1Age-related rhesus macaque models of COVID-19显示文摘Background:Since December 2019,an outbreak of the Corona Virus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus(SARS-CoV-2)in Wuhan,China,has become a public health emergency of international concern.The high fatality of aged cases caused by SARS-CoV-2 was a need to explore the possible age-related phenomena with non-human primate models.Methods:Three 3-5 years old and two 15 years old rhesus macaques were intratracheally infected with SARS-CoV-2,and then analyzed by clinical signs,viral replication,chest X-ray,histopathological changes and immune response.Results:Viral replication of nasopharyngeal swabs,anal swabs and lung in old monkeys was more active than that in young monkeys for 14 days after SARS-CoV-2 challenge.Monkeys developed typical interstitial pneumonia characterized by thickened alveolar septum accompanied with inflammation and edema,notably,old monkeys exhibited diffuse severe interstitial pneumonia.Viral antigens were detected mainly in alveolar epithelial cells and macrophages.Conclusion:SARS-CoV-2 caused more severe interstitial pneumonia in old monkeys than that in young monkeys.Rhesus macaque models infected with SARS-CoV-2 provided insight into the pathogenic mechanism and facilitated the development of vaccines and therapeutics against SARS-CoV-2 infection.Pin Yu Feifei Qi Yanfeng Xu Fengdi Li Peipei Liu Jiayi Liu Linlin Bao Wei Deng Hong Gao Zhiguang Xiang Chong Xiao Qi Lv Shuran Gong Jiangning Liu Zhiqi Song Yajin Qu Jing Xue Qiang Wei Mingya Liu Guanpeng Wang Shunyi Wang Haisheng Yu Xing Liu Baoying Huang Wenling Wang Li Zhao Huijuan Wang Fei Ye Weimin Zhou Wei Zhen Jun Han Guizhen Wu Qi Jin Jianwei Wang Wenjie Tan Chuan Qin 2020Animal Models and Experimental Medicine2020,3,1:22
2Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 4Sequential infection with H1N1 and SARS-CoV-2 aggravated COVID-19 pathogenesis in a mammalian model, and covaccination as an effective method of prevention of COVID-19 and influenza显示文摘Influenza A virus may circulate simultaneously with the SARS-CoV-2 virus,leading to more serious respiratory diseases during this winter.However,the influence of these viruses on disease outcome when both influenza A and SARS-CoV-2 are present in the host remains unclear.Using a mammalian model,sequential infection was performed in ferrets and in K18-MCE2 mice,with SARS-CoV-2 infection following H1N1.We found that co-infection with H1N1 and SARS-CoV-2 extended the duration of clinical manifestation of COVID-19,and enhanced pulmonary damage,but reduced viral shedding of throat swabs and viral loads in the lungs of ferrets.Moreover,mortality was increased in sequentially infected mice compared with single-infection mice.Compared with singlevaccine inoculation,co-inoculation of PiCoVacc(a SARS-CoV-2 vaccine)and the flu vaccine showed no significant differences in neutralizing antibody titers or virus-specific immune responses.Combined immunization effectively protected K18-MCE2 mice against both H1N1 and SARS-CoV-2 infection.Our findings indicated the development of systematic models of co-infection of H1N1 and SARS-CoV-2,which together notably enhanced pneumonia in ferrets and mice,as well as demonstrated that simultaneous vaccination against HINT and SARS-CoV-2 may be an effective prevention strategy for the coming winter.Linlin Bao Wei Deng Feifei Qi Qi Lv Zhiqi Song Jiangning Liu Hong Gao Qiang Wei Pin Yu Yanfeng Xu Yajin Qu Fengdi Li Jing Xue Shuran Gong Mingya Liu Guanpeng Wang Shunyi Wang Binbin Zhao Bin Cong Chuan Qin 2021Signal Transduction and Targeted Therapy2021,6,6:4

6Encoding methods matching the 16×16 pixel CZT detector of a coded aperture gamma camera显示文摘With the advancements in nuclear energy,methods that can accurately obtain the spatial information of radioactive sources have become essential for nuclear energy safety.Coded aperture imaging technology is widely used because it provides two-dimensional distribution information of radioactive sources.The coded array is a major component of a coded aperture gamma camera,and it affects the key performance parameters of the camera.Currently,commonly used coded arrays such as uniformly redundant arrays(URAs)and modified uniformly redundant arrays(MURAs)have prime numbers of rows or columns and may lead to wastage of detector pixels.A 16×16 coded array was designed on the basis of an existing 16×16 multi-pixel position-sensitive cadmium zinc telluride detector.The digital signal-to-noise(SNR)ratio of the point spread function at the center of the array is 25.67.Furthermore,Monte Carlo camera models and experimental devices based on rank-13 MURA and rank-16 URA have been constructed.With the same angular resolution,the field size of view under rank-16 URA is 1.53 times that of under rank-13 MURA.Simulations(Am-241,Co-57,Ir-192,Cs-137)and experiments(Co-57)are conducted to compare the imaging performance between rank-16 URA and rank-13 MURA.The contrast-to-noise ratio of the reconstructed image of the rank-16 array is great and only slightly lower than that of rank-13 MURA.However,as the photon energy increases,the gap becomes almost negligible.Xiao-Lei Shen Pin Gong Xiao-Bin Tang Rui Zhang Jin-Chao Ma 2020Nuclear Science and Techniques2020,31,9:2

8Peptide-RNA complexation-induced fluorescence“turn on”displacement assay for the recognition of small ligands targeting HIV-1 RNA显示文摘The regulator of expression of virion(Rev)protein binds specifically to the Rev-responsive element(RRE)RNA in order to regulate the expression of the human immunodeficiency virus(HIV)-1 genes.Fluorescence indicator displacement assays have been used to identify ligands that can inhibit the ReveRRE interaction;however,the small fluorescence indicators cannot fully replace the Rev peptide or protein.As a result,a single rhodamine B labeled Rev(RB-Rev)model peptide was utilized in this study to develop a direct and efficient ReveRRE inhibitor screening model.Due to photon-induced electron transfer quenching of the tryptophan residue on the RB fluorophore,the fluorescence of RB in Rev was weakened and could be dramatically reactivated by interaction with RRE RNA in ammonium acetate buffer(approximately six times).The interaction could reduce the electron transfer between tryptophan and RB,and RRE could also increase RB fluorescence.The inhibitor screening model was evaluated using three known positive ReveRRE inhibitors,namely,proflavin,6-chloro-9-[3-(2-chloroethylamino)propylamino]-2-methoxyacridine(ICR 191),and neomycin,as well as a negative drug,arginine.With the addition of the positive drugs,the fluorescence of the ReveRRE decreased,indicating the displacement of RB-Rev.This was confirmed using atomic force microscopy(AFM)and the fluorescence was essentially unaffected by the addition of arginine.The results demonstrated that RB-Rev can be used as a fluorescent probe for recognizing small ligands that target RRE RNA.The ReveRRE inhibitor screening model offers a novel approach to evaluating and identifying long-acting Rev inhibitors.Liang Qi Jiayun Zhang Ying Gao Pin Gong Chengyuan Liang Yao Su Qiao Zeng Yafeng Zhang 2022Journal of Pharmaceutical Analysis2022,12,6:0
9Spectrometry analysis based on approximation coefficients and deep belief networks显示文摘A method of spectrometry analysis based on approximation coefficients and deep belief networks was developed. Detection rate and accurate radionuclide identification distance were used to evaluate the performance of the proposed method in identifying radionuclides. Experimental results show that identification performance was not affected by detection time, number of radionuclides, or detection distance when the minimum detectable activity of a single radionuclide was satisfied. Moreover, the proposed method could accurately predict isotopic compositions from the spectra of moving radionuclides. 10Synthesis and Characterization of Dipolar Two-Photon Absorption Chromophores显示文摘 It is a new interest to explore novel organic two-photon absorption (TPA) chromophores with large TPA cross section for their potential applications in various fields, such as three-dimensional optical data storage, micro- and nano-fabrication, biological imaging and optical limiting, and so on. As far as asymmetrical D-π-A type chromophores were concerned, larger TPA cross section can be obtained with the enhancement of electron-donation/electron-acception ability.[1] Recently, we have synthesized a series of new D-π-A type compounds. In these chromophores, stilbene or phenyl cyclo-bridged hexatriene is served as π-bridge, which terminated with a number of amino groups and dicyanomethylene unit (Scheme 1). All the chromophores can be obtained by the condensation reaction of aldehydes and active methyl or methenyl groups in good yields (72%~81%). The chromophores have been characterized by 1H NMR,IR, UV-vis, PL and EA. All these five chromophores in dilute solutions show strong red fluorescence (Table 1).
