1Detection of Grass Carp Hemorrhage Virus (GCHV) from Vietnam and Comparison with GCHV Strain from China显示文摘Grass carp plays an important role in small-scale aquaculture in Vietnam. However, a severe disease, known in Vietnam as 'Red Spot Disease', is causing significant economic loss in grass carp aquaculture. In this study, the tissue samples isolated from the grass carp with Red Spot Disease in Vietnam are investigated and comparied with the control GCHV isolated in China by experimental infection, culture cell infection, serological cross reactivity, and RT-PCR amplification. Infected grass carp exhibits hemorrhage symptoms about 5 days after experimental injection with GCHV-V (Vietnam) strain. The symptoms and lethality induced by the GCHV-V strain are identical to that induced by the Chinese GCHV-9014 strain. The Chinese GCHV-873 strain induces typical cytopathogenic effects in 4 cell lines, such as CIK, CAB, FHM and GCO, from the 6 fish cell lines examined. No cytopathogenic effects are observed in all the 6 examined cell lines, including CAB, FHM, CIK, EPC, CCO and GCO, infected by the GCHV-V strain and GCHV-9014 strain. Immunodiffusion assays demonstrate an obvious cross-reactivity among three GCHV strains. Precipitin lines are clearly observed not only between the anti-GCHV-873 serum and the two strains GCHV-873 and GCHV-9014, but also between the anti-GCHV-873 serum and the GCHV-V strain. GCHV can be detected by immunodiffusion assays after three generations of blind propagations in the cell lines inoculated by GCHV-V strain. This implicates that GCHV-V viruses have been replicated and amplified despite there being no cytopathogenic effects observed in these examined cell lines. Three genome segments of GCHV, including S8, S9 and S10, are amplified by three sets of PCR primers designed according to the segment sequences published recently. The Q8fp and Q8rp primer set specific for genome segment S8 amplifies a 955 bp fragment from the extracted sample of diseased fish with Red Spot Disease, and the fragment size is identical to that amplified by the same primer set from control GCHV-873 strain. Simultaneously, the Q9fp and Q9rp primer set specific for genome segment S9 generates a same 635 bp product, and the Q10fp and Q10rp primer set specific for genome segment S10 produces a same 697 bp fragment from both template samples of diseased fish with Red Spot Disease and control GCHV-873 strain. The RT-PCR amplification and corresponding size comparison data indicate that the three GCHV-V genome segments extracted from the diseased grass carp with Red Spot Disease in Vietnam should be identical to that in control GCHV-873 strain from China. 2Wnt and BMP signaling crosstalk in regulating dental stem cells:Implications in dental tissue engineering显示文摘Tooth is a complex hard tissue organ and consists of multiple cell types that are regulated by important signaling pathways such as Wnt and BMP signaling.Serious injuries and/or loss of tooth or periodontal tissues may significantly impact aesthetic appearance,essential oral functions and the quality of life.Regenerative dentistry holds great promise in treating oral/dental disorders.The past decade has witnessed a rapid expansion of our understanding of the biological features of dental stem cells,along with the signaling mechanisms governing stem cell self-renewal and differentiation.In this review,we first summarize the biological characteristics of seven types of dental stem cells,including dental pulp stem cells,stem cells from apical papilla,stem cells from human exfoliated deciduous teeth,dental follicle precursor cells,periodontal ligament stem cells,alveolar bone-derived mesenchymal stem cells(MSCs),and MSCs from gingiva.We then focus on how these stem cells are regulated by bone morphogenetic protein(BMP)and/or Wnt signaling by examining the interplays between these pathways.Lastly,we analyze the current status of dental tissue engineering strategies that utilize oral/dental stem cells by harnessing the interplays between BMP and Wnt pathways.We also highlight the challenges that must be addressed before the dental stem cells may reach any clinical applications.Thus,we can expect to witness significant progresses to be made in regenerative dentistry in the coming decade.Fugui Zhang Jinlin Song Hongmei Zhang Enyi Huang Dongzhe Song Viktor Tollemar Jing Wang Jinhua Wang Maryam Mohammed Qiang Wei Jiaming Fan Junyi Liao Yulong Zou Feng Liu Xue Hu Xiangyang Qu Liqun Chen Xinyi Yu Hue H.Luu Michael J.Lee Tong-Chuan He Ping Ji 2016Genes & Diseases2016,3,4:16
3Effect of acupotomy on nitric oxide synthase and beta-endorphin in third lumbar vertebrae transverse process syndrome model rats显示文摘OBJECTIVE: To explore the long-term effects and pain relief mechanism of acupotomy by observing changes in nitric oxide synthase(NOS) and beta-endorphin(β-EP) in the hypothalamus, spinal cord,and peripheral blood of rats with third lumbar vertebrae(L3) transverse process syndrome.METHODS:Twenty-eight SD rats were randomly assigned to normal, model, electroacupuncture(EA),and acupotomy group. The last three groups were put through an operation to emulate L3transverse process syndrome. Fourteen days after the simulation operation, EA and acupotomy treatments were applied to the respective groups. Fifty-six days afterthe simulation operation, biochemistry tests and enzyme-linked immunosorbent assay were used to measure NOS and β-EP in the hypothalamus, spinal cord, and peripheral blood.RESULTS: Rats with the simulation operation showed significantly higher levels of NOS and β-EP in the hypothalamus, spinal cord, and peripheral blood than those in the normal group. 5Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. 6Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. 7Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 9Influence of luteolin on the apoptosis of esophageal cancer Eca109 cells and its mechanism of action显示文摘The present study was conducted to verify the influence of luteolin on apoptosis of Eca109 cells and to further investigate the possible mechanisms underlying its effect on apoptosis.The cells were exposed to different concentrations of luteolin(0,40,80,120,160,200,240M)for 24,48,and 72 h respectively.The influence of luteolin on proliferation of Eca109 cells was detected using MTT assay.Eca109 cells were then treated with luteolin(0,40,160,240M)for 24 h.The effect of luteolin on cell cycle progression and apoptosis was assayed by using flow cytometry(FCM).Expression of caspase9 and caspase3 mRNA and protein was analyzed by real-time PCR and Western blot respectively.The results showed that luteolin could inhibit the proliferation of Eca109 cells at all concentrations in a time-dependent manner and the relative inhibition rate showed an inverted U-shaped association with the concentration of luteolin.Further,the cell cycle was arrested in the S phase following treatment with luteolin.Apoptosis analysis indicated that luteolin could induce the apoptosis of Eca109 cells across the three concentration groups,which exhibited a trend of first promotional and then inhibitory with the increases in luteolin concentration.The effect of luteolin on the mRNA and protein expression of caspase 9 and caspase3 first manifested as promotion,then inhibition.Therefore,luteolin may serve a role in promoting cell apoptosis by inducing Eca109 cell apoptosis that involves the expression of caspase3,caspase9 mRNA and protein.This study provides theoretical basis for further study and clinical application of luteolin.The specific mechanism has not yet been clarified and the other activation pathways inducing apoptosis need to be further studied.Shaokang Wang Lingmeng Fu Yi Wu Hongmei Xiao Jing Wang Guiju Sun 2019Food Science and Human Wellness2019,8,2:6
10Sustained high level transgene expression in mammalian cells mediated by the optimized piggyBac transposon system显示文摘Sustained,high level transgene expression in mammalian cells is desired in many cases for studying gene functions.Traditionally,stable transgene expression has been accomplished by using retroviral or lentiviral vectors.However,such viral vector-mediated transgene expression is often at low levels and can be reduced over time due to low copy numbers and/or chromatin remodeling repression.The piggyBac transposon has emerged as a promising nonviral vector system for efficient gene transfer into mammalian cells.Despite its inherent advantages over lentiviral and retroviral systems,piggyBac system has not been widely used,at least in part due to their limited manipulation flexibilities.Here,we seek to optimize piggyBac-mediated transgene expression and generate a more efficient,user-friendly piggyBac system.By engineering a panel of versatile piggyBac vectors and constructing recombinant adenoviruses expressing piggyBac transposase(PBase),we demonstrate that adenovirusmediated PBase expression significantly enhances the integration efficiency and expression level of transgenes in mesenchymal stem cells and osteosarcoma cells,compared to that obtained from co-transfection of the CMV-PBase plasmid.We further determine the drug selection timeline to achieve optimal stable transgene expression.Moreover,we demonstrate that the transgene copy number of piggyBac-mediated integration is approximately 10 times higher than that mediated by retroviral vectors.Using the engineered tandem expression vector,we show that three transgenes can be simultaneously expressed in a single vector with high efficiency.Thus,these results strongly suggest that the optimized piggyBac system is a valuable tool for making stable cell lines with sustained,high transgene expression.Xiang Chen Jing Cui Zhengjian Yan Hongmei Zhang Xian Chen Ning Wang Palak Shah Fang Deng Chen Zhao Nisha Geng Melissa Li Sahitya K.Denduluri Rex C.Haydon Hue H.Luu Russell R.Reid Tong-Chuan He 2015Genes & Diseases2015,2,1:5

12Multicenter phaseⅡstudy of apatinib single or combination therapy in HER2-negative breast cancer involving chest wall metastasis显示文摘Objective:Breast cancer(BC)with chest wall metastasis(CWM)usually shows rich neovascularization.This trial explored the clinical effect of apatinib on human epidermal growth factor receptor 2(HER2)-negative advanced BC involving CWM.Methods:This trial involved four centers in China and was conducted from September 2016 to March 2020.Patients received apatinib 500 mg/d[either alone or with endocrine therapy if hormone receptor-positive(HR+)]until disease progression or unacceptable toxicity.Progression-free survival(PFS)was the primary endpoint.Results:We evaluated 26 patients for efficacy.The median PFS(mPFS)and median overall survival(mOS)were4.9[range:2.0-28.5;95%confidence interval(95%CI):2.1-8.3]months and 18(range:3-55;95%CI:12.9-23.1)months,respectively.The objective response rate(ORR)was 42.3%(11/26),and the disease-control rate was76.9%(20/26).In the subgroup analysis,HR+patients compared with HR-negative patients had significantly improved mPFS of 7.0(95%CI:2.2-11.8)months vs.2.3(95%CI:1.2-3.4)months,respectively(P=0.001);and mPFS in patients without or with chest wall radiotherapy was 6.4(95%CI:1.6-19.5)months vs.3.0(95%CI:1.3-4.6)months,respectively(P=0.041).In the multivariate analysis,HR+status was the only independent predictive factor for favorable PFS(P=0.014).Conclusions:Apatinib was highly effective for BC patients with CWM,especially when combined with endocrine therapy.PFS improved significantly in patients with HR+status who did not receive chest wall radiotherapy.However,adverse events were serious and should be carefully monitored from the beginning of apatinib treatment.Huiping Li Cuizhi Geng Hongmei Zhao Hanfang Jiang Guohong Song Jiayang Zhang Yaxin Liu Xinyu Gui Jing Wang Kun Li Zhongsheng Tong Fangyuan Zhao Junlan Yang Guoliang Chen Qianyu Liu Xu Liang 2021Chinese Journal of Cancer Research2021,33,2:4
13The combination of chidamide with the CHOEP regimen in previously untreated patients with peripheral T-cell lymphoma: a prospective, multicenter, single arm, phase 1b/2 study显示文摘Objective:To assess the efficacy and safety of the novel histone deacetylase inhibitor,chidamide,in combination with cyclophosphamide,doxorubicin,vincristine,etoposide,and prednisone(Chi-CHOEP)for untreated peripheral T-cell lymphoma(PTCL).Methods:A prospective,multicenter,single arm,phase 1 b/2 study was conducted.A total of 128 patients with untreated PTCL(18–70 years of age)were enrolled between March 2016 and November 2019,and treated with up to 6 cycles with the Chi-CHOEP regimen.In the phase 1 b study,3 dose levels of chidamide were evaluated and the primary endpoint was determination of the maximumtolerated dose and recommended phase 2 dose(RP2 D).The primary endpoint of the phase 2 study was 2-year progression-free survival(PFS).Results:Fifteen patients were enrolled in the phase 1 b study and the RP2 D for chidamide was determined to be 20 mg,twice a week.A total of 113 patients were treated at the RP2 D in the phase 2 study,and the overall response rate was 60.2%,with a complete response rate of 40.7%.At a median follow-up of 36 months,the median PFS was 10.7 months,with 1-,2-,and 3-year PFS rates of 49.9%,38.0%,and 32.8%,respectively.The Chi-CHOEP regimen was well-tolerated,with grade 3/4 neutropenia occurring in approximately two-thirds of the patients.No unexpected adverse events(AEs)were reported and the observed AEs were manageable.Conclusions:This large cohort phase 1 b/2 study showed that Chi-CHOEP was well-tolerated with modest efficacy in previously untreated PTCL patients.Wei Zhang Liping Su Lihong Liu Yuhuan Gao Quanshun Wang Hang Su Yuhuan Song Huilai Zhang Jing Shen Hongmei Jing Shuye Wang Xinan Cen Hui Liu Aichun Liu Zengjun Li Jianmin Luo Jianxia He Jingwen Wang O.A.O’Connor Daobin Zhou 2021Cancer Biology & Medicine2021,18,3:4

15Al-Si/Al_2O_3 in situ composite prepared by displacement reaction of CuO/Al system显示文摘Al2O3 particle-reinforced ZL109 composite was prepared by in situ reaction between CuO and Al.The microstructure was observed by means of OM,SEM and TEM.The Al2O3 particles in sub-micron sizes distribute uniformly in the matrix,and the Cu displaced from the in situ reaction forms net-like alloy phases with other alloy elements.The hardness and the tensile strength of the composites at room temperature have a slight increase as compared to that of the matrix.However,the tensile strength at 350 ℃ has reached 90.23 MPa,or 16.92 MPa higher than that of the matrix.The mechanism of the reaction in the CuO/Al system was studied by using of differential scanning calorimetry(DSC) and thermodynamic calculation.The reaction between CuO and Al involves two steps.First,CuO reacts with Al to form Cu2O and Al2O3 at the melting temperature of the matrix alloy,and second,Cu2O reacts with Al to form Cu and Al2O3 at a higher temperature.At ZL109 casting temperature of 750-780 ℃,the second step can also take place because of the effect of exothermic reaction of the first step.Zhang Jing Yu Huashun Chen Hongmei Min Guanghui 2010China Foundry2010,7,1:3
16Comprehensive and high-resolution emission inventory of atmospheric pollutants for the northernmost cities agglomeration of Harbin-Changchun,China:Implications for local atmospheric environment management显示文摘Using a bottom-up estimation method,a comprehensive,high-resolution emission inventory of gaseous and particulate atmospheric pollutants for multiple anthropogenic sectors with typical local sources has been developed for the Harbin-Changchun city agglomeration(HCA).The annual emissions for CO,NO_(x),SO_(2),NH_(3),VOC S,PM_(2.5),PM 10,BC and OC during 2017 in the HCA were estimated to be 5.82 Tg,0.70 Tg,0.34 Tg,0.75 Tg,0.81 Tg,0.67 Tg,1.59 Tg,0.12 Tg and 0.26 Tg,respectively.For PM 10 and SO_(2),the emissions from industry processes were the dominant contributors representing 54.7%and 49.5%,respectively,of the total emissions,while 95.3%and 44.5%of the total NH_(3)and NO x emissions,respectively,were from or associated with agricultural activities and transportation.Spatiotemporal distributions showed that most emissions(except NH_(3))occurred in November to March and were concentrated in the central cities of Changchun and Harbin and the surrounding cities.Open burning of straw made an important contribution to PM_(2.5)in the central regions of the northeastern plain during autumn and spring,while domestic coal combustion for heating purposes was significant with respect to SO_(2)and PM_(2.5)emissions during autumn and winter.Furthermore,based on Principal Component Analysis and Multivariable Linear Regression model,air temperature,relative humidity,electricity and energy consumption,and the urban and rural population were optimized to be representative indicators for rapidly assessing the magnitude of regional atmospheric pollutants in the HCA.Such indicators and equations were demonstrated to be useful for local atmospheric environment management.Mengduo Zhang Weiwei Chen Xiangjin Shen Hongmei Zhao Chengkang Gao Xuelei Zhang Wei Liu Chengjiang Yang Yang Qin Shichun Zhang Jing Fu Daniel Tong Aijun Xiu 2021Journal of Environmental Sciences2021,33,6:3
17Effect of river plume on phytoplankton community structure in Zhujiang River estuary显示文摘To examine the phytoplankton assemblages and the effect of diluted waters on them,a research cruise was conducted from July 19 to August 7,2015 in the Zhujiang(Pearl)River estuary in the northern South China Sea(21°N–23.5°N,111°E–117°E).Samples were collected from 65 stations including one for time-series sampling.A total of 212 phytoplankton taxa were identified from 61 genera belonging to 4 phyla.Among them,122 species identified from 42 genera of Bacillariophyta and 83 species from 15 genera of Pyrrophyta.Chain-forming diatoms dominated the phytoplankton community where Pseudonitzschia delicatissima,Guinardia striata,Thalassionema nitzschioides,and P.pungens comprised about 52%of the total abundance.However,higher cell abundances concentrated on both sides of the estuary,because of low salinity and high nutrients brought by diluted water.In addition,Canonical Correspondence Analysis revealed that salinity and dissolved inorganic nitrogen shaped the species composition in the study area.Furthermore,the Jaccard similarity index showed prevailing high similarity in the distribution of species in low-salinity diluted waters,and the Bray-Curtis similarity depicted distinguished grouping for phytoplankton assemblages along the salinity gradient.However,phytoplankton diel vertical cycles showed maximum abundance occurred at 2:00 am,which was mainly contributed by benthic phytoplankton species Pseudo-nitzschia spp.and T.nitzschioides.Qi ZHONG Bing XUE Md Abu NOMAN Yuqiu WEI Haijiao LIU Hongbin LIU Liping ZHENG Hongmei JING Jun SUN 2021Journal of Oceanology and Limnology2021,39,2:2
18Gene expression profiling under different photoperiod/temperature conditions in a photoperiod-/thermo-sensitive genic male sterile line of rice(Oryza sativa L.)显示文摘Peiai 64S is a photoperiod- and thermo- sensitive genic male sterile (PTSGMS) line of rice, which is male sterile at long day/high temperature and partial fertile at short day/low temperature. A cDNA array representing 3328 unique rice genes was used to profile the gene expression patterns in the young panicles of Peiai 64S under these two condi- tions. The statistical data showed that up to 14.60% of genes exhibited up-or down-regulated expressions in the plants at long day/high temperature compared with plants at short day/low temperature. Only four genes were up-regulated while 482 genes down-regulated. Real-time PCR with all the up-regulated and 9 randomly selected down-regulated genes confirmed the differential expressions detected by the array, indicating that the constructed cDNA array is reliable. These differently expressed genes participated in almost all cell biological responses. Analysis of up- and down-regulated genes revealed distinctive changes between the mRNA abundances of MMK1 and MMK2, both of which are analogs ofMAPK, and significant down-regulation of several transcription factors. 19Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. 20Mechanism of micro-wetting of highly hydrophobic coal dust in underground mining and new wetting agent development显示文摘The internal mechanism of the high hydrophobicity of the coal samples from the Pingdingshan mining area was studied through industrial,element,and surface functional group analysis.Laboratory testing and molecular dynamics simulations were employed to study the impact of three types of surfactants on the surface adsorption properties and wettability of highly hydrophobic bituminous coal.The results show that the surface of highly hydrophobic bituminous coal is compact,rich in inorganic minerals,and poorly wettable and that coal molecules are dominated by hydrophobic functional groups of aromatic rings and aliphatic structures.The wetting performance of surfactants as the intermediate carrier to connect coal and water molecules is largely determined by the interaction force between surfactants and coal(Fs-c)and the interaction force between surfactants and water(Fs-w),which effectively improve the wettability of modified coal dust via modifying its surface electrical properties and surface energy.A new type of wetting agent with a dust removal rate of 89%has been developed through discovery of a compound wetting agent solution with optimal wetting and settling performance.This paper provides theoretical and technical support for removing highly hydrophobic bituminous coal dust in underground mining.Mingzhong Gao Hongmei Li Yun Zhao Yiting Liu Weiqi Zhou Luming Li Jing Xie Jie Deng 2023International Journal of Mining Science and Technology2023,33,1:2
