2Optimization of clay material mixture ratio and filling process in gypsum mine goaf显示文摘Because there is neither waste rock nor mill tailings in the gypsum mine, and the buildings on the goaf of gypsum mine are needed to be protected, the research proposed the scheme of the clay filling technology. Gypsum, cement, lime and water glass were used as adhesive, and the strength of different material ratios were investigated in this study. The influence factors of clay strength were obtained in the order of cement, gypsum, water glass and lime. The results show that the cement content is the determinant influence factor, and gypsum has positive effects, while the water glass can enhance both clay strength and the fluidity of the filing slurry. Furthermore, combining chaotic optimization method with neural network, the optimal ratio of composite cementing agent was obtained. The results show that the optimal ratio of water glass, cement, lime and clay (in quality) is 1.17:6.74:4.17:87.92 in the process of bottom self-flow filling, while the optimal ratio is 1.78:9.58:4.71:83.93 for roof-contacted filling. A novel filling process to fill in gypsum mine goaf with clay is established. 3Allogeneic Vγ9Vδ2 T-cell immunotherapy exhibits promising clinical safety and prolongs the survival of patients with late-stage lung or liver cancer显示文摘Vγ9Vδ2 T cells are promising candidates for cellular tumor immunotherapy.Due to their HLA-independent mode of action,allogeneic Vγ9Vδ2 T cells can be considered for clinical application.To apply allogeneic Vγ9Vδ2 T cells in adoptive immunotherapy,the methodology used to obtain adequate cell numbers with optimal effector function in vitro needs to be optimized,and clinical safety and efficacy also need to be proven.Therefore,we developed a novel formula to improve the expansion of peripheralγδT cells from healthy donors.Then,we used a humanized mouse model to validate the therapeutic efficacy of expandedγδT cells in vivo;furthermore,the expandedγδT cells were adoptively transferred into late-stage liver and lung cancer patients.We found that the expanded cells possessed significantly improved immune effector functions,including proliferation,differentiation,and cancer cell killing,both in vitro and in the humanized mouse model.Furthermore,a phase I clinical trial in 132 late-stage cancer patients with a total of 414 cell infusions unequivocally validated the clinical safety of allogeneic Vγ9Vδ2 T cells.Among these 132 patients,8 liver cancer patients and 10 lung cancer patients who received≥5 cell infusions showed greatly prolonged survival,which preliminarily verified the efficacy of allogeneic Vγ9Vδ2 T-cell therapy.Our clinical studies underscore the safety and efficacy of allogeneic Vγ9Vδ2 T-cell immunotherapy,which will inspire further clinical investigations and eventually benefit cancer patients.Yan Xu Zheng Xiang Mohammed Alnaggar Léonce Kouakanou Jiawei Li Junyi He Jiashuang Yang Yi Hu Yan Chen Li Lin Jianlei Hao Jingxia Li Jibing Chen Man Li Qingling Wu Christian Peters Qinghua Zhou Jianshuang Li Yingqing Liang Xiaohua Wang Baohui Han Meili Ma Dieter Kabelitz Kecheng Xu Wenwei Tu Yangzhe Wu Zhinan Yin 2021Cellular & Molecular Immunology2021,18,2:8
4Efficacy of Shoushen granule on adenosine triphosphate binding cassette transporter A1, proprotein convertase subtilisin/kexin type 9 and toll-like receptor 4/nuclear factor kappa-B signaling pathway in ApoE-knockout mice显示文摘OBJECTIVE: To evaluate the efficacy of Shoushen granule, prepared with four Chinese medicinals, on the targeted regulation of adenosine triphosphate binding cassette transporter A1(ABCA1) through proprotein convertase subtilisin/kexin type 9(PCSK9) and toll-like receptor 4(TLR4)/nuclear factor kappa-B(NF-κB) signaling pathway to affect atherosclerosis(AS) in ApoE-knockout(ApoE-/-) mice.METHODS: ApoE-/-mice fed with a high-fat diet were used for AS modeling and divided into Model,Shoushen, and Atorvastatin groups. C57 BL/6 J mice at the same age and background strain were included in the Control group. Western blot and immunohistochemistry were used to measure ABCA1, PCSK9, TLR4, and NF-κB protein expression in mouse aortas. Enzyme-linked immuno sorbent assay was used to measure mouse serum tumor necrosis factor-α(TNF-α), interleukin-10(IL-10), monocyte chemoattractant protein 1(MCP-1), and intercellular cell adhesion molecule-1(ICAM-1) expression. Serum lipid profiles and histopathology were also assessed. Shoushen granule were composed of Heshouwu(Radix Polygoni Multiflori) 15 g, Gouqizi(Fructus Lycii) 15 g, Sheng shanzha(Raw Fructus Crataegus Pinnatifidae) 10 g, and Sanqi(Radix Notoginseng) 3 g.RESULTS: ApoE-/-mice fed with a high-fat diet had notable AS lesions, with reduced ABCA1 and IL-10 levels, elevated PCSK9, TLR4, NF-κB, TNF-α, MCP-1,and ICAM-1 expression, and increased total cholesterol(TC) and low density lipoprotein cholesterol(LDL-C) contents. With drug interventions, the areas of AS plaques were significantly reduced,the ABCA1 and IL-10 levels were increase, while the PCSK9, TLR4, NF-κB, TC, and LDL-C contents,and the TNF-α, MCP-1, and ICAM-1 expression were reduced.CONCLUSION: Shoushen granule effectively interfered with AS development by antagonizing the expression of key factors of the PCSK9 and TLR4/NF-κB signaling pathway to upregulate ABCA1 expression.Li Shanshan Cao Hui Shen Dingzhu Chen Chuan Xing Sanli Dou Fangfang Jia Qingling 2019Journal of Traditional Chinese Medicine2019,39,4:4

6Regional seismic-damage prediction of buildings under mainshock-aftershock sequence显示文摘Strong aftershocks generally occur following a significant earthquake.Aftershocks further damage buildings weakened by mainshocks.Thus,the accurate and efficient prediction of aftershock-induced damage to buildings on a regional scale is crucial for decision making for post-earthquake rescue and emergency response.A framework to predict regional seismic damage of buildings under a mainshock-aftershock(MS-AS)sequence is proposed in this study based on city-scale nonlinear time-history analysis(THA).Specifically,an MS-AS sequence-generation method is proposed to generate a potential MS-AS sequence that can account for the amplification,spectrum,duration,magnitude,and site condition of a target area.Moreover,city-scale nonlinear THA is adopted to predict building seismic damage subjected to MS-AS sequences.The accuracy and reliability of city-scale nonlinear THA for an MS-AS sequence are validated by as-recorded seismic responses of buildings and simulation results in published literature.The town of Longtoushan,which was damaged during the Ludian earthquake,is used as a case study to illustrate the detailed procedure and advantages of the proposed framework.The primary conclusions are as follows.(1)Regional seismic damage of buildings under an MS-AS sequence can be predicted reasonably and accurately by city-scale nonlinear THA.(2)An MS-AS sequence can be generated reasonably by the proposed MS-AS sequencegeneration method.(3)Regional seismic damage of buildings under different MS-AS scenarios can be provided efficiently by the proposed framework,which in turn can provide a useful reference for earthquake emergency response and scientific decision making for earthquake disaster relief.Xinzheng LU Qingle CHENG Zhen XU Chen XIONG 2021Frontiers of Engineering Management2021,8,1:3
7The Significance of CXCR4 Expression for the Prediction of Lymph Node Metastasis in Breast Cancer Patients显示文摘OBJECTIVE The chemokine receptor(CXCR4)CXC chemokine receptor 4)plays an important role in cancer metastasis.We therefore studied differential expression of the CXCR4,as well as that of the biomarker HER2,so as to evaluate whether these biomarkers can be used to predict axillary lymph node metastasis in breast cancer patients. METHODS Immunohistochemistry was used to evaluate the CXCR4 and HER2 expressions and to examine the paraffin sections of the breast cancers at various stages.Positive lymph node expression was found in 80 of the cases,and in 7 there was negative expression. RESULTS Compared to the cases with negative lymph nodes, there was a high expression of CXCR4(26.3% vs.14.3%,P=0.013), and an over-expression of HER2(28.8% vs.14.3%,P=0.011). Moreover,there was a direct correlation between the CXCR4 and HER2 expressions and the tumor staging(P=0.000)and lymph node metastasis(P=0.032).When the two biomarkers,i.e.CXCR4 and HER2,were concurrently labeled,a high expression of one of the biomarkers could be seen in the cases with positive lymph nodes(51.3% vs.28.6%,P<0.003). 8Bushen Jiangzhi formula reduces atherosclerosis in apoE-/-mice through autophagy显示文摘OBJECTIVE:To study the effect of Bushen Jiangzhi formula(BSJZF)on atherosclerosis(AS)in apolipoprotein E knockout(apoE^-/-)mice and the underlying mechanism.METHODS:We used a high fat diet to induce AS in apoE^-/- mice.The mice were randomly divided into four groups:model,BSJZF,atorvastatin,and 3-methyladenine groups.Syngeneic C57BL/6 mice of the same age were used for the control group.Autophagosomes in the aorta were examined by transmission electron microscopy.Morphology,lipid accumulation,and collagen deposition in the aorta were examined by hematoxylin and eosin,Oil Red O,and Masson's staining,respectively.Serum levels of tumor necrosis factor alpha(TNF-α),interferon gamma(IFN-γ),and interleukin 10(IL-10)were measured by enzyme-linked immunoassays.Protein expression of microtubule-associated protein light chain 3(LC3),Beclin 1,and p62 in the aorta were examined by Western blot analyses.RESULTS:ApoE^-/- mice fed a high fat diet exhibited AS symptoms including less autophagosomes in the aorta,higher serum levels of TNF-α,IFN-γ,and p62,and lower serum levels of IL-10,LC3,and Beclin 1.Treatment with BSJZF significantly reduced the area of the aortic plaque,decreased expression of TNF-α,IFN-γ,and p62,and increased expression of IL-10,LC3,and Beclin 1.CONCLUSION:Our findings suggest that BSJZF promotes autophagy and reduces inflammation by regulating the expression of autophagy-related proteins LC3,Beclin 1,and p62,thereby effectively treating AS.Cao Hui Jia Qingling Shen Dingzhu Yan Li Chen Chuan Xing Sanli Dou Fangfang 2020Journal of Traditional Chinese Medicine2020,40,4:3
9Implantable blood clot loaded with BMP-2 for regulation of osteoimmunology and enhancement of bone repair显示文摘The treatment of large-area bone defects still faces many difficulties and challenges.Here,we developed a blood clot delivery platform loaded with BMP-2 protein(BMP-2@BC)for enhanced bone regeneration.Blood clot gel platform as natural biomaterials can be engineered from autologous blood.Once implanted into the large bone defect site,it can be used for BMP-2 local delivery,as well as modulating osteoimmunology by recruiting a great number of macrophages and regulating their polarization at different stages.Moreover,due to the deep-red color of blood clot gel,mild localized hyperthermia under laser irradiation further accelerated bone repair and regeneration.We find that the immune niche within the bone defect microenvironment can be modulated in a controllable manner by the blood clots implantation and laser treatment.We further demonstrate that the newly formed bone covered almost 95%of the skull defect area by our strategy in both mice and rat disease models.Due to the great biocompatibility,photothermal potential,and osteoimmunomodulation capacity,such technology shows great promise to be used in further clinical translation.Qin Fan Jinyu Bai Huajian Shan Ziying Fei Hao Chen Jialu Xu Qingle Ma Xiaozhong Zhou Chao Wang 2021Bioactive Materials2021,6,11:2
10Soil temperature estimation at different depths,using remotely-sensed data显示文摘Soil temperatures at different depths down the soil profile are important agro-meteorological indicators which are necessary for ecological modeling and precision agricultural activities. In this paper, using time series of soil temperature(ST) measured at different depths(0, 5, 10, 20, and 40 cm) at agro-meteorological stations in northern China as reference data, ST was estimated from land surface temperature(LST) and normalized difference vegetation index(NDVI) derived from AQUA/TERRA MODIS data, and solar declination(Ds) in univariate and multivariate linear regression models. Results showed that when daytime LST is used as predictor, the coefficient of determination(R^2) values decrease from the 0 cm layer to the 40 cm layer. Additionally, with the use of nighttime LST as predictor, the R^2 values were relatively higher at 5, 10 and 15 cm depths than those at 0, 20 and 40 cm depths. It is further observed that the multiple linear regression models for soil temperature estimation outperform the univariate linear regression models based on the root mean squared errors(RMSEs) and R^2. These results have demonstrated the potential of MODIS data in tandem with the Ds parameter for soil temperature estimation at the upper layers of the soil profile where plant roots grow in.
