3Isolation and Identification of Cancer Stem-Like Cells from Murine Melanoma Cell Lines显示文摘In current study,cancer stem-like cells in the murine melanoma B16F10 cells were investigated.CD phenotypes of the B16F10 cells were analyzed by flow cytometry,and the specific CD phenotype cells from the B16F10 cells were isolated by MACS.Then we used colony formation assay in soft agar media,the cell growth assay in serum-free culture media as well as the tumorigenicity investigation of the specific CD phenotype cells in C57BL/6 mice, respectively,to identify cancer stem-like cells in the B16F10 cells.The results showed that the B16F10 cells could form spherical clones in serum-free culture media,and the rate of clonegenesis of CD133 + ,CD44 + and CD44 + CD133 + cells was higher than that of CD133 - ,CD44 - and CD44 + CD133 - cells in soft agar media,respectively. 4Baicalin induces ferroptosis in bladder cancer cells by downregulating FTH1显示文摘Ferroptosis is a non-apoptotic regulated cell death caused by iron accumulation and subsequent lipid peroxidation.Currently,the therapeutic role of ferroptosis on cancer is gaining increasing interest.Baicalin an active component in Scutellaria baicalensis Georgi with anticancer potential various cancer types;however,the effects of baicalein on bladder cancer and the underlying molecular mechanisms remain largely unknown.In the study,we investigated the effect of baicalin on bladder cancer cells5637 and KU-19-19.As a result,we show baicalin exerted its anticancer activity by inducing apoptosis and cell death in bladder cancer cells.Subsequently,we for the first time demonstrate baicalin-induced ferroptotic cell death in vitro and in vivo,accompanied by reactive oxygen species(ROS) accumulation and intracellular chelate iron enrichment.The ferroptosis inhibitor deferoxamine but not necrostatin-1,chloroquine(CQ),N-acetyl-L-cysteine,L-glutathione reduced,or carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone(Z-VAD-FMK) rescued baicalin-induced cell death,indicating ferroptosis contributed to baicalin-induced cell death.Mechanistically,we show that ferritin heavy chain1(FTH1) was a key determinant for baicalin-induced ferroptosis.Overexpression of FTH1 abrogated the anticancer effects of baicalin in both 5637 and KU19-19 cells.Taken together,our data for the first time suggest that the natural product baicalin exerts its anticancer activity by inducing FTH1-dependent ferroptosis,which will hopefully provide a prospective compound for bladder cancer treatment.Na Kong Xiaying Chen Jiao Feng Ting Duan Shuiping Liu Xueni Sun Peng Chen Ting Pan Lili Yan Ting Jin Yu Xiang Quan Gao Chengyong Wen Weirui Ma Wencheng Liu Mingming Zhang Zuyi Yang Wengang Wang Ruonan Zhang Bi Chen Tian Xie Xinbing Sui Wei Tao 2021Acta Pharmaceutica Sinica B2021,11,12:23
5Efficacy and safety of Shaoyang Xibi decoction in patients with knee osteoarthritis:a multi-center,single-blind,randomized controlled trial显示文摘PURPOSE: To observe the efficacy and safety of Shaoyang Xibi decoction(SYXBD) in patients with knee osteoarthritis(KOA), and to verify that the theory of 'Shaoyang dominating bone' in Traditional Chinese Medicine(TCM) can be applied to KOA treatment.METHODS: Participants were randomly allocated to two groups: SYXBD(treatment group, n = 66)and Meloxicam(control group, n = 66). Western Ontario and McMaster Universities Osteoarthritis Index(WOMAC) and 36-Item Short Form Health Survey(SF-36) were used to assess efficacy before the treatment and 8 weeks after the treatment.RESULTS: Baseline data before the treatment between the two groups were similar. The WOMAC scores significantly decreased and the SF-36 scores significantly increased after 8-week treatment in both groups compared with before the treatment(P < 0.05). SYXBD significantly decreased pain scores(P < 0.001), physical function scores(P <0.001) and the total scores(P < 0.001) in WOMAC compared to Meloxicam. SYXBD significantly improved physical function(P = 0.021), bodily pain(P = 0.002) and general health(P = 0.014), with no significant difference in role emotional(P = 0.053),role physical(P = 0.517), vitality(P = 0.241), social function(P = 0.712) and mental health(P = 0.800)in SF-36 compared to Meloxicam. No adverse events were reported in the treatment group while13 adverse events happened in the control group during the study.CONCLUSION: SYXBD, prepared based on the theory of 'Shaoyang dominating bone', has a better curative efficay and safety in patients with KOA compared with Meloxicam. 6Early evaluation of growth traits of Larix kaempferi clones显示文摘Early selection is an important method to shorten the breeding cycle for tree species, which may differ in the time for early selection.To evaluate the early selected time for Larix kaempferi, tree height and diameter at breast height of 57 L.kaempferi clones were measured over many different growth years.The results indicated that, except for age × clone interaction for diameter at breast height(P = 0.741), there were significant differences among all variation sources(P < 0.01).The coefficient of phenotypic variation ranged from 14.89 to 35.65%for height and from 19.17 to 23.86% for diameter at breast height in different growth years.The repeatability of height and of diameter at breast height among clones was high,ranging from 0.6181 to 0.8531(height) and from 0.8443 to0.8497(diameter at breast height), in different growth years.There were significant positive correlations between all pairs of growth traits except between height in the 2 nd growth year and height in the 30 th growth year; and between height in the 2 nd growth year × diameter at breast height in the 30 th growth year.With a comprehensive evaluation method and a selection ratio of 10%, L65,L1, L62, L9, L15, and L78 were selected as excellent clones in the 30 th growth year.Their average values of height and diameter at breast height were 9.81 and 16.57%higher than the overall average, representing genetic gains of 6.46 and 13.99%, respectively.This study provides a theoretical foundation for the genetic improvement of L.kaempferi.Yanyan Pan Shuchun Li Chenglu Wang Wenjun Ma Guiyou Xu Lili Shao Kailong Li Xiyang Zhao Tingbo Jiang 2018Journal of Forestry Research2018,29,4:10
7Synergistic immunoreaction of acupuncture-like dissolving microneedles containing thymopentin at acupoints in immune-suppressed rats显示文摘Dissolving microneedles carried drug molecules can effectively penetrate the stratum corneum of skin to improve the transdermal drug delivery. The traditional Chinese medicine acupuncture is based on the needle stimulation at a specific location(acupoint) to generate and transmit biochemical and physiological signals which alter the pathophysiological state of patients. However, the pain associated with conventional acupuncture needles and the requirement of highly trained professionals limit the development of acupuncture in non-Asian countries. The purpose of this study is to investigate whether the dissolving microneedles can be utilized as a self-administered painless replacement for acupuncture and locally released drug molecules can achieve expected therapeutic outcomes. Immunosuppressive rats were treated with acupuncture at Zusanli(ST36) acupoint using microneedles containing thymopentin.The immune functions and psychological mood of the immunosuppressed animals were examined. The proliferation of splenocytes was examined by CCK-8 assay. CD4 and CD8 expression patterns in spleen cells were detected by flow cytometry. The current study showed that use of either microneedles containing thymopentin or conventional acupuncture both resulted in immune cell proliferation, which was confirmed by flow cytometry. We first define two restricted constraint qualifications and show how these constraint qualifications can be applied to obtain the decomposition properties of the Fréchet, Mordukhovich and Clarke normal cones to the sparsity constrained feasible set. Based on the decomposition properties of the normal cones, we then present and analyze three classes of Karush-KuhnTucker(KKT) conditions for the SNP. 9An ATF24 peptide-functionalized β-elemene-nanostructured lipid carrier combined with cisplatin for bladder cancer treatment显示文摘Objective:In this study,we aimed to develop an amino-terminal fragment(ATF)peptide-targeted liposome carryingβ-elemene(ATF24-PEG-Lipo-β-E)for targeted delivery into urokinase plasminogen activator receptor-overexpressing bladder cancer cells combined with cisplatin(DDP)for bladder cancer treatment.Methods:The liposomes were prepared by ethanol injection and high-pressure microjet homogenization.The liposomes were characterized,and the drug content,entrapment efficiency,andin vitro release were studied.The targeting efficiency was investigated using confocal microscopy,ultra-fast liquid chromatography,and an orthotopic bladder cancer model.The effects of ATF24-PEG-Lipo-β-E combined with DDP on cell viability and proliferation were evaluated by a Cell Counting Kit-8(CCK-8)assay,a colony formation assay,and cell apoptosis and cell cycle analyses.The anticancer effects were evaluated in a KU-19-19 bladder cancer xenograft model.Results:ATF24-PEG-Lipo-β-E had small and uniform sizes(~79 nm),high drug loading capacity(~5.24 mg/mL),high entrapment efficiency(98.37±0.95%),and exhibited sustained drug release behavior.ATF24-PEG-Lipo-β-E had better targeting efficiency and higher cytotoxicity than polyethylene glycol(PEG)ylatedβ-elemene liposomes(PEG-Lipo-β-E).DDP,combined with ATF24-PEG-Lipo-β-E,exerted a synergistic effect on cellular apoptosis and cell arrest at the G2/M phase,and these effects were dependent on the caspase-dependent pathway and Cdc25C/Cdc2/cyclin B1 pathways.Furthermore,thein vivo antitumor activity showed that the targeted liposomes effectively inhibited the growth of tumors,using the combined strategy.Conclusions:The present study provided an effective strategy for the targeted delivery ofβ-elemene(β-E)to bladder cancer,and a combined strategy for bladder cancer treatment.Bingtao Zhai Peng Chen Wengang Wang Shuiping Liu Jiao Feng Ting Duan Yu Xiang Ruonan Zhang Mingming Zhang Xuemeng Han Xiaying Chen Qiujie Li Guohua Li Ying Liu Xingxing Huang Wenzheng Zhang Ting Pan Lili Yan Ting Jin Tian Xie Xinbing Sui 2020Cancer Biology & Medicine2020,17,3:5
10Tourist Behaviors in Wetland Park:A Preliminary Study in Xixi National Wetland Park,Hangzhou,China显示文摘Public education as well as tourism and leisure are the two important functions of national wetland park, and tourist behaviors act as the guiding factor in the national wetland park construction and management.As the first national wetland park in China, Xixi National Wetland Park, located in Hangzhou City, Zhejiang Province, will give constructive instructions to the construction and management of other wetland parks and ecotourism resorts.This paper analyzed the tourist behaviors in Xixi National Wetland Park.By the observation, in-depth interview and questionnaire, the results show that tourists in wetland parks are different from those in the other common destinations, and they are also different from those strict eco-tourists.In fact, the tourists in wetland park have unique characteristics, such as the intense perception to the functions of wetland ecosystem and the environment impact behaviors.Those tourists are preferred to natural landscapes in their motivations and expectations.Wetland landscapes are the main image to those tourists in the Xixi National Wetland Park.Tourist expectation, experience and satisfaction are all critical factors for the success of the planning and management in national wetland parks.Based on the results, some measures for the sustainable development of wetland parks, including to optimize wetland theme landscape and tourism products, to improve tourist facilities and services, to enhance quality of visitor experiences, and to implement total quality management based on tourist characteristics, are given to national wetland park ecotourism.PAN Lili CUI Lijuan WU Ming 2010Chinese Geographical Science2010,20,1:5
11Comparison of viral RNA-host protein interactomes across pathogenic RNA viruses informs rapid antiviral drug discovery for SARS-CoV-2显示文摘In contrast to the extensive research about viral protein–host protein interactions that has revealed major insights about how RNA viruses engage with host cells during infection,few studies have examined interactions between host factors and viral RNAs(vRNAs).Here,we profiled vRNA–host protein interactomes for three RNA virus pathogens(SARS-CoV-2,Zika,and Ebola viruses)using ChIRP-MS.Comparative interactome analyses discovered both common and virus-specific host responses and vRNA-associated proteins that variously promote or restrict viral infection.In particular,SARS-CoV-2 binds and hijacks the host factor IGF2BP1 to stabilize vRNA and augment viral translation.Our interactome-informed drug repurposing efforts identified several FDA-approved drugs(e.g.,Cepharanthine)as broad-spectrum antivirals in cells and hACE2 transgenic mice.A co-treatment comprising Cepharanthine and Trifluoperazine was highly potent against the newly emerged SARS-CoV-2 B.1.351 variant.Thus,our study illustrates the scientific and medical discovery utility of adopting a comparative vRNA-host protein interactome perspective.Shaojun Zhang Wenze Huang Lili Ren Xiaohui Ju Mingli Gong Jian Rao Lei Sun Pan Li Qiang Ding Jianwei Wang Qiangfeng Cliff Zhang 2022Cell Research2022,32,1:5

13Temperature-sensitive splicing is an important molecular regulation mechanism of thermosensitive genic male sterility in rice显示文摘Photoperiod and temperature-sensitive genetic male sterility (PGMS and TGMS) plants have a number of desirable characteristics for hybrid production. Two-line hybrids developed using the PGMS/TGMS system now account for a large proportion of rice production in China. In this paper, we summarize recent advances on molecular regulation mechanisms and genetics of PGMS/TGMS in rice. We suggest that temperature-sensitive splicing, an important posttranscriptional regulatory mechanism in modulating gene expression and eventually development and differentiation, is also an important molecular regulation mechanism of TGMS in rice. We review those factors involved in temperature-sensitive splicing like cis splice site, snRNA, trans premRNA splicing protein and SR proteins, and delineate that splicing could be regulated by a complex cell signaling pathway. 15LncDACH1 promotes mitochondrial oxidative stress of cardiomyocytes by interacting with sirtuin3 and aggravates diabetic cardiomyopathy显示文摘Diabetic cardiomyopathy(DCM)is a common complication in diabetic patients.The molecular mechanisms of DCM remain to be fully elucidated.The intronic long noncoding RNA of DACH1(lnc DACH1)has been demonstrated to be closely associated with heart failure and cardiac regeneration.In this study,we investigated the role of lnc DACH1 in DCM and the underlying molecular mechanisms.The expression of lnc DACH1 was increased in DCM hearts and in high glucose-treated cardiomyocytes.Knockout of lnc DACH1 reduced mitochondrial oxidative stress,cell apoptosis,cardiac fibrosis and hypertrophy,and improved cardiac function in DCM mice.Overexpression of lnc DACH1 exacerbated mitochondria-derived reactive oxygen species(ROS)level and apoptosis,decreased activity of manganese superoxide dismutase(Mn-SOD);while silencing of lnc DACH1 attenuated ROS production,mitochondrial dysfunction,cell apoptosis,and increased the activity of Mn-SOD in cardiomyocytes treated with high glucose.Lnc DACH1 directly bound to sirtuin3(SIRT3)and facilitated its degradation by ubiquitination,therefore promoting mitochondrial oxidative injury and cell apoptosis in mouse hearts.In addition,SIRT3 silencing abrogated the protective effects of lnc DACH1 deficiency in cardiomyocytes.In summary,lnc DACH1 aggravates DCM by promoting mitochondrial oxidative stress and cell apoptosis via increasing ubiquitination-mediated SIRT3 degradation in mouse hearts.Inhibition of lnc DACH1 represents a novel therapeutic strategy for the intervention of diabetic cardiomyopathy.Qi Zhang Danyang Li Xue Dong Xiaowen Zhang Junwu Liu Lili Peng Bo Meng Qi Hua Xinyu Pei Lu Zhao Xiaoxi Hu Yang Zhang Zhenwei Pan Yanjie Lu Baofeng Yang 2022Science China(Life Sciences)2022,65,6:4

17LA-ICP-MS U–Pb geochronology of detrital zircons from the Zhaochigou Formation-complex in the Helan Mountain and its tectonic significance显示文摘This paper carries out studies about LA-ICPMS U–Pb dating and Hf isotopic compositional analysis for detrital zircons in two metasedimentary samples that were taken from the Zhaochigou Formation-complex in the Helan Mountain.The results show that the Zhaochigou Formation-complex formed*1.96 Ga,and it is a part of the Khondalite Series that is widespread in the North China Craton(NCC).The age spectrum of the detrital zircons indicates that the Zhaochigou Formation-complex shared the same provenance with the eastern edge of the Khondalite Series.Products of ancient magmatic activities within the Khondalite Belt may be the major source for the sedimentary materials that formed the Khondalite Series.Our age-dating results suggest that the Khondalite Series in the NCC may have experienced two stages of metamorphism,as the early stage occurred during 1.96–1.95 Ga,and the later stage occurred at about 1.87 Ga.The Hfisotopic data indicate that the Western Block of the NCC endured a stage of crustal growth at 2.5–2.3 Ga.Liming Li Zuoxun Zeng Yanjun Lu Yunxu Wei Shimin Xiang Lili Pan 2014Chinese Science Bulletin2014,59,13:3
18Shenkang injection improves coagulation in patients with chronic kidney disease: a systematic review and Meta-analysis显示文摘OBJECTIVE: To investigate the effect of Shenkang injection(SKI) on chronic kidney disease(CKD).METHODS: Seven databases including Cochrane Central Register of Controlled Trials, PubMed, EMBASE, MEDLINE, China National Knowledge Infrastructure, Wanfang Database, and CQVIP from their inception to March 2018 were searched. Only randomized controlled trials that evaluated conventional treatment and conventional treatment with SKI in CKD patients were investigated. Outcomes such as fibrinogen(FIB), D-dimer, prothrombin time(PT), activated partial thromboplastin time(APTT), and the side effects of SKI were analyzed using Revman 5.3 software. The quality of the studies was assessed using the Cochrane Collaboration’s Risk of Bias tool and the quality of evidence was assessed using GRADEpro.RESULTS: Four randomized controlled trials were investigated in our analysis, and these studies were of moderate quality. For FIB and D-dimer, SKI had a superior effect compared with the control group(mean difference(MD)=-1.23, 95% confidence interval(CI):-1.46,-0.99, P < 0.01;MD =-0.36, 95%CI:-0.51,-0.21, P < 0.01, respectively)SKI increased APTT and PT compared with the control(MD = 7.34, 95% CI: 3.05, 11.62, P < 0.01;MD = 3.40,95% CI: 2.2, 4.61, P < 0.01, respectively). In the four studies, there were no side effects that were related to SKI.CONCLUSION: SKI may be effective in improving coagulation in patients with CKD without obvious adverse reactions. 19A Novel Combinational Convolutional Neural Network for Automatic Food-Ingredient Classification显示文摘With the development of deep learning and Convolutional Neural Networks(CNNs),the accuracy of automatic food recognition based on visual data have significantly improved.Some research studies have shown that the deeper the model is,the higher the accuracy is.However,very deep neural networks would be affected by the overfitting problem and also consume huge computing resources.In this paper,a new classification scheme is proposed for automatic food-ingredient recognition based on deep learning.We construct an up-to-date combinational convolutional neural network(CBNet)with a subnet merging technique.Firstly,two different neural networks are utilized for learning interested features.Then,a well-designed feature fusion component aggregates the features from subnetworks,further extracting richer and more precise features for image classification.In order to learn more complementary features,the corresponding fusion strategies are also proposed,including auxiliary classifiers and hyperparameters setting.Finally,CBNet based on the well-known VGGNet,ResNet and DenseNet is evaluated on a dataset including 41 major categories of food ingredients and 100 images for each category.Theoretical analysis and experimental results demonstrate that CBNet achieves promising accuracy for multi-class classification and improves the performance of convolutional neural networks.Lili Pan Cong Li Samira Pouyanfar Rongyu Chen Yan Zhou 2020Computers, Materials & Continua2020,,2:3
20Evaluation of H2 Influence on the Evolution Mechanism of NOx Storage and Reduction over Pt–Ba–Ce/c-Al2O3 Catalysts显示文摘In this investigation, Pt–Ba–Ce/c-Al2O3 catalysts were prepared by incipient wetness impregnation and experiments were performed to evaluate the influence of H2 on the evolution mechanism of nitrogen oxides (NOx) storage and reduction (NSR). The physical and chemical properties of the Pt–Ba–Ce/c- Al2O3 catalysts were studied using a combination of characterization techniques, which showed that PtOx, CeO2, and BaCO3, whose peaks were observed in X-ray diffraction (XRD) spectra, dispersed well on the c-Al2O3, as shown by transmission electron microscope (TEM), and that the difference between Ce3+ and Ce4+, as detected by X-ray photoelectron spectroscopy (XPS), facilitated the migration of active oxygen over the catalyst. In the process of a complete NSR experiment, the NOx storage capability was greatly enhanced in the temperature range of 250–350℃, and reached a maximum value of 315.3μmol·gcat^-1 at 350℃, which was ascribed to the increase in NO2 yield. In a lean and rich cycling experiment, the results showed that NOx storage efficiency and conversion were increased when the time of H2 exposure (i.e., 30, 45, and 60 s) was extended. The maximum NOx conversion of the catalyst reached 83.5% when the duration of the lean and rich phases was 240 and 60 s, respectively. The results revealed that increasing the content of H2 by an appropriate amount was favorable to the NSR mechanism due to increased decomposition of nitrate or nitrite, and the refreshing of trapping sites for the next cycle of NSR.Pan Wang Jing Yi Chuan Sun Peng Luo Lili Lei 2019Engineering2019,5,3:2
