1Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. 2Methodologic research on TIMP-1,TIMP-2 detection as a new diagnostic index for hepatic fibrosis and its significance显示文摘AIM: To set up a new method to detect tissue inhibitors ofmetalloproteinase1 and -2(TIMP-1 and TIMP-2) in sero ofpatients with hepatic cirrhosis, and to investigate theexpression and location of TIMP-1 and TIMP-2 in liver tissueof patients with hepatic cirrhosis, and the correlationbetween TIMPs in liver and those in sera so as to discusswhether TIMPs can be used ss a diagnosis index of hepaticfibrosisMETHODS: The monoclonal antibodies (McAbs) of TIMP-1and TIMP-2 were used to sensitize erythrocytes, and solid-phase absorption to sensitized erythrocytes (SPASE) wasused to detect TIMP-1 and TIMP-2 in the sera of patients withhepatic cirrhosis. Meanwhile, with the method of in situhybridization and immunohistochemistry, we studied themRNA expression and antigen location of TIMP-1 and TIMP-2in the livers of 40 hepatic cirrhosis patients with pathologicdiagnosis.RESULTS: With SPASE, they were 16.4 % higher in theacute hepatitis group, 33.3 % higher in the chronic hepatitisgroup, and the positive rates were 73.6 % and 61. 2 %respectively in sero of hepatic cirrhosis patients, which wereremarkably higher than those in chronic hepatitis and acutehepatitis group ( P < 0. 001 ). In 40 samples of hepaticcirrhosis tissues, all of them showed positive expression ofTIMP-1 and TIMP-2 mRNA detected withimmunohistochemistry or in situ hybridization (positive ratewas 100 % ). Expression of TIMPs in different degrees couldbe found in liver tissue with cirrhosis. 5Oral health status in Sichuan Province: findings from the oral health survey of Sichuan, 2015–2016显示文摘To investigate oral health status in the residents of Sichuan Province, southwest China, a cross-sectional study was performed using the latest Oral Health Survey Basic Methods recommended by the World Health Organization. A multistage stratified random cluster-sampling method was used to enroll participants from the following three groups: children aged 3–5 years,adolescents aged 12 years, and people aged 65–74 years. In these three groups, the mean numbers of teeth that were affected by caries were 3.28, 0.86 and 5.13, respectively, resulting in a prevalence of 63.47%, 37.20% and 83.20%, respectively.Relative to the high rate of decayed teeth, the prevalence of fillings was very low in all age groups(0.97%, 7.24% and 5.43%,respectively). In the 12-year-old adolescent group, only 3.61% had good pit and fissure sealing. In addition, the rate of dental fluorosis was 24.80%, and the Community Fluorosis Index value was 0.39. In the elder group, the community periodontal index was2.92. The prevalence in the elderly of having lost at least one tooth was 75.54%. Additionally, 4.44% of these participants were edentulous. The incidence of dental prosthesis was 51.75%, the proportion with a removable partial denture, a fixed denture, full dentures, dental implants and an informal fixed bridge was 21.59%, 11.45%, 4.64%, 0 and 16.67%, respectively. In this study,8.2% of the elderly participants were affected by different types of oral mucosal lesions. Among such lesions, recurrent aphthous ulcers were most prevalent(2.69%) and oral lichen planuses were second(1.6%). The conclusion presented in this survey is that oral diseases, especially dental caries and periodontal disease, are frequent and common in Sichuan province, China. 8Performance of the Seasonal Forecasting of the Asian Summer Monsoon by BCC_CSM1.1(m)显示文摘This paper provides a comprehensive assessment of Asian summer monsoon prediction skill as a function of lead time and its relationship to sea surface temperature prediction using the seasonal hindcasts of the Beijing Climate Center Climate System Model, BCC CSM1.1(m). For the South and Southeast Asian summer monsoon, reasonable skill is found in the model's forecasting of certain aspects of monsoon climatology and spatiotemporal variability. Nevertheless, deficiencies such as significant forecast errors over the tropical western North Pacific and the eastern equatorial Indian Ocean are also found. In particular, overestimation of the connections of some dynamical monsoon indices with large-scale circulation and precipitation patterns exists in most ensemble mean forecasts, even for short lead-time forecasts.Variations of SST, measured by the first mode over the tropical Pacific and Indian oceans, as well as the spatiotemporal features over the Ni no3.4 region, are overall well predicted. However, this does not necessarily translate into successful forecasts of the Asian summer monsoon by the model. Diagnostics of the relationships between monsoon and SST show that difficulties in predicting the South Asian monsoon can be mainly attributed to the limited regional response of monsoon in observations but the extensive and exaggerated response in predictions due partially to the application of ensemble average forecasting methods. 9Fast track clinical pathway implications in esophagogastrectomy显示文摘AIM:To investigate the feasibility of fast track clinical pathway for esophageal tumor resections. METHODS:One hundred and fourteen patients with esophageal carcinoma who underwent esophagogastrectomy from January 2006 to October 2007 in our department were studied.Fast track clinical pathway included analgesia control,fluid infusion volume control,early ambulation and enteral nutrition. Nasogastric tube was removed 3 d after operation and chest tube was removed 4 d after operation as a routine,and full liquid diet 5 d after operation. RESULTS:Among 114 patients(84 men and 30 women),26 patients underwent fast track surgery,including 17 patients over 65 years old and 9 under 65(P=0.014);18 patients who had preoperative complications could not bear fast track surgery(P< 0.001).No significant differences in tolerance of fast track surgery were attributed to differences in gender,differentiated degree or stage of tumor,pathological type of tumor,or operative incision.The median length of hospital stay was 7 d(5-28 d),4%patients were readmitted to hospital within 30 d of discharge.Three patients died and postoperative mortality was 2.6%. All 3 patients had no determinacy to fast track surgery approach.CONCLUSION:The majority of patients with esophageal carcinoma can tolerate fast track surgery. 10BMAL1 knockout macaque monkeys display reduced sleep and psychiatric disorders显示文摘Circadian disruption is a risk factor for metabolic, psychiatric and age-related disorders, and non-human primate models could help to develop therapeutic treatments. Here, we report the generation of BMAL1 knockout cynomolgus monkeys for circadian-related disorders by CRISPR/Cas9 editing of monkey embryos. These monkeys showed higher nocturnal locomotion and reduced sleep, which was further exacerbated by a constant light regimen. Physiological circadian disruption was reflected by the markedly dampened and arrhythmic blood hormonal levels. Furthermore, BMAL1-deficient monkeys exhibited anxiety and depression, consistent with their stably elevated blood cortisol, and defective sensory processing in auditory oddball tests found in schizophrenia patients. Ablation of BMAL1 up-regulated transcriptional programs toward inflammatory and stress responses, with transcription networks associated with human sleep deprivation, major depressive disorders, and aging. 14Qingfei Xiaoyan Wan,a traditional Chinese medicine formula, ameliorates Pseudomonas aeruginosa–induced acute lung inflammation by regulation of PI3K/AKT and Ras/MAPK pathways显示文摘Gram-negative pathogen–induced nosocomial infections and resistance are a most serious menace to global public health. Qingfei Xiaoyan Wan(QF), a traditional Chinese medicine(TCM)formula, has been used clinically in China for the treatment of upper respiratory tract infections, acute or chronic bronchitis and pulmonary infection. In this study, the effects of QF on Pseudomonas aeruginosa–induced acute pneumonia in mice were evaluated. The mechanisms by which four typical antiinflammatory ingredients from QF, arctigenin(ATG), cholic acid(CLA), chlorogenic acid(CGA) and sinapic acid(SPA), regulate anti-inflammatory signaling pathways and related targets were investigated using molecular biology and molecular docking techniques. The results showed that pretreatment with QF significantly inhibits the release of cytokines(TNF-α and IL-6) and chemokines(IL-8 and RANTES),reduces leukocytes recruitment into inflamed tissues and ameliorates pulmonary edema and necrosis. In addition, ATG was identified as the primary anti-inflammatory agent with action on the PI3K/AKT and Ras/MAPK pathways. CLA and CGA enhanced the actions of ATG and exhibited synergistic NF-κB inactivation effects possibly via the Ras/MAPK signaling pathway. Moreover, CLA is speculated to target FGFR and MEK firstly. 15Molecular Typing of Leptospira interrogans Strains Isolated from Rattus tanezumi in Guizhou Province,Southwest of China显示文摘Objective To identify and type three leptospires isolated from Rattus tanezumi in Guizhou Province by using three molecular techniques (PFGE, MLVA, and MLST), reveal the molecular characteristic of causative agents of local leptospirosis and evaluate these three molecular methods based on their detection resolution and efficiency. Methods Three Leptospira strains were isolated from the kidney of Rattus tanezumi and cultured with EMJH medium. PFGE, MLVA, and MLST assays were applied to type the three strains isolated from Rattus tanezumi in Guizhou Province. Results PFGE, MLVA, and MLST typing showed that the three leptospiral isolates matched with leptospiral serogroup Icterohaemorrhagiae serovar Lai. The findings of the genotyping methods were consistent. MLVA and MLST defined genotypes, whereas PFGE allowed the recognition of additional subgroups within the genotypes, and the findings of molecular typing were also consistent with those of traditional techniques. 17Substitution of chemical fertilizer by Chinese milk vetch improves the sustainability of yield and accumulation of soil organic carbon in a double-rice cropping system显示文摘The double-rice cropping system is a very important intensive cropping system for food security in China. There have been few studies of the sustainability of yield and accumulation of soil organic carbon (SOC) in the double-rice cropping system following a partial substitution of chemical fertilizer by Chinese milk vetch (Mv). We conducted a 10-year (2008–2017) field experiment in Nan County, South-Central China, to examine the double-rice productivity and SOC accumulation in a paddy soil in response to different fertilization levels and Mv application (22.5 Mg ha^–1). Fertilizer and Mv were applied both individually and in combination (sole chemical fertilizers, Mv plus 100, 80, 60, 40, and 0% of the recommended dose of chemical fertilizers, labeled as F100, MF100, MF80, MF60, MF40, and MF0, respectively). It was found that the grain yields of double-rice crop in treatments receiving Mv were reduced when the dose of chemical fertilizer was reduced, while the change in SOC stock displayed a double peak curve. The MF100 produced the highest double-rice yield and SOC stock, with the value higher by 13.5 and 26.8% than that in the F100. However, the grain yields increased in the MF80 (by 8.4% compared to the F100), while the SOC stock only increased by 8.4%. Analogous to the change of grain yield, the sustainable yield index (SYI) of double rice were improved significantly in the MF100 and MF80 compared to the F100, while there was a slight increase in the MF60 and MF40. After a certain amount of Mv input (22.5 Mg ha^–1), the carbon sequestration rate was affected by the nutrient input due to the stimulation of microbial biomass. Compared with the MF0, the MF100 and MF40 resulted in a dramatically higher carbon sequestration rate (with the value higher by 71.6 and 70.1%), whereas the MF80 induced a lower carbon sequestration rate with the value lower by 70.1% compared to the MF0.
