| 1 | Asphaltenes: Separations, structural analysis and applications显示文摘Asphaltenes, complex aromatic compounds from various carbonaceous sources, could be obtained by solvent dissolution, filtration and adsorption. It was difficult to clarify the molecular structures and chemical properties of asphaltene due to its structural similarity, composition complexity and source dependences. Many techniques, like Mass spectrometry, Chromatography, Nuclear magnetic resonance spectroscopy, Infrared spectroscopy, Roman spectroscopy, Fluorescence spectroscopy, X-ray diffraction analysis and Small-angle scattering techniques and so on, have revealed some valuable descriptions of asphaltenes chemical compositions and fundamental structures. Moreover, advanced Mass spectrometry,Atomic force microscopy and Scanning tunneling microscopy could provide more clear and essential molecular compounds and structures in asphaltenes. In addition, several asphaltenes models have succeeded to illustrate aggregation properties asphaltene. In this work, the progress on asphaltene separation, characterization and application was summarized, and the similarities and differences between coal-derived asphaltenes and petroleum asphaltenes were compared. Furthermore, the reactivity of asphaltenes has been discussed in the aspect of hydroprocessing, pyrolysis and gasification. Asphaltene was excellent carbon precursor for functional carbon material due to its high aromaticity and carbon yield;several porous carbon nanosheets from asphaltenes that would be prospective electrode materials after being graphitized were shown. Pitch-based carbon fiber derived from coal-derived asphaltenes displayed a tensile strength of 1.0 GPa and elastic modulus of 350 MPa, respectively. | 6 | Detection of Target Genes in Viable Bacteria and Extracellular DNA Using Loop-Mediated Isothermal Amplification Assay显示文摘When the loop-mediated isothermal amplification(LAMP)assay is used for detecting target genes,DNA extraction is unnecessary in many cases.Simple pretreatment(e.g.heating)is enough to obtain rather sensitive responses.Even test samples without any pretreatment can be used as template.This feature suggests that LAMP is superior to PCR in developing point-of-care test strategies.In this study,using Stx1 gene from E.coli as model,we verified that viable cells,dead cells and extracellular DNA could function as template in the LAMP assay.In the incubation at 63℃,viable bacteria in the LAMP reaction mixture lysed completely within 2 min,providing DNA template for nucleic acid amplification.The Stx1 gene in diluted culture medium,spiked tap water,spiked seawater and real seawater all could be detected,with or without the step of DNA extraction.We found that the complex substances in real sample(e.g.natural seawater)exhibited considerable inhibitory effect on the sensitivity of the LAMP assay.These outcomes are meaningful for building a point-of-care strategy by employing the LAMP assay for environmental monitoring,bio-resource surveys,food safety,etc.in particular those based on environmental DNA. | YANG Qianqian ZHANG Xuzhi JIANG Xiaoyu LI Yang ZHAO Jun HAO Zhihui WANG Pingping QU Keming | 2020 | 渔业科学进展2020,41,2: | 0 |
| 7 | Developmental competence and ultrastructural changes of heat-stressed mouse early blastocysts produced in vitro显示文摘Mouse early blastocysts were exposed to temperatures of 39℃ and 41℃ for 2 h,respectively,to determine their developmental competence and ultrastructural changes. The results showed that heat stress at 41℃ for 2 h,significantly reduced the percentages of expanded and hatched blastocysts,but not at 39℃ for 2 h. The average cell numbers in expanded blastocysts,which developed from early blastocysts heat-stressed at temperatures of 39℃ and 41℃,were significantly reduced. The average cell numbers in hatched blastocysts subjected to heat stress were no different from those in the control group cultured at 37℃. The mitochondria of the early blastocysts heat-stressed at 39℃ for 2 h,were slightly swollen,but they had recovered after culturing at 37℃ for 2 h. However,the mitochondria in the blastocysts heat stressed at 41℃ for 2 h were severely swollen,and their number increased. The ribosomes shed from the rough endoplasmic reticulum,and the number of secondary lysosomes in the plasma increased. The integrity of desmosomes was disrupted. The space between the nuclear envelope and the perivitelline membrane enlarged. The fibre fraction and the particulate fraction of nucleoli were separated. The heterochromatin in nucleoli was also increased in its quantity. There were some lamellar-shape structures and heterogeneous dense materials exhibiting in the cytoplasm. The ultrastructural changes induced by heat shock at 41℃ for 2 h were not reversible. | 9 | High-throughput measurements of ciprofloxacin, clomipramine and fexofenadine hydrochlorides with an 8-channel electrical titrator显示文摘High-throughput measurements of ciprofloxacin, clomipramine and fexofenadine hydrochlorides were performed by employing an automatic 8-channel electrical titrator. Silver nitrate(AgNO_3) and sodium tetraphenylborate(NaTPB) were used as titrants. When AgNO_3 was used for measuring the drugs in pure form, recoveries were 97.6%–102.0% with RSD values 1.0%; for measuring them in pharmaceutical formulations, recoveries were 96.6%–99.1% with RSD values 1.0%. Batch samples of eight could be measured simultaneously and maximally 30 measurements per minute could be completed. When Na TPB was used for measuring the drugs in pure form, the recoveries were 96.8%–102.6% with RSD values 0.8%; for measuring them in pharmaceutical formulations, the recoveries were 97.5%–102.7% with RSD values 0.9%. For all analyses, no auxiliary devices or chemicals were needed and there was no requirement for changing or cleaning working electrodes between measurements.
