1Identification of a novel coronavirus causing severe pneumonia in human:a descriptive study显示文摘Background:Human infections with zoonotic coronaviruses(CoVs),including severe acute respiratory syndrome(SARS)-CoV and Middle East respiratory syndrome(MERS)-CoV,have raised great public health concern globally.Here,we report a novel batorigin CoV causing severe and fatal pneumonia in humans.Methods:We collected clinical data and bronchoalveolar lavage(BAL)specimens from five patients with severe pneumonia from Wuhan Jinyintan Hospital,Hubei province,China.Nucleic acids of the BAL were extracted and subjected to next-generation sequencing.Virus isolation was carried out,and maximum-likelihood phylogenetic trees were constructed.Results:Five patients hospitalized from December 18 to December 29,2019 presented with fever,cough,and dyspnea accompanied by complications of acute respiratory distress syndrome.Chest radiography revealed diffuse opacities and consolidation.One of these patients died.Sequence results revealed the presence of a previously unknownβ-CoV strain in all five patients,with 99.8%to 99.9%nucleotide identities among the isolates.These isolates showed 79.0%nucleotide identity with the sequence of SARS-CoV(GenBank NC_004718)and 51.8%identity with the sequence of MERS-CoV(GenBank NC_019843).The virus is phylogenetically closest to a bat SARS-like CoV(SL-ZC45,GenBank MG772933)with 87.6%to 87.7%nucleotide identity,but is in a separate clade.Moreover,these viruses have a single intact open reading frame gene 8,as a further indicator of bat-origin CoVs.However,the amino acid sequence of the tentative receptor-binding domain resembles that of SARS-CoV,indicating that these viruses might use the same receptor.Conclusion:A novel bat-borne CoV was identified that is associated with severe and fatal respiratory disease in humans.Li-Li Ren Ye-Ming Wang Zhi-Qiang Wu Zi-Chun Xiang Li Guo Teng Xu Yong-Zhong Jiang Yan Xiong Yong-Jun Li Xing-Wang Li Hui Li Guo-Hui Fan Xiao-Ying Gu Yan Xiao Hong Gao Jiu-Yang Xu Fan Yang Xin-Ming Wang Chao Wu Lan Chen Yi-Wei Liu Bo Liu Jian Yang Xiao-Rui Wang Jie Dong Li Li Chao-Lin Huang Jian-Ping Zhao Yi Hu Zhen-Shun Cheng Un-Lin Liu Zhao-Hui Qian Chuan Qin Qi Jin Bin Cao Jian-Wei Wang 2020Chinese Medical Journal2020,,9:99
2Oceanic plate subduction history in the western Pacific Ocean:Constraint from late Mesozoic evolution of the Tan-Lu Fault Zone显示文摘The NE-to NNE-striking Tan-Lu Fault Zone(TLFZ) is the largest fault zone in East China, and a typical representative for the circum-Pacific tectonics. Its late Mesozoic evolution resulted from subduction of the Paleo-Pacific Plate,and can be used for indication to the subduction history. The TLFZ reactivated at the end of Middle Jurassic since its origination in Middle Triassic. This phase of sinistral motion can only be recognized along the eastern edge of the Dabie-Sulu orogenis,and indicates initiation of the Paleo-Pacific(Izanagi) Plate subduction beneath the East China continent. After the Late Jurassic standstill, the fault zone experienced intense sinistral faulting again at the beginning of Early Cretaceous under N-S compression that resulted from the NNW-ward, low-angle, high-speed subduction of the Izanagi Plate. It turned into normal faulting in the rest of Early Cretaceous, which was simultaneous with the peak destruction of the North China Craton caused by backarc extension that resulted from rollback of the subducting Izanagi Plate. The TLFZ was subjected to sinistral, transpressive displacement again at the end of Early Cretaceous. This shortening event led to termination of the North China Craton destruction. The fault zone suffered local normal faulting in Late Cretaceous due to the far-field, weak backarc extension. The late Mesozoic evolution of the TLFZ show repeated alternation between the transpressive strike-slip motion and normal faulting. 3A novel gene delivery system targeting cells expressing VEGF receptors显示文摘Two ligand oligopeptides GV1 and GV2 were designed according to the putative binding region of VEGF to its receptors. GV1, GV2 and endosome releasing oligopeptide HA20 were conjugated with poly-L-lysine or protamine and the resulting conjugates could interact with DNA in a noncovalent bond to form a complex. Using pSV2-β-galactosidase as a reporter gene, it has been demonstrated that exogenous gene was transferred into bovine aortic arch-derived endothelial cells (ABAE) andhuman malignant melanoma cell lines (A375) in vitro. In vivo experiments, exogenous gene was transferred into tumor vascular endothelial cells and tumor cells of subcutaneously transplanted human colon cancer LOVO, human malignant melanoma A375 and human hepatoma graft in nude mice. This system could also target gene to intrahepatically transplanted human hepatoma injected via portal vein in nude mice. 4Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). 5Visible Kerr comb generation in a high-Q silica microdisk resonator with a large wedge angle显示文摘This paper describes the specially designed geometry of a dry-etched large-wedge-angle silica microdisk resonator that enables anomalous dispersion in the 780 nm wavelength regime. This anomalous dispersion occurs naturally without the use of a mode-hybridization technique to control the geometrical dispersion. By fabricating a1-μm-thick silica microdisk with a wedge angle as large as 56° and an optical Q-factor larger than 107, we achieve a visible Kerr comb that covers the wavelength interval of 700–897 nm. 6The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 7Toll-Like Receptor 4 Deficiency Causes Reduced Exploratory Behavior in Mice Under Approach-Avoidance Conflict显示文摘Abnormal approach-avoidance behavior has been linked to deficits in the mesolimbic dopamine(DA)system of the brain. Recently, increasing evidence has indicated that toll-like receptor 4(TLR4), an important pattern-recognition receptor in the innate immune system,can be directly activated by substances of abuse, resulting in an increase of the extracellular DA level in the nucleus accumbens. We thus hypothesized that TLR4-dependent signaling might regulate approach-avoidance behavior. To test this hypothesis, we compared the novelty-seeking and social interaction behaviors of TLR4-deficient(TLR4^(-/-))and wild-type(WT) mice in an approach-avoidance conflict situation in which the positive motivation to explore a novel object or interact with an unfamiliar mouse was counteracted by the negative motivation to hide in exposed,large spaces. We found that TLR4^(-/-)mice exhibitedreduced novelty-seeking and social interaction in the large open spaces. In less stressful test apparatuses similar in size to the mouse cage, however, TLR4^(-/-)mice performed normally in both novelty-seeking and social interaction tests. The reduced exploratory behaviors under approachavoidance conflict were not due to a high anxiety level or an enhanced fear response in the TLR4^(-/-)mice, as these mice showed normal anxiety and fear responses in the open field and passive avoidance tests, respectively. Importantly,the novelty-seeking behavior in the large open field induced a higher level of c-Fos activation in the nucleus accumbens shell(NAc Sh) in TLR4^(-/-)mice than in WT mice. Partially inactivating the NAc Sh via infusion of GABA receptor agonists restored the novelty-seeking behavior of TLR4^(-/-)mice. These data suggested that TLR4 is crucial for positive motivational behavior under approach-avoidance conflict. 9Synthesis of MoX2 (X =Se or S) monolayers with high-concentration 1T'phase on 4H/fcc-Au nanorods for hydrogen evolution显示文摘Controlled synthesis of transition metal dichalcogenide (TMD) monolayers with unusual crystal phases has attracted increasing attention due to their promising applications in electrocatalysis.However,the facile and large-scale preparation of TMD monolayers with high-concentration unusual crystal phase still remains a challenge.Herein,we report the synthesis of MoX2 (X =Se or S) monolayers with high-concentration semimetallic 1T'phase by using the 4H/face-centered cubic (fcc)-Au nanorod as template to form the 4H/fcc-Au@MoX2 nanocomposite.The concentrations of 1T'phase in the prepared MoSe2 and MoS2 monolayers are up to 86% and 81%,respectively.As a proof-of-concept application,the obtained Au@MoS2 nanocomposite is used for the electrocatalytic hydrogen evolution reaction (HER) in acid medium,exhibiting excellent performance with a low overpotential of 178 mV at the current density of 10 mNcm^2,a small Tafel slope of 43.3 mV/dec,and excellent HER stability.This work paves a way for direct synthesis of TMD monolayers with high-concentration of unusual crystal phase for the electrocatalytic application.Zhengqing Liu Xiao Zhang Yue Gong Qipeng Lu Zhicheng Zhang Hongfei Cheng Qinglang Ma Junze Chen Meiting Zhao Bo Chen Ye Chen Xue-Jun Wu Pengfei Yin Lin Gu Yaping Du Hua Zhang 2019Nano Research2019,12,6:3
10Biomass estimation and uncertainty analysis based on CBERS-02 CCD camera data and field measurement显示文摘The study site is located in Qianyanzhou experimental station and the surrounding area. Based on CBERS-02 satellite data and field measurement, we not only discussed the relationship between NDVI and biomass of two species of coniferous plantations, namely, Pinus massoniana Lamb and Pinus elliottii Engelm, but also introduced the biomass models based on NDVI. The comparison between measured biomass in Qianyanzhou and biomass derived from CBERS-02 CCD data showed that it is feasible to estimate biomass based on NDVI. But its limitations cannot be ignored. This kind of model depends on the dominant vegetation species. There are some effect factors in estimating biomass based on NDVI. This paper analyzes these factors based on fine-resolution CBERS-02 CCD data and some conclusions are drawn: In Qianyanzhou, the area with good vegetation coverage, the nonlinearity of NDVI has little influence on scaling-up of NDVI. As a result of surface heterogeneity, scaling-up can cause NDVI within each pixel to change. 11Jiedu Recipe,a compound Chinese herbal medicine,inhibits cancer stemness in hepatocellular carcinoma via Wnt/β-catenin pathway under hypoxia显示文摘Objective Jiedu Recipe(JR),a Chinese herbal remedy,has been shown to prolong overall survival time and decrease recurrence and metastasis rates in patients with hepatocellular carcinoma(HCC).This work investigated the mechanism of JR in HCC treatment.Methods The chemical constituents of JR were detected using liquid chromatography-mass spectrometry.The potential anti-HCC mechanism of JR was screened using network pharmacology and messenger ribonucleic acid(mRNA)microarray chip assay,followed by experimental validation in human HCC cells(SMMC-7721 and Huh7)in vitro and a nude mouse subcutaneous transplantation model of HCC in vivo.HCC cell characteristics of proliferation,migration and invasion under hypoxic setting were investigated using thiazolyl blue tetrazolium bromide,wound healing and Transwell assays,respectively.Image-i^(TM)Hypoxia Reagent was added to reveal hypoxic conditions.Stem cell sphere formation assay was used to detect the stemness.Epithelial-mesenchymal transition(EMT)markers like E-cadherin,vimentin andα-smooth muscle actin,and pluripotent transcription factors including nanog homeobox,octamer-binding transcription factor 4,and sex-determining region Y box protein 2 were analyzed using Western blotting and real-time polymerase chain reaction.Western blot was performed to ascertain the anti-HCC effect of JR under hypoxia involving the Wnt/β-catenin pathway.Results According to network pharmacology and mRNA microarray chip analysis,JR may potentially act on hypoxia and inhibit the Wnt/β-catenin pathway.In vitro and in vivo experiments showed that JR significantly decreased hypoxia,and suppressed HCC cell features of proliferation,migration and invasion;furthermore,the hypoxia-induced increases in EMT and stemness marker expression in HCC cells were inhibited by JR.Results based on the co-administration of JR and an agonist(LiCl)or inhibitor(IWR-1-endo)verified that JR suppressed HCC cancer stem-like properties under hypoxia by blocking the Wnt/β-catenin pathway.Conclusion JR exerts potent anti-HCC effects by inhibiting cancer stemness via abating the Wnt/β-catenin pathway under hypoxic conditions.Bing-jie Guo Yi Ruan Ya-jing Wang Chu-lan Xiao Zhi-peng Zhong Bin-bin Cheng Juan Du Bai Li Wei Gu Zi-fei Yin 2023Journal of Integrative Medicine2023,21,5:2

14Measurement of the neutron total cross section of carbon at the Back-n white neutron beam of CSNS显示文摘To verify the performance of the neutron total cross-sectional spectrometer, the neutron total cross section of carbon is initially measured in the energy range of 1 eV to 20 MeV using the time-of-flight method. The measurement is performed at the Back-n white neutron source with a 76-m time-of-flight path using the China Spallation Neutron Source. A multilayer fast fission chamber with 235U and 238U is employed as the neutron detector. The diameter and thickness of the natural graphite sample are 70 mm and 40 mm, respectively. Signal waveforms are collected using a data acquisition system. Off-line data processing was used to obtain the neutron time-of-flight spectra and transmissions. The uncertainty of the counting statistics is generally approximately 3% for each bin in the energy range of 1–20 MeV. It is determined that the results for the neutron total cross section of carbon obtained using ^235U cells are in good agreement with the results obtained using 238U cells within limits of statistical uncertainty. We achieve a dry-etched optical microresonator with an intrinsic Q factor as high as 1.94×10^(8) from a 1-mm-diameter silica microdisk with a thickness of 4 μm. 20Pre-transjugular-intrahepatic-portosystemic-shunt measurement of hepatic venous pressure gradient and its clinical application: A comparison study显示文摘BACKGROUND It is controversial whether transjugular intrahepatic portosystemic shunt(TIPS)placement can improve long-term survival.AIM To assess whether TIPS placement improves survival in patients with hepaticvenous-pressure-gradient(HVPG)≥16 mmHg,based on HVPG-related risk stratification.METHODS Consecutive variceal bleeding patients treated with endoscopic therapy+nonselectiveβ-blockers(NSBBs)or covered TIPS placement were retrospectively enrolled between January 2013 and December 2019.HVPG measurements were performed before therapy.The primary outcome was transplant-free survival;secondary endpoints were rebleeding and overt hepatic ence-phalopathy(OHE).RESULTS A total of 184 patients were analyzed(mean age,55.27 years±13.86,107 males;102 in the EVL+NSBB group,82 in the covered TIPS group).Based on the HVPG guided risk stratification,70 patients had HVPG<16 mmHg,and 114 patients had HVPG≥16 mmHg.The median follow-up time of the cohort was 49.5 mo.There was no significant difference in transplant-free survival between the two treatment groups overall(hazard ratio[HR],0.61;95%confidence interval[CI]:0.35-1.05;P=0.07).In the high-HVPG tier,transplant-free survival was higher in the TIPS group(HR,0.44;95%CI:0.23-0.85;P=0.004).In the low-HVPG tier,transplantfree survival after the two treatments was similar(HR,0.86;95%CI:0.33-0.23;P=0.74).Covered TIPS placement decreased the rate of rebleeding independent of the HVPG tier(P<0.001).The difference in OHE between the two groups was not statistically significant(P=0.09;P=0.48).CONCLUSION TIPS placement can effectively improve transplant-free survival when the HVPG is greater than 16 mmHg.Xi-Xuan Wang Xiao-Chun Yin Li-Hong Gu Hui-Wen Guo Yang Cheng Yan Liu Jiang-Qiang Xiao Yi Wang Wei Zhang Xiao-Ping Zou Lei Wang Ming Zhang Yu-Zheng Zhu-Ge Feng Zhang 2023World Journal of Gastroenterology2023,29,22:0
