| 3 | Isolation and characterization of dengue virus serotype 2 from the large dengue outbreak in Guangdong, China in 2014显示文摘Dengue has been well recognized as a global public health threat,but only sporadic epidemics and imported cases were reported in recent decades in China.Since July 2014,an unexpected large dengue outbreak has occurred in Guangdong province,China,resulting in more than 40000 patients including six deaths.To clarify and characterize the causative agent of this outbreak,the acute phase serum from a patient diagnosed with severe dengue was subjected to virus isolation and high-throughput sequencing(HTS).Traditional real-time RT-PCR and HTS with Ion Torrent PGM detected the presence of dengue virus serotype 2(DENV-2).A clinical DENV-2 isolate GZ05/2014 was obtained by culturing the patient serum in mosquito C6/36 cells.The complete genome of GZ05/2014 was determined and deposited in Gen Bank under the access number KP012546.Phylogenetic analysis based on the complete envelope gene showed that the newly DENV-2 isolate belonged to Cosmopolitan genotype and clustered closely with other Guangdong strains isolated in the past decade.No amino acid mutations that are obviously known to increase virulence or replication were identified throughout the genome of GZ05/2014.The high homology of Guangdong DENV-2 strains indicated the possibility of establishment of local DENV-2 circulation in Guangdong,China.These results help clarify the origin of this epidemic and predict the future status of dengue in China. | ZHAO Hui ZHAO Ling Zhai JIANG Tao LI Xiao Feng FAN Hang HONG Wen Xin ZHANG Yu ZHU Qin YE Qing TONG Yi Gang CAO Wu Chun ZHANG Fu Chun QIN Cheng Feng | 2014 | Science China(Life Sciences)2014,57,12: | 21 |
| 4 | Wheel/rail dynamic interaction due to excitation of rail corrugation in high-speed railway显示文摘Characteristics of wheel-rail dynamic interaction due to the rail corrugation in a high-speed railway are analyzed based on the theory of vehicle-track coupled dynamics in this paper.Influences of the corrugation wavelength and depth on the wheel-rail dynamic performance are investigated.The results show that,under the excitation of a measured rail corrugation,the wheel-rail dynamic interaction of high-speed railway is enhanced obviously,and the high-frequency dynamic force between wheel and rail is generated,which has an obvious impact on the vibrations of the wheelset and rail,and little effect on the vibration of the frame and carbody.If the corrugation wavelength is shorter than the sensitive wavelength,the wheel-rail vertical force will increase with the growth of the corrugation wavelength,otherwise,it will decrease.However,the wheel-rail vertical force keeps increasing with the growth of corrugation depth.Furthermore,if the corrugation wavelength is shorter than the sensitive wavelength,the wheel-rail vertical force will increase with the decrease of the running speed,otherwise,it will decrease.It is also found that the critical wavelength of corrugation increases with the growth of the corrugation depth and the running speed,and the critical depth of corrugation is nonlinearly related to the sensitive wavelength. | WANG Kai Yun LIU Peng Fei ZHAI Wan Ming HUANG Chao CHEN Zai Gang GAO JianMin | 2015 | Science China(Technological Sciences)2015,58,2: | 17 |
| 5 | Overlay of a sponge soaked with ropivacaine and multisite infiltration analgesia result in faster recovery after laparoscopic hepatectomy显示文摘BACKGROUND Compared with traditional open surgery,laparoscopic surgery is preferred due to the advantages of less trauma,less pain,and faster recovery.Nevertheless,many patients still suffer from postoperative pain resulting from the surgical incision and associated tissue injury.Many researchers have reported methods to improve postoperative pain control,but there is not a simple and effective method that can be clinically adopted in a widespread manner.We designed this study to prove the hypothesis that application of ropivacaine in the port site and operative site in patients is an effective and convenient method which can decrease postoperative pain and accelerate recovery.AIM To evaluate the effects of ropivacaine on pain control after laparoscopic hepatectomy and its contribution to patient recovery.METHODS From May 2017 to November 2018,146 patients undergoing laparoscopic hepatectomy were randomized to receive infiltration of either 7.5 mg/mL ropivacaine around the trocar insertions,incision,and cutting surface of the liver(with a gelatin sponge soaked with ropivacaine)at the end of surgery(ropivacaine group),or normal saline(5 mL)at the same sites at the end of surgery(control group).The degree of pain,nausea,vomiting,heart rate(HR),and blood pressure were collected.The length of postoperative hospitalization,complications,and the levels of stress hormones were also compared between the two groups.RESULTS Compared with the control group,the ropivacaine group showed reduced postoperative pain at rest within 12 h(P<0.05),and pain on movement was reduced within 48 h.The levels of epinephrine,norepinephrine,and cortisol at 24 and 48 h,HR,blood pressure,and cumulative sufentanil consumption in the ropivacaine group were significantly lower than those in the control group(P<0.05).In the ropivacaine group,hospitalization after operation was shorter,but the difference was not statistically significant.There were no significant differences in postoperative nausea,vomiting,or other complications,including hydrothorax,ascites,peritonitis,flatulence,and venous thrombus(P>0.05),although fewer patients in the ropivacaine group experienced these situations.CONCLUSION Infiltration with ropivacaine in the abdominal wound and covering the cutting surface of the liver with a gelatin sponge soaked with ropivacaine significantly reduce postoperative pain and the consumption of sufentanil. | Hao Zhang Gang Du Yan-Feng Liu Jin-Huan Yang Mu-Guo A-Niu Xiang-Yu Zhai Bin Jin | 2019 | World Journal of Gastroenterology2019,25,34: | 13 |

| 8 | The Large Sky Area Multi-Object Fiber Spectroscopic Telescope (LAMOST)显示文摘The Large Sky Area Multi-Object Fiber Spectroscopic Telescope (LAMOST, also called the Guo Shou Jing Telescope) is a special reflecting Schmidt telescope. LAMOST's special design allows both a large aperture (effective aperture of 3.6 m-4.9 m) and a wide field of view (FOV) (5°). It has an innovative active reflecting Schmidt configuration which continuously changes the mirror's surface that adjusts during the observation process and combines thin deformable mirror active optics with segmented active optics. Its primary mirror (6.67m×6.05 m) and active Schmidt mirror (5.74m×4.40 m) are both segmented, and composed of 37 and 24 hexagonal sub-mirrors respectively. By using a parallel controllable fiber positioning technique, the focal surface of 1.75 m in diameter can accommodate 4000 optical fibers. Also, LAMOST has 16 spectrographs with 32 CCD cameras. LAMOST will be the telescope with the highest rate of spectral acquisition. As a national large scientific project, the LAMOST project was formally proposed in 1996, and approved by the Chinese government in 1997. The construction started in 2001, was completed in 2008 and passed the official acceptance in June 2009. The LAMOST pilot survey was started in October 2011 and the spectroscopic survey will launch in September 2012. Up to now, LAMOST has released more than 480 000 spectra of objects. | 9 | Successful Production of an All-Female Common Carp (Cyprinus carpio L.) Population Using cyp17a1-Deficient Neomale Carp显示文摘Due to sexual dimorphism in the growth of certain cultured fish species,the production of monosex fishes is desirable for the aquaculture industry.Nowadays,the most widely practiced technique available for the mass production of monosex fish populations is sex steroid-induced sex reversal.Here,a novel strategy for the successful production of all-female(AF)common carp(Cyprinus carpio L.),to take advantage of the sexual dimorphism in growth documented in this species,has been developed using genetic engineering via single gene-targeting manipulation without any exogenous hormone treatments.Male and female heterozygous cyp17a1-deficient common carp were first obtained using the clustered regularly interspaced short palindromic repeats/CRISPR-associated endonuclease 9(CRISPR/Cas9)technique.An all-male phenotype for homozygous cyp17a1-deficient carp,regardless of the individuals’sexdetermination genotypes(XY or XX),has been observed.A male-specific DNA marker newly identified in our laboratory was used to screen the neomale carp population with the XX genotype from the cyp17a1-deficient carp.These neomale carp develop a normal testis structure with normal spermatogenesis and sperm capacity.The neomale common carp were then mated with wild-type(WT)females(cyp17a1^(+/+)XX genotype)using artificial fertilization.All the AF offspring sample fish from the neomale-WT female mating were confirmed as having the cyp17a^(1+/-)XX genotype,and normal development of gonads to ovaries was observed in 100.00%of this group at eight months post-fertilization(mpf).A total of 1000 carp fingerlings,500 from the WT male and female and 500 from the neomale and WT female mating,were mixed and reared in the same pond.The average body weight of cyp17a1^(+/-)XX females was higher by 6.60%(8 mpf)and 32.66%(12 mpf)than that of the control common carp.Our study demonstrates the first successful production of a monosex teleost population with the advantages of sexual dimorphism in growth using genetic manipulation targeting a single locus. | Gang Zhai Tingting Shu Kuangxin Chen Qiyong Lou Jingyi Jia Jianfei Huang Chuang Shi Xia Jin Jiangyan He Donghuo Jiang Xueqiao Qian Wei Hu Zhan Yin | 2022 | Engineering2022,8,1: | 7 |
| 10 | Androgen signaling regulates the transcription of anti-Müllerian hormone via synergy with SRY-related protein SOX9A显示文摘Anti-Müllerian hormone(amh) is one of the earliest functional genes expressed during testicular differentiation. It has been suggested that androgen signaling regulates critical genes for the differentiation and development of the testis. To elucidate the exact regulatory mechanisms involved in amh transcription mediated by androgen signaling, androgen signaling was manipulated in zebrafish by cytochrome P45017a1(cyp17a1) knockout and Flutamide treatment. In cyp17a1-deficient and Flutamide-treated testes,up-regulated sry-box9a(sox9a) and down-regulated amh were observed. Moreover, a physical association of the zebrafish androgen receptor(AR) and SOX9 A was found. The interaction between AR and SOX9 A was mediated via the DNA binding domain(DBD) of AR and the transactivation domain(TA) of SOX9 A,and was further enhanced by 5-alpha dihydrotestosterone(DHT), one of the most potent androgens.Intriguingly, together with SOX9 A, androgen signaling synergistically promoted amh transcription,mainly through the proximal 1 kb of the amh promoter region. | 12 | A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. The characteristics of De-NO with plasma and De-NO with plasma-catalyst were investigated comparatively by experiments. | 15 | Hot tearing susceptibility of Fe-20.96Cr-2.13Ni-0.15N-4.76Mn-0.01Mo duplex stainless steel显示文摘The hot tearing susceptibility of a Fe-20.96Cr-2.13Ni-0.15N-4.76Mn-0.01 Mo duplex stainless steel was investigated using method of constrained solidification shrinkage in one dimension.An apparatus for realtime measuring the contraction stress and temperature during solidification was developed,which can achieve the in-situ observation of melting and solidification and avoid the large temperature gradient of casting under the condition of pouring.The results show that the contraction stress increases significantly when the core temperature of casting reaches the liquidus temperature.The contraction stress is released when the core temperature of casting reaches 1456°C.At this temperature,the hot tearing susceptibility of duplex stainless steel is the largest.With decreasing the core temperature to 1363°C,the slope of contraction stress increases,which is related to the ferrite-to-austenite transformation. | Zhi-qiang Wei Xiang-ru Chen Hong-gang Zhong Qi-jie Zhai Gang Wang | 2017 | Journal of Iron and Steel Research(International)2017,24,4: | 3 |

| 17 | The first data release(DR1) of the LAMOST regular survey显示文摘The Large sky Area Multi-Object Fiber Spectroscopic Telescope(LAMOST) general survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects in both the pilot survey and the first year regular survey are included in the LAMOST DR1. The pilot survey started in October 2011 and ended in June 2012, and the data have been released to the public as the LAMOST Pilot Data Release in August 2012. The regular survey started in September 2012, and completed its first year of operation in June 2013. The LAMOST DR1 includes a total of 1202 plates containing 2 955 336 spectra, of which 1 790 879 spectra have observed signalto-noise ratio(SNR) ≥ 10. All data with SNR ≥ 2 are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2 204 696 spectra, of which 1 944 329 are stellar spectra, 12 082 are galaxy spectra and 5017 are quasars. The DR1 not only includes spectra, but also three stellar catalogs with measured parameters: late A,FGK-type stars with high quality spectra(1 061 918 entries), A-type stars(100 073 entries), and M-type stars(121 522 entries). This paper introduces the survey design, the observational and instrumental limitations, data reduction and analysis, and some caveats. | 18 | Prevalence of and risk factors for diabetic macular edema in a northeastern Chinese population显示文摘AIM:To estimate the prevalence of diabetic macular edema(DME) and clinically significant macular edema(CSME),and to assess their risk factors in a population with type 2 diabetic mellitus(T2DM) located in northeast China.METHODS:Patients were included from the Fushun Diabetic Retinopathy Cohort Study(FS-DIRECT),a community-based study conducted in northeast China.The presence of DME and CSME was determined by the Early Treatment Diabetic Retinopathy Study(ETDRS) retinopathy scale of fundus photographs.The age-standardized prevalence of DME and CSME was estimated.The association between DME/CSME and risk factors was analyzed in a multivariate Logistical analysis.RESULTS:A total of 292(15.4%) and 166(8.8%) patients were diagnosed as DME and CSME,yielding the age and sex standardized prevalence of 13.5%(95%CI:11.9%-15.0%),and 7.1%(95%CI:5.9%-8.3%),respectively.Female patients had a higher prevalence of DME compared to their male counterparts(15.7% vs 10.4%,P=0.03).Multivariable Logistic regression analysis showed that younger age,insulin use,proteinuria,longer duration of diabetes,and higher glycosylated hemoglobin A1c,were associated with the prevalence of DME and CSME.Patients with higher fasting plasma glucose,systolic blood pressure,and blood urea nitrogen were also found to be associated with DME.CONCLUSION:Early fundus screening in diabetic patients is invaluable and given the relatively high prevalence of DME and CSME in this study cohort,those with a high risk of sight threatening maculopathy would invariably benefit from earlier detection. | Zhong Lin Feng-Hua Wang Liang Wen Yu Wang Dong Li Xiao-Xia Ding Yu Dou Gang Zhai Yuan-Bo Liang | 2022 | International Journal of Ophthalmology(English edition)2022,15,2: | 2 |
