1A Genomic Variation Map Provides Insights into the Genetic Basis of Spring Chinese Cabbage (Brassica rapa ssp.pekinensis)Selection显示文摘Chinese cabbage is the most consumed leafy crop in East Asian countries.However,premature bolting induced by continuous low temperatures severely decreases the yield and quality of the Chinese cabbage, and therefore restricts its planting season and geographic distribution.In the past 40years,spring Chinese cabbage with strong winterness has been selected to meet the market demand.Here,we report a genome variation map of Chinese cabbage generated from the resequencing data of 194 geographically diverse accessions of three ecotypes.In-depth analyses of the selection sweeps and genome-wide patterns revealed that spring Chinese cabbage was selected from a specific population of autumn Chinese cabbage around the area of Shandong peninsula in northern China.We identified 23 genomic loci that underwent intensive selection,and further demonstrated by gene expression and haplotype analyses that the incorporation of elite alleles of VERNALISATION INSENTIVE 3.1(BrVIN3.1)and FLOWER LOCUS C 1(BrFLC1)is a determinant genetic source of variation during selection.Moreover,we showed that the quantitative response of BrVIN3.1 to cold due to the sequence variations in the cis elements of the BrVlN3.1 promoter significantly contributes to bolting-time variation in Chinese cabbage.Collectively, our study provides valuable insights into the genetic basis of spring Chinese cabbage selection and will facilitate the breeding of bolting-resistant Varieties by molecular-marker-assisted selection,transgenic or gene editingapproaches.Tongbing Su Weihong Wang Peirong Li Bin Zhang Pan Li Xiaoyun Xin Honghe Sun Yangjun Yu Deshuang Zhang Xiuyun Zhao Changlong Wen Gang Zhou Yuntong Wang Hongkun Zheng Shuancang Yu Fenglan Zhang 2018Molecular Plant2018,11,11:6
2Composition variations of the Sinian-Cambrian sedimentary rocks in Hunan and Guangxi and their tectonic significance显示文摘This paper reports the geochemical and zircon U-Pb dating data of the Sinian to Cambrian low-grade metamorphic rocks in the Miaoer Mountain area,Guangxi and the Jinjiling area,Hunan Province.Petrographic and geochemical features indicate that protoliths of these metamorphic rocks are clastic sedimentary rocks with medium weathering,which were formed in the passive continental margin.Geochemistry and zircon U-Pb ages indicate that the Sinian and Cambrian sedimentary rocks in the Jinjiling area have similar detritus components,which are characterized by abundant Grenvillian detrital zircons,suggesting a close affinity with the Cathaysia Block.The Cambrian sedimentary rocks in the Miaoer Mountain area have similar geochemistry and zircon geochronology to those in the Jinjiling area,showing an affinity with the Cathaysia Block.However,the Sinian sedimentary rocks in the Miaoer Mountain area show different geochemical features from the Cambrian sedimentary rocks and those in the Jinjiling area,and are characterized by abundant 840–700 Ma detrital zircons and less^2.0 Ga ones,showing a close affinity with the Yangtze Block.These variations suggest that the Jinjiling area continuously accepted the fragments from the Cathaysia from the Sinian to the Cambrian,whereas the provenance of the Miaoer Mountain sedimentary basin changed from the Yangtze Block to the Cathaysia Block during this interval.This change implies a tectonic movement,which caused the further sinking of the basin in the Miaoer Mountain area and northwestward transferring of the basin center before the Middle Cambrian,so that the Miaoer Mountain basin received the detritus from the Cathaysia Block in the Middle Cambrian.This fact also proves that the Yangtze and Cathaysia blocks have converged at least in Middle Cambrian,and the southwestern boundary between them is located between the Miaoer Mountain and Jinjiling areas.WANG PengMing YU JinHai SUN Tao SHI Yu CHEN PeiRong ZHAO KuiDong CHEN WeiFeng LIU Qian 2013Science China Earth Sciences2013,56,11:6
3Application of backflush and micro-flow techniques to the analysis of C_5—C_(13) hydrocarbons in crude oils and its geochemical significance显示文摘An Agilent 6890N GC equipped with both FID and Agilent 5975 MSD analyzer has been employed to analyze C5—C13 hydrocarbon fractions of crude oils. A technical combination of program temperature volatilizer injection, backflush and micro-flow controller afforded a fine separation of C5—C13 compounds on a PONA column with the heavy part of crude oils being cut off before entering the analytical column. Both GC-FID chromatogram and GC-MS mass chromatograms (MID and full scan) could be obtained at the same time. The retension time differences of nC6—nC13 alkanes between GC and TIC were in the range of 0.02—0.58 minutes. Totally 286 peaks have been assigned group compositions. 4Characteristics and formation mechanism of Changxing Formation-Feixianguan Formation reef-shoal reservoirs in Yuanba Gasfield显示文摘Taking advantage of the successful experience in exploring and discovering the Puguang Gasfield,and targeting the Changxing Formation-Feixianguan Formation organic reef and shoal lithological trap,SINOPEC drilled Well Yuanba 1 in the Bazhong area in Northeast Sichuan in 2006,and discovered the Yuanba Gasfield with a high-production commercial gas flow of 503×10^(3) m^(3)/d.As a normal-pressured lithological gas reservoir with high H_(2)S content,Yuanba Gasfield is characterized by weak tectonic deformation and deep burial,with an average depth of 6,600 m in the central part of the gas reservoir,and is the deepest marine gas field in the Sichuan Basin.Yuanba Gasfield is dominated by large-scale reef-shoal reservoirs of Changxing Formation.The formation of the reservoirs was primarily controlled by early meteoric freshwater dissolution and dolomitization,and less affected by deep-burial dissolution and tectonic movement.A comparative analysis was made on the characteristics of deep reef-shoal reservoirs in the Yuanba and Puguang gas fields so as to explore their formation mechanisms.It is concluded that the reservoir size and early pore development was controlled by early depositional-diagenetic environment.Fracture formation and dissolution were controlled by structure-fluid coupling,pore reworking and preservation is determined by fluid-rock interaction.Yongsheng Ma Xunyu Cai Peirong Zhao 2016Petroleum Research2016,1,2:3
5China's shale gas exploration and development: Understanding and practice显示文摘Through detailed analyses of the distribution characteristics of organic-rich shale, appearance features of high-quality shale, microscopic characteristics of shale reservoir rocks, fracability, and the relationship between preservation conditions and shale gas enrichment in Upper Ordovician Wufeng Formation-Lower Silurian Longmaxi Formation in Sichuan Basin, theoretical understandings and specific suggestions with respect to the exploration and development of shale gas in China are summarized and proposed respectively. Important geological understandings in the exploration and development of shale gas of the Wufeng Formation–Longmaxi Formation in the Sichuan Basin can be summarized into the following aspects: depositional environment and depositional process control the distribution of organic-rich shale; high quality shale in 'sweet spot segments' are commonly characterized by high content of organic carbon, high brittleness, high porosity and gas content; organic pores are important storage space for the enrichment of shale gas; preservation conditions are the key factor for the geological evaluation of shale gas in structurally complex regions; shale gas can be considered as 'artificial gas reservoirs' and the fracability assessment is essential for high-production; nanoscale storage space and the mode of occurrence control the special seepage characteristics of shale gas. 6Formation mechanism of condensates, waxy and heavy oils in the southern margin of Junggar Basin, NW China显示文摘It is a challenge to determine the source and genetic relationship of condensate, waxy and heavy oils in one given complicated petroliferous area, where developed multiple sets of source rocks with different maturity and various chemical features.The central part of southern margin of Junggar Basin, NW China is such an example where there are condensates, light oils, normal density oils, heavy crude oils and natural gases. The formation mechanism of condensates has been seriously debated for long time;however, no study has integrated it with genetic types of waxy and heavy oils. Taking the central part of southern margin of Junggar Basin as a case, this study employs geological and geochemical methods to determine the formation mechanism of condensates,waxy and heavy oils in a complicated petroliferous area, and reveals the causes and geochemical processes of the co-occurrence of different types of crude oils in this region. Based on detailed geochemical analyses of more than 40 normal crude oils, light oils,condensates and heavy oils, it is found that the condensates are dominated by low carbon number n-alkanes and enriched in light naphthenics and aromatic hydrocarbons. Heptane values of these condensates range from 19% to 21%, isoheptane values from1.9 to 2.1, and toluene/n-heptane ratios from 1.5 to 2.0. The distribution of n-alkanes in the condensates presents a mirror image with high density waxy crude oils and heavy oils. Combined with the oil and gas-source correlations of the crude oils, condensates and natural gas, it is found that the condensates are product of evaporative fractionation and/or phase-controlled fractionation of reservoir crude oils which were derived from mature Cretaceous lacustrine source rocks in the relatively early stage. The waxy oils are the intermediate products of evaporative fractionation and/or phase-controlled fractionation of reservoir crude oils, while the heavy oils are in-situ residuals. Therefore, evaporative fractionation and/or phase-controlled fractionation would account for the formation of the condensate, light oil, waxy oil and heavy oil in the central part of southern margin of Junggar Basin, resulting in a great change of the content in terms of light alkanes, naphthenics and aromatics in condensates, followed by great uncertainties of toluene/n-heptane ratios due to migration and re-accumulation. 7Identification of long noncoding RNAs involved in resistance to downy mildew in Chinese cabbage显示文摘Brassica downy mildew,a severe disease caused by Hyaloperonospora brassicae,can cause enormous economic losses in Chinese cabbage(Brassica rapa L.ssp.pekinensis)production.Although some research has been reported recently concerning the underlying resistance to this disease,no studies have identified or characterized long noncoding RNAs involved in this defense response.In this study,using high-throughput RNA sequencing,we analyzed the disease-responding mRNAs and long noncoding RNAs in two resistant lines(T12–19 and 12–85)and one susceptible line(91–112).Clustering and Gene Ontology analysis of differentially expressed genes(DEGs)showed that more DEGs were involved in the defense response in the two resistant lines than in the susceptible line.Different expression patterns and proposed functions of differentially expressed long noncoding RNAs among T12–19,12–85,and 91–112 indicated that each has a distinct disease response mechanism.There were significantly more cis-and trans-functional long noncoding RNAs in the resistant lines than in the susceptible line,and the genes regulated by these RNAs mostly participated in the disease defense response.Furthermore,we identified a candidate resistance-related long noncoding RNA,MSTRG.19915,which is a long noncoding natural antisense transcript of a MAPK gene,BrMAPK15.Via an agroinfiltration-mediated transient overexpression system and virus-induced gene silencing technology,BrMAPK15 was indicated to have a greater ability to defend against pathogens.MSTRG.19915-silenced seedlings showed enhanced resistance to downy mildew,probably because of the upregulated expression of BrMAPK15.This research identified and characterized long noncoding RNAs involved in resistance to downy mildew,laying a foundation for future in-depth studies of disease resistance mechanisms in Chinese cabbage.Bin Zhang Tongbing Su Peirong Li Xiaoyun Xin Yunyun Cao Weihong Wang Xiuyun Zhao Deshuang Zhang Yangjun Yu Dayong Li Shuancang Yu Fenglan Zhang 2021Horticulture Research2021,8,1:3
8Insulin-Enhanced Antitumor Effect of 5-Fluorouracil in vivo显示文摘OBJECTIVE To determine if insulin treatment can enhance the antitumor effect of 5-fluorouracil(5-FU),and to explore the mech- anism of the enhancement of insulin. METHODS S180 sarcoma,H22 liver cancer and human Eca-109 esophageal cancer cells were transplanted into nude mice to evaluate the inhibitory effect on tumor growth of insulin alone or in combination with 5-FU.The levels of serum insulin-like growth factor-I(IGF-I)and insulin-like growth factor binding protein-3 (IGFBP-3)were determined. RESULTS Compared with 5-FU treatment alone,the tumor weight of H22 liver cancer and S180 sarcoma was reduced further with high,medium and low-dose insulin(0.09,0.06,0.03 U/20 g) +5-FU treatment.When a high dosage of insulin+5-FU was ad- ministered,tumor weight was significantly reduced(P<0.05).The inhibitory rate of growth of S180 sarcoma and H22 liver cancer reached 50.2% and 51.4%,respectively,which was significantly higher than 24.9% and 27.9% in the group receiving 5-FU alone (P<0.05).High,medium and low-dose insulin combined with 5-FU significantly inhibited the growth of Eca-109 cancer cells (P<0.05).Compared with the control group,the level of serum IGF-1 decreased(P<0.05),whereas the level of serum IGFBP-3 slightly increased in the 5-FU±insulin groups(P>0.05).In mice with H22 liver cancer and S180 sarcoma the IGF-1 level with high- dose insulin+5-FU treatment was significantly lower compared to treatment with 5-FU alone(P<0.05),but the difference was not significant in mice transplanted with esophageal cancer cells. 9Genome-wide analysis of changes in miRNA and target gene expression reveals key roles in heterosis for chinese cabbage biomass显示文摘Heterosis is a complex phenomenon in which hybrids show better phenotypic characteristics than their parents do.Chinese cabbage(Brassica rapa L.spp.pekinensis)is a popular leafy crop species,hybrids of which are widely used in commercial production;however,the molecular basis of heterosis for biomass of Chinese cabbage is poorly understood.We characterized heterosis in a Chinese cabbage hybrid cultivar and its parental lines from the seedling stage to the heading stage;marked heterosis of leaf weight and biomass yield were observed.Small RNA sequencing revealed 63 and 50 differentially expressed microRNAs(DEMs)at the seedling and early-heading stages,respectively.The expression levels ofthe majority of miRNA clusters in the hybrid were lower than the mid-parent values(MPVs).Using degradome sequencing,we identi fied 1,819 miRNA target genes.Gene ontology(GO)analyses demonstrated that the target genes ofthe MPV-DEMs and low parental expression level dominance(ELD)miRNAs were signi ficantly enriched in leaf morphogenesis,leaf development,and leaf shaping.Transcriptome analysis revealed that the expression levels of photosynthesis and chlorophyll synthesis-related MPV-DEGs(differentially expressed genes)were signi ficantly different in the F_(1) hybrid compared to the parental lines,resulting in increased photosynthesis capacity and chlorophyll content in the former.Furthermore,expression of genes known to regulate leaf development was also observed at the seedling stage.Arabidopsis plants overexpressing BrGRF4.2 and bra-miR396 presented increased and decreased leaf sizes,respectively.These results provide new insight into the regulation of target genes and miRNA expression patterns in leaf size and heterosis for biomass of B.rapa.Peirong Li Tongbing Su Deshuang Zhang Weihong Wang Xiaoyun Xin Yangjun Yu Xiuyun Zhao Shuancang Yu Fenglan Zhang 2021Horticulture Research2021,8,1:1
10Research Progress in the Petroleum and Natural Gas Geological Theory of China显示文摘The Chinese landmass, as a composite region, consists of multiple small continental blocks, such as Sino-Korea, Yangtze, Tarim, etc., and orogenic belts. Because of its distinctive tectonosedimentary evolution, China's oil/gas-bearing regions differ remarkably from that elsewhere in the world. For instance, in comparison to the Middle East oil/gas-bearing regions which are characterized by Mesozoic-Cenozoic marine oil/gas-bearing beds, early oil and gas discoveries in China are distributed mainly in Mesozoic-Cenozoic continental sedimentary basins. Generation of oil from terrestrial organic matter, or terrestrial oil generation, and the formation of large oil/gas fields in continental sedimentary basins were previously the major characteristics of petroleum geology of China. However, in the past 20 years, a series of major oil and gas discoveries from marine strata have been made. Marine oil/gas fields in China are mainly distributed in the Tarim, Sichuan, and Ordos basins, which are tectonically stable and covered with Mesozoic-Cenozoic deposits. In these basins, hydrocarbon-bearing strata are of old age and the oil/gas fields are commonly deeply-buried. Cumulatively, 995 oil/gas fields have been found so far, making China the fourth largest oil-producing country and the sixth largest gas-producing country in the world. In terms of petroleum and natural gas geology, theories of hydrocarbon generation from continental strata, such as terrestrial oil generation and coal-generated hydrocarbons, etc., have been established. Significant progress has been made in research on the sequence stratigraphy of continental strata, formation mechanisms of ultra-deep clastic reservoirs, and hydrocarbon accumulation in the continental subtle reservoir. Regarding research on the marine petroleum geology of China, with respect to the major characteristics, such as deeply-buried reservoirs, old strata, and multiple phases of transformation, important advances have been made, in areas such as the multiple-elements of hydrocarbon supply, formation of reservoirs jointly controlled by deposition, tectonic activities, and diagenetic fluid-rock reactions, and oil/gas reservoirs formed through superimposed multi-stage hydrocarbon accumulation. 11Practice and theoretical and technical progress in exploration and development of Shunbei ultra-deep carbonate oil and gas field,Tarim Basin,NW China显示文摘In this review on the exploration and development process of the Shunbei ultra-deep carbonate oil and gas field in the Tarim Basin, the progress of exploration and development technologies during the National 13th Five-Year Plan of China has been summarized systematically, giving important guidance for the exploration and development of ultra-deep marine carbonate reservoirs in China and abroad. Through analyzing the primary geological factors of “hydrocarbon generation-reservoir formation-hydrocarbon accumulation” of ancient and superposed basin comprehensively and dynamically, we point out that because the Lower Cambrian Yuertusi Formation high-quality source rocks have been located in a low-temperature environment for a long time, they were capable of generating hydrocarbon continuously in late stage, providing ideal geological conditions for massive liquid hydrocarbon accumulation in ultra-deep layers. In addition, strike-slip faults developed in tectonically stable areas have strong control on reservoir formation and hydrocarbon accumulation in this region. With these understandings, the exploration focus shifted from the two paleo-uplifts located in the north and the south to the Shuntuoguole lower uplift located in between and achieved major hydrocarbon discoveries. Through continuing improvement of seismic exploration technologies for ultra-deep carbonates in desert, integrated technologies including seismic acquisition in ultra-deep carbonates,seismic imaging of strike-slip faults and the associated cavity-fracture systems, detailed structural interpretation of strike-slip faults, characterization and quantitative description of fault-controlled cavities and fractures, description of fault-controlled traps and target optimization have been established. 14Petrogenesis of Early Cretaceous adakitic granodiorite: Implication for a crust thickening event within the Cathaysia Block, South China显示文摘Adakitic rocks in continental settings are commonly considered to be formed by partial melting of thickened or delaminated lower crust. Investigations on this kind of rocks can provide important information about crustal evolution complementary to information from other rocks. This paper reports adakitic granodiorite of the Lingxi pluton in the interior of the Cathayisa Block. LA-ICP-MS zircon U-Pb dating shows that it was formed in the late Early Cretaceous(100±1 Ma). The granodiorite has geochemical features of adakitic rocks derived from partial melting of the thickened lower crust, e.g., high SiO2(mainly ranging from 64.4 to 68.9 wt.%) and Sr(624–894 ppm) contents, Sr/Y(49.9–60.8) and La/Yb(23.4–42.8) values, low Y(10.3–17.1 ppm), Ni(5.62–11.8 ppm) and MgO(mostly from 0.86 wt.% to 1.57 wt.%) contents and weak Eu anomaly. It has initial 87Sr/86 Sr ratios of 0.7086–0.7091, εNd(t) values of.6.2 to.5.9 and zircon εHf(t) values mostly of.10.1 to.7.6. Based on the geochemical characteristics and simple modelling, it is suggested that the most likely generation mechanism of the Lingxi granodiorite is partial melting of a thickened Proterozoic lower continental crust at a pressure ≥12 kbar(or crust thickness ≥40km), leaving a garnet-bearing amphibolite residue. Combining our results and previous studies of the tectonic evolution of the Cathaysia Block, we propose that the crust was thickened to over 40 km by a compressive event occurring during the late Early Cretaceous, which is supported by the observation that there is an angular unconformity between the Upper Cretaceous Series and the early Lower Cretaceous or the Jurassic rocks. After this event, the Cathaysia Block experienced a lithospheric extension and thinning probably driven by the high-angle paleo-Pacific subduction. With the attenuation of lithosphere, the lower crust was heated to partial melting by upwelling asthenospheric materials, resulting in generation of the Lingxi granodiorite and other coeval granitoids in the Cathaysia Block. 15Cloning, tissue expression pattern, and cnromosome localization of human protein kinase By gene显示文摘Protein kinase B (PKB) is a member of the second messenger-regulated subfamily of protein kinases, and plays a key role in cell-cycle regulation, glucose uptake and promotion of cell differentiation. Evidence shows that PKB undergoes activation in some human tumors and is involved in Ras pathway, which implies that PKB can trigger a pathway to induce oncogenic transformation. A nucleotide sequence of mouse Pkbywas used as a probe to screen homolog in a human liver cDNA library. A fragment of 1998 bp containing a 1440 bp ORF encoding 479 amino acid residues was obtained. Then the 3’-terminal of this fragment was extended to 2788 bp by ’electronic walking’ screening, and the extended fragment was confirmed by PCR amplification. The protein deduced by the gene had a high identity of 83% and 78% to the human PKBα and β, respectively, and was designated as human PKBγ. 17Constant light in early life induces fear-related behavior in chickens with suppressed melatonin secretion and disrupted hippocampal expression of clock-and BDNF-associated genes显示文摘Background: Light management plays an important role in the growth and behavior of broiler chickens. Constant light in early post hatch stage has been a common practice in broiler industry for improving growth performance,while whether and how constant light in early life affects the behavior of broiler chickens is rarely reported.Results: In this study, newly hatched chicks were kept in either constant(24 L:0 D, LL) or(12 L:12 D, LD) photoperiod for 7 d and then maintained in 12 L:12 D thereafter until 21 days of age. Constant light increased the average daily feed intake but not the body weight, which led to higher feed conversion ratio. Chickens in LL group exhibited fear-related behaviors, which was associated with higher corticosterone, lower melatonin and 5-HT levels. Concurrently, constant light exposure increased the mRNA expression of clock-related genes and suppressed the expression of antioxidative genes in the hippocampus. 18Isolation and polymorphic frequency detection of a novel STR on human chromosome 14q24.3显示文摘A single copy fragment (FD14_ca 1) containing 22 CA repetitive units was isolated with (CA) 15 oligonucleotide probe from a human chromosome 14q24 3 probe pool generated by microdissection, and proved to be a new short tandem repeat (STR) sequence through querying in GenBank of NCBI. The STR has 11 alleles in Chinese, and its polymorphic information content (PIC) is 0 85. Mendelian segregation was shown in 2 Chinese pedigrees with two generations. This STR has been accurately relocalized on chromosome 14q24 3 by fluorescence in situ hybridization (FISH). The accession numbers of this STR in GDB and GenBank database are D14S1435 and G31413 respectively. 20Cloning, tissue expression pattern characterization and chromosome localization of human peptide methionine sulfoxide reductase cDNA显示文摘Oxidation and reduction of some amino acids are one of the molecular mechanisms for regulating the function of proteins. The oxidation of methionine (Met) to methionine sulfoxide (Met(O)) results in decreasing or loss of the biological activity of related proteins. It was found that peptide methionine sulfoxide reductase (msrA) can reduce Met(O) to Met and therefore restored the biological function of the oxidized proteins. To reveal the methionine oxidation-reduction mechanism in human body, in this study, the cDNA sequence of bovine msrA was used as an information-probe to screen the human EST database. Based on a contig assembled from homologous ESTs, a 1 256-bp human MSRA cDNA was cloned from several human cDNA libraries. The cDNA contains an open reading frame (ORF) of 705 bp in length, which encodes 235 amino acid residues. Homology comparison revealed that human MSRA shares 88% and 61% identities with bovine and Escherichia coli msrA protein respectively.
