Construction of sperm-specific lactate dehydrogenase DNA vaccine and experimental study of its immuno-contraceptive effect on mice显示文摘Lactate dehydrogenase C4 (LDHC4) is a key enzyme for sperm metabolism. It is distributed specifically in testis and is highly immunogenic. In this study, two DNA vaccines pVAX1-hLDHC and pVAX1-mLDHC were constructed by inserting coding sequences of human and mice LDHC4 into the eukaryotic ex-pression vector pVAX1. The production of LDHC4 specific antibodies was induced in the sera of vac-cinated mice and the reproductive tract secretions of vaccinated female mice through immunization by mucosal surface instillation. Furthermore, the antibody titer increased with the times of immunization. In the mating experiment, the number of newborns of the vaccinated mice reduced significantly and some immunized female mice even lost the ability to bear any offsprings, suggesting that the difference between the immunized and control mice was statistically significant. Sperm agglutination analysis indicated that both the antisera from immunized mice and the reproductive tract secretions of vacci-nated female mice could agglutinate normal sperms. Results of immunohistochemistry showed that the antibodies present in the sera of immunized mice and the reproductive tract secretions of vaccinated female mice could specifically react with LDHC4 antigen, which mainly locates in the cytoplasm, acrosome membrane externa and acrosome capsule of the sperm. Taken together, our results indicated that the constructed contraceptive DNA vaccines did yield immunocontraceptive effects on mice and this would enable clinical trials in near future.CHEN Yong*, ZHANG Duo*, XIN Na, XIONG YongZhong, CHEN Ping, LI Bo, TU XiangDong & LAN FengHua Research Laboratory of Medical Genetics, PLA Institute of Technology for Birth Control, Fetal Health and Child Growth, Fuzhou General Hospital of Nanjing Military Region, Fuzhou 350025, China 2008Science China(Life Sciences)2008,51,4:3
Small molecule induces mitochondrial fusion for neuroprotection via targeting CK2 without affecting its conventional kinase activity显示文摘Mitochondrial fusion/hission dynamics plays a fundamental role in neuroprotection;however, there is till a severe lack oftherapeutic targets for this biological process. Here,we found that the naturally derived small molecule echinacoside(ECH)signifhcantly promotes mitochondrial fusion progression.ECH selectively binds to the previously uncharacterized casein kinase 2(CK2)α'subunit(CK2a)as a direct cellular target, and genetic knockdown of CK2α'abolishes ECH-mediated mitochondrial fusion.Mechanistically,ECH allosterically regulates CK2α'conformation to recruit basic transcription factor 3(BTF 3)to form a binary proteincomplex. Then, the CK2α'/BTF3 complex facilitatesβ-catenin nuclear translocation to activate TCF/AEF transcription factors andstimulate transcription of the mitochondrial fusion gene Mfn2.trikingly,in a mouse middle cerebral artery occlusion(MCAO)model,ECH administration was found to significantly improve cerebral injuries and behavioral deficits by enhancing Mfn2expression in wild-type but not CK2α'^(+/-)mice.Taken together, our findings reveal, for the first time, that CK2 is essential forpromoting mitochondrial fusion in a Wnt/B-catenin-dependent manner and suggest that pharmacologically targeting CK2 is apromising therapeutic strategy for ischemic stroke.Ke-Wu Zeng Jing-Kang Wang Li-Chao Wang Qiang Guo Ting-Ting Liu Fu-Jiang Wang Na Feng Xiao-Wen Zhang Li-Xi Liao Mei-Mei Zhao Dan Liu Yong Jiang Pengfei Tu 2021Signal Transduction and Targeted Therapy2021,6,3:3

Molecular basis of vasohibins-mediated detyrosination and its impact on spindle function and mitosis显示文摘α-Tubulin detyrosination, largely catalyzed by vasohibins, is involved in many microtubule (MT)-related cellular events. In this study, we identified a core heterodimeric complex of human small vasohibin-binding protein (SVBP) and vasohibin 1 (VASH1)(hereafter denoted as SVBP-VASH1) that catalyzes the detyrosination of a peptide derived from C-terminus of α-tubulin. We further solved the crystal structures of the SVBP-VASH1 heterodimer alone and in complex with either an inhibitor or a mutant substrate peptide. Our structural research, complemented by biochemical and mutagenesis experiments, resulted in identification of the key residues for VASH1 binding to SVBP and α-tubulin substrate. Our in vivo experiments reveal that MT detyrosination in general, as well as the interactions between SVBP, VASH1, and α-tubulin, are critical for spindle function and accurate chromosome segregation during mitosis. Furthermore, we found that the phenotypes caused by the depletion of vasohibins were largely rescued upon co-depletion of kinesin13/MCAK, suggesting the coordination between the MT depolymerase and MT detyrosination during mitosis. Thus our work not only provides structural insights into the molecular mechanism of α-tubulin detyrosination catalyzed by SVBP-bound vasohibins, but also uncovers the key role of vasohibins-mediated MT detyrosination in spindle morphology and chromosome segregation during mitosis.Shanhui Liao Girish Rajendraprasad Na Wang Susana Eibes Jun Gao Huijuan Yu Gao Wu Xiaoming Tu Hongda Huang Marin Barisic Chao Xu 2019Cell Research2019,29,7:2

Al homogeneous deposition induced by N-containing functional groups for enhanced cycling stability of Al-ion battery negative electrode显示文摘Rechargeable Al-ion batteries(AIBs)are considered as one of the most fascinating energy storage systems due to abundant Al resource and low cost. However, the cycling stability is subjected to critical problems for using Al foil as negative electrode, including Al dendrites, corrosion and pulverization. For addressing these problems, here a lightweight self-supporting N-doped carbon rod array(NCRA)is demonstrated for a long-life negative electrode in AIBs.Experimental analysis and first-principle calculations reveal the storage mechanism involving the induced deposition of N-containing function groups to Al as well as the ideal skeleton of the NCRA matrix for Al plating/stripping,which is favorable for regulating Al nucleation and suppressing dendrites growth.Compared with the Al foil, the NCRA exhibits lower areal mass density(∼72%of Al foil),smaller thickness(40%of Al foil),but much longer cycle life(>4 times of Al foil).Benefiting from the remarkable stability of the array structure, symmetric cells show excellent cycling stability with small voltage hysteresis(∼80 mV)and meanwhile there are no corrosion and pulverization problems even after cycled for 120 hours.Besides, full cells also manifest long lifespan(1,500 cycles)and increased Coulombic efficiency(100±1%).Handong Jiao Shuqiang Jiao Wei-Li Song Xiang Xiao Dongmei She Na Li Haosen Chen Jiguo Tu Mingyong Wang Daining Fang 2021Nano Research2021,14,3:1

Effects of Toluene on the Development of the Inner Ear and Lateral Line Sensory System of Zebrafish显示文摘Objective The aim of this study was to explore the ototoxicity of toluene in the early development of zebrafish embryos/larvae.Methods Zebrafish were utilized to explore the ototoxicity of toluene. Locomotion analysis, immunofluorescence, and q PCR were used to understand the phenotypes and molecular mechanisms of toluene ototoxicity. Results The results demonstrated that at 2 mmol/L, toluene induced zebrafish larvae death at 120 hours post fertilization(hpf) at a rate of 25.79% and inhibited the rate of hatching at 72 hpf.Furthermore, toluene exposure inhibited the distance travelled and average swimming velocity of zebrafish larvae while increasing the frequency of movements. As shown by fluorescence staining of hair cells, toluene inhibited the formation of lateral line neuromasts and middle line 1(Ml1) neuromasts in 3 days post fertilization larvae in a concentration-dependent manner. Toluene altered the expression level of genes involved in ear development/function in zebrafish, among which the m RNA levels of cd164 l2,tekt3, and pcsk5 a were upregulated, while the level of otofb was downregulated, according to the q PCR results. Conclusion This study indicated that toluene may affect the development of both the inner ear and lateral line systems in zebrafish, while the lateral line system may be more sensitive to toluene than the inner ear.LI Xu Dong TU Hong Wei HU Ke Qi LIU Yun Gang MAO Li Na WANG Feng Yan QU Hong Ying CHEN Qing 2021Biomedical and Environmental Sciences2021,34,2:1
The role of crm-1 in ionizing radiation-induced nervous system dysfunction in Caenorhabditis elegans显示文摘Ionizing radiation can cause changes in nervous system function. However, the underlying mechanism remains unclear. In this study,Coenorhabditis elegans(C.elegans)was irradiated with 75 Gy of ^(60)Co whole-body γ radiation.Behavioral indicators(head thrashes, touch avoidance, and foraging),and the development of dopaminergic neurons related to behavioral function, were evaluated to assess the effects of ionizing radiation on nervous system function in C.elegans.Various behaviors were impaired after whole-body irradiation and degeneration of dopamine neurons was observed. This suggests that 75 Gy of γ radiation is sufficient to induce nervous system dysfunction. The genes nhr-76 and crm-1,which are reported to be related to nervous system function in human and mouse, were screened by transcriptome sequencing and bioinformatics analysis after irradiation or sham irradiation. The expression levels of these two genes were increased after radiation.Next,RNAi technology was used to inhibit the expression of crm-1,a gene whose homologs are associated with motor neuron development in other species.Downregulation of crm-1 expression effectively alleviated the deleterious effects of ionizing radiation on head thrashes and touch avoidance. It was also found that the expression level of crm-1 was regulated by the nuclear receptor gene nhr-76.The results of this study suggest that knocking down the expression level of nhr-76 can reduce the expression level of crm-1,while down-regulating the expression level of crm-1 can alleviate behavioral disorders induced by ionizing radiation. Therefore, inhibition of crm-1 may be of interest as a potential therapeutic target for ionizing radiation-induced neurological dysfunction.Hui-Qiang Long Jin Gao Shu-Qing He Jian-Fang Han Yu Tu Na Chen 2023Neural Regeneration Research2023,18,6:1

Responses of plant diversity and soil microorganism diversity to nitrogen addition in the desert steppe,China显示文摘Nitrogen(N)deposition is a significant aspect of global change and poses a threat to terrestrial biodiversity. The impact of plant-soil microbe relationships to N deposition has recently attracted considerable attention.Soil microorganisms have been proven to provide nutrients for specific plant growth, especially in nutrient-poor desert steppe ecosystems. However, the effects of N deposition on plant-soil microbial community interactions in such ecosystems remain poorly understood. To investigate these effects,we conducted a 6-year N-addition field experiment in a Stipa breviflora Griseb.desert steppe in Inner Mongolia Autonomous Region,China.Four N treatment levels(N0,N30,N50,and N100,corresponding to 0,30,50,and 100 kg N/(hm2•a),respectively)were applied to simulate atmospheric N deposition. The results showed that N deposition did not significantly affect the aboveground biomass of desert steppe plants.N deposition did not significantly reduce the alfa-diversity of plant and microbial communities in the desert steppe, and low and mediate N additions(N30 and N50)had a promoting effect on them. The variation pattern of plant Shannon index was consistent with that of the soil bacterial Chao1 index.N deposition significantly affected the beta-diversity of plants and soil bacteria, but did not significantly affect fungal communities. In conclusion,N deposition led to co-evolution between desert steppe plants and soil bacterial communities, while fungal communities exhibited strong stability and did not undergo significant changes. These findings help clarify atmospheric N deposition effects on the ecological health and function of the desert steppe.YE He HONG Mei XU Xuehui LIANG Zhiwei JIANG Na TU Nare WU Zhendan 2024Journal of Arid Land2024,16,3:0

Rationally engineered Co and N co-doped WS_(2) as bifunctional catalysts for pH-universal hydrogen evolution and oxidative dehydrogenation reactions显示文摘In the field of electrolysis of water, the design and synthesis of catalysts over a wide pH range have attracted extensive attentions. In this paper,Co and N are co-introduced into the structural unit of tungsten disulfide(WS_(2)),and the hydrogen evolution reaction(HER)performances of different WS_(2)-based catalysts are theoretically predicted and systematically studied by density functional theory(DFT)calculations. With the guidance of DFT calculations,an evaporation-pyrolysis strategy is applied to prepare Co and N co-doped WS_(2)(Co,N-WS_(2))flower-like nanosheets, which exhibits excellent HER performance over a wide pH range. In addition, the DFT calculations show that the active sites in Co,N-WS_(2) have a good ability of hydrogen adsorption after the introduction of Co and N,suggesting that such a co-doping system will be an ideal catalyst for oxidative dehydrogenation(ODH).The following experiment results indeed evidence that the Co,N-WS_(2) catalyst displays a high activity in the ODH of 1,2,3,4-tetrahydroquinoline(4H-quinoline)and its derivatives. Therefore, this work provides a good example for the rational design and accurate preparation of functional catalysts, which enables it possible to develop other efficient catalysts with multiple functions.Min Ling Na Li Binbin Jiang Renyong Tu Tao Wu Pingli Guan Yin Ye Weng-Chon(Max)Cheong Kaian Sun Shoujie Liu Konglin Wu Aijian Huang Xianwen Wei 2022Nano Research2022,15,3:0
