| 1 | Analysis of DNA methylation variation in wheat genetic background after alien chromatin introduction based on methylation-sensitive amplification polymorphism显示文摘During the process of alien germplasm introduced into wheat genome by chromosome engineering, extensive genetic variations of genome structure and gene expression in recipient could be induced. In this study, we performed GISH (genome in situ hybridization) and AFLP (amplified fragment length po-lymorphism) on wheat-rye chromosome translocation lines and their parents to detect the identity in genomic structure of different translocation lines. The results showed that the genome primary struc-ture variations were not obviously detected in different translocation lines except the same 1RS chromosome translocation. Methylation sensitive amplification polymorphism (MSAP) analyses on genomic DNA showed that the ratios of fully-methylated sites were significantly increased in translo-cation lines (CN12, 20.15%; CN17, 20.91%; CN18, 22.42%), but the ratios of hemimethylated sites were significantly lowered (CN12, 21.41%; CN17, 23.43%; CN18, 22.42%), whereas 16.37% were fully-me-thylated and 25.44% were hemimethylated in case of their wheat parent. Twenty-nine classes of me-thylation patterns were identified in a comparative assay of cytosine methylation patterns between wheat-rye translocation lines and their wheat parent, including 13 hypermethylation patterns (33.74%), 9 demethylation patterns (22.76%) and 7 uncertain patterns (4.07%). | 2 | Fabrication and Thermal Structural Characteristics of Ultra-high Temperature Ceramic Struts in Scramjets显示文摘ZrB_2-SiC based ultra-high temperature ceramic(UHTC) struts were firstly proposed and fabricated with the potential application in the combustor of scramjets for fuel injection and flame-holding for their machinability and excellent oxidation/ablation resistance in the extreme harsh environment. The struts were machined with electrospark wire-electrode cutting techniques to form UHTC into the desired shape, and with laser drilling to drill tiny holes providing the channels for fuel injection. The integrated thermal-structural characteristic of the struts was evaluated in high-temperature combustion environment by the propane-oxygen free jet facility, subject to the heat flux of 1.5 MW/m^2 lasting for 300 seconds, and the struts maintained integrity during and after the first experiment. The experiments were repeated for verifying the reusability of the struts. Fracture occurred during the second repeated experiment with the crack propagating through the hole. Finite element analysis(FEA) was carried out to study the thermal stress distribution in the UHTC strut. The simulation results show a high thermal stress concentration occurs at the hole which is the crack initiation position. The phenomenon is in good agreement with the experimental results. | 3 | Comparative FISH and molecular identification of new stripe rust resistant wheat-Thinopyrum intermedium ssp. trichophorum introgression lines显示文摘Thinopyrum intermedium(2 n = 6 x = 42, JJJ^sJ^sStSt) has been hybridized extensively with common wheat and has proven to be a valuable germplasm source for improving disease resistance, quality attributes, and yield potential in wheat. We characterized new disease resistant wheat-Th. intermedium derivatives A1082 and A5-5 using sequential multi-color fluorescence in situ hybridization(mc-FISH), genomic in situ hybridization(GISH), PCRbased landmark unique gene(PLUG) and intron targeting(IT) markers. A1082 was identified as a wheat-Th. intermedium 3J disomic addition line, and A5-5 was a T4BS·5J^sL homozygous Robertsonian translocation line. Seventy-one and 106 pairs of primers amplified Th.intermedium-specific bands allowing chromosomes 3J and 5J^s to be tracked, respectively. A new oligonucleotide probe, Oligo-6 H-2-100, was developed for FISH labeling of the subterminal region of the long arm of chromosome 5J^s. Both lines were highly resistant to stripe rust pathogen races prevalent in Chinese field screening nurseries. A5-5 also displayed a significant increase in tiller number compared to its wheat parent. | 4 | Maternal genetic history of ancient Tibetans over the past 4000 years显示文摘The settlement of the Tibetan Plateau epitomizes human adaptation to a high-altitude environment that poses great challenges to human activity.Here,we reconstruct a 4000-year maternal genetic history of Tibetans using 128 ancient mitochondrial genome data from 37 sites in Tibet.The phylogeny of haplotypes M9a1a,M9a1b,D4g2,G2a’c,and D4i show that ancient Tibetans share the most recent common ancestor with ancient Middle and Upper Yellow River populations around the Early and Middle Holocene.In addition,the connections between Tibetans and Northeastern Asians vary over the past 4000 years,with a stronger matrilineal connection between the two during 4000 BPe3000 BP,and a weakened connection after 3000 BP,that are coincident with climate change,followed by a reinforced connection after the Tubo period(1400 BPe1100 BP).Besides,an over 4000-year matrilineal continuity is observed in some of the maternal lineages.We also find the maternal genetic structure of ancient Tibetans is correlated to the geography and interactions between ancient Tibetans and ancient Nepal and Pakistan populations.Overall,the maternal genetic history of Tibetans can be characterized as a long-term matrilineal continuity with frequent internal and external population interactions that are dynamically shaped by geography,climate changes,as well as historical events. | Ganyu Zhang Can Cui Shargan Wangdue Hongliang Lu Honghai Chen Lin Xi Wei He Haibing Yuan Tinley Tsring Zujun Chen Feng Yang Tashi Tsering Shuai Li Norbu Tashi Tsho Yang Yan Tong Xiaohong Wu Linhui Li Yuanhong He Peng Cao Qingyan Dai Feng Liu Xiaotian Feng Tianyi Wang Ruowei Yang Wanjing Ping Ming Zhang Xing Gao Yichen Liu Wenjun Wang Qiaomei Fu | 2023 | Journal of Genetics and Genomics2023,50,10: | 1 |
