2Reverse vaccinology assisted designing of multiepitope-based subunit vaccine against SARS-CoV-2显示文摘Background:Coronavirus disease 2019(COVID-19)linked with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)cause severe illness and life-threatening pneumonia in humans.The current COVID-19 pandemic demands an effective vaccine to acquire protection against the infection.Therefore,the present study was aimed to design a multiepitope-based subunit vaccine(MESV)against COVID-19.Methods:Structural proteins(Surface glycoprotein,Envelope protein,and Membrane glycoprotein)of SARS-CoV-2 are responsible for its prime functions.Sequences of proteins were downloaded from GenBank and several immunoinformatics coupled with computational approaches were employed to forecast B-and T-cell epitopes from the SARS-CoV-2 highly antigenic structural proteins to design an effective MESV.Results:Predicted epitopes suggested high antigenicity,conserveness,substantial interactions with the human leukocyte antigen(HLA)binding alleles,and collective global population coverage of 88.40%.Taken together,276 amino acids long MESV was designed by connecting 3 cytotoxic T lymphocytes(CTL),6 helper T lymphocyte(HTL)and 4 B-cell epitopes with suitable adjuvant and linkers.The MESV construct was non-allergenic,stable,and highly antigenic.Molecular docking showed a stable and high binding affinity of MESV with human pathogenic toll-like receptors-3(TLR3).Furthermore,in silico immune simulation revealed significant immunogenic response of MESV.Finally,MEV codons were optimized for its in silico cloning into the Escherichia coli K-12 system,to ensure its increased expression.Conclusion:The MESV developed in this study is capable of generating immune response against COVID-19.Therefore,if designed MESV further investigated experimentally,it would be an effective vaccine candidate against SARS-CoV-2 to control and prevent COVID-19.Muhammad Tahir ul Qamar Farah Shahid Sadia Aslam Usman Ali Ashfaq Sidra Aslam Israr Fatima Muhammad Mazhar Fareed Ali Zohaib Ling-Ling Chen 2020Infectious Diseases of Poverty2020,9,5:2

4COVID-19 and comorbidities of hepatic diseases in a global perspective显示文摘The worldwide outbreak of coronavirus disease 2019(COVID-19) has challenged the priorities of healthcare system in terms of different clinical management and infection transmission, particularly those related to hepatic-disease comorbidities. Epidemiological data evidenced that COVID-19 patients with altered liver function because of hepatitis infection and cholestasis have an adverse prognosis and experience worse health outcomes. COVID-19-associated liver injury is correlated with various liver diseases following a severe acute respiratory syndrome-coronavirus type 2(SARS-CoV-2) infection that can progress during the treatment of COVID-19 patients with or without pre-existing liver disease. SARS-CoV-2 can induce liver injury in a number of ways including direct cytopathic effect of the virus on cholangiocytes/hepatocytes, immune-mediated damage, hypoxia, and sepsis. Indeed, immediate cytopathogenic effects of SARSCoV-2 via its potential target, the angiotensin-converting enzyme-2 receptor, which is highly expressed in hepatocytes and cholangiocytes, renders the liver as an extra-respiratory organ with increased susceptibility to pathological outcomes. But, underlying COVID-19-linked liver disease pathogenesis with abnormal liver function tests(LFTs) is incompletely understood. Hence, we collated COVID-19-associated liver injuries with increased LFTs at the nexus of pre-existing liver diseases and COVID-19, and defining a plausible pathophysiological triad of COVID-19, hepatocellular damage, and liver disease. 9Estimation of Genetic Divergence and Character Association Studies in Local and Exotic Diversity Panels of Soybean(Glycine max L.)Genotypes显示文摘The availability of favorable genetic diversity is a thriving vitality for the success of a breeding program.It provides a firm basis of selecting superior breeding lines for the development of high yielding crop genotypes.In this context,present investigation aimed to generate information on genetic divergence and character association in a diversity panel of 123 local and exotic soybean genotypes.Analysis of variance revealed significant response of the evaluated genotypes based on studied attributes.It depicted the probability of selecting desirable soybean genotypes by focusing on character association studies and genetic diversity analysis.Correlation analysis revealed that seed yield per plant showed high positive correlation with 100-seed weight followed by pods per plant and plant height.Furthermore,path coefficient analysis exposed that pods per plant had maximum direct contribution in seed yield per plant followed by 100-seed weight,days to flowering and SPAD measurement.Genotype named“G-10”showed maximum yield per plant followed by 24607,G-52,24593,Arisoy,24566,17426,A-3127,24570 and 24567.Genetic diversity analysis grouped the evaluated germplasm into 17 clusters.All clusters showed zero intra-cluster variability;while inter-cluster divergence ranged from 9.00 to 91.11.Cluster V showed maximum inter-cluster distance with cluster XII followed by that of between V and VIII.Moreover,cluster IV with superior genotypes(G-10,24607,24593 and 24566),VI(17426 and 24567),XIII(24570)and X(Arisoy and G-52)showed above mean values for most of the studied characters.Overall,the results of hybridization between the superior genotypes of these cluster pairs might be useful for soybean breeding with improving agronomic traits and adaptability.Syed Ali Zafar Muhammad Aslam Haroon Zaman Khan Sehrish Sarwar Rao Saad Rehman Mariam Hassan Ramala Masood Ahmad Rafaqat AGill Basharat Ali Ibrahim Al-Ashkar Abdullah Ibrahim Md Atikur Rahman Ayman El Sabagh 2023Phyton-International Journal of Experimental Botany2023,92,6:0
10Treatment patterns of primary care physicians vs specialists prior to subspecialty urogynaecology referral for women suffering from pelvic floor disorders显示文摘BACKGROUND There are approximately 25% of women in the United States suffering from pelvic floor disorders (PFDs) and this number is predicted to rise. The potential complications and increasing healthcare costs that exist with an operation indicate the importance of conservative treatment options prior to attempting surgery. Considering the prevalence of PFDs, it is important for primary care physician and specialists (obstetricians and gynecologists) to be familiar with the initial work-up and the available conservative treatment options prior to subspecialist (urogynecologist) referral. AIM To assess the types of treatments that specialists attempted prior to subspecialty referral and determine the differences in referral patterns. METHODS This is a retrospective cohort study of 234 patients from a community teaching hospital referred to a single female pelvic medicine and reconstructive surgery (FPMRS) provider for PFD. Specialist vs primary care provider (PCP) referrals were compared. Number, length and treatment types were studied using descriptive statistics. RESULTS There were 184 referrals (78.6%) by specialists and 50 (21.4%) by PCP. Treatment (with Kegel exercises, pessary placements, and anticholinergic medications) was attempted on 51%(n = 26) of the PCP compared to 48%(n = 88) of the specialist referrals prior to FPMRS referral (P = 0.6). There was no significant difference in length of treatment prior to referral for PCPs vs specialists (14 mo vs 16 mo, respectively, P = 0.88). However, there was a significant difference in the patient’s average time with the condition prior to referral (35 mo vs 58 mo for PCP compared to specialist referrals)(P = 0.02). CONCLUSION One half of the patients referred to FPMRS clinic received treatment prior to referral. 13Role of Organic Amendments to Mitigate Cd Toxicity and Its Assimilation in Triticum aestivum L.显示文摘In soil biota,higher and enduring concentration of heavy metals like cadmium(Cd)is hazardous and associated with great loss in growth,yield,and quality parameters of most of the crop plants.Recently,in-situ applications of eco-friendly stabilizing agents in the form of organic modifications have been utilized to mitigate the adverse effects of Cd-toxicity.This controlled experiment was laid down to appraise the imprints of various applied organic amendments namely poultry manure(PM),farmyard manure(FYM),and sugarcane press mud(PS)to immobilize Cd in polluted soil.Moreover,phytoavailability of Cd in wheat was also accessed under an alkaline environment.Results revealed that the addition of FYM(5–10 ton ha^(-1))in Cd-contaminated soil significantly increased germination rate,leaf chlorophyll content,plant height,spike length,biological and grain yield amongst all applied organic amendments.Moreover,the addition of FYM(5–10 ton ha^(-1))also reduced the phytoavailability of Cd by 73–85%in the roots,57–83%in the shoots,and 81–90%in grains of wheat crop.Thus,it is affirmed that incorporation of FYM(5–10 ton ha^(-1))performed better to enhance wheat growth and yield by remediating Cd.Thus,the application of FYM(5–10 ton ha^(-1))reduced the toxicity induced by Cd to plants by declining its uptake and translocation as compared to all other applied organic amendments to immobilize Cd under sandy alkaline polluted soil.Tauqeer Ahmad Yasir Sobia Aslam Muhammad Shahid Rizwan Allah Wasaya Muhammad Ateeq Muhammad Naeem Khan Sikander Khan Tanveer Walid Soufan Basharat Ali Allah Ditta Arpna Kumari Ayman EL Sabagh 2022Phyton-International Journal of Experimental Botany2022,91,11:0
14Graphene Nanosheets Decorated with Copper Oxide Nanoparticles for the Photodegradation of Methylene Blue显示文摘Textile industries extensively use colorants,such as methylene blue,and if disposed off untreated,they contaminate the effluent streams,causing a severe impact on the environment and aquatic life.Photocatalytic degradation has been found as an inevitable approach to treat them.Herein,we decorated the copper oxide nanoparticles on graphene nanosheets during the reflux process.The resultant copper oxide/graphene nanocomposites were analyzed for structural and functional attributes.It was observed that on increasing the copper oxide contents,the z-average size of the resultant nanocomposites decreased.The X-ray diffraction analysis demonstrated the crystalline nature of the nanocomposite.The surface morphology of the copper oxide nanoparticles appeared to be spherical and that of the copper oxide/graphene composite somehow wrinkled.The infrared analysis indicated successful intercalation of precursors in the nanocomposite.The bandgap of copper oxide/graphene nanocomposites varied in the range of 1.03—1.30 eV,which indicated their effective photocatalytic activity.The results demonstrated that after 120 min of exposure,the methylene blue removal efficiency reached 94.0%,92.2%,and 89.4%(mass fraction)on the copper oxide/graphene nanocomposite at copper oxide nanoparticles to graphene nanosheets ratios of 1:1,1.5:1,and 2:1(mass ratio),respectively.The photodegradation performance of the prepared nano-catalyst was found satisfactory even after five cycles.Samavia RAFIQ Zulfiqar Ali RAZA Muhammad ASLAM Muhammad Junaid BAKHTIYAR 2022Chemical Research in Chinese Universities2022,38,6:0

16Analysis and Forecasting COVID-19 Outbreak in Pakistan Using Decomposition and Ensemble Model显示文摘COVID-19 has caused severe health complications and produced a substantial adverse economic impact around the world.Forecasting the trend of COVID-19 infections could help in executing policies to effectively reduce the number of new cases.In this study,we apply the decomposition and ensemble model to forecast COVID-19 confirmed cases,deaths,and recoveries in Pakistan for the upcoming month until the end of July.For the decomposition of data,the Ensemble Empirical Mode Decomposition(EEMD)technique is applied.EEMD decomposes the data into small components,called Intrinsic Mode Functions(IMFs).For individual IMFs modelling,we use the Autoregressive Integrated Moving Average(ARIMA)model.The data used in this study is obtained from the official website of Pakistan that is publicly available and designated for COVID-19 outbreak with daily updates.Our analyses reveal that the number of recoveries,new cases,and deaths are increasing in Pakistan exponentially.Based on the selected EEMD-ARIMA model,the new confirmed cases are expected to rise from 213,470 to 311,454 by 31 July 2020,which is an increase of almost 1.46 times with a 95%prediction interval of 246,529 to 376,379.The 95%prediction interval for recovery is 162,414 to 224,579,with an increase of almost two times in total from 100802 to 193495 by 31 July 2020.On the other hand,the deaths are expected to increase from 4395 to 6751,which is almost 1.54 times,with a 95%prediction interval of 5617 to 7885.Thus,the COVID-19 forecasting results of Pakistan are alarming for the next month until 31 July 2020.They also confirm that the EEMD-ARIMA model is useful for the short-term forecasting of COVID-19,and that it is capable of keeping track of the real COVID-19 data in nearly all scenarios.The decomposition and ensemble strategy can be useful to help decision-makers in developing short-term strategies about the current number of disease occurrences until an appropriate vaccine is developed.Xiaoli Qiang Muhammad Aamir Muhammad Naeem Shaukat Ali Adnan Aslam Zehui Shao 2021Computers, Materials & Continua2021,,7:0
