| 1 | The cGAS-STING signaling in cardiovascular and metabolic diseases: Future novel target option for pharmacotherapy显示文摘The cyclic GMP-AMP synthase(cGAS)-stimulator of interferon genes(STING) signaling exert essential regulatory function in microbial-and onco-immunology through the induction of cytokines, primarily type I interferons. Recently, the aberrant and deranged signaling of the cGAS-STING axis is closely implicated in multiple sterile inflammatory diseases, including heart failure,myocardial infarction, cardiac hypertrophy, nonalcoholic fatty liver diseases, aortic aneurysm and dissection, obesity, etc. This is because of the massive loads of damage-associated molecular patterns(mitochondrial DNA, DNA in extracellular vesicles) liberated from recurrent injury to metabolic cellular organelles and tissues, which are sensed by the pathway. Also, the cGAS-STING pathway crosstalk with essential intracellular homeostasis processes like apoptosis, autophagy, and regulate cellular metabolism.Targeting derailed STING signaling has become necessary for chronic inflammatory diseases. Meanwhile, excessive type I interferons signaling impact on cardiovascular and metabolic health remain entirely elusive. In this review, we summarize the intimate connection between the cGAS-STING pathway and cardiovascular and metabolic disorders. We also discuss some potential small molecule inhibitors for the pathway. | 2 | Zacopride selectively activates the Kir2.1 channel via a PKA signaling pathway in rat cardiomyocytes显示文摘We recently reported that zacopride is a selective inward rectifier potassium current (IK1 ) channel agonist, suppressing ventricular arrhythmias without affecting atrial arrhythmias. The present study aimed to investigate the unique pharmacological properties of zacopride. The whole-cell patch-clamp technique was used to study IK1 currents in rat atrial myocytes and Kir2.x currents in human embryonic kidney (HEK)-293 cells transfected with inward rectifier potassium channel (Kir)2.1, Kir2.2, Kir2.3, or mutated Kir2.1 (at phosphorylation site S425L). Western immunoblots were performed to estimate the relative protein expression levels of Kir2.x in rat atria and ventricles. Results showed that zacopride did not affect the IK1 and transmembrane potential of atrial myocytes. In HEK293 cells, zacopride increased Kir2.1 homomeric channels by 40.7%±9.7% at 50 mV, but did not affect Kir2.2 and Kir2.3 homomeric channels, and Kir2.1-Kir2.2, Kir2.1-Kir2.3 and Kir2.2-Kir2.3 heteromeric channels. Western immunoblots showed that similar levels of Kir2.3 protein were expressed in rat atria and ventricles, but atrial Kir2.1 protein level was only 25% of that measured in the ventricle. In addition, 5-hydroxytryptamine (5-HT) 3 receptor was undetectable, whereas 5-HT 4 receptor was weakly expressed in HEK293 cells. The Kir2.1-activating effect of zacopride in these cells was abolished by inhibition of protein kinase A (PKA), but not PKC or PKG. | 3 | The three-dimensional structure and seasonal variation of the North Pacific meridional overturning circulation显示文摘The three-dimensional structure and the seasonal variation of the North Pacific meridional overturning circulation (NPMOC) are analyzed based on the Simple Ocean Data Assimilation data and Argo profiling float data.The NPMOC displays a multi-cell structure with four cells in the North Pacific altogether.The TC and the STC are a strong clockwise meridional cell in the low latitude ocean and a weaker clockwise meridional cell between 7°N and 18°N,respectively, while the DTC and the subpolar cell are a weaker anticlockwise meridional cell between 3°N and 15°N and a weakest anticlockwise meridional cell between 35°N and 50°N,respectively. The DTC,the TC and the STC are all of very strong seasonal variations.As to the DTC, the southward transport is strongest in fall and weakest in spring.For the TC,the northward transport is strongest in winter and weakest in spring,while the southward transport is strongest in fall and weakest in spring, which is associated with the strong southward flow of the DTC in fall.As the STC,the northward transport is strongest in winter and weakest in summer,while the southward transport is strongest in summer and weakest in spring. | 4 | The study on seasonal characteristics of water masses in the western East China Sea shelf area显示文摘On the basis of the CTD data and the modeling results in the winter and summer of 2009, the seasonal characteristics of the water masses in the western East China Sea shelf area were analyzed using a cluster analysis method. The results show that the distributions and temperature-salinity characteristics of the water masses in the study area are of distinct seasonal difference. In the western East China Sea shelf area, there are three water masses during winter, i.e., continental coastal water(CCW), Taiwan Warm Current surface water(TWCSW) and Yellow Sea mixing water(YSMW), but four ones during summer, i.e., the CCW, the TWCSW, Taiwan Warm Current deep water(TWCDW) and the YSMW. Of all, the CCW, the TWCSW and the TWCDW are all dominant water masses. The CCW, primarily characterized by a low salinity, has lower temperature, higher salinity and smaller spatial extent in winter than in summer. The TWCSW is warmer, fresher and smaller in summer than in winter, and it originates mostly from the Kuroshio surface water(KSW) northeast of Taiwan, China and less from the Taiwan Strait water during winter, but it consists of the strait water and the KSW during summer. | 9 | Maize MS2 encodes an ATP-binding cassette transporter that is essential for anther development显示文摘The anther cuticle and pollen exine play a critical role in male gametophyte development. The sporopollenin precursors and cuticular lipid monomers are transported to the surface of the microspores and the epidermis by lipid transport proteins(LTPs) and ATP-binding cassette G(ABCG) transporters for the formation of the pollen wall and anther cuticle, respectively. However, the function of ABCG transporters in maize anther development is unclear. Here, we cloned the MS2 gene from the maize male sterile2 mutant using map-based cloning and determined that it encodes an ABCG transporter. MS2 protein was experimentally confirmed to be located on the cell membrane. The quantitative real-time PCR(qRT-PCR)results showed that MS2 was ubiquitously expressed in all vegetative and reproductive tissues, whereas a high transcriptional level of MS2 was observed in anthers, especially at the young microspore stage. Gas chromatography-mass spectrometry(GC–MS) analysis showed decreased accumulation of cutin and wax components in ms2 anthers, indicating that MS2 plays a role in the transport of lipid molecules to anther cuticle and pollen exine. | 11 | Porous-hollow nanorods constructed from alternate intercalation of carbon and MoS2 monolayers for lithium and sodium storage显示文摘Weak ion diffusion and electron transport due to limited interlayer spacing and poor electrical conductivity have been identified as critical roadbacks for fast and abundant energy storage of both MoS2-based lithium ion batteries (LIBs) and sodium ion batteries (SIBs). In this work, MoS2 porous-hollow nanorods (MoS2/m-C800) have been designed and synthesized via an annealing-followed chemistry-intercalated strategy to solve the two issues. They are uniformly assembled from ultrathin MoS2 nanosheets, deviated to the rod-axis direction, with expanded interlayer spacing due to alternate intercalation of N-doped carbon monolayers between the adjacent MoS2 monolayers. Electrochemical studies of the MoS2/m-C800 sample, as an anode of LIBs, demonstrate that the superstructure can deliver a reversible discharge capacity of 1,170 mAh·g^-1 after 100 cycles at 0.2 A·g^-1 and maintain a reversible capacity of 951 mAh·g^-1 at 1.25 A·g^-1 after 350 cycles. | 16 | Annual and interannual variations of the Western Pacific Warm Pool volume and sources of warm water revealed by Argo data显示文摘Based on gridded Argo profile data from January 2004 to December 2010, together with the P-vector inverse method, the three-dimensional structure, annual and inter-annual variations in volume of the Western Pacific Warm Pool(WPWP) are studied. The variations of latitudinal and longitudinal warm water flowing into and out of the WPWP and the probable mechanism of warm water maintenance are also discussed. From the surface to the bottom, climatic WPWP tilts southward and its area decreases. The maximum depth could extend to 120 m, such that its volume could attain 1.86×1015 m3. Annual variation of the WPWP volume shows two obvious peaks that occur in June and October, whereas its inter-annual variations are related to ENSO events. Based on a climatic perspective, the warm water flowing latitudinally into the pool is about 52 Sv, which is mainly through upper layers and via the eastern boundary. Latitudinally, warm water flowing outward is about 49 Sv, and this is mainly through lower layers and via the western boundary. In contrast, along the latitude, warm water flowing into and out of the pool is about 28 Sv and 23 Sv, respectively. Annual and inter-annual variations of the net transportation of the warm water demonstrate that the WPWP mainly loses warm water in the west-east direction, whereas it receives warm water from the north-south direction. The annual variation of the volume of WPWP is highly related to the annual variation of the net warm water transportation, however, they are not closely related on inter-annual time scale. On the inter-annual time scale, influences of ENSO events on the net warm water transportation in the north-south direction are much more than that in the west-east direction.
