| 1 | Abundance and biomass of heterotrophic microbes in the Kongsfjorden, Svalbard显示文摘Abundance and biomass of heterotrophic microbes were investigated using epifluorescense microscopy in August 2004 along a transect in the Kongsfjorden, a glacial fjord in the high arctic, located in the north-western part of Svalbard. A bacterial abundance was low at outer fjord stations (on average 2.29×108 ind./dm3) and was higher at inner fjord stations (on average 3.52×108 ind./dm3).The biomass of bacteria ranged from 3.69 to 8.01 mg/m3.The abundance of bacteria was low at the surface of water(0 m), whereas the higher abundances often occurred at depths of 30~50 m.The abundance of heterotrophic flagellates varied from 2.785 to 8.891×104 ind./dm3.The assemblage was numerically dominated by less than 5 μm heterotrophic flagellates (78.67%~94.80%).The biomass of heterotrophic flagellates varied from 0.37 to 2.30 mg/m3.The mean biomass distribution for all stations was 4.91% for less than 5 μm heterotrophic flagellates, 86.74% for 6~20 μm flagellates and 8.35% for greater than 20 μm sized flagellates.The low biomass of heterotrophic flagellates often occurred at the surface of water(0 m),while the maximum value of biomass occurred at different depths. | 2 | Introduction of antisense waxy gene into main parent lines of indica hybrid rice显示文摘Amylose content in rice endosperm is one of the key determinants of rice eating and cooking quality, and the poor quality of indica hybrid rice is closely related to the high amylose level in rice grains. In order to improve the grain quality of the indica hybrid rice by genetic engineering, an antisense fragment of rice waxy gene, driven by the 5’-franking sequences of the rice waxy gene, was successfully introduced into three major parent lines of indica hybrid rice, all contain a high amylose level in the grains, via Agrobacte-rium, and more than 100 hygromycin-resistant plants were regenerated. The analysis of PCR amplification and Southern blots indicated that the T-DNA containing the antisense waxy gene had been integrated into the genome of transgenic rice plants. Most of the primary transgenic rice plants grew normally, and the mature seeds from these transgenic plants were performed for analysis of the amylose content. | 3 | FBG Arrays for Quasi-Distributed Sensing: A Review显示文摘Fiber Bragg grating (FBG) array is a powerful technique for quasi-distributed sensing along the entire length of sensing fiber with fast response and high precision. It has been widely used for temperature, strain, and vibration monitoring. In this review work, an overview on the recent advances of FBG arrays is conducted. Firstly, the fabrication methods of FBG array are reviewed, which include femtosecond laser system and online writing technique. Then, the demodulation techniques for FBG arrays are presented and discussed. Distributed static sensing can be performed by demodulating wavelength shift of each FBG, while phase demodulation techniques with low noise are employed for dynamic vibration sensing. Simultaneous distributed dynamic and static sensing system based on FBG array is also outlined. Finally, possible future directions are discussed and concluded. | 4 | Spatial heterogeneity of a microbial community in Kongsfjorden,Svalbard during late summer 2006 and its relationship to biotic and abiotic factors显示文摘The 16S and 18S ribosomal ribonucleic acid genes of microbial organisms collected from the contrasting environments (temperature,salinity,silicate,phosphate and nitrate,p <0.05) of the inner and outer basins of Kongsfjorden (Spitsbergen,Arctic) were studied using polymerase chain reaction-denaturing gradient gel electrophoresis(DGGE) fingerprinting.Comparison of the microbial fingerprints and the physicochemical parameters revealed that molecular methodology exhibited a greater sensitivity.Sequences obtained from bacterial DGGE were affiliated with four main phylogenetic groups of bacteria:Proteobacteria(Alpha,Beta and Gamma),Bacteroidetes,Verrucomicrobia and Cyanobacteria.The relationships between the genotype distribution of these microbes and associated biotic/abiotic factors,revealed by canonical correspondence analysis,showed that Station 1 at 30 m (outer fjord) was grouped separately from the other sites.This difference could be a consequence of the thermocline and base of the euphotic layer at this depth where the Atlantic and Arctic-type waters overlapped. | GUO ChaoYing HE JianFeng ZHANG Fang CAI MingHong WANG GuiZhong | 2011 | Chinese Journal of Polar Science2011,22,1: | 3 |
| 5 | Flow cytometry investigation of picoplankton across latitudes and along the circum Antarctic Ocean显示文摘Using a flow cytometer (FCM) onboard the R/V Xuelong during the 24th Chinese Antarctic cruise, picoplankton community structure and biomass in the surface water were examined along the latitude and around the Antarctic Ocean. Salinity and temperature were automatically recorded and total Chl a was determined. Along the cruise, the abundance of Synechococcus, Prochlorococcus, pico-eukaryotes and heterotrophic bacteria ranged in 0.001-1.855 × 10 8 ind./L, 0.000-2.778 × 10 8 ind./L, 0.002-1.060 × 10 8 ind./L and 0.132-27.073 × 10 8 ind./L, respectively. Major oceanic distribution of Synechococcus and Prochlorococcus appeared between latitudes 30 N and 30 S. Prochlorococcus was mainly influenced by water temperature, water mass combination and freshwater inflow. Meanwhile, Synechococcus distribution was significantly associated with landing freshwater inflow. Pico-eukaryotes and heterotrophic bacteria were distributed all over the oceans, but with a relatively low abundance in the high latitudes of the Antarctic Ocean. | 6 | Concentration and distribution of 17 organochlorine pesticides (OCPs) in seawater from the Japan Sea northward to the Arctic Ocean显示文摘Seventeen classic organochlorine pesticides in surface seawater were studied in terms of their composition pattern as well as their distribution pattern in the areas covering the Japan Sea,Okhotsk Sea,Bering Sea,Chukchi Sea and Arctic Ocean.Their concentrations varied,but roughly two levels were seen with one ranging between 0.1 and 1 ng L-1 for most HCH isomers and the other lower than 0.1 ng L-1 for other chemicals.Of the 17 target compounds,HCHs were dominant with a total concentration percentage generally more than 50%,and a relatively high concentration percentage of heptachlor and aldrine was also observed at scattered stations.The historical long-term trend of several target chemicals in the five sea zones considered was discussed in comparison with previous reports.Inter-sea zone comparison was carried out for individual chemicals by comparing the concentration variation in all five sea zones.A higher variation in concentration was generally found in the northern sea zones,namely the Bering Sea,Chukchi Sea and Arctic Ocean,for most target compounds.The sum concentration of the 17 target chemicals displayed a general trend of increasing northward from the Japan Sea to the Okhotsk Sea to the Bering Sea to the Arctic Ocean.Different latitudinal trends were found forγ-HCH andγ-HCH,and the reason of this difference was discussed by considering their divergence of thermodynamic properties,which could contribute to a slightly different fractionation effect during their northward transport driven by atmospheric long range transport.The source of the HCHs was identified by analyzing theγ-HCH/γ-HCH ratio,which was less than 4 without exception,indicating a component characteristic featuring a mixture of technical HCHs and lindane.In addition,the vertical distribution ofγ-HCH,γ-HCH and their ratio at station B80 was discussed.Different patterns were found in the upper 300 m while in layers from 300 m downward to 3500 m the patterns were fairly comparable.The vertical profiles were used to correlate the water mass distribution,based on which four layers were tentatively classified at station B80. | CAI MingGang 1,2,QIU CanRong 1,SHEN Yuan 1,CAI MingHong 3,HUANG ShuiYing 1,QIAN BiHua 1,SUN JiongHui 1 & LIU XiaoYan 1 1Department of Oceanography,College of Oceanography and Environmental Science,Xiamen University,Xiamen 361005,China 2State Key Laboratory of Marine Environmental Science,Xiamen University,Xiamen 361005,China 3State Oceanic Administration Key Laboratory for Polar Science,Polar Research Institute of China,Shanghai 200136,China | 2010 | Science China Chemistry2010,53,5: | 3 |

| 13 | Characterization of the summer pack ice biotic community of Canada Basin显示文摘Summer pack ice biotic community of the Canada Basin was characterized during the Second Chinese National Arctic Research Expedition (CHINARE-2003, 20 August-5 September 2003). Bacteria, ice algae (diatoms and autotrophic flagellates) and protozoa (mainly heterotrophic flagellates) were observed throughout the whole ice column. The vertical distribution of biotic taxa varied among sites. The integrated biomass ranged from 48.4 and 58.1 mg/m2, with an average of 55.2 mg/m2. Bacteria were the dominant of the assemblage in pack ice, accounted for 84.1% of the integrated, and ice algae, which usually dominate the ice biotic community, constituted only 3.5% of the total. Considering the quick environmental changes of the Arctic Ocean in recent years, we suggested that quick melting of pack ice in summer was suggested, which caused such change of pack ice biotic community. The low salinity throughout the whole ice column and the continuous melting of the pack ice cumbered the formation of ice algae bloom in summer, finally resulting in the dominance of microbial food web with bacteria and heterotrophic flagellates as the most obvious characteristics. | 15 | Denaturing gradient gel electrophoresis fingerprinting of soil bacteria in the vicinity of the Chinese Great Wall Station, King George Island, Antarctica显示文摘Bacterial diversity was investigated in soil samples collected from 13 sites around the Great Wall Station, Fildes Peninsula, King George Island, Antarctica, using denaturing gradient gel electrophoresis (DGGE) of 16S rRNA genes. The classes α-, β-, and γ-Proteobacteria, as well as the phylum Actinobacteria, were found to be the dominant bacteria in the soils around the Great Wall Station. Although the selected samples were not contaminated by oil, a relationship between soil parameters, microbial biodiversity, and human impact was still seen. Sample sites in human impacted areas showed lower bacterial biodiversity (average H′= 2.65) when compared to nonimpacted sites (average H′= 3.05). There was no statistically significant correlation between soil bacterial diversity and total organic carbon (TOC), total nitrogen, or total phosphorus contents of the soil. Canonical correlation analysis showed that TOC content was the most important factor determining bacterial community profiles among the measured soil parameters.
