| 1 | Natural history of covert hepatic encephalopathy: An observational study of 366 cirrhotic patients显示文摘AIM To explore the natural history of covert hepatic encephalopathy(CHE) in absence of medication intervention.METHODS Consecutive outpatient cirrhotic patients in a Chinese tertiary care hospital were enrolled and evaluated for CHE diagnosis. They were followed up for a mean of 11.2 ± 1.3 mo. Time to the first cirrhosis-related complications requiring hospitalization, including overt HE(OHE), resolution of CHE and death/transplantation, were compared between CHE and no-CHE patients. Predictors for complication(s) and death/transplantation were also analyzed.RESULTS A total of 366 patients(age: 47.2 ± 8.6 years, male: 73.0%) were enrolled. CHE was identified in 131 patients(35.8%). CHE patients had higher rates of death and incidence of complications requiring hospitalization, including OHE, compared to unimpaired patients. Moreover, 17.6% of CHE patients developed OHE, 42.0% suffered persistent CHE, and 19.8% of CHE spontaneously resolved. In CHE patients, serum albumin < 30 g/L(HR = 5.22, P = 0.03) was the sole predictor for developing OHE, and blood creatinine > 133 μmol/L(HR = 4.75, P = 0.036) predicted mortality. Child-Pugh B/C(HR = 0.084, P < 0.001) and OHE history(HR = 0.15, P = 0.014) were predictors of spontaneous resolution of CHE.CONCLUSION CHE exacerbates, persists or resolves without medication intervention in clinically stable cirrhosis. | 2 | Brucine Inhibits Bone Metastasis of Breast Cancer Cells by Suppressing Jagged1/Notch1 Signaling Pathways显示文摘Objective: To examine the effects of brucine on the invasion, migration and bone resorption of receptor activator of nuclear factor-kappa B ligand(RANKL)-induced osteoclastogenesis. Methods: The osteoclastogenesis model was builded by co-culturing human breast tumor MDA-MB-231 and mouse RAW264.7 macrophages cells. RANKL(50 ng/m L) and macrophage-colony stimulating factor(50 ng/m L) were added to this system, followed by treatment with brucine(0.02, 0.04 and 0.08 mmol/L), or 10 μmol/L zoledronic acid as positive control. The migration and bone resorption were measured by transwell assay and in vitro bone resorption assay. The protein expressions of Jagged1 and Notch1 were investigated by Western blot. The expressions of transforming growth factor-β1(TGF-β1), nuclear factor-kappa B(NF-κB) and Hes1 were determined by enzyme-linked immunosorbent assay. Results: Compared with the model group, brucine led to a dose-dependent decrease on migration of MDA-MB-231 cells, inhibited RANKL-induced osteoclastogenesis and bone resorption of RAW264.7 cells(P <0.01). Furthermore, brucine decreased the protein levels of Jagged1 and Notch1 in MDA-MB-231 cells and RAW264.7 cells co-cultured system as well as the expressions of TGF-β1, NF-κB and Hes1(P<0.05 or P<0.01). | 3 | Inulin Can Alleviate Metabolism Disorders in ob/ob Mice by Partially Restoring Leptinrelated Pathways Mediated by Gut Microbiota显示文摘Inulin has been used as a prebiotic to alleviate glucose and lipid metabolism disorders in mice and humans by modulating the gut microbiota. However, the mechanism underlying the alleviation of metabolic disorders by inulin through interactions between the gut microbiota and host cells is unclear. We use ob/ob mice as a model to study the effect of inulin on the cecal microbiota by16 S rRNA gene amplicon sequencing and its interaction with host cells by transcriptomics. The inulin-supplemented diet improved glucose and lipid metabolism disorder parameters in ob/ob mice,alleviating fat accumulation and glucose intolerance. The a diversity of gut microbial community of ob/ob mice was reduced after inulin treatment, while the b diversity tended to return to the level of wild type mice. Interestingly, Prevotellaceae UCG 001(family Prevotellaceae) was obviously enriched after inulin treatment. A comparative analysis of the gene expression profile showed that the cecal transcriptome was changed in leptin gene deficiency mice, whereas the inulin-supplemented diet partially reversed the changes in leptin gene-related signaling pathways, especially AMPK signaling pathway, where the levels of gene expression became comparable to those in wild type mice.Further analysis indicated that Prevotellaceae UCG 001 was positively correlated with the AMPK signaling pathway, which was negatively correlated with markers of glycolipid metabolism disorders. | 8 | Strengths, weaknesses, opportunities and threats (SWOT) analysis of reinitiation into methamphetamine abusers: qualitative findings from an exploration of methamphetamine abusers in Shanghai, China显示文摘Background Despite high relapse rate among methamphetamine (MA) abusers, there still have been little empirical data to date detecting the risk factors related to craving and relapse from the perspective of MA abusers. Therefore, the aim of this study was to use qualitative research methods exploring the strengths, weaknesses, opportunities and threat factors that could facilitate or impede individuals' abstinence in a real-life context from the perspectives of MA abusers. Aim To use qualitative research methods exploring the strengths, weaknesses, opportunities and threat factors relating to individuals' abstinence in a real-life context. Method Semistructured interviews were conducted with 32 MA abusers recruited in Shanghai guided by open-ended questions on narrating the real-life catalysts and inhibitors related to craving, initiation, relapse and abstinence. All data were transcribed verbatim and analysed using a strengths, weaknesses, opportunities and threats (SWOT) analysis. Results The results of the SWOT analysis revealed that contextual factors including peer influence, prevalence and availability of MA, familiar venue of MA use, discrimination, sexual behaviours, alcohol, emotional states and their attitudes towards smoking MA were important factors that contribute to reinitiation and relapse. Surveillance systems, antidrug social workers, vocational skills trainings, moving to another city and family responsibility might serve as counter measures targeti叩 those mentioned weaknesses and threats above. Conclusion This SWOT analysis highlights the complex nature of relapse. | 9 | Repeatability and Reproducibility of Quantitative Assessment of the Retinal Microvasculature Using Optical Coherence Tomography Angiography Based on Optical Microangiography显示文摘Objective The aim of this study was to determine the repeatability and reproducibility of optical coherence tomography angiography(OCTA) based on optical microangiography(OMAG) measurements of macular vessels in normal eyes. Methods In this prospective cohort study, 40 eyes of 40 healthy volunteers underwent repeated OCTA(Cirrus HD-OCT 5000 angiography system, Carl Zeiss Meditec, Inc.) scans on two separate visit days. On each visit day, the eyes were scanned three times. The following parameters were used to quantitatively describe the OCTA images of the superficial vascular network: vessel area density(VAD), vessel skeleton density(VSD), vessel diameter index(VDI), vessel perimeter index(VPI), vessel complexity index(VCI), flux, and foveal avascular zone(FAZ). Coefficient of variation(CV) and intraclass correlation coefficient(ICC) were calculated for evaluating intravisit and intervisit repeatability, as well as interobserver reproducibility. Results The measurements showed high repeatability [CVs ≤ 4.2%(intravisit) and ≤ 4.6%(intervisit)] and interobserver reproducibility(ICCs ≥ 0.923) for all parameters. | 14 | An adjustment method for active reflector of large high-frequency antennas considering gain and boresight显示文摘The design of the Qitai 110 m Radio Telescope(QTT) with large aperture and very high working frequency(115 GHz) was investigated in Xinjiang, China. The results lead to a main reflector with high surface precision and high pointing precision. In this paper, the properties of active surface adjustment in a deformed parabolic reflector antenna are analyzed. To assure the performance of large reflector antennas such as gain and boresight, which can be obtained by utilizing an electromechanical coupling model, and satisfy them simultaneously, research on active surface adjustment applied to a new parabolic reflector as target surface has been done. Based on the initial position of actuators and the relationship between adjustment points and target points, a novel mathematical model and a program thatdirectly calculates the movements of actuators have been developed for guiding the active surface adjustment of large reflector antennas. This adjustment method is applied to an 8 m reflector antenna,in which we only consider gravity deformation. | 15 | Using ultrasound to improve the sequential post-synthesis modification method for making mesoporous Y zeolites显示文摘Mesoporous Y zeolites were prepared by the sequential chemical dealumination(using chelating agents such as ethylenediaminetetraacetic acid,H EDTA,and citric acid aqueous solutions)and alkaline desilication(using sodium hydroxide,NaOH,aqueous solutions)treatments.Specifically,the ultrasound-assisted alkaline treatment(i.e.,ultrasonic treatment)was proposed as the alternative to conventional alkaline treatments which are performed under hydrothermal conditions.In comparison with the hydrothermal alkaline treatment,the ultrasonic treatment showed the comparatively enhanced efficiency(with the reduced treatment time,i.e.,5 min vs.30 min,all with 0.2 mol·L NaOH at 65°C)in treating the dealuminated Y zeolites for creating mesoporosity.For example,after the treatment of a dealuminated zeolite Y(using 0.1 mol·L H EDTA at 100°C for 6 h),the ultrasonic treatment produced the mesoporous zeolite Y with the specific external surface area()of 160 m·g and mesopore volume()of 0.22 cm·g,being slightly higher than that by the conventional method(i.e.,=128 m·g and=0.19 cm·g).The acidic property and catalytic activity(in catalytic cracking of-octane)of mesoporous Y zeolites obtained by the two methods were comparable.The ultrasonic desilication treatment was found to be generic,also being effective to treat the dealuminated Y zeolites by citric acid.Additionally,the first step of chemical dealumination treatment was crucial to enable the effective creation of mesopores in the parent Y zeolite(with a silicon-to-aluminium ratio,Si/Al=2.6)regardless of the subsequent alkaline desilication treatment(i.e.,ultrasonic or hydrothermal).Therefore,appropriate selection of the condition of the chemical dealumination treatment based on the property of parent zeolites,such as Si/Al ratio and crystallinity,is important for making mesoporous zeolites effectively. | Rongxin Zhang Peinan Zhong Hamidreza Arandivan Yanan Guan Jinmin Liu Na Wang Yilai Jiao Xiaolei Fan | 2020 | Frontiers of Chemical Science and Engineering2020,14,2: | 5 |

| 19 | Genetic Characteristics of 2009 Pandemic H1N1 Influenza A Viruses Isolated from China's Mainland显示文摘A total of 100 H1N1 flu real-time-PCR positive throat swabs collected from fever patients in Zhejiang, Hubei and Guangdong between June and November 2009, were provided by local CDC laboratories. After MDCK cell culture, 57 Influenza A Pandemic (H1N1) viruses were isolated and submitted for whole genome sequencing. A total of 39 HA sequences, 52 NA sequences, 36 PB2 sequences, 31 PB1 sequences, 40 PA sequences, 48 NP sequences, 51 MP sequences and 36 NS sequences were obtained, including 20 whole genome sequences. Sequence comparison revealed they shared a high degree of homology (96%~99%) with known epidemic strains (A/California/04/2009(H1N1). Phylogenetic analysis showed that although the sequences were highly conserved, they clustered into a small number of groups with only a few distinct strains. Site analysis revealed three substitutions at loop 220 (221-228) of the HA receptor binding site in the 39 HA sequences: A/Hubei/86/2009 PKVRDQEG→PKVRDQEA, A/Zhejiang/08/2009 PKVRDQEG→PKVRDQER, A/Hubei/75/2009 PKVRDQEG→PKVRDQGG, the A/Hubei/75/2009 was isolated from an acute case, while the other two were from patients with mild symptoms. Other key sites such as 119, 274, 292 and 294 amino acids of NA protein,627 of PB2 protein were conserved. Meanwhile, all the M2 protein sequences possessed the Ser32Asn mutation, suggesting that these viruses were resistant to adamantanes.
