1BMAL1 knockout macaque monkeys display reduced sleep and psychiatric disorders显示文摘Circadian disruption is a risk factor for metabolic, psychiatric and age-related disorders, and non-human primate models could help to develop therapeutic treatments. Here, we report the generation of BMAL1 knockout cynomolgus monkeys for circadian-related disorders by CRISPR/Cas9 editing of monkey embryos. These monkeys showed higher nocturnal locomotion and reduced sleep, which was further exacerbated by a constant light regimen. Physiological circadian disruption was reflected by the markedly dampened and arrhythmic blood hormonal levels. Furthermore, BMAL1-deficient monkeys exhibited anxiety and depression, consistent with their stably elevated blood cortisol, and defective sensory processing in auditory oddball tests found in schizophrenia patients. Ablation of BMAL1 up-regulated transcriptional programs toward inflammatory and stress responses, with transcription networks associated with human sleep deprivation, major depressive disorders, and aging. 2Filamin B:The next hotspot in skeletal research?显示文摘Filamin B(FLNB)is a large dimeric actin-binding protein which crosslinks actin cytoskeleton filaments into a dynamic structure.Up to present,pathogenic mutations in FLNB are solely found to cause skeleta deformities,indicating the important role of FLNB in skeletal development.FLNB-related disorders are classifiedasspondylocarpotarsalsynostosis(SCT),Larsensyndrome(LS),atelosteogenesis(AO)boomerang dysplasia(BD),and isolated congenital talipes equinovarus,presenting with scoliosis,shortlimbed dwarfism,clubfoot,joint dislocation and other unique skeletal abnormalities.Several mecha?nisms of FLNB mutations causing skeletal malformations have been proposed,including delay of ossification in long bone growth plate,reduction of bone mineral density(BMD),dysregulation of muscle differentiation,ossification of intervertebral disc(IVD),disturbance of proliferation,differentiation and apoptosis in chondrocytes,impairment of angiogenesis,and hypomotility of osteoblast,chondrocyte and fibroblast.Interventions on FLNB-related diseases require prenatal surveillance by sonography,gene testing in high-risk carriers,andproper orthosis or orthopedic surgeries to correct malformations including scoliosis,cervical spine instability,large joint dislocation,and clubfoot.Gene and cell therapies for FLNB-related diseases are also promising but require further studies.Qiming xu Nan Wu Lijia Cui Zhihong Wu Guixing Qiu 2017Journal of Genetics and Genomics2017,44,7:5
3Co-implantation of magnesium and zinc ions into titanium regulates the behaviors of human gingival fibroblasts显示文摘Soft tissue sealing around implants acts as a barrier between the alveolar bone and oral environment,protecting implants from the invasion of bacteria or external stimuli.In this work,magnesium(Mg)and zinc(Zn)are introduced into titanium by plasma immersed ion implantation technology,and their effects on the behaviors of human gingival fibroblasts(HGFs)as well as the underlying mechanisms are investigated.Surface characterization confirms Mg and Zn exist on the surface in metallic and oxidized states.Contact angle test suggests that surface wettability of titanium changes after ion implantation and thus influences protein adsorption of surfaces.In vitro studies disclose that HGFs on Mg ion-implanted samples exhibit better adhesion and migration while cells on Zn ion-implanted samples have higher proliferation rate and amounts.The results of immunofluorescence staining and real-time reverse-transcriptase polymerase chain reaction(RT-PCR)suggest that Mg mainly regulates the motility and adhesion of HGFs through activating the MAPK signal pathway whereas Zn influences HGFs proliferation by triggering the TGF-βsignal pathway.The synergistic effect of Mg and Zn ions ensure that HGFs cultured on co-implanted samples possessed both high proliferation rate and motility,which are critical to soft tissue sealing of implants.Lanyu Wang Qiming Luo Xianming Zhang Jiajun Qiu Shi Qian Xuanyong Liu 2021Bioactive Materials2021,6,1:4

8Cronobacter spp.,foodborne pathogens threatening neonates and infants显示文摘Cronobacter spp.(formerly Enterobacter sakazakii) are special foodborne pathogens. Cronobacter infection can cause necrotizing enterocolitis, sepsis and meningitis in all age groups, especially neonates and infants, with a high fatality of up to 80%, although the infection is rare. Outbreaks of Cronobacter infection are epidemiologically proven to be associated with contaminated powdered infant formula(PIF). Cronobacter spp.can resist dry environments and survive for a long period in food with low water activity. Therefore, Cronobacter spp.have become serious pathogens of neonates and infants, as well as in the dairy industry. 9Used for the Study of Nitrogen Fertilizer Slow-Release Carrier Life Sludge显示文摘After the sewage treatment, putting the wet sludge in which the heavy metal content is extremely low, corrupt and broken straw, bentonite, urea in proportioning according to the certain ratio, mixing well-distributed, taking the shape of Nitrogen slow-release fertilizers, doing the dynamic bioleaching test by the method of Artificial rainfall simulation, researching the slow-release characteristic, water retention. When the mass ratio of the wet sludge whose water content is 82.5%: bentonite: corrupt and broken straw: urea is 62.5: 12.5: 12.5: 12.5, drip washing the 10g Nitrogen slow-release fertilizers by the 80ml distilled water after 48h, the residue rate of urea is 29.63%; Under the room temperature of 25 ℃, 77%RH, moisture evaporate 46.32% after 60 h. The results demonstrate that the slow-release fertilizer has a good release-effect of nitrogen and water conservation effect. 11Blockade of integrin signaling reduces chemotherapy-induced premature senescence in collagen cultured bladder cancer cells显示文摘Background:Diminished sensitivity towards chemotherapy remains the major impediment to the clinical treatment of bladder cancer.However,the critical elements in control of chemotherapy resistance remain obscure.Methods:We adopted improved collagen gels and performed cytotoxicity analysis of doxorubicin(DOX)and mitomycin C(MMC)of bladder cancer cells in a 3D culture system.We then detected the expression of multidrug resistant gene ABCB1,dormancy-associated functional protein chicken ovalbumin upstream-transcription factor 1(COUPTF1),cell proliferation marker Ki-67,and cellular senescence marker senescence-associatedβ-galactosidase(SA-β-Gal)in these cells.We further tested the effects of integrin blockade or protein kinase B(AKT)inhibitor on the senescent state of bladder cancer.Also,we examined the tumor growth and survival time of bladder cancer mouse models given the combination treatment of chemotherapeutic agents and integrinα2β1 ligand peptide TFA(TFA).Results:Collagen gels played a repressive role in bladder cancer cell apoptosis induced by DOX and MMC.In mechanism,collagen activated the integrinβ1/AKT cascade to drive bladder cancer cells into a premature senescence state via the p21/p53 pathway,thus attenuating chemotherapy-induced apoptosis.In addition,TFA had the ability to mediate the switch from senescence to apoptosis of bladder cancer cells in xenograft mice.Meanwhile,TFA combined with chemotherapeutic drugs produced a substantial suppression of tumor growth as well as an extension of survival time in vivo.Conclusions:Based on our finding that integrinβ1/AKT acted primarily to impart premature senescence to bladder cancer cells cultured in collagen gel,we suggest that integrinβ1 might be a feasible target for bladder cancer eradication.Linghui Deng Kun Jin Xianghong Zhou Zilong Zhang Liming Ge Xingyu Xiong Xingyang Su Di Jin Qiming Yuan Chichen Zhang Yifan Li Haochen Zhao Qiang Wei Lu Yang Shi Qiu 2022Precision Clinical Medicine2022,5,2:0
12Data-driven intelligent modeling framework for the steam cracking process显示文摘Steam cracking is the dominant technology for producing light olefins,which are believed to be the foundation of the chemical industry.Predictive models of the cracking process can boost production efficiency and profit margin.Rapid advancements in machine learning research have recently enabled data-driven solutions to usher in a new era of process modeling.Meanwhile,its practical application to steam cracking is still hindered by the trade-off between prediction accuracy and computational speed.This research presents a framework for data-driven intelligent modeling of the steam cracking process.Industrial data preparation and feature engineering techniques provide computational-ready datasets for the framework,and feedstock similarities are exploited using k-means clustering.We propose LArge-Residuals-Deletion Multivariate Adaptive Regression Spline(LARD-MARS),a modeling approach that explicitly generates output formulas and eliminates potentially outlying instances.The framework is validated further by the presentation of clustering results,the explanation of variable importance,and the testing and comparison of model performance.Qiming Zhao Kexin Bi Tong Qiu 2023Chinese Journal of Chemical Engineering2023,61,9:0

14Complete androgen blockade vs.medical castration alone as adjuvant androgen deprivation therapy for prostate cancer patients following radical prostatectomy:a retrospective cohort study显示文摘Background:Till date,the optimal treatment strategy for delivering adjuvant androgen deprivation therapy(ADT)in localized and locally advanced prostate cancer(PCa),as a lower stage in PCa progression compared with metastatic PCa,is still unclear.This study compares the efficacy of castration alone with complete androgen blockade(CAB)as adjuvant ADT in patients with localized and locally advanced PCa undergoing radical prostatectomy(RP).Methods:Patients diagnosed with PCa,without lymph node or distant metastasis,who received RP in West China Hospital between January 2009 and April 2019,were enrolled in this study.We performed survival,multivariable Cox proportional hazard regression,and subgroup analyses.Results:A total of 262 patients were enrolled,including 107 patients who received castration alone and 155 patients who received CAB.The survival analysis revealed that there was no significant difference between the two groups(hazard ratios[HR]=1.07,95%confidence intervals[95%CI]=0.60-1.90,P=0.8195).Moreover,the multivariable Cox model provided similarly negative results before and after adjustment for potential covariant.Similarly,there was no significant difference in the clinical recurrence between the two groups in both non-adjusted and adjusted models.Furthermore,our subgroup analysis showed that CAB achieved better biochemical recurrence(BCR)outcomes than medical castration alone as adjuvant ADT for locally advanced PCa(P for interaction=0.0247,HR=0.37,95%CI=0.14-1.00,P=0.0497).Conclusion:Combined androgen blockade achieved better BCR outcomes compared with medical castration alone as adjuvant ADT for locally advanced PCa without lymph node metastasis.Di Jin Kun Jin Bo Chen Xianghong Zhou Qiming Yuan Zilong Zhang Qiang Wei Shi Qiu 2022Chinese Medical Journal2022,,7:0
