2Latent fingerprint enhancement on conductive substrates using electrodeposition of copper显示文摘We presented a novel method for the development of a latent fingerprint by selective electrodeposition of a copper thin film from sulfate solution onto the conductive substrate between fingerprint ridges to generate a negative image of the fingerprint deposit. After optimizing the parameters(deposition time, deposition potential, and copper concentration), the preferential electrodeposition of copper films allowed latent fingerprints on six kinds of conductive surfaces(indium/tin oxide-coated glass, silver sheet, platinum sheet, gold sheet, copper sheet, and a stainless steel coin) to be successfully developed with high resolution. In addition, this technique could also be exploited to visualize latent fingerprints on rough and dirty surfaces. 5Evaluation of Cell Type Annotation R Packages on Single-cell RNA-seq显示文摘Annotating cell types is a critical step in single-cell RNA sequencing(scRNA-seq)data analysis.Some supervised or semi-supervised classification methods have recently emerged to enable automated cell type identification.However,comprehensive evaluations of these methods are lacking.Moreover,it is not clear whether some classification methods originally designed for analyzing other bulk omics data are adaptable to scRNA-seq analysis.In this study,we evaluated ten cell type annotation methods publicly available as R packages.Eight of them are popular methods developed specifically for single-cell research,including Seurat,scmap,SingleR,CHETAH,SingleCellNet,scID,Garnett,and SCINA.The other two methods were repurposed from deconvoluting DNA methylation data,i.e.,linear constrained projection(CP)and robust partial correlations(RPC).We conducted systematic comparisons on a wide variety of public scRNA-seq datasets as well as simulation data.We assessed the accuracy through intra-dataset and inter-dataset predictions;the robustness over practical challenges such as gene filtering,high similarity among cell types,and increased cell type classes;as well as the detection of rare and unknown cell types.Overall,methods such as Seurat,SingleR,CP,RPC,and SingleCellNet performed well,with Seurat being the best at annotating major cell types.Additionally,Seurat,SingleR,CP,and RPC were more robust against downsampling.However,Seurat did have a major drawback at predicting rare cell populations,and it was suboptimal at differentiating cell types highly similar to each other,compared to SingleR and RPC.All the code and data are available from http://gffzz188fe103f8f1460asofwv6kcqccw06wuk.ffgz.tsg.suse.edu.cn/qianhuiSenn/scRNA_cell_deconv_benchmark.Qianhui Huang Yu Liu Yuheng Du Lana X.Garmire 2021Genomics, Proteomics & Bioinformatics2021,19,2:1
6Housing Market Speculation and Firm Productivity:Evidence from China显示文摘An appreciation in the price of housing could generate speculation opportunities for enterprises.Many firms try to seize these opportunities for quick returns.This may harm their productivity.We use the real estate boom and the investment activities of Chinese manufacturing firms during 2006–2017 to examine the processes and the net effect of housing market speculation on firm productivity.We find that housing market speculation negatively affects firm productivity by crowding out investments in research and development and productive fixed assets,and the positive effect brought about by a strengthened balance sheet and better financing capacity is outweighed by the crowding-out effect.This negative impact is more evident in state-owned firms,but is mitigated in regions and periods with better business environments.Set in the overheated housing market in China,our study contributes to the existing literature by exploring how firms’investments outside of their main business influence firm behavior and productivity.Qianhui Yu Yanying Chen Feng Helen Liang 2021China & World Economy2021,29,5:0
7MFSD7c functions as a transporter of choline at the blood–brain barrier显示文摘Mutations in the orphan transporter MFSD7c(also known as Flvcr2),are linked to Fowler syndrome.Here,we used Mfsd7c knockout(Mfsd7c–/–)mice and cell-based assays to reveal that MFSD7c is a choline transporter at the blood–brain barrier(BBB).We performed comprehensive metabolomics analysis and detected differential changes of metabolites in the brains and livers of Mfsd7c–/–embryos.Particularly,we found that choline-related metabolites were altered in the brains but not in the livers of Mfsd7c–/–embryos.Thus,we hypothesized that MFSD7c regulates the level of choline in the brain.Indeed,expression of human MFSD7c in cells significantly increased choline uptake.Interestingly,we showed that choline uptake by MFSD7c is greatly increased by choline-metabolizing enzymes,leading us to demonstrate that MFSD7c is a facilitative transporter of choline.Furthermore,single-cell patch clamp analysis showed that the import of choline by MFSD7c is electrogenic.Choline transport function of MFSD7c was shown to be conserved in vertebrates,but not in yeasts.We demonstrated that human MFSD7c is a functional ortholog of HNM1,the yeast choline importer.We also showed that several missense mutations identified in patients exhibiting Fowler syndrome had abolished or reduced choline transport activity.Mice lacking Mfsd7c in endothelial cells of the central nervous system suppressed the import of exogenous choline from blood but unexpectedly had increased choline levels in the brain.Stable-isotope tracing study revealed that MFSD7c was required for exporting choline derived from lysophosphatidylcholine in the brain.Collectively,our work identifies MFSD7c as a choline exporter at the BBB and provides a foundation for future work to reveal the disease mechanisms of Fowler syndrome.Xuan Thi Anh Nguyen Thanh Nha Uyen Le Toan Q.Nguyen Hoa Thi Thuy Ha Anna Artati Nancy C.P.Leong Dat T.Nguyen Pei Yen Lim Adelia Vicanatalita Susanto Qianhui Huang Ling Fam Lo Ngah Leong Isabelle Bonne Angela Lee Jorge L.Granadillo Catherine Gooch Dejie Yu Hua Huang Tuck Wah Soong Matthew Wook Chang Markus R.Wenk Jerzy Adamski Amaury Cazenave-Gassiot Long N.Nguyen 2024Cell Research2024,34,3:0

12COATS:Comprehensive observation on the atmospheric boundary layer three-dimensional structure during haze pollution in the North China Plain显示文摘The North China Plain(NCP)is troubled by severe haze pollution and the evolution of haze pollution is closely related to the atmospheric boundary layer(ABL).However,experimental and theoretical studies on the physical-chemical processes of the ABL in the NCP are lacking,with many scientific problems to be addressed.To solve these problems,the Comprehensive Observation on the Atmospheric boundary layer Three-dimensional Structure(COATS)during haze pollution was carried out in the NCP from 2016 to 2020.The COATS experiment adopted a'point-line-surface'spatial layout,obtaining both spatial-temporal profiles of the meteorological and environmental elements in the ABL and the turbulent transport data of fine particulate matter(PM_(2.5))in winter and summer.The research achievements are as follows.The spatial-temporal distribution characteristics of the ABL structure and PM_(2.5)concentrations in NCP were determined.The typical thermal structure of persistent heavy haze events and the pollutant removal mechanism by low-level jets were revealed.It was determined that the spatial structure of the ABL adjusted by the Taihang Mountains is responsible for the heterogeneous distribution of haze pollution in the NCP,and that mountain-induced vertical circulations can promote the formation of elevated pollution layers.The restraints of the atmospheric internal boundaries on horizontal diffusion of pollutants were emphasized.The contribution of the ABL to haze pollution in winter and summer was qualitatively compared and quantitatively estimated.The turbulent transport nature behind the relationship between the atmospheric boundary layer height(ABLH)and surface PM_(2.5)concentrations was analyzed.The concept of'aerosol accumulation layer'was defined,and the applicability of the material method in determining ABLH was clarified.A measurement system for obtaining the turbulent flux of PM_(2.5)concentrations was developed,and the turbulence characteristics of PM_(2.5)concentrations were demonstrated.The COATS experiment is of great theoretical significance for thoroughly understanding the physical mechanisms of the ABL during haze pollution and filling the knowledge gap on the impacts of the ABL three-dimensional structure on haze pollution.The results of this study are conducive to the improvement and development of ABL parameterization schemes and serve as a scientific basis for formulating regional pollution prevention and control measures.Qianhui LI Hongsheng ZHANG Xiaoye ZHANG Xuhui CAI Xipeng JIN Lu ZHANG Yu SONG Ling KANG Fei HU Tong ZHU 2023Science China Earth Sciences2023,66,5:0
