| 1 | Screening and breeding of high taxol producing fungi by genome shuffling显示文摘To apply the fundamental principles of genome shuffling in breeding of taxol-producing fungi, Nodulisporium sylviform was used as starting strain in this work. The procedures of protoplast fusion and genome shuffling were studied. Three hereditarily stable strains with high taxol production were obtained by four cycles of genome shuffling. The qualitative and quantitative analysis of taxol produced was confirmed using thin-layer chromatography (TLC), high performance liquid chromatography (HPLC) and LC-MS. A high taxol producing fungus, Nodulisporium sylviform F4-26, was obtained, which produced 516.37 μg/L taxol. | 2 | Apelin and vascular endothelial growth factor are associated with mobilization of endothelial progenitor cells after acute myocardial infarction显示文摘This study was designed to determine the levels of early endothelial progenitor cells(EPCs),apelin,vascu-lar endothelial growth factor(VEGF) and stromal cell-derived growth factor-1(SDF-1) after acute myocardial infarction(AMI),and to investigate the relationships between these cytokines and early EPCs.Early EPCs,de-fined as CD133+,KDR+,and CD34+ cells,were quantified by flow cytometry.The levels of early EPCs and those cytokines in AMI patients were significantly different from those with coronary artery disease or controls(P < 0.05).Plasma apelin levels were inversely correlated with Gensini score and early EPCs(both P < 0.01).Early EPCs,VEGF and SDF-1 showed different patterns of changes in AMI patients during the first 24 h.The trend in the change of early EPCs was proportionally correlated with that of VEGF(P < 0.05).AMI patients exhibited in-creased early EPCs with remarkably decreased apelin levels and enhanced VEGF levels. | Jiaxin Ye Ping Ni Lina Kang Biao Xu | 2012 | The Journal of Biomedical Research2012,26,6: | 16 |
| 3 | Electrochemical performance of MCMB/(AC+LiFePO_4) lithium-ion capacitors显示文摘Lithium-ion capacitors(LICs) were fabricated using mesocarbon microbeads(MCMB) as a negative electrode and a mixture of activated carbon(AC) and LiFePO4 as a positive electrode(abbreviated as LAC).The phase structure and morphology of LAC samples were characterized by X-ray diffraction(XRD) and field emission scanning electron microscopy(FESEM).The electrochemical performance of the LICs was studied using cyclic voltammetry,charge-discharge rate measurements,and cycle performance testing.A LIC with 30 wt% LiFePO4 was found to have the best electrochemical performance with a specific energy density of 69.02 W h kg-1 remaining at 4 C rate after 100 cycles.Compared with an AC-only positive electrode system,the ratio of practical capacity to theoretical calculated capacity of the LICs was enhanced from 42.22% to 56.59%.It was proved that adding LiFePO4 to AC electrodes not only increased the capacity of the positive electrode,but also improved the electrochemical performances of the whole LICs via Li+ pre-doping. | PING LiNa ZHENG JiaMing SHI ZhiQiang QI Jie WANG ChengYang | 2013 | Chinese Science Bulletin2013,58,6: | 12 |

| 6 | Short-term link quality prediction using nonparametric time series analysis显示文摘Wireless link quality prediction(LQP) is the foundation for proactive operations and is therefore a key technique in alleviating network performance degradation. However, accurate LQP is difficult because of the dynamic nature of wireless environments. A recent study found that fluctuations in intermediate quality links often show dynamics on a sub-second granularity, making the task even more challenging. In order to leverage the intermediate links, as well as fine-tune upper-layer protocols, we propose to use nonparametric modeling in nonlinear time series analysis that predicts short-term link quality online. Unlike existing studies,we do not define any new experimental or hypothetical models, or train models using a set of training data.Functional-coefficient autoregression is employed to predict the link dynamics at high time resolutions. We apply our approach and a local linear regression-based LQP(a typical parametric modeling approach) to both NS-2 simulation and empirical packet traces. | 8 | Effects of “Bioactive” amino acids leucine, glutamate, arginine and tryptophan on feed intake and mRNA expression of relative neuropeptides in broiler chicks显示文摘Feed intake control is vital to ensuring optimal nutrition and achieving full potential for growth and development in poultry. The aim of the present study was to investigate the effects of L-leucine, L-glutamate, L-tryptophan and L-arginine on feed intake and the mRNA expression levels of hypothalamic Neuropeptide involved in feed intake regulation in broiler chicks. Leucine, glutamate, tryptophan or arginine was intra-cerebroventricularly (ICV) administrated to 4d-old broiler chicks respectively and the feed intake were recorded at various time points. Quantitative PCR was performed to determine the hypothalamic mRNA expression levels of Neuropeptide Y (NPY), agouti related protein (AgRP), pro-opiomelanocortin (POMC), melanocortin receptor 4 (MC4R) and corticotrophin releasing factor (CRF). Our results showed that ICV administration of L-leucine (0.15 or 1.5 μmol) significantly (P<0.05) increased feed intake up to 2 h post-administration period and elevated both hypothalamic NPY and AgRP mRNA expression levels. In contrast, ICV administration of L-glutamate (1.6μmol) significantly (P<0.05) decreased feed intake 0.25, 0.5 and 2 h post-injection, and increased hypothalamic CRF and MC4R mRNA expression levels. Meanwhile, both L-tryptophan (10 or 100μg) and L-arginine (20 or 200μg) had no significant effect on feed intake. | 9 | Effect of diacylglycerol acyltransferase 2overexpression in 3T3-L1 is associated to an increase in mono-unsaturated fatty acid accumulation显示文摘Background:Fatty acid(FA) composition is the most important parameter affecting the flavor and nutritional value of the meat.The final and the only committed step in the biosynthesis of triglycerides is catalyzed by diacylglycerol acyltransferase 2(DGAT2).The role of DGAT2 in lipid accumulation has been demonstrated in adipocytes,However,little is known about the effect of DGAT2 on the FA composition of these cells.Methods:To investigate the role of DGAT2 in regulating lipid accumulation,FA composition and the expression of adipogenic genes,we cloned the open reading frame of the porcine DGAT2 gene and established 3T3-L1 cells that overexpressed DGAT2.Cells were then cultured in differentiation medium(DM) without FA,with a mixture of FAs(FA-DM),or containing a ^(13)C stable isotope-labeled FA mixture(IFA-DM).The FA composition of adipocytes was analyzed by gas chromatography-mass spectrometry and gas chromatography-isotope ratio mass spectrometry.Quantitative PCR and western blotting were employed to detect expression of adipogenic genes in 3T3-L1 adipocytes cultured with FA-DM for 12 d.Results:The triacylglyceride(TAG) content was significantly higher in 3T3-L1 adipocytes overexpressing DGAT2 than in control cells.When cultured in DM or FA-DM for 12 d,cells overexpressing DGAT2 showed a higher proportion of unsaturated FAs(C16:1 and C18:1).However,when cells overexpressing DGAT2 were cultured with FA-DM for30 min,the FA composition was almost identical to that of controls.Further,the proportion of stable isotope-labeled FAs were similar in 3T3-L1 adipocytes overexpressing DGAT2 and control cells cultured in IFA-DM for 12 d.These results collectively indicate that the higher proportion of mono-unsaturated FAs,C16:1 and C18:1,may originate from de novo FA synthesis but not from the uptake of specific FAs from the medium.This hypothesis is further supported by evidence that both mRNA and protein expression of genes involved in FA synthesis(ACACA,FASN,SCD1,and A-FABP)were significantly higher in cells overexpressing DGAT2 than in control cells.Conclusions:In conclusion,our study revealed that TAG accumulation,the proportion of MUFAs,and the expression of adipogenic genes were higher in 3T3-L1 cells overexpressing DGAT2 than in control cells. | Zhiqi Zhang Gang Shu Xiaotong Zhu Junming Guo Han Cai Songbo Wang Lina Wang Ping Gao Qianyun Xi Yongliang Zhang Li Yuan Qingyan Jiang | 2014 | Journal of Animal Science and Biotechnology2014,5,4: | 3 |
| 10 | Colonization of endophyte Acremonium sp. D212 in Panax notoginseng and rice mediated by auxin and jasmonic acid显示文摘Endophytic fungi can be beneficial to plant growth. However, the molecular mechanisms under-lying colonization of Acremonium spp. remain unclear. In this study, a novel endophytic Acremonium strain was isolated from the buds of Panax notoginseng and named Acremonium sp. D212. The Acremonium sp. D212 could colonize the roots of P. notoginseng, enhance the resistance of P. notoginseng to root rot disease, and promote root growth and saponin bio-synthesis in P. notoginseng. Acremonium sp. D212 could secrete indole-3-acetic acid (IAA) and jasmonic acid (JA), and inoculation with the fungus increased the endogenous levels of IAA and JA in P. noto-ginseng. Colonization of the Acremonium sp. D212 in the roots of the rice line Nipponbare was dependent on the concentration of methyl jasmonate (MeJA) (2–15μmol/L) and 1-naphthalenacetic acid (NAA) (10–20μmol/L). Moreover, the roots of the JA signaling-defective coi1-18 mutant were colonized by Acremonium sp. D212 to a lesser degree than those of the wild-type Nipponbare and miR393b-overexpressing lines, and the colonization was res-cued by MeJA but not by NAA. It suggests that the cross-talk between JA signaling and the auxin biosynthetic pathway plays a crucial role in the colonization of Acremonium sp. | 12 | Sex-specific association between coffee consumption and incident chronic kidney disease: a population-based analysis of 359,906 participants from the UK Biobank显示文摘Background:The risk for chronic kidney disease(CKD)is influenced by genetic predisposition,sex,and lifestyle.Previous research indicates that coffee is a potentially protective factor in CKD.The current study aims to investigate whether sex disparity exists in the coffee–CKD association,and whether genetic risk of CKD or genetic polymorphisms of caffeine metabolism affect this association.Methods:A total of 359,906 participants from the UK Biobank who were enrolled between 2006 and 2010 were included in this prospective cohort study,which aimed to estimate the hazard ratios for coffee intake and incident CKD using a Cox proportional hazard model.Allele scores of CKD and caffeine metabolism were additionally adjusted for in a subsample with qualified genetic data(n=255,343).Analyses stratified by genetic predisposition,comorbidities,and sex hormones were performed.Tests based on Bayesian model averaging were conducted to ascertain the robustness of the results.Results:Coffee was inversely associated with CKD in a dose-dependent manner.The effects of coffee did not differ across different strata of genetic risk for CKD,but were more evident among slower genetically predicted caffeine metabolizers.Significant sex disparity was observed(P value for interaction=0.013),in that coffee drinking was only associated with the risk reduction of CKD in females.Subgroup analysis revealed that testosterone and sex hormone-binding globulin(SHBG),but not estradiol,modified the coffee–CKD association.Conclusions:In addition to the overall inverse coffee–CKD association that was observed in the general population,we could also establish that a sex disparity existed,in that females were more likely to experience the benefit of the association.Testosterone and SHBG may partly account for the sex disparity. | Lei Tang Lina Yang Wenwen Chen Chunyang Li Yu Zeng Huazhen Yang Yao Hu Yuanyuan Qu Huan Song Xiaoxi Zeng Ping Fu | 2022 | Chinese Medical Journal2022,,12: | 2 |

| 15 | Interfacial Interaction Between Polyurethane/Poly(Methacrylateco-Styrene) Coating and the Regenerated Cellulose Film显示文摘Water-resistant films were prepared by coating the surface or regenerated cellulose films withcastor oil-based polyurethane (PU) / poly-(methacrylate-co-styrene) [P(MA-St)]. The erfects of the ratioof PU to P(MA-St) copolymer on tensile strength (dry and wet states), vapor permeability, size stability,and water resistivity of the coated films were studied. The interracial interaction between cellulose and thePU/P (MA-St) coating was analyzed using infrared (IR), ultraviolet (UV), scanning electron microscopy(SEM), transmission electron microscopy (TEM), differential thermal analysis (DTA), and electron probemicroanalysis (EPMA). The results indicated that the mechanical properties and water resistlvity of the coated rilms significantly enhanced, and the biodegradability was displayed, when the ratio or PU to P(MA-St)was 8: 2 by weight. The chemical bonds and hydrogen bonds between the cellulose, PU, and the copolymerexist in the coated films. | 17 | Tauroursodeoxycholic acid(TUDCA)improves intestinal barrier function associated with TGR5-MLCK pathway and the alteration of serum metabolites and gut bacteria in weaned piglets显示文摘Background:Tauroursodeoxycholic acid(TUDCA),a hydrophilic bile acid,is the main medicinal component of bear bile and is commonly used to treat a variety of hepatobiliary diseases.Meanwhile,TUDCA has been shown to modulate the intestinal barrier function and alleviate DSS-induced colitis in mice.However,the effect of TUDCA on the intestinal barrier of weaned piglets remains largely unclear.Methods:The weaned piglets and porcine IPEC-J2 intestinal epithelial cells were used to investigate the effects of TUDCA on intestinal barrier function in weaned piglets and explore the possible underlying mechanisms.In vivo,72 healthy weaned piglets were randomly allocated into 2 groups according to their gender and body weight,and piglets were fed the basal diet with 0(control,CON)and 200 mg/kg TUDCA for 30 d,respectively.Three female and three male piglets reflecting the average bodyweight were slaughtered in each group and samples were collected.In vitro,IPEC-J2 cells were subjected to 100μmol/L TUDCA to explore the possible underlying mechanisms.Results:Our results demonstrated that dietary TUDCA supplementation significantly reduced the diarrhea incidence of weaned piglets,possibly attributing to the TUDCA-enhanced intestinal barrier function and immunity.In addition,TUDCA supplementation altered serum metabolites and the relative abundance of certain gut bacteria,which might contribute to the improved intestinal barrier function.Furthermore,the in-vitro results showed that TUDCA improved the E.coli-induced epithelial barrier impairment of IPEC-J2 cells and increased Takeda G-coupled protein receptor 5(TGR5)protein expression.However,knockdown of TGR5 and inhibition of myosin light chain kinase(MLCK)pathway abolished the TUDCA-improved epithelial barrier impairment in E.coli-treated IPEC-J2 cells,indicating the involvement of TGR5-MLCK in this process.Conclusions:These findings showed that TUDCA improved intestinal barrier function associated with TGR5-MLCK pathway and the alteration of serum metabolites and gut bacteria in weaned piglets,suggesting the potential application of TUDCA in improving gut health in piglet production. | Min Song Fenglin Zhang Yiming Fu Xin Yi Shengchun Feng Zhichang Liu Dun Deng Qiang Yang Miao Yu Canjun Zhu Xiaotong Zhu Lina Wang Ping Gao Gang Shu Xianyong Ma Qingyan Jiang Songbo Wang | 2022 | Journal of Animal Science and Biotechnology2022,13,6: | 1 |
