1Cloning and sequence characteristics of the genomic gene of a rice metallothionein显示文摘Northern blot analysis showed that a metallothionein gene, ricMT is expressed strongly in the stem of rice with an expression level that could be more than 100-fold stronger than in leaf blades. The results suggest that the 5’ upstream region flanking the coding sequence of the ricMT may contain a fairly strong promoter To elucidatr its regulation and promoter structure, the genomic clones of ricMT were screened out from a rice genomic library and a fragment of about 4084 bp was sequenced. The fragment included a 5’ upstream region of ca. 2970 bp, a transcription region of ca. 690 bp and a 3’ downstream region of ca. 420 bp. Computer analysis of the sequence homology showed that the 5’ upstream region included a putative TATA box, a putative CAAT box, and a typical metal-responsive element TGCGCGCG. 2The mitochondrial function was impaired in APP knockout mouse embryo fibroblast cells显示文摘The amyloid precursor protein (APP) is recognized as the source of Aβ, which plays an important role in Alzheimer’s disease. However, the biological function of APP is obscure. Previous studies showed that mitochondria could be a target of APP. In this work, APP knockout mouse embryo fibroblast (MEF) cells were used to test if APP plays any role in maintaining the mitochondrial function. As the result, APP knockout MEF cells (APP-/- cells) showed the abnormal mitochondrial function, including slower cell proliferation, lower mitochondrial membrane potential, lower intracellular ROS, higher mitochondrial membrane fluidity and lower cytochrome c oxidase activity than their wild-type counterparts. 3Progress in artificial control system for gene expression显示文摘Along with the increasingly wide application of transgenic techniques, new stricter criteria have been raised for controlling the expression of exogenous genes. For these demands, a series of artificial control systems for gene expression have been developed and testified in recent years, which can control exogenous genes expression in exact time and certain level by administration of a specific drug or hormone. 13Slip-Control Strategy of Dual Independent Electric Drive Tracked Vehicle显示文摘In order to improve the brake performance of a dual independent electric drive tracked vehicle,a dynamic model for braking situation was established.Then,a sliding model controller(SMC)with an auxiliary system was designed to control the slip and its effectiveness was proved.A hardware-in-loop simulation through MATLAB/XPC was compared with the normal SMC and normal integral sliding mode controller(ISMC),the results show that SMC with the auxiliary system has a better performance:a smaller overshoot and steady state error.The disturbance is suppressed effectively.In the initial speed of 65.km/h,the brake distance was shortened by 3.4%and 6.8%compared with the other two methods,respectively.Finally,initial speeds of 30-36.km/h tests was carried out on a flat soil road.Compared with a no-control brake,the displacement was shortened by 1.8.m.It demonstrates the effectiveness of the slip-control strategy.In the same situation,the error between the simulation and test is 18.1%,which validates the accuracy of models.Shuyang Zhang Yunan Zhang Keyan Ning Nanming Yan Qingfeng Fang 2019Journal of Beijing Institute of Technology2019,28,4:1

18Endogenously generated amyloid β increases membrane fluidity in neural 2a cells显示文摘The effect of endogenously generated amyloid β on membrane fluidity was investigated in Neural 2a cells stably expressing Swedish mutant amyloid precursor protein (APPswe). Membrane fluidity was studied by fluorescence polarizability using 1,6-Diphenyl-1,3,5-Hexatriene (DPH) as the fluorescence probe. It was found that the membrane fluidity in APPswe cells was significantly higher than that in its wild type counterparts. Alleviating the effect of amyloid β either by γ secretase activity inhibition or by amyloid antibody treatment decreased membrane fluidity, which indicated an important role of amyloid β in increasing membrane fluidity.
