1Inhibiting effect of antisense oligonucleotides phosphorthioate on gene expression of TIMP-1 in rat liver fibrosis显示文摘AIM To observe the inhibition of antisenseoligonucleotides (asON) phosphorthioate to thetissue inhibitors metalloproteinase-1 (TIMP-1)gene and protein expression in the liver tissue ofimmunologically induced hepatic fibrosis rats.The possibility of reversing hepatic fibrosisthrough gene therapy was observed.METHODS Human serum albumin (HSA) wasused to attack rats, as hepatic fibrosis model, inwhich asONs were used to block the gene andprotein expressing TIMP-1. According to theanalysis of modulator, structure protein, codingseries of TIMP-1 genome, we designed fourdifferent asONs. These asONs were injected intothe hepatic fibrosis models through coccygealvein. The results was observed by RT-PCR formeasuring TIMP-1 mRNA expression,immunohistochemistry and in situ hybridizationfor collagen Ⅰ, Ⅲ, special staining of collagenfiber, and electron microscopic examination.RESULTS Hepatic fibrosis could last within 363days in our modified model. The expressinglevel of TIMP-1 was high during hepatic fibrosisprocess. It has been proved by theimmunohistochemical and the electronmicroscopic examination that the asONphosphorthioate of TIMP-1 could exactly expressin vivo. The effect of colchicine wasdemonstrated to inhibit the expressing level ofmRNA and the content of collagen Ⅰ, Ⅲ in theliver of experimental hepatic fibrosis rats.However, the electron microscopy research andthe pathologic grading of hepatic fibrosisshowed that there was no significant differencebetween the treatment group and the modelgroup (P>0.05).CONCLUSION The experimental rat model ofhepatic fibrosis is one of the preferable modelsto estimate the curative effect of anti-hepaticfibrosis drugs. The asON phosphorthioate ofTIMP-1 could block the gene and proteinexpression of TIMP-1 in the liver of experimentalhepatic fibrosis rats at the mRNA level. It ispossible to reverse hepatic fibrosis, and it isexpected to study a new drug of anti-hepaticfibrosis on the genetic level. 4Paeoniflorin Attenuates Myocardial Fibrosis in Isoprenaline-induced Chronic Heart F ailure Rats via Inhibiting P38 MA PK Pathway显示文摘Paconiflorin(Pae)is a monoterpenoid glycoside compound and has many biological activitics,such as immunosuppression,anti-inflammation and anti-cell proliferation.However,the effects and mechanisms of Pae on chronic heart failure(CHF)remain unclear.This study was conducted to assess the effects and mechanisms of Pae on myocardial fibrosis in isoprenaline(Iso)-induced CHF rats.Pae(20 mgkg)was intragastrically administrated to CHF rats for 6 weeks.Cardiac structure and function were assessed.The protein and mRNA levels of transforming growth factorβ1(TGF-β1)and p38 were detected.C ompared to Iso group,Pae could alleviate myocardial fbrosis and improve cardiac function in CHF rats.The levels of collagen volume fraction(13.75%+3.77%vs.30.97%+4.22%,P<0.001)and perivascular collagen volume area(14.32%+2.50%v8.28.31%+3.16%,P<0.001)were significantly reduced in Pae group as compared with those in Iso group.The expression of TGF-BI protein(0.30+0.07 vs.0.66+0.07,P<0.05)and mRNA(3.51+0.44 vs.7.58+0.58,P<0.05)decreased significantly in Pac group as compared with that in Iso group.The expression of p38 protein(0.36+0.12 vs.0.81+0.38,P<0.05)and mRNA(3.84+0.05 vs.4.40+0.17,P<0.05)also decreased markedly in Pae group as compared with that in Iso group.Pae could attenuate myocardial fibrosis and improve cardiac function in CHF rats by down-regulating the p38 MAPK signaling pathway.Mao LIU Jie FENG Qian DU Jiao AI Zhan LV 2020Current Medical Science2020,40,2:11
5Multicenter study of the clinicopathological features and recurrence risk prediction model of early-stage breast cancer with low-positive human epidermal growth factor receptor 2 expression in China (Chinese Society of Breast Surgery 021)显示文摘Background:Breast cancer with low-positive human epidermal growth factor receptor 2(HER2)expression has triggered further refinement of evaluation criteria for HER2 expression.We studied the clinicopathological features of early-stage breast cancer with low-positive HER2 expression in China and analyzed prognostic factors.Methods:Clinical and pathological data and prognostic information of patients with early-stage breast cancer with low-positive HER2 expression treated by the member units of the Chinese Society of Breast Surgery and Chinese Society of Surgery of Chinese Medical Association,from January 2015 to December 2016 were collected.The prognostic factors of these patients were analyzed.Results:Twenty-nine hospitals provided valid cases.From 2015 to 2016,a total of 25,096 cases of early-stage breast cancer were treated,7642(30.5%)of which had low-positive HER2 expression and were included in the study.After ineligible cases were excluded,6486 patients were included in the study.The median follow-up time was 57 months(4-76 months).The disease-free survival rate was 92.1%at 5 years,and the overall survival rate was 97.4%at 5 years.At the follow-up,506(7.8%)cases of metastasis and 167(2.6%)deaths were noted.Multivariate Cox regression analysis showed that tumor stage,lymphvascular invasion,and the Ki67 index were related to recurrence and metastasis(P<0.05).The recurrence risk prediction model was established using a machine learning model and showed that the area under the receiving operator characteristic curve was 0.815(95%confidence interval:0.750-0.880).Conclusions:Early-stage breast cancer patients with low-positive HER2 expression account for 30.5%of all patients.Tumor stage,lymphvascular invasion,and the Ki67 index are factors affecting prognosis.The recurrence prediction model for breast cancer with low-positive HER2 expression based on a machine learning model had a good clinical reference value for predicting the recurrence risk at 5 years.Trial registration:ChiCTR.org.cn,ChiCTR2100046766.Ling Xin Qian Wu Chongming Zhan Hongyan Qin Hongyu Xiang Ling Xu Jingming Ye Xuening Duan Yinhua Liu Chinese Society of Breast Surgery(CSBrS) Chinese Society of Surgery of Chinese Medical Association 2022Chinese Medical Journal2022,,6:9

7Successful Production of an All-Female Common Carp (Cyprinus carpio L.) Population Using cyp17a1-Deficient Neomale Carp显示文摘Due to sexual dimorphism in the growth of certain cultured fish species,the production of monosex fishes is desirable for the aquaculture industry.Nowadays,the most widely practiced technique available for the mass production of monosex fish populations is sex steroid-induced sex reversal.Here,a novel strategy for the successful production of all-female(AF)common carp(Cyprinus carpio L.),to take advantage of the sexual dimorphism in growth documented in this species,has been developed using genetic engineering via single gene-targeting manipulation without any exogenous hormone treatments.Male and female heterozygous cyp17a1-deficient common carp were first obtained using the clustered regularly interspaced short palindromic repeats/CRISPR-associated endonuclease 9(CRISPR/Cas9)technique.An all-male phenotype for homozygous cyp17a1-deficient carp,regardless of the individuals’sexdetermination genotypes(XY or XX),has been observed.A male-specific DNA marker newly identified in our laboratory was used to screen the neomale carp population with the XX genotype from the cyp17a1-deficient carp.These neomale carp develop a normal testis structure with normal spermatogenesis and sperm capacity.The neomale common carp were then mated with wild-type(WT)females(cyp17a1^(+/+)XX genotype)using artificial fertilization.All the AF offspring sample fish from the neomale-WT female mating were confirmed as having the cyp17a^(1+/-)XX genotype,and normal development of gonads to ovaries was observed in 100.00%of this group at eight months post-fertilization(mpf).A total of 1000 carp fingerlings,500 from the WT male and female and 500 from the neomale and WT female mating,were mixed and reared in the same pond.The average body weight of cyp17a1^(+/-)XX females was higher by 6.60%(8 mpf)and 32.66%(12 mpf)than that of the control common carp.Our study demonstrates the first successful production of a monosex teleost population with the advantages of sexual dimorphism in growth using genetic manipulation targeting a single locus.Gang Zhai Tingting Shu Kuangxin Chen Qiyong Lou Jingyi Jia Jianfei Huang Chuang Shi Xia Jin Jiangyan He Donghuo Jiang Xueqiao Qian Wei Hu Zhan Yin 2022Engineering2022,8,1:7
8EXPERIMENTAL STUDY OF FRICTION FACTOR FOR GROUNDWATER FLOW IN A SINGLE ROUGH FRACTURE显示文摘To study the relationships between the friction factor f and the flow type in a single rough fracture, the formulae of f for both unconfined and confined flows are deduced based on previous studies. The relationships between f and the Reynolds number ( Re ) for different relative roughnesses are investigated experimentally. The Moody-type diagram, based on the deduced formula of f , is also plotted and the hydraulic characteristics of the flow in a rough fracture are analyzed. Results show that the Moody-type diagram of the experiment has a similar distribution to that of the conventional Moody diagram. It is found that the value of f in the experiment is much smaller than that of the conventional Moody diagram and turbulent flow appears easier for rough fractures, which can be explained by the separation phenomenon in boundary layers. The critical Re ranging from 650 to 700 in rough fractures is concluded based on the experimental results. 9Enhancement of the imbibition recovery by surfactants in tight oil reservoirs显示文摘Hydraulic fracturing technology can significantly increase oil production from tight oil formations, but performance data show that production declines rapidly. In the long term, it is necessary to increase the development efficiency of block matrix, surfactant-aided imbibition is a potential way. The current work aimed to explain comprehensively how surfactants can enhance the imbibition rate. Laboratory experiments were performed to investigate the effects of wettability, interfacial tension(IFT), and relative permeability as the key parameters underlying surfactant solution imbibition. Two different types of surfactants, sodium dodecyl sulfate and polyethylene glycol octylphenol ether, at varied concentrations were tested on reservoir rocks. Experimental results showed that the oil recovery rate increased with increased wettability alteration and IFT and decreased residual oil saturation. A mechanistic simulator developed in previous studies was used to perform parametric analysis after successful laboratory-scale validation. Results were proven by parametric studies. 10Influence of lanthanum on enhancement of mechanical and electrical properties of Cu-Al_2O_3 composites显示文摘Two kinds of Cu-Al_2O_3 composites(with and without La) were prepared via mechanical alloying-spark plasma sintering(MA-SPS) method. Microstructure, mechanical properties and electrical resistivity were investigated systematically using metallography, scanning electron microscopy, transmission electron microscopy, mechanical and electrical properties testing. The results indicate that an appropriate amount of La can homogenize the distribution of Al_2O_3. As such, yield strength, ultimate tensile strength and elongation of Cu-Al_2O_3-La are greatly increased. Some semi-coherent interface between Cu and Al_2O_3 is found, which means a low interface energy. The grain shape of Cu changes to irregular band with the addition of La. This change results in a density decrease of grain boundary and reduces electrical resistance. 12The role of substantia nigra sonography in the differentiation of Parkinson’s disease and multiple system atrophy显示文摘Background:The differential diagnosis of Parkinson’s disease(PD)and multiple system atrophy(MSA)remains a challenge,especially in the early stage.Here,we assessed the value of transcranial sonography(TCS)to discriminate non-tremor dominant(non-TD)PD from MSA with predominant parkinsonism(MSA-P).Methods:Eighty-six MSA-P patients and 147 age and gender-matched non-TD PD patients who had appropriate temporal acoustic bone windows were included in this study.All the patients were followed up for at least 2 years to confirm the initial diagnosis.Patients with at least one substantia nigra(SN)echogenic size≥18 mm^(2) were classified as hyperechogenic,those with at least one SN echogenic size≥25 mm^(2) was defined as markedly hyperechogenic.Results:The frequency of SN hyperechogenicity in non-TD PD patients was significantly higher than that in MSA-P patients(74.1%vs.38.4%,p<0.001).SN hyperechogenicity discriminated non-TD PD from MSA-P with sensitivity of 74.1%,specificity of 61.6%,and positive predictive value of 76.8%.If marked SN hyperechogenicity was used as the cutoff value(≥25 mm^(2)),the sensitivity decreased to 46.3%,but the specificity and positive predictive value increased to 80.2 and 80.0%.Additionally,in those patients with SN hyperechogenicity,positive correlation between SN hyperechogenicity area and disease duration was found in non-TD PD rather than in MSA-P patients.In this context,among early-stage patients with disease duration≤3 years,the sensitivity,specificity and positive predictive value of SN hyperechogenicity further declined to 69.8%,52.2%,and 66.7%,respectively.Conclusions:TCS could help discriminate non-TD PD from MSA-P in a certain extent,but the limitation was also obvious with relatively low specificity,especially in the early stage.Hai-Yan Zhou Pei Huang Qian Sun Juan-Juan Du Shi-Shuang Cui Yun-Yun Hu Wei-Wei Zhan Ying Wang Qin Xiao Jun Liu Yu-Yan Tan Sheng-Di Chen 2018Translational Neurodegeneration2018,7,1:6
13The histone H3 lysine-27 demethylase Jmjd3 plays a critical role in specific regulation of Th17 cell differentiation显示文摘Interleukin(IL)17-producing T helper(Th17)cells play critical roles in the clearance of extracellular bacteria and fungi as well as the pathogenesis of various autoimmune diseases,such as multiple sclerosis,psoriasis,and ulcerative colitis.Although a global transcriptional regulatory network of Th17 cell differentiation has been mapped recently,the participation of epigenetic modifications in the differentiation process has yet to be elucidated.We demonstrated here that histone H3 lysine-27(H3K27)demethylation,predominantly mediated by the H3K27 demethylase Jmjd3,crucially regulated Th17 cell differentiation.Activation of naı¨ve CD41 T cells immediately induced high expression of Jmjd3.Genetic depletion of Jmjd3 in CD41 T cells specifically impaired Th17 cell differentiation both in vitro and in vivo.Ectopic expression of Jmjd3 largely rescued the impaired differentiation of Th17 cells in vitro in Jmjd3-deficientCD41 T cells.Importantly,Jmjd3-deficient mice were resistant to the induction of experimental autoimmune encephalomyelitis(EAE).Furthermore,inhibition of the H3K27 demethylase activity with the specific inhibitor GSK-J4 dramatically suppressed Th17 cell differentiation in vitro.At the molecular level,Jmjd3 directly bound to and reduced the level of H3K27 trimethylation(me3)at the genomic sites ofRorc,which encodes the masterTh17 transcription factorRorgt,and Th17 cytokine genes such as Il17,Il17f,and Il22.Therefore,our studies established acritical role of Jmjd3-mediatedH3K27demethylation inTh17 cell differentiation andsuggest that Jmjd3 can be a novel therapeutic target for suppressing autoimmune responses.Zhi Liu Wei Cao Longxia Xu Xi Chen Yu Zhan Qian Yang Sanhong Liu Pengfei Chen Yuhang Jiang Xiaohua Sun Yu Tao Yiming Hu Cuifeng Li Qi Wang Ying Wang Charlie Degui Chen Yufang Shi Xiaoren Zhang 2015Journal of Molecular Cell Biology2015,7,6:6
14A blind watermarking algorithm for 3D mesh models based on vertex curvature显示文摘We propose a robust blind watermarking algorithm for three-dimensional(3D)mesh models based on vertex curvature to maintain good robustness and improve visual masking in 3D mesh models.In the embedding process,by using the local window of vertex,the root mean square curvature is calculated for every vertex of the 3D mesh model and an ordered set of fluctuation values is obtained.According to the ordered fluctuation values,the vertices are separated into bins.In each bin the fluctuation values are normalized.Finally,the mean of the root mean square curvature fluctuation values of the vertices in each bin is modulated to embed watermark information.In watermark detection,the algorithm uses a blind watermark extraction technique to extract the watermark information.The experimental results show that the algorithm has a very good performance for visual masking of the embedded model and that it can resist a variety of common attacks such as vertex rearrangement,rotation,translating,uniform scaling,noise,smoothing,quantization,and simplification.Yong-zhao ZHAN Yan-ting LI Xin-yu WANG Yi QIAN 2014Journal of Zhejiang University-Science C(Computers and Electronics)2014,15,5:6

16Vegetation changes in the agricultural-pastoral areas of northern China from 2001 to 2013显示文摘Climate change and human activity have resulted in increasing change of vegetation growth globally. Numerous studies have been conducted on extreme climate events and analyses of ecological environment evolution. However, such studies have placed little emphasis on vegetation change and spatial variation in this type of ecotone. Accordingly, this study analyzed the changes in vegetation type and growth using the 16-d composite MOD13A1 product with 1-km resolution and MOD12Q1 product with 1-km resolution. We used the mean, maximum, standard deviation normalized-difference vegetation index(NDVI) values, and the rate of change(ROC) of NDVI value to explain vegetation changes within the studied ecotone. Our results showed that significant vegetation type and growth changes have occurred in the study area. From 2001 to 2013, for example, with the exception of 2001, 2004 and 2009, a certain extent of grassland area was converted to cropland. Drought severity index(DSI) results indicate that there exists drought in 2001, 2004 and 2009. Such temporal changes in cropland and grassland area confirmed the ecological vulnerability of the ecotone. At the same time, vegetation varied spatially from west to east and from south to north. 17Research progress on gel polymer electrolytes for lithium-sulfur batteries显示文摘Lithium-sulfur(Li-S)batteries have become a promising candidate for advanced energy storage system owing to low cost and high theoretical specific energy.In the last decade,in pursuit of Li-S batteries with enhanced safety and energy density,the investigation on the electrolytes has leaped form liquid organic electrolytes to solid polymer ones.However,such solid-state Li-S battery system is greatly limited by unfavorable ionic conductivity,poor interfacial contact and narrow electrochemical windows on account of the absence of any liquid components.To address these issues,gel polymer electrolytes(GPEs),the incorporation of liquid electrolytes into solid polymer matrixes,have been newly developed.Although the excellent ionic transport and low interfacial resistance provided by GPEs have prompted numerous researchers to make certain progress on high-performance Li-S coins,a comprehensive review on GPEs for Li-S batteries remains vacant.Herein,this review focuses on recent development and progress on GPEs in view of their physical and chemical properties for the applications in Li-S batteries.Studies on the components including solid hosts,liquid solutions and fillers of GPEs are systematically summarized with particular emphasis on the relationship between components and performance.Finally,current challenges and directional outlook for fabricating GPEs-based Li-S batteries with outstanding performance are outlined.Jie Qian Biyu Jin Yuanyuan Li Xiaoli Zhan Yang Hou Qinghua Zhang 2021Journal of Energy Chemistry2021,30,5:5
18Targeted methylation sequencing reveals dysregulated Wnt signaling in Parkinson disease显示文摘Parkinson disease(PD) is a progressive neurodegenerative movement disorder. Both environmental and genetic factors play important roles in PD etiology. A number of environmental toxins cause parkinsonism in human and animal models. Genetic studies of rare early onset familial PD cases resulted in identification of disease-linked mutations in multiple genes. Nevertheless, the potential interaction between environment and genetics in PD pathogenesis remains largely unknown. We hypothesized that environmental factors induce abnormal epigenetic regulation that is involved in the pathogenesis of both familial and sporadic PD. We determined the global methylation status of 80,000e110,000 Cp G sites in each of the five sporadic PD patient brains and five age and postmodern interval matched control brains utilizing bisulfite padlock sequencing. Multiple genes involved in neurogenesis, particularly the ones in the Wnt signaling pathway, were hypermethylated in PD brains compared to their matched control brains. Consistent with the DNA methylation changes, marked reduction of protein expression was observed for four Wnt and neurogenesis related genes(FOXC1, NEURG2, SPRY1, and CTNNB1) in midbrain dopaminergic(DA) neurons of PD. The treatment of low concentration of 1-methyl-4-phenylpyridinium(MPPt) for cells resulted in downregulation of Wnt related genes. 19Tuning performance of Pd/Sn_(0.9)Ce_(0.1)O_2 catalyst for methane combustion by optimizing calcination temperature of support显示文摘A series of 0.2 wt% Pd/Sn_(0.9)Ce_(0.1)O_2 catalysts were prepared by impregnation method based on the presynthesis of Sn_(0.9)Ce_(0.1)O_2 support prepared by co-precipitation method, and then characterized by Brunauer–Emmett–Teller(BET), X-ray diffraction(XRD), X-ray photoelectron spectroscopy(XPS), Raman, CO chemical adsorption and hydrogen temperature-programmed reduction(H_2-TPR) techniques. The effect of calcination temperature of the composite oxide support on the catalytic performances of the Pd/Sn_(0.9)Ce_(0.1)O_2 catalyst for the CH_4 total oxidation was studied. It is found that the catalytic activity of the Pd/Sn_(0.9)Ce_(0.1)O_2 catalyst increases with the increase in calcination temperature of the Pd/Sn_(0.9)Ce_(0.1)O_2 support. The 0.2 wt% Pd/Sn_(0.9)Ce_(0.1)O_2/1100 catalyst(the Pd/Sn_(0.9)Ce_(0.1)O_2 support was calcined at 1100 ℃) exhibits the best reactive activity(T_(10)= 255 ℃). 20Effects of Structural Changes in Subchondral Bone on Articular Cartilage in a Beagle Dog Model显示文摘Objective Using MR T2-mapping and histopathologic score for articular cartilage to evaluate the effect of structural changes in subchondral bone on articular cartilage. Methods Twenty-four male Beagle dogs were randomly divided into a subchondral bone defect group(n = 12) and a bone cement group(n = 12). Models of subchondral bone defectin the medial tibial plateau and subchondral bone filled with bone cement were constructed. In all dogs, the left knee joint was used as the experimental sideand the right knee as the sham side. The T2 value for articular cartilage at the medial tibial plateau was measured at postoperative weeks 4, 8, 16, and 24. The articular cartilage specimens were stained with hematoxylin and eosin, and evaluated using the Mankin score. Results There was a statistically significant difference(P < 0.05) in Mankin score between the bone defect group and the cement group at postoperative weeks 16 and 24. There was a statistically significant difference in the T2 values between the bone defect group and its sham group(P < 0.05) from week 8, and between the cement group and its sham group(P < 0.05) from week 16. There was significant difference in T2 values between the two experimental groups at postoperative week 24(P < 0.01). The T2 value for articular cartilage was positively correlated with the Mankin score(ρ = 0.758, P < 0.01). Conclusion Structural changes in subchondral bone can lead to degeneration of the adjacent articular cartilage. Defects in subchondral bone cause more severe degeneration of cartilage than subchondral bone filled with cement. The T2 value for articular cartilage increases with the extent of degeneration.
