1Oxidative stress and diabetes:antioxidative strategies显示文摘Diabetes mellitus is one of the major public health problems worldwide.Considerable recent evidence suggests that the cellular reduction-oxidation(redox)imbalance leads to oxidative stress and subsequent occurrence and development of diabetes and related complications by regulating certain signaling pathways involved in p-cell dysfunction and insulin resistance.Reactive oxide species(ROS)can also directly oxidize certain proteins(defined as redox modification)involved in the diabetes process.There are a number of potential problems in the clinical application of antioxidant therapies including poor solubility,storage instability and nonselectivity of antioxidants.Novel antioxidant delivery systems may overcome pharmacokinetic and stability problem and improve the selectivity of scavenging ROS.We have therefore focused on the role of oxidative stress and antioxidative therapies in the pathogenesis of diabetes mellitus.Precise therapeutic interventions against ROS and downstream targets are now possible and provide important new insights into the treatment of diabetes.Pengju Zhang Tao Li Xingyun Wu Edouard C.Nice Canhua Huang Yuanyuan Zhang 2020Frontiers of Medicine2020,14,5:29

4The cytotoxicity and protective effects of Astragalus membranaceus extracts and butylated hydroxyanisole on hydroxyl radical-induced apoptosis in fish erythrocytes显示文摘Erythrocytes play an essential role in transporting O_2 and CO_2 for respiration in fish. However,erythrocytes continuously suffer from reactive oxygen species(ROS)-induced oxidative stress and apoptosis. Thus, it is essential to expand our knowledge of how to protect erythrocytes against ROS-induced oxidative stress and apoptosis in fish. In this study, we explored the cytotoxicity and the effects of butylated hydroxyanisole(BHA), ethyl ether extracts, ethyl acetate extracts, acetone extracts(AE), ethanol extracts, and aqueous extracts of Astragalus membranaceus(EAm) on hydroxyl radical(·OH)-induced apoptosis in carp erythrocytes. The rat hepatocytes and carp erythrocytes were incubated with different concentrations of BHA or EAm(0.125 to 1 mg/mL). The toxicity in rat hepatocytes and carp erythrocytes was then measured using a 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide(MTT) assay and a haemolysis assay,respectively. The carp erythrocytes were treated with BHA or EAm in the presence of 40 μmol/L FeSO4 and20 μmol/L H_2 O_2 at 37℃, except for the control group. Oxidative stress and apoptosis parameters in the carp erythrocytes were then evaluated using the commercial kit. The results indicated that at high concentrations, BHA and EAm could induce toxicity in rat hepatocytes and fish erythrocytes. However, BHA was more toxic than EAm at the same concentrations. Moreover, the toxicity order of BHA and EAm in the fish erythrocytes approximately agreed with that for the rat hepatocytes. Butylated hydroxyanisole and EAm suppressed the ·OH-induced phosphatidylserine exposure and DNA fragmentation(the biomarkers of apoptosis) by decreasing the generation of ROS, inhibiting the oxidation of cellular components, and restoring the activities of antioxidants in carp erythrocytes. Of all of the examined EAm, the AE showed the strongest effects. The effects of AE on superoxide anion, H_2 O_2, met-haemoglobin and reduced glutathione levels, as well as glutathione reductase activity and apoptosis were equivalent to or stronger than those of BHA. 5Surgical intervention combined with weight-bearing walking training improves neurological recoveries in 320 patients with clinically complete spinal cord injury:a prospective self-controlled study显示文摘Although a large number of trials in the SCI field have been conducted,few proven gains have been realized for patients.In the present study,we determined the efficacy of a novel combination treatment involving surgical intervention and long-term weight-bearing walking training in spinal cord injury(SCI)subjects clinically diagnosed as complete or American Spinal Injury Association Impairment Scale(AIS)Class A(AIS-A).A total of 320 clinically complete SCI subjects(271 male and 49 female),aged 16–60 years,received early(≤7 days,n=201)or delayed(8–30 days,n=119)surgical interventions to reduce intraspinal or intramedullary pressure.Fifteen days post-surgery,all subjects received a weight-bearing walking training with the“Kunming Locomotion Training Program(KLTP)”for a duration of 6 months.The neurological deficit and recovery were assessed using the AIS scale and a 10-point Kunming Locomotor Scale(KLS).We found that surgical intervention significantly improved AIS scores measured at 15 days post-surgery as compared to the pre-surgery baseline scores.Significant improvement of AIS scores was detected at 3 and 6 months and the KLS further showed significant improvements between all pair-wise comparisons of time points of 15 days,3 or 6 months indicating continued improvement in walking scores during the 6-month period.In conclusion,combining surgical intervention within 1 month post-injury and weight-bearing locomotor training promoted continued and statistically significant neurological recoveries in subjects with clinically complete SCI,which generally shows little clinical recovery within the first year after injury and most are permanently disabled.This study was approved by the Science and Research Committee of Kunming General Hospital of PLA and Kunming Tongren Hospital,China and registered at ClinicalTrials.gov(Identifier:NCT04034108)on July 26,2019.Yansheng Liu Jia-Xin Xie Fang Niu Zhexi Xu Pengju Tan Caihong Shen Hongkun Gao Song Liu Zhengwen Ma Kwok-Fai So Wutian Wu Chen Chen Sujuan Gao Xiao-Ming Xu Hui Zhu 2021Neural Regeneration Research2021,16,5:3

8Quasi-variable Discount Rates and Market Volatility: An Empirical Study显示文摘This paper proposes an assumption of quasi-variable discount rates to explain the excess volatility puzzle of stock market. Under the assumption, the ARMAX model is derived based on the CCAPM model and CRRA utility function to describe the linear relationship between the discount rate and the consumption growth rate. We conducted empirical research on this model using historical data of the US stock market. The results confirm a significantly negative relationship between consumption growth rate and discount rate. 9The archaeal KEOPS complex possesses a functional Gon7 homolog and has an essential function independent of the cellular t^(6)A modification level显示文摘Kinase,putative Endopeptidase,and Other Proteins of Small size(KEOPS)is a multisubunit protein complex conserved in eukaryotes and archaea.It is composed of Pcc1,Kae1,Bud32,Cgi121,and Gon7 in eukaryotes and is primarily involved in N^(6)-threonylcarbamoyl adenosine(t^(6)A)modification of transfer RNAs(tRNAs).Recently,it was reported that KEOPS participates in homologous recombination(HR)repair in yeast.To characterize the KEOPS in archaea(aKEOPS),we conducted genetic and biochemical analyses of its encoding genes in the hyperthermophilic archaeon Saccharolobus islandicus.We show that aKEOPS also possesses five subunits,Pcc1,Kae1,Bud32,Cgi121,and Pcc1-like(or Gon7-like),just like eukaryotic KEOPS.Pcc1-like has physical interactions with Kae1 and Pcc1 and can mediate the monomerization of the dimeric subcomplex(Kae1-Pcc1-Pcc1-Kae1),suggesting that Pcc1-like is a functional homolog of the eukaryotic Gon7 subunit.Strikingly,none of the genes encoding aKEOPS subunits,including Pcc1 and Pcc1-like,can be deleted in the wild type and in a t^(6)A modification complementary strain named TsaKI,implying that the aKEOPS complex is essential for an additional cellular process in this archaeon.Knock-down of the Cgi121 subunit leads to severe growth retardance in the wild type that is partially rescued in TsaKI.These results suggest that aKEOPS plays an essential role independent of the cellular t^(6)A modification level.In addition,archaeal Cgi121 possesses dsDNA-binding activity that relies on its tRNA 3ʹCCA tail binding module.Our study clarifies the subunit organization of archaeal KEOPS and suggests an origin of eukaryotic Gon7.The study also reveals a possible link between the function in t^(6)A modification and the additional function,presumably HR.Pengju Wu Qi Gan Xuemei Zhang Yunfeng Yang Yuanxi Xiao Qunxin She Jinfeng Ni Qihong Huang Yulong Shen 2023mLife2023,2,1:0
10Identification of optimal reference genes in golden Syrian hamster with ethanol-and palmitoleic acid-induced acute pancreatitis using quantitative real-time polymerase chain reaction显示文摘Background:Acute pancreatitis(AP)is a severe disorder that leads to high morbidity and mortality.Appropriate reference genes are important for gene analysis in AP.This study sought to study the expression stability of several reference genes in the golden Syrian hamster,a model of AP.Methods:AP was induced in golden Syrian hamster by intraperitoneal injection of ethanol(1.35 g/kg)and palmitoleic acid(2 mg/kg).The expression of candidate genes,including Actb,Gapdh,Eef2,Ywhaz,Rps18,Hprt1,Tubb,Rpl13a,Nono,and B2m,in hamster pancreas at different time points(1,3,6,9,and 24 h)posttreatment was analyzed using quantitative polymerase chain reaction.The expression stability of these genes was calculated using Best Keeper,Comprehensive Delta CT,Norm Finder,and ge Norm algorithms and Ref Finder software.Results:Our results show that the expression of these reference genes fluctuated during AP,of which Ywhaz and Gapdh were the most stable genes,whereas Tubb,Eef2,and Actb were the least stable genes.Furthermore,these genes were used to normalize the expression of TNF-αmessenger ribonucleic acid in inflamed pancreas.Conclusions:In conclusion,Ywhaz and Gapdh were suitable reference genes for gene expression analysis in AP induced in Syrian hamster.Jinxin Miao Le Kang Tianfeng Lan Jianyao Wang Siqing Wu Yifan Jia Xia Xue Haoran Guo Pengju Wang Yan Li 2023Animal Models and Experimental Medicine2023,6,6:0
11Effects of substrate types on the transformation of heavy metal speciation and bioavailability in an anaerobic digestion system显示文摘Chemical speciation can fundamentally affect the potential toxicity and bioavailability of heavy metals.The transformation of heavy metal speciation and change of bioavailability were investigated in an anaerobic digestion(AD)system using four different substrates(pig manure(PM),cattle manure(CAM),chicken manure(CHM)and rice straw(RS)).The results obtained indicated that the total contents of heavy metals in PM,CHM and GAM were higher than in RS and decreased in the order Zn>Cu>Ni>Pb>As>Cd in all substrates.Moreover,the speciation with the largest proportion for each heavy metal was the same both in the different substrates and the biogas residues.Among them,Zn,Ni,Cd and As were mainly in the reducible fraction(F2),while Cu was mainly in the oxidizable fraction(F3)and Pb occurred predominantly in the residual fraction(F4).Our results further indicated that the AD process had a greater effect on the speciation of heavy metals in CHM and PM,but less on CAM and RS.The rates of change in bioavailability followed the order PM>CHM>CAM>RS.Changes in organic matter,humic acid or local metal ion environment as a result of AD were inferred as likely mechanisms leading to the transformation of heavy metal speciation.These results enhanced our understanding of the behavior of heavy metals in AD and provided a new perspective for the treatment and disposal of the substrates.Xiarong Zheng Kuanhuai Wu Pengju Sun Siyu Zhouyang Yuanpeng Wang Haitao Wang Yanmei Zheng Qingbiao Li 2021Journal of Environmental Sciences2021,33,3:0
12Nanocellulose-based reusable liquid metal printed electronics fabricated by evaporation-induced transfer printing显示文摘Reusable electronics have received widespread attention and are urgently needed. Here, nanocellulosebased liquid metal(NC-LM) printed circuit has been fabricated by the evaporation-induced transfer printing technology. In this way, the liquid metal pattern is embedded into the nanocellulose membrane, which is beneficial for the stability of the circuit during use. Besides, the NC-LM circuit is ultrathin with just tens of microns. In particular, the finished product is environmentally friendly because it can be completely dissolved by water, and both the liquid metal ink and the nanocellulose membrane can be easily recollected and reused, thereby reducing waste and pollution to the environment. Several examples of flexible circuits have been designed to evaluate their performance. The mechanism of evaporation-induced transfer printing technology involves the deposition, aggregation, and coverage tightly of the nanosized cellulose fibrils as the water evaporated.
