1Prognostic factors of refractory NSCLC patients receiving anlotinib hydrochloride as the third-or further-line treatment显示文摘Objective:Anlotinib hydrochloride is a multitarget tyrosine kinase inhibitor that targets vascular endothelial growth factor receptor,fibroblast growth factor receptor,platelet-derived growth factor receptor,c-Kit,and c-MET;therefore,it exhibits both antitumor and anti-angiogenetic activities.A phase III trial has shown that anlotinib improved progression-free survival(PFS)and overall survival(OS)in patients with advanced non-small cell lung cancer(NSCLC),who presented with progressive disease or intolerance after standard chemotherapy.This study aimed to analyze the characteristics of patients receiving anlotinib treatment to determine the dominant populations who are fit for the treatment.Methods:Data were collected from March 2015 to January 2017 from a randomized,double-blind,placebo-controlled,multicenter,phase III trial of anlotinib(ALTER0303).A total of 437 patients were enrolled and randomly allocated(2:1)to the anlotinib and placebo groups.Kaplan–Meier analysis and log-rank test were performed to compare PFS and OS.Cox proportional hazards model was adopted for multivariate prognostic analysis.Results:Multivariate analysis indicated that high post-therapeutic peripheral blood granulocyte/lymphocyte ratio and elevated alkaline phosphatase levels were independent risk factors for PFS.Meanwhile,elevated thyroid-stimulating hormone,blood glucose,and triglyceride levels;hypertension;and hand–foot syndrome were independent protective factors of PFS.High posttherapeutic peripheral blood granulocyte/lymphocyte ratio,an Eastern Cooperative Oncology Group(ECOG)score≥2,and the sum of the maximal target lesion length at baseline were independent risk factors of OS,and hypertriglyceridemia was an independent protective factor of OS.Conclusions:This study preliminarily explored the possible factors that affected PFS and OS after anlotinib treatment in patients with advanced refractory NSCLC,and the baseline characteristics of the therapeutically dominant populations were then identified.Jing Wang Yizhuo Zhao Qiming Wang Li Zhang Jianhua Shi Zhehai Wang Ying Cheng Jianxing He Yuankai Shi Hao Yu Yang Zhao Weiqiang Chen Yi Luo Xiuwen Wang Kejun Nan Faguang Jin Jian Dong Baolan Li Zhujun Liu Baohui Han Kai Li 2018Cancer Biology & Medicine2018,15,4:46
2Effects of dietary arginine levels on growth performance,body composition, serum biochemical indices and resistance ability against ammonia-nitrogen stress in juvenile yellow catfish(Pelteobagrus fulvidraco)显示文摘This experiment was conducted to investigate the effects of dietary arginine levels on growth performance, body composition, serum biochemical indices and resistance ability against ammonia-nitrogen stress in juvenile yellow catfish(Pelteobagrus fulvidraco). Five isonitrogenous and isolipidic diets(42%protein and 9% lipid) were formulated to contain graded levels of arginine(2.44%, 2.64%, 2.81%, 3.01% and3.23% of diet), by supplementing L-Arginine HCI. Seven hundred juvenile yellow catfish with an initial average body weight of 1.13 ± 0.02 g were randomly divided into 5 groups with 4 replicates of 35 fish each and each group was fed one of the diets. After 56 d feeding, fish were exposed to 100 mg/L of ammonia-nitrogen for 72 h. The results showed that weight gain(WG) and specific growth rate(SGR) in2.64% and 2.81% groups were significantly higher than those in 3.23% group(P < 0.05). The feed conversation ratio(FCR) in 2,64%, 2,81% and 3.01% groups was significantly decreased when compared with3,23% group. The protein efficiency ratio(PER) in 2.64% group was significantly higher than that in 2.44%and 3.23% groups(P < 0.05). The condition factor(CF) of fish was significantly higher in 2.81% group than that in 2,44% group(P < 0.05). Dietary arginine levels had no significant effect on hepatosomatic index(HSI), viscerosomatic index(VSI), and whole-body dry matter, crude protein, crude lipid, ash contents, as well as serum total protein(TP), triglyceride(TG), glucose(GLU), urea nitrogen(UN) contents and aspartate aminotransferase(AST), alanine aminotransferase(ALT) activities(P> 0.05). After the fish were challenged to ammonia-nitrogen for 72 h, their cumulative mortality rate in 2.81% group was significantly lower than that in 2.44% group(P < 0.05). The results suggested that dietary arginine level at 2.81%could optimize anti-ammonia-nitrogen stress ability of juvenile yellow catfish and a level of 3.23%arginine seemed to depress the growth performance of fish and decreased their tolerance to the ammonia-nitrogen stress under current study. 3Spatial–temporal distribution and potential ecological risk assessment of nonylphenol and octylphenol in riverine outlets of Pearl River Delta, China显示文摘The aquatic environments of the Pearl River Delta in Southern China are subjected to contamination with various industrial chemicals from local industries. In this paper, the occurrence, seasonal variation and spatial distribution of alkylphenol octylphenol(OP) and nonylphenol(NP) in river surface water and sediments in the runoff outlets of the Pearl River Delta were investigated. NP and OP were detected in all water and sediment samples and their mean concentrations in surface water during the dry season ranged from 810 to 3366 ng/L and85.5 to 581 ng/L, respectively, and those in sediments ranged from 14.2 to 95.2 ng/g dw and 0.4to 3.0 ng/g dw, respectively. In surface water, much higher concentrations were detected in the dry season than those in the wet season. In sediments, the concentrations in the dry season were also mostly higher. High concentrations of NP and OP were found in Humen outlet, likely due to high levels of domestic and industrial wastewater discharges. 4A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. 5DYNAMIC VARIATIONS OF WATER QUALITY IN TAIHU LAKE AND MULTIVARIATE ANALYSIS OF ITS INFLUENTIAL FACTORS显示文摘Dynamic variation of water quality in Meiliang Bay and part of West Taihu Lake has been analysed based on data from 1991 to 1992. Principal Component Analysis is used to reveal the mutual relationships of various factors. It is shown that there existis an obvious spatial and temporal variation in the main factors of water quality. Annual values of TP, CON, TN, Chl-a and conductivity decrease evidently from inner Meiliang Bay to the outer from north to south. TP and TN fluctuate seasonally with much higher value in winter. This is particularly true for the mouth of Liangxi River. In addition, the Chl-1 has a synchronous variation with water temperature, although being lagged a little, and closely relates to TP and TN. 6Nanoprinted high-neuron-density optical linear perceptrons performing near-infrared inference on a CMOS chip显示文摘Optical machine learning has emerged as an important research area that,by leveraging the advantages inherent to optical signals,such as parallelism and high speed,paves the way for a future where optical hardware can process data at the speed of light.In this work,we present such optical devices for data processing in the form of single-layer nanoscale holographic perceptrons trained to perform optical inference tasks.We experimentally show the functionality of these passive optical devices in the example of decryptors trained to perform optical inference of single or whole classes of keys through symmetric and asymmetric decryption.The decryptors,designed for operation in the near-infrared region,are nanoprinted on complementary metal-oxide-semiconductor chips by galvo-dithered two-photon nanolithography with axial nanostepping of 10 nm achieving a neuron density of>500 million neurons per square centimetre.This power-efficient commixture of machine learning and on-chip integration may have a transformative impact on optical decryption3,sensing4,medical diagnostics5 and computing6,7.Elena Goi Xi Chen Qiming Zhang Benjamin P.Cumming Steffen schoenhardt Haitao Luan Min Gu 2021Light(Science & Applications)2021,10,3:8
7Breach mechanism and numerical simulation for seepage failure of earth-rock dams显示文摘In this study,a numerical model,which can capture the full process of the development of seepage passages,the collapse of dams and the failure due to overtopping,is proposed for earth-rock dams.The critical incipient velocity for the occurrence of seepage failure is derived by analyzing the forces acting on soil particles in the seepage passage.The sediment transport formula is proposed to simulate the erosion process and the evolution of breach within the dam.In this model,the grain size distribution,the compaction density and the strength of dam materials are reasonably accounted for.Furthermore,the influences of the direction of seepage paths,the slope of the dam and the velocity of water flow on the amount of erosion are also taken into consideration.The proposed model and the corresponding numerical programs are employed to simulate the development of breaches and discharge of two typical cases due to seepage failure.The development of breaches,the history of discharge and the peak flood flux predicted by the numerical models are rather comparable to the measured data,which confirms the validity of the proposed model and the feasibility of applying the model in evaluating the disaster consequences and preparing the emergency counter measurements in the case of dam collapse.CHEN ShengShui ZHONG QiMing CAO Wei 2012Science China(Technological Sciences)2012,55,6:8
8Successful Applications of Generic Atmospheric Correction Online Service for InSAR (GACOS) to the Reduction of Atmospheric Effects on InSAR Observations显示文摘The tremendous development of Synthetic Aperture Radar(SAR)missions in recent years facilitates the study of smaller amplitude ground deformation over greater spatial scales using longer time series.However,this poses greater challenges for correcting atmospheric effects due to the wider coverage of SAR imagery than ever.Previous attempts have used observations from Global Positioning System(GPS)and Numerical Weather Models(NWMs)to separate atmospheric delays,but they are limited by(1)The availability(and distribution)of GPS stations;(2)The low spatial resolution of NWM;And(3)The difficulties in quantifying their performance.To overcome these limitations,we have developed the Generic Atmospheric Correction Online Service for InSAR(GACOS)which utilizes the high-resolution European Centre for Medium-Range Weather Forecasts(ECMWF)products using an Iterative Tropospheric Decomposition(ITD)model.This enables the reduction of the coupling effects of the troposphere turbulence and stratification and hence achieves equivalent performances over flat and mountainous terrains.GACOS comprises a range of notable features:(1)Global coverage;(2)All-weather,all-time usability;(3)Available with a maximum of two-day latency;And(4)Indicators available to assess the model’s performance and feasibility.In this paper,we demonstrate some successful applications of the GACOS online service to a variety of geophysical studies.Chen YU Zhenhong LI Lin BAI Jan-Peter MULLER Jingfa ZHANG Qiming ZENG 2021Journal of Geodesy and Geoinformation Science2021,4,1:8
9A 3-DIMENSIONAL DATA MODEL FOR VISUALIZING CLOVERLEAF JUNCTION IN A CITY MODEL显示文摘The research work has been seldom done about cloverleaf junction expression in a 3-dimensional city model (3DCM). The main reason is that the cloverleaf junction is often in a complex and enormous construction. Its main body is bestraddle in air,and has aerial intersections between its parts. This complex feature made cloverleaf junction quite different from buildings and terrain, therefore, it is difficult to express this kind of spatial objects in the same way as for buildings and terrain. In this paper,authors analyze spatial characteristics of cloverleaf junction, propose an all-constraint points TIN algorithm to partition cloverleaf junction road surface, and develop a method to visualize cloverleaf junction road surface using TIN. In order to manage cloverleaf junction data efficiently, the authors also analyzed the mechanism of 3DCM data management, extended BLOB type in relational database, and combined R-tree index to manage 3D spatial data. 10Multifunctional oral delivery systems for enhanced bioavailability of therapeutic peptides/proteins显示文摘In last few years, therapeutic peptides/proteins are rapidly growing in drug market considering their higher efficiency and lower toxicity than chemical drugs. However, the administration of therapeutic peptides/proteins is mainly limited in parenteral approach. Oral therapy which was hampered by harsh gastrointestinal environment and poorly penetrating epithelial barriers often results in low bioavailability(less than 1%–2%). Therefore, delivery systems that are rationally designed to overcome these challenges in gastrointestinal tract and ameliorate the oral bioavailability of therapeutic peptides/proteins are seriously promising. 11Anlotinib as third- or further-line therapy for short-term relapsed small-cell lung cancer: subgroup analysis of a randomized phase 2 study (ALTER1202)显示文摘Patients with small-cell lung cancer (SCLC) relapse within months after completing previous therapies. This study aimed to investigate the efficacy and safety of anlotinib as third- or further-line therapy in patients with short-term relapsed SCLC from ALTER1202. Patients with short-term relapsed SCLC (disease progression within 3 months after completing ≥ two lines of chemotherapy) in the anlotinib (n = 67) and placebo (n = 34) groups were analyzed. The primary endpoint was progression-free survival (PFS). The secondary endpoints included overall survival, objective response rate (ORR), disease control rate, and safety. Anlotinib significantly improved median PFS/OS (4.0 vs. 0.7 months, P < 0.0001)/(7.3 vs. 4.4 months, P = 0.006) compared with placebo. The ORR was 4.5%/2.9% in the anlotinib/placebo group (P = 1.000). The DCR in the anlotinib group was higher than that in the placebo group (73.1% vs. 11.8%, P < 0.001). The most common adverse events (AEs) were hypertension (38.8%), loss of appetite (28.4%), and fatigue (22.4%) in the anlotinib group and gamma-glutamyl transpeptidase elevation (20.6%) in the placebo group. No grade 5 AEs occurred. For patients with short-term relapsed SCLC, third- or further-line anlotinib treatment was associated with improved survival benefit. 12CircRNA-SORE mediates sorafenib resistance in hepatocellular carcinoma by stabilizing YBX1显示文摘Sorafenib is the first-line chemotherapeutic therapy for advanced hepatocellular carcinoma(HCC).However,sorafenib resistance significantly limits its therapeutic efficacy,and the mechanisms underlying resistance have not been fully clarified.Here we report that a circular RNA,circRNA-SORE(a circular RNA upregulated in sorafenib-resistant HCC cells),plays a significant role in sorafenib resistance in HCC.We found that circRNA-SORE is upregulated in sorafenib-resistant HCC cells and depletion of circRNA-SORE substantially increases the cell-killing ability of sorafenib.Further studies revealed that circRNA-SORE binds the master oncogenic protein YBX1 in the cytoplasm,which prevents YBX1 nuclear interaction with the E3 ubiquitin ligase PRP19 and thus blocks PRP19-mediated YBX1 degradation.Moreover,our in vitro and in vivo results suggest that circRNA-SORE is transported by exosomes to spread sorafenib resistance among HCC cells.Using different HCC mouse models,we demonstrated that silencing circRNA-SORE by injection of siRNA could substantially overcome sorafenib resistance.Our study provides a proof-of-concept demonstration for a potential strategy to overcome sorafenib resistance in HCC patients by targeting circRNA-SORE or YBX1.Junjie Xu Lin Ji Yuelong Liang Zhe Wan Wei Zheng Xiaomin Song Kirill Gorshkov Qiming Sun Hui Lin Xueyong Zheng Jiang Chen Ren-an Jin Xiao Liang Xiujun Cai 2020Signal Transduction and Targeted Therapy2020,5,1:5

16LIN28 coordinately promotes nucleolar/ribosomal functions and represses the 2C-like transcriptional program in pluripotent stem cells显示文摘LIN28 is an RNA binding protein with important roles in early embryo development,stem cell differentiation/re-programming,tumorigenesis and metabolism.Previous studies have focused mainly on its role in the cytosol where it interacts with Let-7 microRNA precursors or mRNAs,and few have addressed LIN28's role within the nucleus.Here,we show that LIN28 displays dynamic temporal and spatial expression during murine embryo development.Maternal LIN28 expression drops upon exit from the 2-cell stage,and zygotic LIN28 protein is induced at the forming nucleolus during 4-cell to blas-tocyst stage development,to become dominantly expressed in the cytosol after implantation.In cultured pluripotent stem cells(PSCs),loss of LIN28 led to nucleolar stress and activation of a 2-cell/4-cell-like transcriptional program characterized by the expression of endogenous retrovirus genes.Mechanistically,LIN28 binds to small nucleolar RNAs and rRNA to maintain nucleolar integrity,and its loss leads to nucleolar phase separation defects,ribosomal stress and activation of P53 which in turn binds to and activates 2C transcription factor Dux.LIN28 also resides in a complex containing the nucleolar factor Nucleolin(NCL)and the transcrip-tional repressor TRIM28,and LIN28 loss leads to reduced occupancy of the NCL/TRIM28 complex on the Dux and rDNA loci,and thus de-repressed Dux and reduced rRNA expression.Lin28 knockout cells with nucleolar stress are more likely to assume a slowly cycling,translationally inert and anabolically inactive state,which is a part of previously unappreciated 2C-like transcriptional program.These findings elucidate novel roles for nucleolar LIN28 in PSCs,and a new mechanism linking 2C program and nucleolar functions in PSCs and early embryo development.Zhen Sun Hua Yu Jing Zhao Tianyu Tan Hongru Pan Yuqing Zhu Lang Chen Cheng Zhang Li Zhang Anhua Lei Yuyan Xu Xianju Bi Xin Huang Bo Gao Longfei Wang Cristina Correia Ming Chen Qiming Sun Yu Feng Li Shen Hao Wu Jianlong Wang Xiaohua Shen George Q.Daley Hu Li Jin Zhang 2022Protein & Cell2022,13,7:4

18A new type of ERGIC–ERES membrane contact mediated by TMED9 and SEC12 is required for autophagosome biogenesis显示文摘Under stress,the endomembrane system undergoes reorganization to support autophagosome biogenesis,which is a central step in autophagy.How the endomembrane system remodels has been poorly understood.Here we identify a new type of membrane contact formed between the ER-Golgi intermediate compartment(ERGIC)and the ER-exit site(ERES)in the ER-Golgi system,which is essential for promoting autophagosome biogenesis induced by different stress stimuli.The ERGIC-ERES contact is established by the interaction between TMED9 and SEC12 which generates a short distance opposition(as close as 2-5 nm)between the two compartments.The tight membrane contact allows the ERES-located SEC12 to transactivate COPII assembly on the ERGIC.In addition,a portion of SEC12 also relocates to the ERGIC.Through both mechanisms,the ERGIC-ERES contact promotes formation of the ERGIC-derived COPII vesicle,a membrane precursor of the autophagosome.The ERGIC-ERES contact is physically and functionally different from the TFG-mediated ERGIC-ERES adjunction involved in secretory protein transport,and therefore defines a unique endomembrane structure generated upon stress conditions for autophagic membrane formation.Shulin Li Rui Yan Jialu Xu Shiqun Zhao Xinyu Ma Qiming Sun Min Zhang Ying Li Jun-Jie Gogo Liu Liangyi Chen Sai Li Ke Xu Liang Ge 2022Cell Research2022,32,2:3
