| 1 | Whole genome sequencing of 10K patients with acute ischaemic stroke or transient ischaemic attack: design, methods and baseline patient characteristics显示文摘Background and purpose Stroke is the second leading cause of death worldwide and the leading cause of mortality and long-term disability in China,but its underlying risk genes and pathways are far from being comprehensively understood.We here describe the design and methods of whole genome sequencing(WGS)for 10914 patients with acute ischaemic stroke or transient ischaemic attack from the Third China National Stroke Registry(CNSR-III).Methods Baseline clinical characteristics of the included patients in this study were reported.DNA was extracted from white blood cells of participants.Libraries are constructed using qualified DNA,and WGS is conducted on BGISEQ-500 platform.The average depth is intended to be greater than 30×for each subject.Afterwards,Sentieon software is applied to process the sequencing data under the Genome Analysis Toolkit best practice guidance to call genotypes of single nucleotide variants(SNVs)and insertion-deletions.For each included subject,21 fingerprint SNVs are genotyped by MassARRAY assays to verify that DNA sample and sequencing data originate from the same individual.The copy number variations and structural variations are also called for each patient.All of the genetic variants are annotated and predicted by bioinformatics software or by reviewing public databases.Results The average age of the included 10914 patients was 62.2±11.3 years,and 31.4%patients were women.Most of the baseline clinical characteristics of the 10914 and the excluded patients were balanced.Conclusions The WGS data together with abundant clinical and imaging data of CNSR-III could provide opportunity to elucidate the molecular mechanisms and discover novel therapeutic targets for stroke. | Si Cheng Zhe Xu Yang Liu Jinxi Lin Yong Jiang Yilong Wang Xia Meng Anxin Wang Xinying Huang Zhimin Wang Guohua Chen Songdi Wu Zhengchang Jia Yongming Chen Xuerong Qiu Jun Wu Binbin Song Weizhong Ji Zhongping An Wenjun Xue Lili Zhao Yu Geng Hongyan Li Hao Li Yongjun Wang | 2021 | Stroke & Vascular Neurology2021,6,2: | 8 |

| 4 | Somatic mutation contributing to clonal haematopoiesis is a risk factor of recurrent stroke in first-ever acute ischaemic stroke:a prospective cohort study显示文摘Background Somatic mutation contributes to clonal haematopoiesis of indeterminate potential(CHIP)is related to age and associated with a higher risk of stroke and atherosclerotic cardiovascular disease.Here,we investigated the prognostic significance of CHIP in a large first-ever acute ischaemic stroke(AIS)cohort and explored the underlying mechanisms.Methods We studied a prospective cohort of 6016 patients who had a first-ever AIS in China.Whole-genome sequencing was performed to identify CHIP.High-sensitivity C reactive protein(hs-CRP)levels above 3 mg/L at baseline were defined as hyperinflammation.Recurrent stroke during the 3-month follow-up was the primary outcome.Results Among the 6016 patients who had a first-ever AIS,with a median age was 62 years(IQR,54.0‒70.0),3.70%were identified as CHIP carriers.The most common mutations occurred in the DNMT3A(30.0%)and TET2(11.4%)genes.During a follow-up of 3 months,the presence of CHIP was associated with recurrent stroke(HR 1.62,95%CI 1.04 to 2.51,p=0.03),recurrent ischaemic stroke(HR 1.64,95%CI 1.04 to 2.58,p=0.03)and combined vascular events(HR 1.58,95%CI 1.02 to 2.44,p=0.04)after adjusting for hsCRP levels at baseline in patients who had a first-ever AIS.Subgroup analysis demonstrated that CHIP was only associated with recurrent stroke when patients under hyperinflammation(OR 3.10,95%CI 1.92 to 5.00,p<0.001)but not in those without hyperinflammation(OR 0.18,95%CI 0.03 to 1.04,p=0.06,Pinteraction=0.002).Conclusion Our results suggest that somatic mutations contributing to CHIP increase the risk of short-term recurrent stroke in patients who had a first-ever AIS.Hyperinflammation may be important in the relationship between CHIP and recurrent stroke. | Xin Qiu Yalun Dai Si Cheng Hong-Qiu Gu Yong Jiang Xia Meng Yilong Wang Xingquan Zhao Yingyu Jiang Zhe Xu Xinying Huang Meng Wang Tian Jie Lyu Yubo Wang Jiaxu Weng Lingyun Cui Yi Shangguan Hao Li Yongjun Wang Zixiao Li | 2023 | Stroke & Vascular Neurology2023,8,2: | 1 |
| 5 | Polarization-insensitive 3D conformal-skin metasurface cloak显示文摘Electromagnetic metasurface cloaks provide an alternative paradigm toward rendering arbitrarily shaped scatterers invisible.Most transformation-optics(TO)cloaks intrinsically need wavelength-scale volume/thickness,such that the incoming waves could have enough long paths to interact with structured meta-atoms in the cloak region and consequently restore the wavefront.Other challenges of TO cloaks include the polarization-dependent operation to avoid singular parameters of composite cloaking materials and limitations of canonical geometries,e.g.,circular,elliptical,trapezoidal,and triangular shapes.Here,we report for the first time a conformal-skin metasurface carpet cloak,enabling to work under arbitrary states of polarization(SOP)at Poincarésphere for the incident light and arbitrary conformal platform of the object to be cloaked.By exploiting the foundry three-dimensional(3D)printing techniques to fabricate judiciously designed meta-atoms on the external surface of a conformal object,the spatial distributions of intensity and polarization of its scattered lights can be reconstructed exactly the same as if the scattering wavefront were deflected from a flat ground at any SOP,concealing targets under polarization-scanning detections.Two conformal-skin carpet cloaks working for partial-and full-azimuth plane operation are respectively fabricated on trapezoid and pyramid platforms via 3D printing.Experimental results are in good agreement with numerical simulations and both demonstrate the polarization-insensitive cloaking within a desirable bandwidth.Our approach paves a deterministic and robust step forward to the realization of interfacial,free-form,and full-polarization cloaking for a realistic arbitrary-shape target in real-world applications. | He-Xiu Xu Guangwei Hu Yanzhao Wang Chaohui Wang Mingzhao Wang Shaojie Wang Yongjun Huang Patrice Genevet Wei Huang Cheng-Wei Qiu | 2021 | Light(Science & Applications)2021,10,5: | 1 |

| 11 | Effect of light and nutrients on interspecific interactions between submerged macrophytes:implications for restoration of multispecies aquatic vegetation in eutrophic lakes显示文摘Constructing multispecies submerged vegetation systems and maintaining stable seasonal succession is crucial for restoring shallow eutrophic lakes.However,little is known about the interactions between successional and existing species of different growth forms,particularly under the low light and high nutrient conditions of eutrophic lakes.We measured the functional traits of mature Vallisneria natans(Lour.)Hara plants and Potamogeton crispus L.shoots in monoculture and mosaic patterns under different light and nutrient conditions.The effect of light on functional traits of the submerged macrophyte species was more significant than that of nutrients,but the reverse was true for P.crispus biomass allocation.Moreover,interspecific interactions affected only the submerged macrophytes under the low light condition and varied with species.Specifically,the interaction of P.crispus to V.natans was biased towards competition,while the interaction of V.natans to P.crispus was converted from facilitation to competition by eutrophication,particularly in the homogenous mosaic growth pattern.This study demonstrates that sufficient light is a prerequisite and patch planting is an effective means to form a multispecies submerged vegetation system.In addition,we emphasize that the coexistence of eutrophication and low light will likely result in a competition between submerged macrophytes thus simplifying the vegetation,even if their growth forms and growing seasons are different.These findings help explain the collapse of multispecies submerged vegetation and guide the restoration of aquatic plants in eutrophic lakes. | Yiping WANG Xiaoguang XU Dujun LI Yongjun LU Xinhou ZHANG Chuyao YANG Qiu JIN Guoxiang WANG | 2023 | Journal of Oceanology and Limnology2023,41,5: | 0 |
