1Preliminary Study on Magnetic Induction Intensity Induced by Plasma During Hypervelocity Impact显示文摘An experimental system has been built to produce and measure the magnetic field in the backward ejected matter during hypervelocity impact.The designs of measurement system and coil,the choice of associated equipment,and the system calibration are also described in detail.The measurement of magnetic induction intensity for different given coil positions and azimuth angles are performed with two-stage light-gas gun.On condition that impact velocities are approximately equal and incidence angles are 45°,60° and 90° respectively,the relationship between average magnetic induction intensity and impact angle at different time spans is obtained.Experimental results show that the average magnetic induction intensity with incidence angle of 90° is larger than those with incidence angles of 45°and 60°.Tang Enling Zhang Qingming Zhang Jian 2009Chinese Journal of Aeronautics2009,22,4:13

3Compact, submilliwatt, 2 × 2 silicon thermo-optic switch based on photonic crystal nanobeam cavities显示文摘We propose and experimentally demonstrate a 2×2 thermo-optic(TO) crossbar switch implemented by dual photonic crystal nanobeam(PCN)cavities within a silicon-on-insulator(SOI)platform.By thermally tuning the refractive index of silicon,the resonance wavelength of the PCN cavities can be red-shifted.With the help of the ultrasmall mode volumes of the PCN cavities,only~0.16 mW power is needed to change the switching state.With a spectral passband of 0.09 nm at the 1583.75 nm operation wavelength,the insertion loss(IL)and crosstalk(CT)performances were measured as IL(bar)=-0.2 dB,CT(bar)=-15 dB,IL(cross)=-1.5 dB,and CT(cross)=-15 dB.Furthermore,the thermal tuning efficiency of the fabricated device is as high as1.23 nm/mW.HUANYING ZHOU CIYUAN QIU XINHONG JIANG QINGMING ZHU YU HE YONG ZHANG YIKAI SU RICHARD SOREF 2017Photonics Research2017,5,2:8
4Fabrication technique of micro/nano-scale speckle patterns with focused ion beam显示文摘The fabrication technique of micro/nano-scale speckle patterns with focused ion beam (FIB) system is studied for digital image correlation (DIC) measurement under a scanning electron microscope (SEM).The speckle patterns are fabricated by directly etching the counterpart of the specimen to the black part of a template.Mean intensity gradient is used to evaluate the quality of these SEM images of speckle patterns fabricated based on different templates to select an optimum template.The pattern size depending on the displacement measurement sensitivity is adjusted by altering the magnification of FIB according to the relation curve of the etching size versus magnification.The influencing factors including etching time and ion beam current are discussed.Rigid body translation tests and rotation tests are carried out under SEM to verify the reliability of the fabricated speckle patterns.The calculated values are in good agreement with the imposed ones.LI YanJie XIE HuiMin LUO Qiang GU ChangZhi HU ZhenXing CHEN PengWan ZHANG QingMing 2012Science China(Physics,Mechanics & Astronomy)2012,55,6:7
5Oxygenation,inflammatory response and lung injury during one lung ventilation in rabbits using inspired oxygen fraction of 0.6 vs.1.0显示文摘Maintaining adequate oxygenation during one-lung ventilation(OLV) requires high inspired oxygen fraction(FiO_2).However,high FiO_2 also causes inflammatory response and lung injury.Therefore,it remains a great interest to clinicians and scientists to optimize the care of patients undergoing OLV.The aim of this study was to determine and compare oxygenation,inflammatory response and lung injury during OLV in rabbits using FiO_2 of 0.6 vs.1.0.After 30 minutes of two-lung ventilation(TLV) as baseline,30 rabbits were randomly assigned to three groups receiving mechanical ventilation for 3 hours:the sham group,receiving TLV with 0.6 FiO_2;the 1.0 FiO_2 group,receiving OLV with 1.0 FiO_2;the 0.6 FiO_2 group,receiving OLV with 0.6 FiO_2.Pulse oximetry was continuously monitored and arterial blood gas analysis was intermittently conducted.Histopathologic study of lung tissues was performed and inflammatory cytokines and the mRNA and protein of nuclear factor kappa B(NF-κB) p65 were determined.Three of the 10 rabbits in the 0.6 FiO_2 group suffered hypoxemia,defined by pulse oximetric saturation(SpO_2) less than 90%.Partial pressure of oxygen(PaO_2),acute lung injury(ALI) score,myeloperoxidase(MPO),tumor necrosis factor-a(TNF-α),interleukin-6(IL-6),mRNA and protein of NF-kB p65 were lower in the 0.6 FiO_2group than in the 1.0 FiO_2 group.In conclusion,during OLV,if FiO_2 of 0.6 can be tolerated,lung injury associated with high FiO_2 can be minimized.Further study is needed to validate this finding in human subjects.Zeping Xu Lianbing Gu Qingming Bian Pengyi Li Lijun Wang Jingyuan Zhang Yanning Qian 2017The Journal of Biomedical Research2017,31,1:7
6Phase I study of CAR-T cells with PD-1 and TCR disruption in mesothelin-positive solid tumors显示文摘Programmed cell death protein-1(PD-1)-mediated immunosuppression has been proposed to contribute to the limited clinical efficacy of chimeric antigen receptor T(CAR-T)cells in solid tumors.We generated PD-1 and T cell receptor(TCR)deficient mesothelin-specific CAR-T(MPTK-CAR-T)cells using CRISPR-Cas9 technology and evaluated them in a dose-escalation study.A total of 15 patients received one or more infusions of MPTK-CAR-T cells without prior lymphodepletion.No dose-limiting toxicity or unexpected adverse events were observed in any of the 15 patients.The best overall response was stable disease(2/15 patients).Circulating MPTK-CAR-T cells peaked at days 7–14 and became undetectable beyond 1 month.TCR-positive CAR-T cells rather than TCR-negative CAR-T cells were predominantly detected in effusion or peripheral blood from three patients after infusion.We further confirmed the reduced persistence of TCR-deficient CAR-T cells in animal models.Our results establish the preliminary feasibility and safety of CRISPR-engineered CAR-T cells with PD-1 disruption and suggest that the natural TCR plays an important role in the persistence of CAR-T cells when treating solid tumors.Zhenguang Wang Na Li Kaichao Feng Meixia Chen Yan Zhang Yang Liu Qingming Yang Jing Nie Na Tang Xingying Zhang Chen Cheng Lianjun Shen Jiaping He Xun Ye Wei Cao Haoyi Wang Weidong Han 2021Cellular & Molecular Immunology2021,18,9:7
7On-chip silicon photonic 2 × 2 mode-and polarization-selective switch with low inter-modal crosstalk显示文摘Mode-and polarization-division multiplexing offer new dimensions to increase the transmission capacity of optical communications. Selective switches are key components in reconfigurable optical network nodes. An on-chip silicon 2 × 2 mode-and polarization-selective switch that can route four data channels on two modes and two polarizations simultaneously is proposed and experimentally demonstrated for the first time, to the best of our knowledge. The overall insertion losses are lower than 8.6 d B. To reduce the inter-modal crosstalk, polarization beam splitters are added to filter the undesired polarizations or modes. 8Improvement of water resistance of long afterglow SrAl_2O_4: Dy, Eu phosphor by coating with SiO_2 film显示文摘The long afterglow SrAl2O4:Dy,Eu phosphor is liable to hydrolyze in water with deterioration of the luminescent property. SrAl2O4:Dy,Eu phosphors were therefore heated at 60-90℃ in TEOS sol to form a surface gel and then heat-treated at 400℃ to obtain SiO2 coated phosphors. Observation by Transmission Electron Microscope (TEM) and X-ray photoelectron spectroscopy (XPS) shows that a thin silica film forms on the surface of the phosphors. 9Structure Analysis of Locking Mechanism of Gear-Rack Typed Ship-Lift显示文摘Contact nonlinear theory was researched. Contact problem was transformed into optimization problem containing Lagrange multiplier, and unsymmetrical stiffness matrix was transformed into symmetrical stiffness matrix. A finite element analysis (FEA) model defining more than 300 contact pairs for long nut-short screw locking mechanism of a large-scale vertical gear-rack typed ship-lift was built. Using augmented Lagrange method and symmetry algorithm of contact element stiffness, the FEA model was analyzed, and the contact stress of contact interfaces and the von Mises stress of key parts were obtained. 10Metastatic status of sentinel lymph nodes in breast cancer determined with photoacoustic microscopy via dual-targeting nanoparticles显示文摘Detection of sentinel lymph nodes(SLNs)is critical to guide the treatment of breast cancer.However,distinguishing metastatic SLNs from normal and inflamed lymph nodes(LNs)during surgical resection remains a challenge.Here,we report a CD44 and scavenger receptor class B1 dual-targeting hyaluronic acid nanoparticle(5K-HA-HPPS)loaded with the near-infra-red fluorescent dye DiR-BOA for SLN imaging in breast cancer.The small sized(~40 nm)self-assembled 5K-HA-HPPSs accumulated rapidly in the SLNs after intradermal injection.Compared with normal popliteal LNs(N-LN),there were ~3.2-fold and ~2.4-fold increases in fluorescence intensity in tumour metastatic SLNs(T-MLN)and inflamed LNs(Inf-LN),respectively,6 h after nanoparticle inoculation.More importantly,photoacoustic microscopy(PAM)of 5KHA-HPPS showed a significantly distinct distribution in T-MLN compared with N-LN and Inf-LN.Signals were mainly distributed at the centre of T-MLN but at the periphery of N-LN and Inf-LN.The ratio of PA intensity(R)at the centre of the LNs compared with that at the periphery was 5.93±0.75 for T-MLNs of the 5K-HA-HPPS group,which was much higher than that for the Inf-LNs(R=0.2±0.07)and N-LNs(R=0.45±0.09).These results suggest that 5K-HA-HPPS injection combined with PAM provides a powerful tool for distinguishing metastatic SLNs from pLNs and inflamed LNs,thus guiding the removal of SLNs during breast cancer surgery.Yanfeng Dai Xiang Yu Jianshuang Wei Fanxin Zeng Yiran Li Xiaoquan Yang Qingming Luo Zhihong Zhang 2020Light(Science & Applications)2020,9,1:6
11An analytical biomarker for treatment of patients with recurrent B-ALL after remission induced by infusion of anti-CD19 chimeric antigen receptor T(CAR-T) cells显示文摘Anti-CD19 chimeric antigen receptor-modified T(CAR-T-19) cells have emerged as a powerful targeted immunotherapy for B-cell lineage acute lymphoblastic leukemia with a remarkable clinical response in recent trials. Nonetheless, few data are available on the subsequent clinical monitoring and treatment of the patients, especially those with disease recurrence after CAR-T-19 cell infusion. Here, we analyzed three patients who survived after our phase I clinical trial and who were studied by means of biomarkers reflecting persistence of CAR-T-19 cells in vivo and predictive factors directing further treatment. One patient achieved 9-week sustained complete remission and subsequently received an allogeneic hematopoietic stem cell transplant. Another patient who showed relapse after 20 weeks without detectable leukemia in the cerebrospinal fluid after CAR-T-19 cell treatment was able to achieve a morphological remission under the influence of stand-alone low-dose chemotherapeutic agents. The third patient gradually developed extensive extramedullary involvement in tissues with scarce immune-cell infiltration during a long period of hematopoietic remission after CAR-T-19 cell therapy. Long-term and discontinuous increases in serum cytokines(mainly interleukin 6 and C-reactive protein) were identified in two patients(Nos. 1 and 6) even though only a low copy number of CAR molecules could be detected in their peripheral blood. This finding was suggestive of persistent functional activity of CAR-T-19 cells. 13High-resolution mapping of brain vasculature and its impairment in the hippocampus of Alzheimer’s disease mice显示文摘Accumulating evidence indicates the critical importance of cerebrovascular dysfunction in the pathogenesis of Alzheimer’s disease(AD).However,systematic comparative studies on the precise brain vasculature of wild-type and AD model mice are still rare.Using an image-optimization method for analysing Micro-Optical Sectioning Tomography(MOST)data,we generated cross-scale whole-brain 3 D atlases that cover the entire vascular system from large vessels down to smallest capillaries at submicron resolution,for both wild-type mice and a transgenic(APP/PS1)mouse model of AD.In addition to distinct vascular patterns in different brain regions,we found that the main vessels of the molecular layer of the hippocampal dentate gyrus(DG-ml)undergo abrupt changes in both diameter and branch angle,spreading a unique comb-like pattern of capillaries.By using a quantitative analysis workflow,we identified in the hippocampus of AD mice an overall reduction of the mean vascular diameter,volume fraction and branch angle,with most significant impairment in the DG-ml.In addition,virtual endoscopy revealed irregular morphological features in the vessel lumen of the AD mice,potentially contributing to the impairment of blood flow.Our results demonstrate the capability of high-resolution cross-scale evaluation of brain vasculature and underscore the importance of studying hippocampal microcirculation for understanding AD pathogenesis.Xiaochuan Zhang Xianzhen Yin Jingjing Zhang Anan Li Hui Gong Qingming Luo Haiyan Zhang Zhaobing Gao Hualiang Jiang 2019National Science Review2019,6,6:4
14Iterative Solution of Regularization to Ill-conditioned Problems in Geodesy and Geophysics显示文摘In geodesy and geophysics,many large-scale over-determined linear equations need to be solved which are often ill-conditioned.When the conjugate gradient method is used,their ill-conditioning effects to the solutions must be overcome,which is studied in this paper.Through the regularization ideas,the conjugate gradient method is improved,and the regularization iterative solution based on controlling condition number is put forward.Firstly by constructing the interference source vector,a new equation is derived with ill-condition diminished greatly,which has the same solution to the original normal equation.Then the new equation is solved by conjugate gradient method.Finally the effectiveness of the new method is verified by some numerical experiments of airborne gravity downward to the earth surface.In the numerical experiments the new method is compared with LS,CG and Tikhonov methods,and its accuracy is the highest.Yongwei GU Qingming GUI Xuan ZHANG Songhui HAN 2019Journal of Geodesy and Geoinformation Science2019,2,1:4
15A digitally generated ultrafine optical frequency comb for spectral measurements with 0.01-pm resolution and 0.7-μs response time显示文摘Optical spectral measurements are crucial for optical sensors and many other applications,but the prevailing methods,such as optical spectrum analysis and tunable laser spectroscopy,often have to make compromises among resolution,speed,and accuracy.Optical frequency combs are widely used for metrology of discrete atomic and molecular spectral lines.However,they are usually generated by optical methods and have large comb spacing,which limits the resolution for direct sampling of continuous spectra.To overcome these problems,this paper presents an original method to digitally generate an ultrafine optical frequency comb(UFOFC)as the frequency ruler for spectral measurements.Each comb line provides one sampling point,and the full spectrum can be captured at the same time using coherent detection.For an experimental demonstration,we adopted the inverse fast Fourier transform to generate a UFOFC with a comb spacing of 1.46 MHz over a 10-GHz range and demonstrated its functions using a Mach–Zehnder refractive index sensor.The UFOFC obtains a spectral resolution of 0.01 pm and response time of 0.7 μs;both represent 100-fold improvements over the state of the art and could be further enhanced by several orders of magnitude.The UFOFC presented here could facilitate new label-free sensor applications that require both high resolution and fast speed,such as measuring binding kinetics and single-molecule dynamics.Yuan Bao Xingwen Yi Zhaohui Li Qingming Chen Jianping Li Xudong Fan Xuming Zhang 2015Light(Science & Applications)2015,4,1:4

17Distinct proteins in cortex of rats with closed traumatic brain injury detected by a WCX-2 protein chip显示文摘BACKGROUND: Mechanical injury can cause the changes of polygene expression spectrum in rat cerebral cortical nerve cells,and then result in the changes of intracellular protein expression. At present,dielectrophoresis is combined with mass spectrum technique to detect the expression of different proteins in rat cortex after brain injury,but the protein chip technique requires further investigation. OBJECTIVE: To analyze the differences of protein expression spectrum in rat cerebral cortex before and after closed traumatic brain injury using WCX-2 protein chip technique. DESIGN: A randomized controlled animal experiment. SETTING: Training Division of the Medical College of Chinese People's Armed Police Force. MATERIALS: Seventy-two male SD rats of clean degree,350-450 g,were provided by the Experimental Animal Center,Academy of Military Medical Sciences of Chinese PLA. Urea,trifluoroacetic acid,CHAPS and Tris (Sigma,USA); WCX-2 (Ciphergen,USA). Ultra-high speed hypothermia centrifuger (Bechman,USA); Rotary tissue microtome (Keuca,Germany); Biochip processor and PBSⅡ-C protein chip reader (Ciphergen,USA). METHODS: The experiments were carried out in the Institute of Molecular Pathology,Central Laboratory,and Department of Pathology,Medical College of Chinese People's Armed Police Force from June 2005 to March 2006. ① Grouping and treatment: The experiments were completed in molecular pathological institute,central laboratory and pathological department. ① The rats were randomly divided into control group (n =12) and brain injury group (n =60). Marmarou's weight-dropping models were duplicated at different time points in the brain injury group. In the control group,the rats were only treated by incising the skin of head top,without fixing the stainless steel hitting backup plate at the vault of skull,and obtain brain cortex for pathological and protein chip research,and they were killed after 24 hours. The rats in the brain injury group were killed at 4,8,12,24 and 48 hours after model establishment. ② Pathological observation: Longitudinal section was made on cerebral cortex,and sections of 5 μm were prepared,then stained with hematoxylin and eosin (HE). ③ Protein chip analysis: 100 mg cerebral cortex was collected from each rat,and the protein content in sample was detected with Bradford method,meanwhile,WCX-2 protein chip was used to analyze the protein spectrum. The data were automatically collected with Ciphergen proteinchip 3.0 software,and the results were analyzed using Biomarker Wizard software to compare the differences of protein spectrum in rat cortex between the groups. MAIN OUTCOME MEASURES: Results of the pathological observation of cerebral cortex and the protein spectrum analysis. RESULTS: ① Pathological changes of cerebral cortex: In the control group,no necrosis and edema was observed. In the brain injury group,injures of different severity occurred at different time points; After 4 hours,focal or scattered red nerve cells could be observed,the size of some cells was increased,cytoplasm was lightly stained,and only nuclear fragments were seen; After 8 hours,the necrotic nerve cells were increased,and the number of nerve cells was reduced,astrocytes (neuronophagia) could be seen in partial cytoplasm; there was small vascular dilatation,and endothelial cell proliferation; interstitial edema,regional rarefaction lightly stained. After 12-48 hours,the necrotic nerve cells were reduced,and astrocytes proliferated. ② Results of protein spectrum analysis: The WCX-2 experiment found that the expressions of 5 639,3 212 and 7 536 u proteins in cerebral cortex changed after injury in the brain injury group. The peak intensity of 5 639 u protein in the brain injury group at 8 hours after injury was higher than that in the control group (P < 0.05); The peak intensity of 3 212 u protein in the brain injury group at 48 hours after injury was higher than that in the control group (P < 0.05); The peak intensity of 7 536 u protein at 24 hours after injury was higher than that in the control group (P < 0.05). 18Urban-rural disparities in mild cognitive impairment and its functional subtypes among community- dwelling older residents in central China显示文摘background Substantial variations in the prevalence of mild cognitive impairment(MCI)and its subtypes have been reported,although mostly in geographically defined developed countries and regions.Less is known about MCI and its subtypes in rural areas of less developed central China.Aims The study aimed to compare the prevalence of MCI and its subtypes in residents aged 65 years or older in urban and rural areas of Hubei Province,China.Methods Participants aged 65 years or older were recruited between 2018 and 2019.Inperson structured interviews and clinical and neuropsychological assessments were performed at city health community centres and township hospitals.results Among 2644 participants without dementia,735 had MCI,resulting in a prevalence of 27.8%for total MCI,20.9%for amnestic MCI(aMCI)and 6.9%for non-amnestic MCI(naMCI).The prevalence of MCI in urban and rural areas was 20.2%and 44.1%,respectively.After adjusting for demographic factors,the prevalence of total MCI,aMCI and naMCI differed significantly between rural and urban areas(adjusted odds ratio(OR)2.10,1.44 and 3.76,respectively).Subgroup analysis revealed an association between rural socioeconomic and lifestyle disadvantage and MCI and its subtypes.Conclusions Our findings suggest that the prevalence of MCI among urban residents in central China is consistent with that in other metropolis areas,such as Shanghai,but the prevalence in rural areas is twice that in urban areas.Prospective studies and dementia prevention in China should focus on rural areas.Dan Liu Lin Li Lina An Guirong Cheng Cong Chen Mingjun Zou Bo Zhang Xuguang Gan Lang Xu Yangming Ou Qingming Wu Ru Wang Yan Zeng 2021General Psychiatry2021,34,5:3
19A novel random phase-shifting digital holographic microscopy method显示文摘This paper proposes a new method that reconstructs the information of specimen by using random phase shift step in digital holographic microscopy (DHM). The principles of the method are described and discussed in detail. In practical experiment, because the phase shifter is neither perfectly linear nor calibrated, digital holograms with inaccurate phase shift step are recorded by the charge-coupled device (CCD). The phase could be accurately reconstructed from the recorded digital holograms by using the random phase-shifting algorithm, which makes up for reconstructed phase error caused by ordinary phase-shifting algorithm. The phase aberration compensation is also discussed. In order to verify the flexibility of the proposed method, numerical simulation of random phase-shifting DHM was carried out.
