Physical-biogeochemical interactions and potential effects on phytoplankton and Ulva prolifera in the coastal waters off Qingdao(Yellow Sea, China)显示文摘In recent years, the spectacular massive green tide of Ulva prolifera has become a recurrent phenomenon appearing every summer in the coastal waters off Qingdao(Yellow Sea, China), attracting the attention of scientists and local government. Based on multidisciplinary data collected during summer and winter, this study focuses on the hydrological characteristics and regional biogeochemical processes in coastal waters off Qingdao. The results show that the boundary of the Yellow Sea Cold Water Mass(YSCWM) can reach the Qingdao coastal region in summer and is locally raised to the upper layers to form coastal upwelling beyond tidal mixing and favorable wind. The regional summer upwelling off the Qingdao coast effectively enriches the nutrient concentrations in the upper water column and thus promotes growth of phytoplankton but reduces the dissolved oxygen(DO) concentration and pH value in the bottom. The regional summer upwelling off Qingdao coast may facilitate the growth and regional blooming of the U. prolifera that migrate to this region with the southerly wind.Additionally, the effects of the front on the aggregation of U. prolifera may be significant. In winter, the Yellow Sea Warm Current(YSWC) extends and spreads along the offshore region off the Subei Shoal towards the Qingdao coastal sea. This tongue-shaped warm water meets the cold coastal water off Qingdao, which leads to the formation of a physical front. As a consequence, remarkable fronts of nutrient and chlorophyll a(Chl a) also form between the shoreward warm water and the cold coastal water. This study increases the understanding of the interactions between the regional physical, chemical, and biological processes off the Qingdao coast. | Qinsheng Wei Baodong Wang Qingzhen Yao Zhigang Yu Mingzhu Fu Junchuan Sun Bochao Xu Linping Xie Ming Xin | 2019 | Acta Oceanologica Sinica2019,38,2: | 4
Spatiotemporal variability of physical-biogeochemical processes and intrinsic correlations in the semi-enclosed South Yellow Sea显示文摘Investigating the spatiotemporal variability of biogeochemical processes and ecological responses under multiple physical controls in shelf seas is of great importance for obtaining an in-depth understanding of marine ecosystem.Based on a compiled data set of historical observations and remote sensing data, the spatiotemporal variability and heterogeneity of physical-biogeochemical processes in the semi-enclosed South Yellow Sea(SYS)are investigated, and the intrinsic connectivity among different subregions and the associated mechanisms are examined. The results show that the seasonal alternation between southward transport in cold seasons and upwelling-induced vertical delivery in warm seasons is the primary physical control of the biogeochemical processes and primary production off Shidao and in the area adjacent to the Haizhou Bay.The northeastward expansion of coastal waters in the Subei Shoal constitutes an important physical driver for the offshore transport of Ulva prolifera in summer.Stratification significantly affects the biogeochemical processes in the Yellow Sea Cold Water Mass(YSCWM)-dominated area during warm seasons, and nutrients can accumulate in bottom waters from spring to autumn, making the Yellow Sea Cold Water Mass(YSCWM)be an important nutrient pool.Upwelling around the YSCWM boundary in the stratified season leads to consistency among the high chlorophyll a(Chl a)area, high primary productivity region and low-temperature upwelling zone.During cold seasons, the interactions of the southward cold waters in the western nearshore area and the northward warm waters in the central region lead to an“S”-shaped front in the SYS.In summer, upwelling can extract nutrients from the YSCWM;thus, the biogeochemical-ecological processes inside the cold-water mass and in the frontal zone are well connected via upwelling, and three typical physical-biogeochemical coupling regions are generated, namely, the Shidao coast, the area beyond the Haizhou Bay and the area off the Subei Shoal. This work refines and integrates studies on regional oceanography in the SYS and provides a comprehensive and systematic framework of physical-biogeochemical-ecological processes. | Qinsheng Wei Baodong Wang Mingzhu Fu Junchuan Sun Qingzhen Yao Ming Xin Zhigang Yu | 2020 | Acta Oceanologica Sinica2020,39,10: | 3

Phytoplankton pigment pattern in the subsurface chlorophyll maximum in the South Java coastal upwelling system, Indonesia显示文摘Upwelling occurs on the coast of Java between June and October, forced by local alongshore winds associated with the southeasterly monsoon. This causes variations in phytoplankton community composition in the upwelling zone compared with the surrounding offshore area. Based on pigments analysis with subsequent calculations of group contributions to total chlorophyll a(Chl a) using CHEMTAX, we studied the distribution and composition of phytoplankton assemblages in the subsurface chlorophyll maximum along the south coast of Java and the influence of upwelling. Nineteen phytoplankton pigments were identified using high-performance liquid chromatography, and CHEMTAX analysis associated these to ten major phytoplankton groups. The phytoplankton community in the coastal area influenced by upwelling was characterized by high Chl a and fucoxanthin concentrations, indicating the dominance of diatoms. In contrast, in the offshore area, the Chl a and fucoxanthin concentrations declined to very low levels and the community was dominated by haptophytes represented by 19′-Hexanoyloxyfucoxanthin. Accordingly, microphytoplankton was found to be the major size class in the coastal area influenced by upwelling, while nanophytoplankton was most abundant in the offshore area. Low concentrations of other accessory pigments indicated less contribution from dinoflagellates, prasinophytes, chlorophytes and cryptophytes. Photo-pigment indices revealed that photosynthetic carotenoids(PSCs) were the largest component of the pigment pool, exceeding the proportion of Chl a, with the average PSCTP up to 0.62. These distribution trends can mainly be explained by phytoplankton adaption strategies to upwelling and subsurface conditions by changing species composition and adjusting the pigment pool. | GAO Chunlei FU Mingzhu SONG Hongjun WANG Lei WEI Qinsheng SUN Ping LIU Lin ZHANG Xuelei | 2018 | Acta Oceanologica Sinica2018,37,12: | 1
Does reduced sediment load contribute to increased outbreaks of harmful algal blooms off the Changjiang Estuary?显示文摘Harmful algal blooms(HABs) have been increasingly frequent in coastal waters around the world over the last several decades. Accelerated coastal eutrophication, resulting from the increased anthropogenic loadings of nutrients, is commonly assumed to be the primary cause of this increase. However, although important, accelerated coastal eutrophication may not be the only explanation for the increasing blooms or toxic outbreaks in estuarine waters. Changes in riverine material fluxes other than nutrients, such as sediment load, may significantly affect biological activities and HAB incidence in estuarine and coastal waters. A case study off the Changjiang(Yangtze River) Estuary indicated that with the increasing riverine loadings of nutrients, the sediment load from the Changjiang River has been reduced by 70% over the past four decades. A comparison of long-term data revealed that the phytoplankton biomass maximum has expanded to a region of much lower salinity due to the drastic reduction in riverine sediment load and the subsequent improvement in light penetration in the Changjiang River plume. Furthermore, there was an apparent mirror-image relationship between the sediment load from the Changjiang River and the HAB incidence off the Changjiang Estuary over the past four decades, and the number of HAB incidents was significantly negatively correlated with the sediment load. Therefore, it is argued that the drastic decline in sediment load from the Changjiang River reduced turbidity in the Changjiang Estuary and thus contributed to the increased frequency of HABs in the buoyant discharge plumes. | WANG Baodong XIN Ming SUN Xia WEI Qinsheng ZHANG Xuelei | 2016 | Acta Oceanologica Sinica2016,35,8: | 1
Spatial Distribution and Seasonal Variation of Hypoxic Zone in the Eastern Equatorial Indian Ocean显示文摘The spatial distribution and seasonal variations of the hypoxic zone in the eastern equatorial Indian Ocean were investigated using survey data collected from four cruises from 2013 to 2018.Results showed that hypoxic zone occurred all year round in the eastern equatorial Indian Ocean, and it spread southward in the shape of a double tongue at two depths with one at subsurface centered at a depth of 150 m and the other in intermediate water centered at a depth of 800 m.The southward expansion and maximum thickness of the hypoxic zone were greatest in the spring inter-monsoon and least in the summer monsoon. The hypoxic zone originated from the southward expansion of the hypoxic water in the Bay of Bengal and its spatial distribution was driven by southward output flux of mid-deep(100–1000 m)hypoxic water from the Bay of Bengal. The hypoxia southward expansion was blocked near the equator in the subsurface layer, because of mixing with multiple zonal circulations(e.g.,Wyrtki Jets and the equatorial undercurrent),which meant that the hypoxic zone extended over a smaller area than in the intermediate water. These new findings will contribute to an improved understanding of the hypoxic zone and will contribute to circulation research, particularly about intermediate circulation in the eastern equatorial Indian Ocean. | XIE Linping WANG Baodong XIN Ming WANG Ying SUN Xia WEI Qinsheng LIU Lin YUAN Chao | 2023 | Journal of Ocean University of China2023,22,4: | 0
Combined inhibition of HDAC and DNMT1 induces p85α/MEKmediated cell cycle arrest by dual target inhibitor 208 in U937 cells显示文摘Multiple histone deacetylase inhibitors (HDACi) and DNA methyltransferase inhibitors (DNMTi) have been developed for cancer therapy. However, the research on their mechanisms of action is not sophisticated enough. In this study, we reported a dual HDAC and DNMT inhibitor 208 and found it induced G1 cell cycle arrest and apoptosis in U937 cells. Proteome and bioinformatic analyses revealed that the combined inhibition of DNMT1 and HDAC by 208 affected the expression of a series of proteins involved in many biological processes. We observed that several proteins associated with G1 cell cycle arrest and apoptosis were down regulated after 208 treatment, including p85α, MEK, and CDK4, suggesting that 208 induces cell cycle arrest and apoptosis through the p85α/MEK-mediated pathway in U937 cells. Moreover, biological function analysis showed that the combined epigenetic inhibition influenced various processes, including the synthesis and processing of RNA, translation, protein transport, and DNA repair. These findings provide novel insight into the potential mechanisms of multifunctional epigenetic inhibitors, which supports their further improvement and development. | Yue Ren Qinsheng Sun Zigao Yuan Yuyang Jiang | 2019 | Chinese Chemical Letters2019,30,6: | 0
ABI5 promotes heat stress-induced chlorophyll degradation by modulating the stability of MYB44 in cucumber显示文摘The yellowing of leaves caused by the decomposition of chlorophyll(Chl)is a characteristic event during senescence, which can be induced by various environmental stresses. However, the molecular mechanisms of high temperature-induced Chl degradation in horticultural plants remain poorly understood. Here,we found that heat stress induced Chl degradation and the expression of ABI5 and MYB44 in cucumber.Silencing of ABI5 compromised heat stress-induced Chl degradation, and the transcription of pheophytinase(PPH)and pheophorbide a oxygenase(PAO),two key genes in Chl catabolic pathway, but silencing of MYB44 exhibited the opposite results.Furthermore,ABI5 interacted with MYB44 in vitro and in vivo.ABI5 positively regulated heat stress-induced Chl degradation through two pathways.ABI5 directly bound to PPH and PAO promoters to promote their expression, leading to accelerating Chl degradation. On the other hand, the interaction between ABI5 and MYB44 reduced the binding of MYB44 to PPH and PAO promoters and led to the ubiquitination-depended protein degradation of MYB44,thereby alleviating the transcription inhibitory effect of MYB44 on PPH and PAO.Taken together, our findings propose a new regulatory network for ABI5 in regulating heat stress-induced Chl degradation. | Weikang Liu Guangling Chen Mingming He Jianqiang Wu Wenxu Wen Qinsheng Gu Shirong Guo Yu Wang Jin Sun | 2023 | Horticulture Research2023,10,6: | 0

Design, synthesis and antitumor evaluations of nucleoside base hydroxamic acid derivatives as DNMT and HDAC dual inhibitors显示文摘DNA methyl transferase(DNMT) and histone deacetylase(HDAC) are well recognized epigenetic targets for discovery of antitumor agents. In this study,we designed and synthesized a series of nucleoside base hydroxamic acid derivatives as DNMT and HDAC dual inhibitors.MTT assays and enzymatic inhibitory activity tests indicated that compound 204 exhibited potent DNMT1 and HDAC1/6 inhibitory potency simultaneously in enzymatic levels and at cellular levels, inducing hypomethylation of p16 and hyperacetylation of histones H_(3) K9 and H4 K8.Besides,204 remarkably inhibited proliferation against cancer cells U937 by prompting G0/G1 cell cycle arrest.Molecular docking models explained the functional mechanism of 204 inhibiting DNMT1 and HDAC.Preliminary studies on metabolic profiles revealed that 204 showed desirable stability in liver microsomes. Our study suggested that 204 inhibiting DNMT and HDAC concurrently can be a potential lead compound for epigenetic cancer therapy. | Qinsheng Sun Qiuzi Dai Cunlong Zhang Yan Chen Lei Zhao Zigao Yuan Yuyang Jiang | 2021 | Chinese Chemical Letters2021,32,8: | 0
