stimulated carbon uptake,and the warming hiatus,which lessened the decrease in the NEP trend.Compared to the contribution of 56.3%by the climate effect,atmospheric CO2 concentration and nitrogen deposition had relatively small contributions(8.6 and 11.3%,respectively)to the shift.In conclusion,within the context of the global-warming hiatus,the strengthening of the summer monsoon is a critical climate factor that enhances carbon uptake in China due to the asymmetric response of photosynthesis and respiration.Our study not only revealed the shift in ecosystem carbon sequestration in China in recent decades,but also provides some insight for understanding ecosystem carbon dynamics in other monsoonal areas. | Honglin He Shaoqiang Wang Li Zhang Junbang Wang Xiaoli Ren Lei Zhou Shilong Piao Hao Yan Weimin Ju Fengxue Gu Shiyong Yu Yuanhe Yang Miaomiao Wang Zhongen Niu Rcmg Ge Huimin Yan Mei Huang Guoyi Zhou Yongfei Bai Zongqiang Xie Zhiyao Tang Bingfang Wu Leiming Zhang Nianpeng He Qiufeng Wang Guirui Yu | 2019 | National Science Review2019,6,3: | 24 |
| 2 | The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. Dipeptidyl peptidase Ⅳ inhibition improves the impaired insulin secretion and decrease postprandial concentrations of glucagon by enhancing the incretin hormone levels lucagon-like peptide-1(GLP-1) and glucose-dependent insulinotropic polypeptide/gastric inhibitory polypeptide(GIP). Recently, DPP Ⅳ inhibitors have attracted more and more attention, several of which have entered pre-clinical and clinical trials, and one has received approval for use as an anti-diabetic agent. Among the DPP Ⅳ inhibitors, two leading agents(sitagliptin and vildagliptin) have been shown to be effective in reducing glycosylated hemoglobin(HbA1c) and fasting plasma glucose(FPG) in patients with type 2 diabetes. | 5 | The role of GLI2-ABCG2 signaling axis for 5Fu resistance in gastric cancer显示文摘Gastric cancer is a leading cause of cancer-related mortality worldwide, and options to treat gastric cancer are limited. Fluorouracil(5Fu)-based chemotherapy is frequently used as a neoadjuvant or an adjuvant agent for gastric cancer therapy. Most patients with advanced gastric cancer eventually succumb to the disease despite the fact that some patients respond initially to chemotherapy. Thus, identifying molecular mechanisms responsible for chemotherapy resistance will help design novel strategies to treat gastric cancer. In this study, we discovered that residual cancer cells following 5Fu treatment have elevated expression of hedgehog(Hg) target genes GLI1 and GLI2, suggestive of Hh signaling activation. Hh signaling, a pathway essential for embryonic development, is an important regulator for putative cancer stem cells/residual cancer cells. We found that high GLI1/GLI2 expression is associated with some features of putative cancer stem cells, such as increased side population. We demonstrated that GLI2 knockdown sensitized gastric cancer cells to 5Fu treatment, decreased ABCG2 expression, and reduced side population. Elevated GLI2 expression is also associated with an increase in tumor sphere size, another marker for putative cancer stem cells. We believe that GLI2 regulates putative cancer stem cells through direct regulation of ABCG2. ABCG2 can rescue the GL12 shRNA effects in 5Fu response, tumor sphere formation and side population changes, suggesting that ABCG2 is an important mediator for GLI2-associated 5Fu resistance. The relevance of our studies to gastric cancer patient care is reflected by our discovery that high GLI1/GLI2/ABCG2 expression is associated with a high incidence of cancer relapse in two cohorts of gastric cancer patients who underwent chemotherapy(containing 5Fu). | 8 | AFM quantitative analysis and determination of shear angle of {259}_f martensitic surface relief显示文摘An attentive observation and quantitative analysis of {259}f martensitic surface relief in an Fe-23Ni-0.55C alloy are made by means of an atomic force microscope (AFM), and different martensitic variants' shear angles are determined in this paper. The experiments show that {259}f martensitic surface relief exhibits regular shape in many cases, which is in agreement with the prediction of invariant plane strain (IPS). Generally, {259}f martensitic surface relief appears to be 'N'-shaped, but it is tent-shaped in the case of zigzag-shaped martensite. The compressed deformation of parent phase diminishes the surface relief in size but with little change of its relief angle. {259}f martensitic surface relief, large or small, has approximately the same relief angles, exhibiting a good 'self-similar fractal'. | 9 | Tumor shrinkage by cyclopamine tartrate through inhibiting hedgehog signaling显示文摘The link of hedgehog (Hh) signaling activation to human cancer and synthesis of a variety of Hh signaling inhibitors raise great expectation that inhibiting Hh signaling may be effective in human cancer treatment. Cyclopamine (Cyc), an alkaloid from the Veratrum plant, is a specific natural product inhibitor of the Hh pathway that acts by targeting smoothened (SMO) protein. However, its poor solubility, acid sensitivity, and weak potency relative to other Hh antagonists prevent the clinical development of Cyc as a therapeutic agent. Here, we report properties of cyclopamine tartrate salt (CycT) and its activities in Hh signaling-mediated cancer in vitro and in vivo. Unlike Cyc, CycT is water soluble (5-10 mg/mL). The median lethal dose (LD50) of CycT was 62.5 mg/kg body weight compared to 43.5 mg/kg for Cyc, and the plasma half-life (T1/2) of CycT was not significantly different from that of Cyc. We showed that CycT had a higher inhibitory activity for Hh signaling-dependent motor neuron differentiation than did Cyc (IC50 = 50 nmol/L for CycT vs. 300 nmol/L for Cyc). We also tested the antitumor effectiveness of these Hh inhibitors using two mouse models of basal cell carcinomas (K14cre:Ptch1neo/neo and K14cre:SmoM2YFP). After topical application of CycT or Cyc daily for 21 days, we found that all CycT-treated mice had tumor shrinkage and decreased expression of Hh target genes. Studying the genes that predominantly or even specifically expressed in stamens will aid in understanding the molecular mechanism of stamen development. The structure of a rice CHS-like gene (D5) which predominantly expressed in anthers has been analysized, showing high structural similarity with two other anther-specific genes (A1 and BA42) from Brassica napus. Northern analysis also reveals that the three genes share a similar expression pattern and that the expression of D5 may relate to the early development of pollens. | 14 | Interface engineering of high performance all-inorganic perovskite solar cells via low-temperature processed TiO2 nanopillar arrays显示文摘All-inorganic perovskite solar cells suffer from low performance due to unsatisfactory carrier transport and light harvesting efficiency.Semiconductor nanopillar arrays can reduce light reflection loss and suppress exciton recombination dynamics in optoelectronic devices.In all-inorganic perovskite solar cells,few studies employing TiO_(2)nanopillar arrays(TiO_(2)NaPAs)have been reported to improve the device performance.Herein,well-arranged TiO_(2)NaPAs are chosen to enhance the interfacial contact between perovskite and electron transporting layers for improving the carrier transport.Notably,TiO_(2)NaPAs can be directly fabricated on rigid/flexible substrates at roughly room temperature by unique glancing angle deposition,which is more available than high-temperature hydrothermal/solvothermal methods.By embedding TiO_(2)NaPAs into chemical processable CsPbI2Br layers,continuous and intimate films are readily formed,guaranteeing large physical contact for facilitating more effective electron injection and charge separation.The vertically grown TiO_(2)NaPAs also provide a straightforward electron transporting path to electrodes.In addition,TiO_(2)NaPAs can guide the incident light and enhance the light-harvesting ability of CsPbI2Br films.As a result,the solar cell with TiO_(2)NaPAs displays a power conversion efficiency of 11.35%higher than planar control of 10.04%,and exhibits better long-term thermal stability.This strategy provides an opportunity by constructing direct interfacial regulation towards the performance improvement of inorganic perovskite solar cells. | Bingkun Pan Jiahao Gu Xiaoli Xu Lingbo Xiao Jie Zhao Guifu Zou | 2021 | Nano Research2021,14,10: | 2 |
| 15 | Rice FLOURY ENDOSPERM22, encoding a pentatricopeptide repeat protein, is involved in both mitochondrial RNA splicing and editing and is crucial for endosperm development显示文摘Most of the reported P-type pentatricopeptide repeat(PPR) proteins play roles in organelle RNA stabilization and splicing. However, P-type PPRs involved in both RNA splicing and editing have rarely been reported, and their underlying mechanism remains largely unknown. Here, we report a rice floury endosperm22(flo22) mutant with delayed amyloplast development in endosperm cells. Map-based cloning and complementation tests demonstrated that FLO22 encodes a mitochondrion-localized P-type PPR protein.Mutation of FLO22 resulting in defective transsplicing of mitochondrial nad1 intron 1 and perhaps causing instability of mature transcripts affected assembly and activity of complex Ⅰ, and mitochondrial morphology and function. RNA-seq analysis showed that expression levels of many genes involved in starch and sucrose metabolism were significantly down-regulated in the flo22mutant compared with the wild type, whereas genes related to oxidative phosphorylation and the tricarboxylic acid cycle were significantly upregulated. In addition to involvement in splicing as a P-type PPR protein, we found that FLO22 interacted with DYW3, a DYW-type PPR protein, and they may function synergistically in mitochondrial RNA editing. | 20 | Genetic Evidence for XPC-KRAS Interactions During Lung Cancer Development显示文摘Lung cancer causes more deaths than breast, colorectal and prostate cancers combined. Despite major advances in targeted therapy in a subset of lung adenocarcinomas, the overall 5-year survival rate for lung cancer worldwide has not significantly changed for the last few decades. DNA repair deficiency is known to contribute to lung cancer development. In fact, human polymorphisms in DNA repair genes such as xeroderma pigmentosum group C(XPC) are highly associated with lung cancer incidence. However, the direct genetic evidence for the role of XPC for lung cancer development is still lacking. Mutations of the Kirsten rat sarcoma viral oncogene homolog(Kras) or its downstream effector genes occur in almost all lung cancer cells, and there are a number of mouse models for lung cancer with these mutations. Using activated Kras, Kras LA1, as a driver for lung cancer development in mice, we showed for the first time that mice with Kras LA1 and Xpc knockout had worst outcomes in lung cancer development, and this phenotype was associated with accumulated DNA damage. Using cultured cells, we demonstrated that induced expression of oncogenic KRAS G12 V led to increased levels of reactive oxygen species(ROS) as well as DNA damage, and both can be suppressed by anti-oxidants.
