4Recent advancements in metal-organic frameworks for green applications显示文摘A series of environmental and energy issues,such as global warming,water pollution,acid rain,and energy shortage,have to be settled urgently.Metal-organic frameworks(MOFs)are compounds consisting of metal ions or clusters coordinated to organic ligands,which show great promise for alleviating or mitigating these challenges owing to their outstanding physical and chemical properties.In this review,we summarize the recent advances of MOFs in the fields of green applications,including carbon capture,harmful gas removal,sewage treatment,and green energy storage.In addition,the challenges and prospects of the large-scale commercialized use of MOFs in handling environmental issues are also discussed.Chongxiong Duan Yi Yu Jing Xiao Yuanyuan Li Pengfei Yang Fei Hu Hongxia Xi 2021Green Energy & Environment2021,6,1:6
5CD146 as a promising therapeutic target for retinal and choroidal neovascularization diseases显示文摘Blood vessel dysfunction causes several retinal diseases,including diabetic retinopathy,familial exudative vitreoretinopathy,macular degeneration and choroidal neovascularization in pathological myopia.Vascular endothelial growth factor(VEGF)-neutralizing proteins provide benefits in most of those diseases,yet unsolved haemorrhage and frequent intraocular injections still bothered patients.Here,we identified endothelial CD146 as a new target for retinal diseases.CD146 expression was activated in two ocular pathological angiogenesis models,a laser-induced choroid neovascularization model and an oxygeninduced retinopathy model.The absence of CD146 impaired hypoxia-induced cell migration and angiogenesis both in cell lines and animal model.Preventive or therapeutic treatment with anti-CD146 antibody AA98 significantly inhibited hypoxia-induced aberrant retinal angiogenesis in two retinal disease models.Mechanistically,under hypoxia condition,CD146 was involved in the activation of NFκB,Erk and Akt signalling pathways,which are partially independent of VEGF.Consistently,anti-CD146therapy combined with anti-VEGF therapy showed enhanced impairment effect of hypoxia-induced angiogenesis in vitro and in vivo.Given the critical role of abnormal angiogenesis in retinal and choroidal diseases,our results provide novel insights into combinatorial therapy for neovascular fundus diseases.Bai Xue Ping Wang Wenzhen Yu Jing Feng Jie Li Rulian Zhao Zhenglin Yang Xiyun Yan Hongxia Duan 2022Science China(Life Sciences)2022,65,6:3
6Expression of nerve growth factor precursor, mature nerve growth factor and their receptors during cerebral ischemia-reperfusion injury显示文摘We investigated nerve growth factor precursor (proNGF) and mature NGF expression in ischemic and non-ischemic cortices after cerebral ischemia-reperfusion injury. In both ischemic and non-ischemic cortices, proNGF was found to be present in the extracellular space and cytoplasm. In addition, mature NGF was expressed in extracellular space, but with a very low signal. In ischemic cortex only, proNGF was significantly decreased, reaching a minimal level at 1 day. Mature NGF was increased at 4 hours, then reached a minimal level at 3 days. The p75 neurotrophin receptor (p75NTR) was significantly decreased after ischemia, and increased at 3 days after ischemia. These results confirmed that proNGF was the predominant form of NGF during the pathological process of cerebral ischemia-reperfusion injury. 7Establishment of a rhesus monkey model of middle cerebral artery ischemia and reperfusion using a microcatheter embolization method显示文摘Nonhuman primates are closest to humans in terms of lineage,and middle cerebral artery ischemia/reperfusion responses of nonhuman primates are most similar to ischemic stroke in humans.Therefore,nonhuman primates could be utilized to simulate the process of ischemic stroke in the human.Few studies,however,have reported the use of endovascular technology to establish a rhesus monkey stroke model.In the present study,seven adult,male,rhesus monkeys were selected and,following anesthesia,a microcatheter was inserted into one side of the middle cerebral artery via the femoral artery to block blood flow,thereby resulting in middle cerebral artery occlusion.After 2 hours,the microcatheter was withdrawn to restore the middle cerebral artery blood flow and to establish ischemia/reperfusion.Results from angiography and magnetic resonance angiography confirmed occlusion and reopening of the middle cerebral artery.Magnetic resonance imaging revealed the existence of ischemic brain lesions,and neurological examination showed sustained functional deficits following surgery.The rhesus monkey middle cerebral artery ischemia/reperfusion models established by microcatheter embolization had the advantage of non-craniotomy invasion and reproducibility.The scope and degree of ischemic damage using this model was controllable.Therefore,this nonhuman primate model is an ideal model for cerebral ischemia and reperfusion.Jie Yang Jian Guo Hongbo Zheng Muke Zhou Li He Wenying Cao Li Xiao Jiachuan Duan Qiyong Gong Xiaoqi Huang Hongxia Li Li Wang Jingqiu Cheng 2010Neural Regeneration Research2010,5,15:2

9Targeting the CD146/Galectin-9 axis protects the integrity of the blood-brain barrier in experimental cerebral malaria显示文摘Cerebral malaria(CM)is a life-threatening diffuse encephalopathy caused by Plasmodium falciparum,in which the destruction of the blood–brain barrier(BBB)is the main cause of death.However,increasing evidence has shown that antimalarial drugs,the current treatment for CM,do little to protect against CM-induced BBB damage.Therefore,a means to alleviate BBB dysfunction would be a promising adjuvant therapy for CM.The adhesion molecule CD146 has been reported to be expressed in both endothelial cells and proinflammatory immune cells and mediates neuroinflammation.Here,we demonstrate that CD146 expressed on BBB endothelial cells but not immune cells is a novel therapeutic target in a mouse model of experimental cerebral malaria(eCM).Endothelial CD146 is upregulated during eCM development and facilitates the sequestration of infected red blood cells(RBCs)and/or proinflammatory lymphocytes in CNS blood vessels,thereby promoting the disruption of BBB integrity.Mechanistic studies showed that the interaction of CD146 and Galectin-9 contributes to the aggregation of infected RBCs and lymphocytes.Deletion of endothelial CD146 or treatment with the anti-CD146 antibody AA98 prevents severe signs of eCM,such as limb paralysis,brain vascular leakage,and death.In addition,AA98 combined with the antiparasitic drug artemether improved the cognition and memory of mice with eCM.Taken together,our findings suggest that endothelial CD146 is a novel and promising target in combination with antiparasitic drugs for future CM therapies.Hongxia Duan Shuai Zhao Jianquan Xiang Chenhui Ju Xuehui Chen Irene Gramaglia Xiyun Yan 2021Cellular & Molecular Immunology2021,18,10:2
