1MicroRNA-125b Induces Cancer Cell Apoptosis Through Suppression of Bcl-2 Expression显示文摘MicroRNAs(miRNAs) are small,noncoding RNAs which can often act as an oncogene or a tumor suppressor.Several miRNAs are associated with the development of hepatocellular carcinoma(HCC).We demonstrated that miR-125b significantly suppresses HCC cell proliferation and promotes apoptosis by inhibiting the gene expression of the anti-apoptotic protein,Bcl-2.Bioinformatic analysis indicated that the 3'UTR of Bcl-2 has binding sites for miR-125b.Luciferase reporter assay confirmed the ability of miR-125b to dramatically suppress Bcl-2 transcription,suggesting that Bcl-2 is a target gene for miR-125b.We concluded that miR-125b acts as a tumor suppressor in hepatic tumor development by targeting Bcl-2 and inducing cancer cell apoptosis.Aihua Zhao Quan Zeng Xiaoyan Xie Junnian Zhou Wen Yue Yali Li Xuetao Pei 2012Journal of Genetics and Genomics2012,39,1:30

3Electroacupuncture preconditioning protects against focal cerebral ischemia/reperfusion injury via suppression of dynamin-related protein 1显示文摘Electroacupuncture preconditioning at acupoint Baihui(GV20) can reduce focal cerebral ischemia/reperfusion injury. However, the precise protective mechanism remains unknown. Mitochondrial fission mediated by dynamin-related protein 1(Drp1) can trigger neuronal apoptosis following cerebral ischemia/reperfusion injury. Herein, we examined the hypothesis that electroacupuncture pretreatment can regulate Drp1, and thus inhibit mitochondrial fission to provide cerebral protection. Rat models of focal cerebral ischemia/reperfusion injury were established by middle cerebral artery occlusion at 24 hours after 5 consecutive days of preconditioning with electroacupuncture at GV20(depth 2 mm, intensity 1 m A, frequency 2/15 Hz, for 30 minutes, once a day). Neurological function was assessed using the Longa neurological deficit score. Pathological changes in the ischemic penumbra on the injury side were assessed by hematoxylin-eosin staining. Cellular apoptosis in the ischemic penumbra on the injury side was assessed by terminal deoxyribonucleotidyl transferase-mediated d UTP-digoxigenin nick end labeling staining. Mitochondrial ultrastructure in the ischemic penumbra on the injury side was assessed by transmission electron microscopy. Drp1 and cytochrome c expression in the ischemic penumbra on the injury side were assessed by western blot assay. Results showed that electroacupuncture preconditioning decreased expression of total and mitochondrial Drp1, decreased expression of total and cytosolic cytochrome c, maintained mitochondrial morphology and reduced the proportion of apoptotic cells in the ischemic penumbra on the injury side, with associated improvements in neurological function. 6Allergy-related Evidences in Relation to Serum IgE:Data from the China State Key Laboratory of Respiratory Disease,2008-2013显示文摘Objective To investigate the serum total IgE(tlgE) and specific IgE(slgE) to common allergens among allergic patients in Guangzhou,China.Methods 7 085 patients were examined for tlgE and slgE to 15 allergens,based on the protocols of reversed enzyme allergosorbent test and the sandwich enzyme-linked immunosorbent assay.Results 3 758(53.04%) patients tested positive for tlgE,and 4 640(65.49%) for slgE.Der pteronyssinus,Der farinae,eggs,and cow's milk were the most common allergens leading to higher positive rates of slgE responses.Several peaks of sensitization were:Der pteronyssinus,Der farinae,and Blomia tropicalis at age 10-12;cow's milk at age below 3;eggs at age 4-6.The mean level and positive rate of tlgE tended to increase in subjects sensitized to more allergens.Sensitization to Der pteronyssinus(OR,1.6;P<0.05),Der farinae(OR,1.5;P<0.05),Blomia tropicalis(OR,1.4;P<0.05),Blattella germanica(OR,1.5;P<0.05),cow's milk(OR,1.3;P<0.05),and soy beans(OR,2.0;P<0.05) were independently correlated with allergy-related conditions in preliminary diagnosis.Conclusion The major allergens in Guangzhou include Der pteronyssinus,Der farinae,cow's milk,and eggs.Sensitization to these allergens appears to be predictors of allergy-related disorder.SUN Bao Qing CHEN De Hui ZHENG Pei Yan HUANG Hui Min LUO Wen Ting ZENG Guang Qiao ZHANG Xiao Wen 2014Biomedical and Environmental Sciences2014,27,7:11

8Hierarchical Magnetic Network Constructed by CoFe Nanoparticles Suspended Within “Tubes on Rods” Matrix Toward Enhanced Microwave Absorption显示文摘Hierarchical magnetic-dielectric composites are promising functional materials with prospective applications in microwave absorption(MA)field.Herein,a three-dimension hierarchical“nanotubes on microrods,”core–shell magnetic metal–carbon composite is rationally constructed for the first time via a fast metal–organic frameworksbased ligand exchange strategy followed by a carbonization treatment with melamine.Abundant magnetic CoFe nanoparticles are embedded within one-dimensional graphitized carbon/carbon nanotubes supported on micro-scale Mo2N rod(Mo2N@CoFe@C/CNT),constructing a special multi-dimension hierarchical MA material.Ligand exchange reaction is found to determine the formation of hierarchical magnetic-dielectric composite,which is assembled by dielectric Mo2N as core and spatially dispersed CoFe nanoparticles within C/CNTs as shell.Mo2N@CoFe@C/CNT composites exhibit superior MA performance with maximum reflection loss of−53.5 dB at 2 mm thickness and show a broad effective absorption bandwidth of 5.0 GHz.The Mo2N@CoFe@C/CNT composites hold the following advantages:(1)hierarchical core–shell structure offers plentiful of heterojunction interfaces and triggers interfacial polarization,(2)unique electronic migration/hop paths in the graphitized C/CNTs and Mo2N rod facilitate conductive loss,(3)highly dispersed magnetic CoFe nanoparticles within“tubes on rods”matrix build multi-scale magnetic coupling network and reinforce magnetic response capability,confirmed by the off-axis electron holography.Chunyang Xu Lei Wang Xiao Li Xiang Qian Zhengchen Wu Wenbin You Ke Pei Gang Qin Qingwen Zeng Ziqi Yang Chen Jin Renchao Che 2021Nano-Micro Letters2021,13,3:10

10Expression of miRNA-140 in Chondrocytes and Synovial Fluid of Knee Joints in Patients with Osteoarthritis显示文摘Objective To investigate the expression of miRNA-140 in chondrocytes and synovial fluid of osteoarthritis(OA) patients, and explore the relationship between the miRNA-140 expression and OA severity. Methods This study enrolled 30 OA patients who underwent total knee arthroplasty for chondrocytes sampling and 30 OA patients who underwent intra-articular injection for synovial fluid sampling. All OA patients were grouped into mild [Kellgren and Lawrence(KL) grade 1-2], moderate(KL grade 3) and severe(KL grade 4), with 10 in each subgroups for each sampling purposes. 7 non-OA patients and 10 patients with knee injury were collected for cartilage and synovial fluid sampling respectively as control groups. Chondrocytes were isolated from the cartilage tissue and cultured in vitro. Quantitative real time PCR for miRNA-140 in chondrocytes and synovial fluid were performed, and the U6 sn RNA was used as internal control. The expression difference of miRNA-140 among groups and correlation between the expression and the KL grade of OA were analysed using one-way ANOVA and Spearman test respectively. Results The expression of miRNA-140 in chondrocytes of knees in OA patients was reduced than that in normal knees, and the between-group difference was statistically significant(F=305.464, P<0.001). miRNA-140 could be detected in synovial fluid of both normal knees and OA knees, its relative expression level was reduced in synovial fluid of OA group compared with normal group, and the between-group difference was statistically significant as well(F=314.245, P<0.001). The relative expression level of miRNA-140 in both chondrocytes and synovial fluid were negatively correlated with the KL grade of OA(r=-0.969, P<0.001; r=-0.970, P<0.001). 11Manipulation of Band Degeneracy and Lattice Strain for Extraordinary PbTe Thermoelectrics显示文摘Maximizing band degeneracy and minimizing phonon relaxation time are proven to be successful for advancing thermoelectrics.Alloying with monotellurides has been known to be an effective approach for converging the valence bands of PbTe for electronic improvements,while the lattice thermal conductivity of the materials remains available room for being further reduced.It is recently revealed that the broadening of phonon dispersion measures the strength of phonon scattering,and lattice dislocations are particularly effective sources for such broadening through lattice strain fluctuations.In this work,a fine control of MnTe and EuTe alloying enables a significant increase in density of electron states near the valence band edge of PbTe due to involvement of multiple transporting bands,while the creation of dense in-grain dislocations leads to an effective broadening in phonon dispersion for reduced phonon lifetime due to the large strain fluctuations of dislocations as confirmed by synchrotron X-ray diffraction.The synergy of both electronic and thermal improvements successfully leads the average thermoelectric figure of merit to be higher than that ever reported for p-type PbTe at working temperatures.Yixuan Wu Pengfei Nan Zhiwei Chen Zezhu Zeng Siqi Lin Xinyue Zhang Hongliang Dong Zhiqiang Chen Hongkai Gu Wen Li Yue Chen Binghui Ge Yanzhong Pei 2020Research2020,,1:6

13Hypoxia inducible factor-1alpha mediates protection of DL-3-n-butylphthalide in brain microvascular endothelial cells against oxygen glucose deprivation-induced injury显示文摘Studies have demonstrated that DL-3-n-butylphthalide can significantly alleviate oxygen glucose deprivation-induced injury of human umbilical vein endothelial cells at least partly associated with its enhancement on oxygen glucose deprivation-induced hypoxia inducible factor-1α expression.In this study,we hypothesized that DL-3-n-butylphthalide can protect against oxygen glucose deprivation-induced injury of newborn rat brain microvascular endothelial cells by means of upregulating hypoxia inducible factor-1α expression.MTT assay and Hoechst staining results showed that DL-3-n-butylphthalide protected brain microvascular endothelial cells against oxygen glucose deprivation-induced injury in a dose-dependent manner.Western blot and immunofluorescent staining results further confirmed that the protective effect was related to upregulation of hypoxia inducible factor-1α.Real-time RT-PCR reaction results showed that DL-3-n-butylphthalide reduced apoptosis by inhibiting downregulation of pro-apoptotic gene caspase-3 mRNA expression and upregulation of apoptosis-executive protease bcl-2 mRNA expression;however,DL-3-n-butylphthalide had no protective effects on brain microvascular endothelial cells after knockdown of hypoxia inducible factor-1α by small interfering RNA.These findings suggest that DL-3-n-butylphthalide can protect brain microvascular endothelial cells against oxygen glucose deprivation-induced injury by upregulating bcl-2 expression and downregulating caspase-3 expression though hypoxia inducible factor-1α pathway.Weihong Yang Ling Li Ruxun Huang Zhong Pei Songjie Liao Jinsheng Zeng 2012Neural Regeneration Research2012,7,12:6
14Analysis of CD8^+CD28^-T-suppressor cells in living donor liver transplant recipients显示文摘BACKGROUND:Human CD8 + CD28 - T-suppressor(Ts) cells have been considered to indicate a reduced need for immunosuppression in pediatric liver-intestine transplant recipients and recipients of deceased heart-kidney transplants.However,in adult-to-adult living donor liver transplantation(A-A LDLT)little information is available and the clinical significance is still unknown. METHODS:Flow cytometry was used to detect the population of CD8+CD28 -Ts cells present in peripheral blood in A-A LDLT recipients(n=31),patients with end- stage liver disease(n=24)and healthy controls(n=19). Meanwhile,we tested the graft function and trough levels of immunosuppression in recipients.The clinical and follow- up data of 31 transplant recipients were analyzed. RESULTS:Compared with diseased controls(P=0.007) and healthy individuals(P=0.000),a notable expansion of CD8 + CD28 - Ts cells was found in recipients of A-A LDLT.This was associated with graft function,levels of immunosuppression and rejection episodes. CONCLUSIONS:To monitor the CD8 + CD28 - Ts cells levels is important to evaluate the immune state of recipients. 15Divergent Projection Patterns Revealed by Reconstruction of Individual Neurons in Orbitofrontal Cortex显示文摘The orbitofrontal cortex(OFC)is involved in diverse brain functions via its extensive projections to multiple target regions.There is a growing understanding of the overall outputs of the OFC at the population level,but reports of the projection patterns of individual OFC neurons across different cortical layers remain rare.Here,by combining neuronal sparse and bright labeling with a whole-brain florescence imaging system(fMOST),we obtained an uninterrupted three-dimensional whole-brain dataset and achieved the full morphological reconstruction of 25 OFC pyramidal neurons.We compared the wholebrain projection targets of these individual OFC neurons in different cortical layers as well as in the same cortical layer.We found cortical layer-dependent projections characterized by divergent patterns for information delivery.Our study not only provides a structural basis for understanding the principles of laminar organizations in the OFC,but also provides clues for future functional and behavioral studies on OFC pyramidal neurons.Junjun Wang Pei Sun Xiaohua Lv Sen Jin Anan Li Jianxia Kuang Ning Li Yadong Gang Rui Guo Shaoqun Zeng Fuqiang Xu Yu-Hui Zhang 2021Neuroscience Bulletin2021,37,4:5
16Chromatographic fingerprinting and quantitative analysis for the quality evaluation of Xinkeshu tablet显示文摘A simple, sensitive and accurate method based on high performance liquid chromatography (HPLC) with diode array detector (DAD) was developed and validated for systematic quality evaluation of one type of traditional Chinese medicine preparations named Xinkeshu (XKS) tablet. In this study, the chromatographic fingerprints of XKS tablet were developed first, 23 peaks were selected as the common peaks to evaluate the similarities among different batches of XKS samples, which were manufactured in a long time span of three years. Additionally, simultaneous quantification of six markers in XKS tablet, including Danshensu, Protocatechuic aldehyde, Puerarin, Daidzin, Salvianolic acid B and Daidzein, was performed. The validation results showed that the developed method was specific, accurate, precise and robust. The preliminary explanation on why a close similarity between fingerprints did not exactly mean similar contents of chemical components in samples was given. The contribution of each chromatographic peak to similarity was also evaluated. 17Sub-10 nm fabrication:methods and applications显示文摘Reliable fabrication of micro/nanostructures with sub-10 nm features is of great significance for advancing nanoscience and nanotechnology.While the capability of current complementary metal-oxide semiconductor(CMOS)chip manufacturing can produce structures on the sub-10 nm scale,many emerging applications,such as nano-optics,biosensing,and quantum devices,also require ultrasmall features down to single digital nanometers.In these emerging applications,CMOS-based manufacturing methods are currently not feasible or appropriate due to the considerations of usage cost,material compatibility,and exotic features.Therefore,several specific methods have been developed in the past decades for different applications.In this review,we attempt to give a systematic summary on sub-10 nm fabrication methods and their related applications.In the first and second parts,we give a brief introduction of the background of this research topic and explain why sub-10 nm fabrication is interesting from both scientific and technological perspectives.In the third part,we comprehensively summarize the fabrication methods and classify them into three main approaches,including lithographic,mechanics-enabled,and post-trimming processes.The fourth part discusses the applications of these processes in quantum devices,nano-optics,and high-performance sensing.Finally,a perspective is given to discuss the challenges and opportunities associated with this research topic.Yiqin Chen Zhiwen Shu Shi Zhang Pei Zeng Huikang Liang Mengjie Zheng Huigao Duan 2021International Journal of Extreme Manufacturing2021,3,3:4
18Highly sensitive solar-blind deep ultraviolet photodetector based on graphene/PtSe_(2)/β-Ga_(2)O_(3)2D/3D Schottky junction with ultrafast speed显示文摘There is an emerging need for high-sensitivity solar-blind deep ultraviolet(DUV)photodetectors with an ultra-fast response speed.Although nanoscale devices based on Ga_(2)O_(3)nanostructures have been developed,their practical applications are greatly limited by their slow response speed as well as low specific detectivity.Here,the successful fabrication of two-/three-dimensional(2D/3D)graphene(Gr)/PtSe2/β-Ga_(2)O_(3)Schottky junction devices for high-sensitivity solar-blind DUV photodetectors is demonstrated.Benefitting from the high-quality 2D/3D Schottky junction,the vertically stacked structure,and the superior-quality transparent graphene electrode for effective carrier collection,the photodetector is highly sensitive to DUV light illumination and achieves a high responsivity of 76.2 mA/W,a large on/off current ratio of~105,along with an ultra-high ultraviolet(UV)/visible rejection ratio of 1.8×104.More importantly,it has an ultra-fast response time of 12µs and a remarkable specific detectivity of~1013 Jones.Finally,an excellent DUV imaging capability has been identified based on the Gr/PtSe2/β-Ga_(2)O_(3)Schottky junction photodetector,demonstrating its great potential application in DUV imaging systems.Di Wu Zhihui Zhao Wei Lu Lukas Rogée Longhui Zeng Pei Lin Zhifeng Shi Yongtao Tian Xinjian Li Yuen Hong Tsang 2021Nano Research2021,14,6:4
