| 2 | Bushen Qiangjin capsule(补肾强筋胶囊) inhibits the Wnt/β-catenin pathway to ameliorate papain-induced knee osteoarthritis in rats显示文摘OBJECTIVE:To evaluate the molecular mechanism underlying the beneficial effect of Bushen Qiangjin capsule(补肾强筋胶囊,BSQJ),a Traditional Chinese Medicine,on knee osteoarthritis(KOA).METHODS:In the present study,32 female Sprague-Dawley rats were randomly divided into four groups:control,KOA,high-dose BSQJ(H-BSQJ),and low-dose BSQJ(L-BSQJ).After successfully establishing the KOA model by intra-articular injection of papain,H-BSQJ and L-BSQJ groups were intragastrically administered 0.243 and 0.122 g/kg BSQJ,respectively,daily for 6 weeks.At the end of the experiment,knee articular cartilage tissues of rats were collected for evaluation by hematoxylin and eosin staining,Safranin O-Fast Green staining,and terminal deoxynucleotidyl transferase-mediated d UTP nick-end labeling assay.Serum interleukin-1βand tumor necrosis factor-αlevels of rats were detected with an enzyme-linked immunosorbent assay method.Gene expression of Wnt-4,β-catenin,Frizzled-2,glycogen synthase kinase-3β(GSK-3β),cysteinyl aspartate-specific proteinases 3 and 9(caspases 3 and 9),collagen typeⅡalpha 1(Col2 a1),and matrix metalloproteinases 1 and 13(MMP-1 and MMP-3)of rat knee articular cartilage was quantified by reverse transcription-quantitative polymerase chain reaction analysis.Wnt-4,β-catenin,Frizzled-2,GSK-3β,cleaved caspase-3,and cleaved caspase-9 protein expression in rat knee articular cartilage was determined by western blot analysis.RESULTS:BSQJ obviously reduced pathological damage and matrix degradation of articular cartilage in KOA rats.Compared with the KOA group,H-BSQJ rats exhibited downregulated m RNA and protein expression of Wnt-4,β-catenin,Frizzled-2,and caspase-3,as well as upregulated m RNA and protein expression of GSK-3β.In addition,H-BSQJ significantly increased m RNA expression of Col2 a1 and decreased m RNA expression of MMP-1 and MMP-13.CONCLUSION:BSQJ exerted a beneficial effect on KOA by a mechanism involving downregulation of the Wnt/β-catenin pathway,which inhibited both cartilage extracellular matrix degradation and chondrocyte apoptosis to ameliorate KOA in rats. | YAO Nan CHEN Guocai LU Yanyan XU Xuemeng ZHAO Chuanxi HUANG Xuejun LIU Wengang PENG Sha WU Huai | 2021 | Journal of Traditional Chinese Medicine2021,41,6: | 5 |

| 4 | Quantitative detection of ING1 mRNA under different gene regulation based on molecular beacon显示文摘The effects of drug treatment, gene transfection, and p53 RNA interference (RNAi) on level of ING1 mRNA in tumor cells were quantitatively detected with the help of ING1 molecular beacon (MB)/cRNA standard curve based on MB detection technology. Results showed that the level of ING1 mRNA could be upregulated by 5-FU treatment or ING1 transfection in low expression cell line of MCF-7 and be inhibited by silencing p53 with RNAi technology in normal expression cell line of CNE2. The level of ING1 varied from 7.7×10?16 to 53.4× 10?16 mol/μg total RNA of tumor cells. | 9 | Perspective of Calcium Imaging Technology Applied to Acupuncture Research显示文摘Acupuncture, a therapeutic treatment defined as the insertion of needles into the body at specific points(ie, acupoints), has growing in popularity world-wide to treat various diseases effectively, especially acute and chronic pain. In parallel, interest in the physiological mechanisms underlying acupuncture analgesia, particularly the neural mechanisms have been increasing. Over the past decades, our understanding of how the central nervous system and peripheral nervous system process signals induced by acupuncture has developed rapidly by using electrophysiological methods. However, with the development of neuroscience, electrophysiology is being challenged by calcium imaging in view field, neuron population and visualization in vivo. Owing to the outstanding spatial resolution, the novel imaging approaches provide opportunities to enrich our knowledge about the neurophysiological mechanisms of acupuncture analgesia at subcellular, cellular, and circuit levels in combination with new labeling, genetic and circuit tracing techniques. Therefore, this review will introduce the principle and the method of calcium imaging applied to acupuncture research. | 10 | A two-year(2020–2021)observation of marine phycotoxins in phytoplankton in typical mariculture areas of East China Sea显示文摘To learn the marine phycotoxins(MPTs)contamination status in mariculture areas in the East China Sea(ECS),from May 2020 to October 2021,80 net-concentrated phytoplankton samples were collected from 12 stations in three typical areas with high incidence of red tides,namely,Gouqi Island,Sandu’ao-Lianjiang,and Zhangzhou-Dongshan Island in ECS,and MPTs of the samples were detected.Six types of toxins were detected in 24 samples from 9 stations.Lipophilic marine toxins(LMTs)were more common and diverse in these areas.Pectenotoxin 2(PTX2)was the main lipophilic marine toxin(LMT)in the concentrated phytoplankton samples and the occurrence showed seasonal differences from north to south.According to the potential risks of pectenotoxin(PTXs)to seafood safety,it is suggested to execute regular monitoring on PTXs in ECS and a mandatory standard should be formulated based on the comprehensive analysis of in-situ monitoring and lab research.Meanwhile,contamination risks of cyclic imine toxins in the north and domoic acid(DA)in the south of ECS should also be paid with attention to.Only 2 paralytic shellfish toxins(PSTs),N-sulfocarbamoyl toxin C2(C2),and decarbamoyl gonyatoxin 3(dcGTX3),were found from spring samples in the north of ECS.As the biggest mussel culture county of China,Gouqi Island showed higher toxin diversity and the toxin detection rate was higher than the other two areas.In Gouqi Island area,PSTs were a serious potential threat in spring,and LMTs instead of PSTs became the main risk in summer-autumn seasons.To ensure the safety of seafood and marine environmental health,it is recommended to conduct long-term targeted tracking and monitoring of MPTs in this and similar mariculture areas. | Xiaoqing TIAN Chengqi FAN Yunyu TANG Haiyan ZHANG Wei KANG Sha CHEN Chongbin LI Ya’nan LU | 2022 | Journal of Oceanology and Limnology2022,40,6: | 1 |

| 16 | VOC emission caps constrained by air quality targets based on response surface model:A case study in the Pearl River Delta Region,China显示文摘Because of the recent growth in ground-level ozone and increased emission of volatile organic compounds(VOCs),VOC emission control has become a major concern in China.In response,emission caps to control VOC have been stipulated in recent policies,but few of them were constrained by the co-control target of PM_(2.5)and ozone,and discussed the factor that influence the emission cap formulation.Herein,we proposed a framework for quantification of VOC emission caps constrained by targets for PM_(2.5)and ozone via a new response surface modeling(RSM)technique,achieving 50%computational cost savings of the quantification.In the Pearl River Delta(PRD)region,the VOC emission caps constrained by air quality targets varied greatly with the NOxemission reduction level.If control measures in the surrounding areas of the PRD region were not considered,there could be two feasible strategies for VOC emission caps to meet air quality targets(160μg/m^(3)for the maximum 8-hr-average 90th-percentile(MDA8-90%)ozone and 25μg/m^(3)for the annual average of PM_(2.5)):a moderate VOC emission cap with<20%NOxemission reductions or a notable VOC emission cap with>60%NOxemission reductions.If the ozone concentration target were reduced to 155μg/m^(3),deep NOxemission reductions is the only feasible ozone control measure in PRD.Optimization of seasonal VOC emission caps based on the Monte Carlo simulation could allow us to gain higher ozone benefits or greater VOC emission reductions.If VOC emissions were further reduced in autumn,MDA8-90%ozone could be lowered by 0.3-1.5μg/m^(3),equaling the ozone benefits of 10%VOC emission reduction measures.The method for VOC emission cap quantification and optimization proposed in this study could provide scientific guidance for coordinated control of regional PM_(2.5)and O_(3)pollution in China. | Ya’nan Hu Bowen Shi Xin Yuan Chuanzeng Zheng Qing’e Sha Yufan Yu Zhijiong Huang Junyu Zheng | 2023 | Journal of Environmental Sciences2023,,1: | 0 |

| 20 | Inhibition of self-renewal and differentiation of HT-29 cells-derived cancer stem-like cells by scutellarin via Hedgehog signaling pathway显示文摘OBJECTIVE To investigate the inhibitory effect of scutellarin on the self-renewal and differentiation of HT-29 cells-derived cancer stem-like cells(HT-29CSC)in vitro and in vivo,and to explore its mechanism.METHODS The effect of scutellarin on the growth of HT-29CSC was determined by 3D Culture assay.The effect of scutellarin on growth and transformation of HT-29CSC was probed by soft agar colony formation assay.The effect of scutellarin on the differentiation of HT-29CSC was determined by serum induction differentiation assay in vitro.The effects of scutellarin on the expressions of marker gene Lgr5,target gene c-Myc,proliferation gene CK20 and Nanog gene were measured by quantitative real-time RT-PCR.Investigate the effect of scutellarin on the expression of c-Myc,Gli1,and Lgr5 protein by Western blotting.A subcutaneous xenograft model of colon cancer in nude mice was established and administered by intraperitoneal injection.The change of body weight and tumor size of nude mice were observed every two days.Investi⁃gate the effects of scutellarin on the growth of xenograft tumors in nude mice.The expression of CD133,Lgr5,Gli1,Ptch1,c-Myc,Ki67,CK20,Nanog gene in tumors were measured by quantitative real-time RT-PCR.The expression of c-Myc,Gli1,Lgr5,CD133,Ki67 protein were measured by Western blotting.RESULTS Scutellarin can inhibit the growth of HT-29CSC in 3D culture.Compared with the solvent control group,scutellarin can significantly inhibit the growth and transformation and differentiation of HT-29CSC in vitro(P<0.01).The expression levels of marker genes Lgr5,target gene c-Myc,proliferation gene CK20 and Nanog in HT-29CSC were down-regulated by scutellarin.Scutellarin can reduce the expression of c-Myc,Gli1,and Lgr5 protein in HT-29CSC.Scutellarin can inhibit the growth of colon cancer xenografts,lower CD133,Lgr5,Gli1,Ptch1,c-Myc,Ki67,CK20,and Nanog mRNA level of xenograft tumors,reduce the expression of c-Myc,Gli1,Lgr5,CD133,and Ki67 protein of xenograft tumors in nude mice.CONCLUSION Scutellarin,which is the main component of scutellaria barbata,can inhibit the differentiation of HT-29CSC and the mechanism is to inhibit the activity of Hedgehog signaling pathway. | LEI Nan XIONG Si-hui TAN Li HE Man ZHANG Meng SUN Qiang ZENG Sha CHEN Li XU Hai-bo | 2019 | 中国药理学与毒理学杂志2019,33,9: | 0 |
