2Genome Analysis of Dasineura jujubifolia Toursvirus 2,A Novel Ascovirus显示文摘So far,ascoviruses have only been identified from Lepidoptera host insects and their transmission vectors-endoparasitic wasps.Here,we reported the first finding of a complete novel ascovirus genome from a Diptera insect,Dasineura jujubifolia.Initially,sequence fragments with homology to ascoviruses were incidentally identified during metagenomic sequencing of the mitochondria of D.jujubifolia(Cecidomyiidae,Diptera)which is a major pest on Ziziphus jujuba.Then a full circular viral genome was assembled from the metagenomic data,which has an A+T percentage of 74%and contains 142,600 bp with 141 open reading frames(ORFs).Among the 141 ORFs,37 were conserved in all sequenced ascoviruses(core genes)including proteins predicted to participate in DNA replication,gene transcription,protein modification,virus assembly,lipid metabolism and apoptosis.Multi-gene families including those encode for baculovirus repeated open reading frames(BROs),myristylated membrane proteins,RING/U-box E3 ubiquitin ligases,and ATP-binding cassette(ABC)transporters were found in the virus genome.Phylogenetic analysis showed that the newly identified virus belongs to genus Toursvirus of Ascoviridae,and is therefore named as Dasineura jujubifolia toursvirus 2(DjTV-2a).The virus becomes the second reported species of the genus after Diadromus pulchellus toursvirus 1(DpTV-1a).The genome arrangement of DjTV-2a is quite different from that of DpTV-1a,suggesting these two viruses separated in an early time of evolution.The results suggest that the ascoviruses may infect a much broader range of hosts than our previous knowledge,and shed lights on the evolution of ascoviruses and particularly on that of the toursviruses.Jun Wang Minglu Yang Haibing Xiao Guo-Hua Huang Fei Deng Zhihong Hu 2020Virologica Sinica2020,35,2:4
3Near-zero-dispersion soliton and broadband modulational instability Kerr microcombs in anomalous dispersion显示文摘The developing advances of microresonator-based Ker cavity solitons have enabled versatile applications ranging from communication,signal processing to high-precision measurements.Resonator dispersion is the key factor determining the Kerr comb dynamics.Near the zero group-velocity-dispersion(GVD)regime,low-noise and broadband microcomb sources are achievable,which is crucial to the application of the Kerr soliton.When the GVD is almost vanished,higher-order dispersion can significantly affect the Kerr comb dynamics.Although many studies have investigated the Kerr comb dynamics near the zero-dispersion regime in microresonator or fiber ring system,limited by dispersion profles and dispersion perturbations,the near-zero-dispersion soliton structure pumped in the anomalous dispersion side is still elusive so far.Here,we theoretically and experimentally investigate the microcomb dynamics in fiber-based Fabry-Perot microresonator with ultra-small anomalous GVD.We obtain 2/3-octave-spaning microcombs with~10 GHz spacing,>84 THz span,and>8400 comb lines in the modulational instability(MI)state,without any external nonlinear spectral broadening.Such widely-spanned Ml combs are also able to enter the soliton state.Moreover,we report the first observation of anomalous-dispersion based near-zero-dispersion solitons,which exhibits a local repetition rate up to 8.6 THz,an individual pulse duration<100 fs,a span>32 THz and>3200 comb lines.These two distinct comb states have their own advantages.The broadband MI combs possess high conversion efficiency and wide existing range,while the near-zero-dispersion soliton exhibits relatively low phase noise and ultra-high local repetition rate.This work complements the dynamics of Kerr cavity soliton near the zero-dispersion regime,and may stimulate cross-disciplinary inspirations ranging from dispersion-controlled microresonators to broadband coherent comb devices.Zeyu Xiao Tieying Li Minglu Cai Hongyi Zhang Yi Huang Chao Li Baicheng Yao Kan Wu Jianping Chen 2023Light(Science & Applications)2023,12,2:2

5Efficient control of chronic LCMV infection by a CD4 T cell epitope-based heterologous prime-boost vaccination in a murine model显示文摘CD4^(+)T cells are essential for sustaining CD8^(+)T cell responses during a chronic infection.The adoptive transfer of virus-specific CD4^(+)T cells has been shown to efficiently rescue exhausted CD8^(+)T cells.However,the question of whether endogenous virus-specific CD4^(+)T cell responses can be enhanced by certain vaccination strategies and subsequently reinvigorate exhausted CD8^(+)T cells remains unexplored.In this study,we developed a CD4^(+)T cell epitope-based heterologous prime-boost immunization strategy and examined the efficacy of this strategy using a mouse model of chronic lymphocytic choriomeningitis virus(LCMV)infection.We primed chronically LCMV-infected mice with a Listeria monocytogenes vector that expressed the LCMV glycoprotein-specific I-Ab-restricted CD4^(+)T cell epitope GP61–80(LM-GP61)and subsequently boosted the primed mice with an influenza virus A(PR8 strain)vector that expressed the same CD4^(+)T cell epitope(IAV-GP61).This heterologous prime-boost vaccination strategy elicited strong anti-viral CD4^(+)T cell responses,which further improved both the quantity and quality of the virusspecific CD8^(+)T cells and led to better control of the viral loads.The combination of this strategy and the blockade of the programmed cell death-1(PD-1)inhibitory pathway further enhanced the anti-viral CD8^(+)T cell responses and viral clearance.Thus,a heterologous prime-boost immunization that selectively induces virus-specific CD4^(+)T cell responses in conjunction with blockade of the inhibitory pathway may represent a promising therapeutic approach to treating patients with chronic viral infections.Ran He Xinxin Yang Cheng Liu Xiangyu Chen Lin Wang Minglu Xiao Jianqiang Ye Yuzhang Wu Lilin Ye 2018Cellular & Molecular Immunology2018,15,9:1
6Single-cell RNA Sequencing Reveals Sexually Dimorphic Transcriptome and Type 2 Diabetes Genes in Mouse Islet β Cells显示文摘Type 2 diabetes(T2D)is characterized by the malfunction of pancreaticβcells.Susceptibility and pathogenesis of T2D can be affected by multiple factors,including sex differences.However,the mechanisms underlying sex differences in T2D susceptibility and pathogenesis remain unclear.Using single-cell RNA sequencing(scRNA-seq),we demonstrate the presence of sexually dimorphic transcriptomes in mouseβcells.Using a high-fat diet-induced T2D mouse model,we identified sex-dependent T2D altered genes,suggesting sex-based differences in the pathological mechanisms of T2D.Furthermore,based on islet transplantation experiments,we found that compared to mice with sexmatched islet transplants,sex-mismatched islet transplants in healthy mice showed down-regulation of genes involved in the longevity regulating pathway ofβcells.Moreover,the diabetic mice with sex-mismatched islet transplants showed impaired glucose tolerance.These data suggest sexual dimorphism in T2D pathogenicity,indicating that sex should be considered when treating T2D.We hope that our findings could provide new insights for the development of precision medicine in T2D.Gang Liu Yana Li Tengjiao Zhang Mushan Li Sheng Li Qing He Shuxin Liu Minglu Xu Tinghui Xiao Zhen Shao Weiyang Shi Weida Li 2021Genomics, Proteomics & Bioinformatics2021,19,3:1
7Numerical Analysis of the Tunnel-Train-Air Interaction Problem in a Tunnel with a Double-Hat Oblique Hood显示文摘The tunnel-train-air interaction problem is investigated by using a numerical method able to provide relevant information about pressure fluctuations,aerodynamic drag characteristics and the“piston wind”effect.The method relies on a RNG k-εtwo-equation turbulence model.It is shown that although reducing the oblique slope can alleviate the pressure gradient resulting from initial compression waves at the tunnel entrance,the pressure fluctuations in the tunnel are barely affected;however,a large reduction of micro-pressure wave amplitudes is found outside the tunnel.In comparison to the case where no tunnel hood is present,the amplitudes of micro-pressure waves at 40 m from the tunnel reach an acceptable range.The aerodynamic drag of the head and tail fluctuates greatly while that of the intermediate region undergoes only limited variations when the high-speed train passes through the double-hat oblique tunnel.It is shown that the effects of the oblique slope of the portal on the aerodynamic drag can almost be ignored while the train speed plays an important role.Zongfa Zhang Minglu Zhang Xinbiao Xiao 2023Fluid Dynamics & Materials Processing2023,19,2:0
