| 1 | De novo characterization of the root transcriptome of a traditional Chinese medicinal plant Polygonum cuspidatum显示文摘Various active components have been extracted from the root of Polygonum cuspidatum.However,the genetic basis for their activity is virtually unknown.In this study,25600002 short reads (2.3 Gb) of P.cuspidatum root transcriptome were obtained via Illumina HiSeq 2000 sequencing.A total of 86418 unigenes were assembled de novo and annotated.Twelve,18,60 and 54 unigenes were respectively mapped to the mevalonic acid (MVA),methyl-D-erythritol 4-phosphate (MEP),shikimate and resveratrol biosynthesis pathways,suggesting that they are involved in the biosynthesis of pharmaceutically important anthraquinone and resveratrol.Eighteen potential UDP-glycosyltransferase unigenes were identified as the candidates most likely to be involved in the biosynthesis of glycosides of secondary metabolites.Identification of relevant genes could be important in eventually increasing the yields of the medicinally useful constituents of the P.cuspidatum root.From the previously published transcriptome data of 19 non-model plant taxa,1127 shared orthologs were identified and characterized.This information will be very useful for future functional,phylogenetic and evolutionary studies of these plants. | HAO DaCheng MA Pei MU Jun CHEN ShiLin XIAO PeiGen PENG Yong HUO Li XU LiJia SUN Chao | 2012 | Science China(Life Sciences)2012,55,5: | 36 |

| 4 | Efficacy and safety evaluation of icotinib in patients with advanced non-small cell lung cancer显示文摘Objective: To evaluate the efficacy and safety of icotinib hydrochloride in patients with advanced non-small cell lung cancer (NSCLC). Methods: A total of 89 patients with stage ⅢB or IV NSCLC received icotinib at a dose of 125 mg administered 3 times a day. Icotinib treatment was continued until disease progression or development of unacceptable toxicity. Results: A total of 89 patients were assessable. In patients treated with icotinib, the overall response rate (RR) was 36.0% (32/89), and the disease control rate (DCR) was 69.7% (62/89). RR and DCR were significantly improved in patients with adenocarcinoma versus non-adenocarcinoma (P<0.05). The symptom improvement rate was 57.3% (51/89), and the main symptoms improved were cough, pain, chest distress, dyspnea, and Eastern Cooperative Oncology Group performance status. The main toxic effects were rash [30/89 (33.7%)] and diarrhea [15/89 (16.9%)]. The level of toxicity was typically low. | 5 | A novel gene delivery system targeting cells expressing VEGF receptors显示文摘Two ligand oligopeptides GV1 and GV2 were designed according to the putative binding region of VEGF to its receptors. GV1, GV2 and endosome releasing oligopeptide HA20 were conjugated with poly-L-lysine or protamine and the resulting conjugates could interact with DNA in a noncovalent bond to form a complex. Using pSV2-β-galactosidase as a reporter gene, it has been demonstrated that exogenous gene was transferred into bovine aortic arch-derived endothelial cells (ABAE) andhuman malignant melanoma cell lines (A375) in vitro. In vivo experiments, exogenous gene was transferred into tumor vascular endothelial cells and tumor cells of subcutaneously transplanted human colon cancer LOVO, human malignant melanoma A375 and human hepatoma graft in nude mice. This system could also target gene to intrahepatically transplanted human hepatoma injected via portal vein in nude mice. | 6 | Melatonin alleviates intervertebral disc degeneration by disrupting the IL-1β/NF-κB-NLRP3 inflammasome positive feedback loop显示文摘The inflammatory response is induced by the overexpression of inflammatory cytokines, mainly interleukin(IL)-1β, and is one of the main causes of intervertebral disc degeneration(IVDD). NLR pyrin domain containing 3(NLRP3) inflammasome activation is an important source of IL-1β. As an anti-inflammatory neuroendocrine hormone, melatonin plays various roles in different pathophysiological conditions. However, its roles in IVDD are still not well understood and require more examination. First, we demonstrated that melatonin delayed the progression of IVDD and relieved IVDD-related low back pain in a rat needle puncture IVDD model;moreover, NLRP3 inflammasome activation(NLRP3, p20, and IL-1β levels) was significantly upregulated in severely degenerated human discs and a rat IVDD model. Subsequently, an IL-1β/NF-κB-NLRP3 inflammasome activation positive feedback loop was found in nucleus pulposus(NP) cells that were treated with IL-1β. In these cells, expression of NLRP3 and p20 was significantly increased, NF-κB signaling was involved in this regulation, and mitochondrial reactive oxygen species(mt ROS)production increased. Furthermore, we found that melatonin disrupted the IL-1β/NF-κB-NLRP3 inflammasome activation positive feedback loop in vitro and in vivo. Melatonin treatment decreased NLRP3, p20, and IL-1β levels by inhibiting NF-κB signaling and downregulating mt ROS production. Finally, we showed that melatonin mediated the disruption of the positive feedback loop of IL-1β in vivo. In this study, we showed for the first time that IL-1β promotes its own expression by upregulating NLRP3 inflammasome activation. | 7 | Multimorbidity patterns and association with mortality in 0.5 million Chinese adults显示文摘Background:Few studies have assessed the relationship between multimorbidity patterns and mortality risk in the Chinese population.We aimed to identify multimorbidity patterns and examined the associations of multimorbidity patterns and the number of chronic diseases with the risk of mortality among Chinese middle-aged and older adults.Methods:We used data from the China Kadoorie Biobank and included 512,723 participants aged 30 to 79 years.Multimorbidity was defined as the presence of two or more of the 15 chronic diseases collected by self-report or physical examination at baseline.Multimorbidity patterns were identified using hierarchical cluster analysis.Cox regression was used to estimate the associations of multimorbidity patterns and the number of chronic diseases with all-cause and cause-specific mortality.Results:Overall,15.8%of participants had multimorbidity.The prevalence of multimorbidity increased with age and was higher in urban than rural participants.Four multimorbidity patterns were identified,including cardiometabolic multimorbidity(diabetes,coronary heart disease,stroke,and hypertension),respiratory multimorbidity(tuberculosis,asthma,and chronic obstructive pulmonary disease),gastrointestinal and hepatorenal multimorbidity(gallstone disease,chronic kidney disease,cirrhosis,peptic ulcer,and cancer),and mental and arthritis multimorbidity(neurasthenia,psychiatric disorder,and rheumatoid arthritis).During a median of 10.8 years of follow-up,49,371 deaths occurred.Compared with participants without multimorbidity,cardiometabolic multimorbidity(hazard ratios[HR]=2.20,95%confidence intervals[CI]:2.14-2.26)and respiratory multimorbidity(HR=2.13,95%CI:1.97-2.31)demonstrated relatively higher risks of mortality,followed by gastrointestinal and hepatorenal multimorbidity(HR=1.33,95%CI:1.22-1.46).The mortality risk increased by 36%(HR=1.36,95%CI:1.35-1.37)with every additional disease.Conclusion:Cardiometabolic multimorbidity and respiratory multimorbidity posed the highest threat on mortality risk and deserved particular attention in Chinese adults. | Junning Fan Zhijia Sun Canqing Yu Yu Guo Pei Pei Ling Yang Yiping Chen Huaidong Du Dianjianyi Sun Yuanjie Pang Jun Zhang Simon Gilbert Daniel Avery Junshi Chen Zhengming Chen Jun Lyu Liming Li On Behalf of the China Kadoorie Biobank Collaborative Group | 2022 | Chinese Medical Journal2022,,6: | 17 |

| 9 | Numerical and Experimental Study on Flow-induced Noise at Blade-passing Frequency in Centrifugal Pumps显示文摘With the increasing noise pollution,low noise optimization of centrifugal pimps has become a hot topic.However,experimental study on this problem is unacceptable for industrial applications due to unsustainable cost.A hybrid method that couples computational fluid dynamics(CFD)with computational aeroacoustic software is used to predict the flow-induced noise of pumps in order to minimize the noise of centrifugal pumps in actual projects.Under Langthjem’s assumption that the blade surface pressure is the main flow-induced acoustic source in centrifugal pumps,the blade surface pressure pulsation is considered in terms of the acoustical sources and simulated using CFX software.The pressure pulsation and noise distribution in the near-cutoff region are examined for the blade-passing frequency(BPF)noise,and the sound pressure level(SPL)reached peaks near the cutoff that corresponded with the pressure pulsation in this region.An experiment is performed to validate this prediction.Four hydrophones are fixed to the inlet and outlet ports of the test pump to measure the flow-induced noise from the four-port model.The simulation results for the noise are analyzed and compared with the experimental results.The variation in the calculated noise with changes in the flow agreed well with the experimental results.When the flow rate was increased,the SPL first decreased and reached the minimum near the best efficient point(BEP);it then increased when the flow rate was further increased.The numerical and experimental results confirmed that the BPF noise generated by a blade-rotating dipole roughly reflects the acoustic features of centrifugal pumps.The noise simulation method in current study has a good feasibility and suitability,which could be adopted in engineering design to predict and optimize the hydroacoustic behavior of centrifugal pumps. | YANG Jun YUAN Shouqi YUAN Jianping SI Qiaorui PEI Ji | 2014 | Chinese Journal of Mechanical Engineering2014,27,3: | 16 |
| 10 | Overexpression of HMGB1 A-box reduced lipopolysaccharide-induced intestinal inflammation via HMGB1/TLR4 signaling in vitro显示文摘AIM: To investigate the inhibitory effects and mechanism of high mobility group box(HMGB)1 A-box in lipopolysaccharide(LPS)-induced intestinal inflammation.METHODS: Overexpression of HMGB1 A-box in human intestinal epithelial cell lines(SW480 cells) was achieved using the plasmid p EGFP-N1. HMGB1 A-box-overexpressing SW480 cells were stimulated with LPS and co-culturing with human monocyte-like cell lines(THP-1 cells) using a Transwell system, compared with another HMGB1 inhibitor ethyl pyruvate(EP). The m RNA and protein levels of HMGB1/toll-like receptor(TLR) 4 signaling pathways [including HMGB1, TLR4, myeloid differentiation factor88(MYD88), Phosphorylated Nuclear Factor κB(p NF-κB) p65] in the stimulated cells were determined by realtime polymerase chain reaction and Western blotting. The levels of the proinflammatory mediators [including HMGB1, interleukin(IL)-1β, IL-6 and tumor necrosis factor(TNF)-α] in the supernatants of the stimulated cells were determined by ELISA.RESULTS: EP downregulated the m RNA and protein levels of HMGB1, inhibited the TLR4 signaling pathways(TLR4, MYD88 and p NF-κB p65) and reduced the secretion of proinflammatory mediators(HMGB1, IL-1β, IL-6 and TNF-α) in the SW480 and THP-1 cells activated by LPS but not in the unstimulated cells. Activated by LPS, the overexpression of HMGB1 A-box in the SW480 cells also inhibited the HMGB1/TLR4 signaling pathways and reduced the secretion of these proinflammatory mediators in the THP-1 cells but not in the transfected and unstimulated cells. Moreover, neurotrophic factors play a key role in the regeneration and repair of peripheral nerve. However, whether craniocerebral injury alters the repair of peripheral nerve injuries remains poorly understood. Rat injury models were established by transecting the left sciatic nerve and using a free-fall device to induce craniocerebral injury. Compared with sciatic nerve injury alone after 6–12 weeks, rats with combined sciatic and craniocerebral injuries showed decreased sciatic functional index, increased recovery of gastrocnemius muscle wet weight, recovery of sciatic nerve ganglia and corresponding spinal cord segment neuron morphologies, and increased numbers of horseradish peroxidase-labeled cells. | 13 | Variation of cyanobacteria with different environmental conditions in Nansi Lake,China显示文摘Nansi Lake is located on the east line of the South-to-North Water Diversion Project in China.A comprehensive study was carried out to investigate the spatial and temporal distribution of cyanobacteria in the lake from June 2008 to May 2011 based on monthly sample monitoring from five stations.The effect of environmental factors on cyanobacterial abundance was also evaluated.The cyanobacterial community contained 15 genera and 23 species.The cyanobacterial abundance of each monitoring station ranged from 0 to 1.53×10 7 cells/L with an average of 1.45×10 6 cells/L,which accounted for 11.66% of the total phytoplankton abundance.The dominant species of cyanobacteria were Pseudanabaena(32.94%) and Merismopedia(19.85%),not the bloom-forming algae such as Microcystis and Anabaena.In addition,the cyanobacterial community structure and water quality variables changed substantially over the survey period.Redundancy analysis(RDA) suggested that temperature and phosphorus were important environmental factors that affected cyanobacteria.Temperature was the most important factor affecting cyanobacterial abundance.The effect of phosphorus on cyanobacterial abundance was more notable in warm periods than in periods with low temperature. | Chang Tian Haiyan Pei Wenrong Hu Jun Xie | 2012 | Journal of Environmental Sciences2012,24,8: | 14 |
| 14 | Plant diversity reduces the effect of multiple heavy metal pollution on soil enzyme activities and microbial community structure显示文摘It is unclear whether certain plant species and plant diversity could reduce the impacts of multiple heavy metal pollution on soil microbial structure and soil enzyme activities. Random amplified polymorphic DNA (RAPD) was used to analyze the genetic diversity and microbial similarity in planted and unplanted soil under combined cadmium (Cd) and lead (Pb) pollution. A metal hyper- accumulator, Brassica juncea, and a common plant, Festuca arundinacea Schreb, were used in this research. The results showed that microorganism quantity in planted soil significantly increased, compared with that in unplanted soil with Cd and Pb pollution. The order of microbial community sensitivity in response to Cd and Pb stress was as follows: actinomycetes 〉 bacteria 〉 fungi. Respiration, phosphatase, urease and dehydrogenase activity were significantly inhibited due to Cd and Pb stress. Compared with unplanted soil, planted soils have frequently been reported to have higher rates of microbial activity due to the presence of additional surfaces for microbial colonization and organic compounds released by the plant roots. Two coexisting plants could increase microbe population and the activity of phosphatases, dehydrogenases and, in particular, ureases. Soil enzyme activity was higher in B. juncea phytoremediated soil than in F. arundinacea planted soil in this study. Heavy metal pollution decreased the richness of the soil microbial community, but plant diversity increased DNA sequence diversity and maintained DNA sequence diversity at highlevels. | 17 | Experimental Investigation on the Flow-induced Noise under Variable Conditions for Centrifugal Pumps显示文摘With extensively using of centrifugal pumps,noise generation in these pumps is increasingly receiving research attention in recent years.The noise sources in centrifugal pumps are mainly composed of mechanical noise and flow-induced noise.And the study of flow-induced noise has become a hotspot and important domain in the field.The flow-induced noise closely related to the inner pressure pulses and vibration of volute in pumps,therefore,it is necessary to research the interaction and mechanism among them.To investigate the relationships,a test system is designed which includes a test loop and a measurement system.The hydrophones and pressure sensors are installed on the outlet of the pump and vibration acceleration sensors are disposed on the pump body.Via these instruments,the signals of noise,pressure pulses and vibration are collected and analyzed.The results show that the level of flow-induced noise becomes smaller as the flow increment during low flow rate operations,and it is steadily close to the design point,then it increases with the growing of flow rate in high flow rate conditions.Furthermore,there are some similar peak points in the power spectrum charts of noise,pressure pulses and vibration.The broadband noise at low flow rate is mostly focused on the region of 0-40 times shaft frequency,which is mostly made by rotating stall and vortex;while the noise at high flow rate conditions is focused on the region of 60-100 times shaft frequency,which may be mostly made by cavitations.The proposed research is of practical and academic significance to the study of noise reduction for centrifugal pumps. | YUAN Shouqi YANG Jun YUAN Jianping LUO Yin PEI Ji | 2012 | Chinese Journal of Mechanical Engineering2012,25,3: | 12 |

Due to progressiveness and complexity of modern combat aircraft, the existing vulnerability modeling and computation methods cannot meet the current engineering application requirements. Therefore, a vulnerability modeling and computation method based on product structure and CATIA is proposed in sufficient consideration of the design characteristics of modern combat aircraft. This method directly constructs the aircraft vulnerability model by CATIA or the digital model database, and manages all the product components of the vulnerability model via aircraft product structure. Using CAA second development, the detailed operations and computation methods of vulnerability analysis are integrated into CATIA software environment. Comprehensive assessment data and visual kill probability Iso-contours can also be presented, which meet the vulnerability analysis requirements of modern combat aircraft effectively. | 19 | Risk factors and outcomes of acute kidney injury after intracoronary stent implantation显示文摘BACKGROUND:Acute kidney injury following percutaneous coronary intervention(PCI) is associated with a worse outcome.However,the risk factors and outcomes of acute kidney injury(AKI) in patients after intracoronary stent implantation are still unknown.METHODS:A retrospective case control study was done in 325 patients who underwent intracoronary stent implantation from January 2010 to March 2011 at the Drum Tower Hospital of Nanjing University School of Medicine.Those were excluded from the study if they had incomplete clinical data.The patients were divided into a normal group and a AKI group according to the standard of post-operation day 7 to identify AKI.The parameters of the patients included:1) pre-operative ones:age,gender,hypertension,diabetes mellitus,cerebrovascular disease,left ventricular insufficiency,peripheral angiopathy,creatinine,urea nitrogen,estimated glomerular filtration rate(eGFR),hyperuricemia,proteinuria,emergency operation,hydration,medications(ACEI/ARBs,statins);2) intraoperative ones:dose of contrast media,operative time,hypotension;and 3) postoperative one:hypotension.The parameters were analyzed with univariate analysis and multivariate logistical regression analysis.RESULTS:Of the 325 patients,51(15.7%) developed AKI.Hospital day and in-hospital mortality were increased significantly in the AKI-group.Univariate analysis showed that age,pre-operative parameters(left ventricular insufficiency,peripheral angiopathy,creatinine,urea nitrogen,estimated glomerular filtration rate,hyperuricemia,proteinuria,hydration),emergency operation,intraoperative parameters(operative time,hypotension) and postoperative hypotension were significantly different.However,multivariate logistic regression analysis revealed that increased age(OR=0.253,95%CI=0.088-0.727),pre-operative proteinuria(OR=5.351,95%CI=2.128-13.459),pre-operative left ventricular insufficiency(OR=8.704,95%CI=3.170-23.898),eGFR<60 ml/min/1.73 m^2(OR=6.677,95%CI = 1.167-38.193),prolonged operative time,intraoperative hypotension(OR=25.245,95%CI=1.001-1.034) were independent risk factors of AKI.CONCLUSIONS:AKI is a common complication and associated with ominous outcome following intracoronary stent implantation.Increased age,pre-operative proteinuria,pre-operative left ventricular insufficiency,pre-operative low estimated glomerular filtration rate,prolonged operative time,intraoperative hypotension were the significant risk factors of AKI. | Fei He Jun Zhang Zhong-qiu Lu Qing-ling Gao Du-juan Sha Li-gang Pei Guo-feng Fan | 2012 | World Journal of Emergency Medicine2012,3,3: | 11 |
| 20 | Selective lateral lymph node dissection after neoadjuvant chemoradiotherapy in rectal cancer显示文摘BACKGROUND Lateral lymph node metastasis is one of the leading causes of local recurrence in patients with advanced mid or low rectal cancer.Neoadjuvant chemoradiotherapy(NCRT)can effectively reduce the postoperative recurrence rate;thus,NCRT with total mesorectal excision(TME)is the most widely accepted standard of care for rectal cancer.The addition of lateral lymph node dissection(LLND)after NCRT remains a controversial topic.AIM To investigate the surgical outcomes of TME plus LLND,and the possible risk factors for lateral lymph node metastasis after NCRT.METHODS This retrospective study reviewed 89 consecutive patients with clinical stage II-III mid or low rectal cancer who underwent TME and LLND from June 2016 to October 2018.In the NCRT group,TME plus LLND was performed in patients with short axis(SA)of the lateral lymph node greater than 5 mm.In the non-NCRT group,TME plus LLND was performed in patients with SA of the lateral lymph node greater than 10 mm.Data regarding patient demographics,clinical workup,surgical procedure,complications,and outcomes were collected.Multivariate logistic regression analysis was performed to evaluate the possible risk factors for lateral lymph node metastasis in NCRT patients.RESULTS LLN metastasis was pathologically confirmed in 35 patients(39.3%):26(41.3%)in the NCRT group and 9(34.6%)in the non-NCRT group.The most common site of metastasis was around the obturator nerve(21/35)followed by the internal iliac artery region(12/35).In the NCRT patients,46%of patients with SA of LLN greater than 7 mm were positive.The postoperative 30-d mortality rate was 0%.Two(2.2%)patients suffered from lateral local recurrence in the 2-year follow up.Multivariate analysis showed that cT4 stage(odds ratio[OR]=5.124,95%confidence interval[CI]:1.419-18.508;P=0.013),poor differentiation type(OR=4.014,95%CI:1.038-15.520;P=0.044),and SA≥7 mm(OR=7.539,95%CI:1.487-38.214;P=0.015)were statistically significant risk factors associated with LLN metastasis.CONCLUSION NCRT is not sufficient as a stand-alone therapy to eradicate LLN metastasis in lower rectal cancer patients and surgeons should consider performing selective LLND in patients with greater LLN SA diameter,poorer histological differentiation,or advanced T stage.Selective LLND for NCRT patients can have a favorable oncological outcome. | Jia-Nan Chen Zheng Liu Zhi-Jie Wang Shi-Wen Mei Hai-Yu Shen Juan Li Wei Pei Zheng Wang Xi-Shan Wang Jun Yu Qian Liu | 2020 | World Journal of Gastroenterology2020,26,21: | 11 |
