1A novel gene delivery system targeting cells expressing VEGF receptors显示文摘Two ligand oligopeptides GV1 and GV2 were designed according to the putative binding region of VEGF to its receptors. GV1, GV2 and endosome releasing oligopeptide HA20 were conjugated with poly-L-lysine or protamine and the resulting conjugates could interact with DNA in a noncovalent bond to form a complex. Using pSV2-β-galactosidase as a reporter gene, it has been demonstrated that exogenous gene was transferred into bovine aortic arch-derived endothelial cells (ABAE) andhuman malignant melanoma cell lines (A375) in vitro. In vivo experiments, exogenous gene was transferred into tumor vascular endothelial cells and tumor cells of subcutaneously transplanted human colon cancer LOVO, human malignant melanoma A375 and human hepatoma graft in nude mice. This system could also target gene to intrahepatically transplanted human hepatoma injected via portal vein in nude mice. 4Over-expression of fibroblast growth factor receptor 3 in human hepatocellular carcinoma显示文摘AIM: To describe the significant over-expression of fibroblast growth factor receptor 3 (FGFR3), which is a signal transduction and cell proliferation related gene in hepatocellular carcinoma (HCC).METHODS: Following DNA microarray, Northern blot and quantitative real-time PCR were employed to confirm FGFR3 expression difference in HCC tissues and surrounding non-neoplastic liver tissue. FGFR3 expression levels were further determined by immunohistochemical study in 43 cases of HCC.RESULTS: Northern blot results showed the significant over-expression of FGFR3 in HCC tissues, which was consistent with that from DNA microarray. Quantitative real-time PCR demonstrated that the mean ratio of FGFR3 mRNA to glyceraldehyde-3-phosphate dehydrogenase (GADPH) mRNA in HCC tissue was 0.250, whereas the ratio in non-neoplastic liver tissue was 0.014. Statistical analyses of 43 cases of HCC revealed that HCC scored higher than the matched non-neoplastic liver tissues.Examination of clinicopathological features revealed a strong correlation of over-expression of FGFR3 with poor tumor differentiation and high nuclear grade.CONCLUSION: Over-expression of FGFR3 may play an important role in liver carcinogenesis. 6Effects of Heyan Kuntai Capsule(和颜坤泰胶囊)on Follicular Development and Oocyte Cohesin Levels in Aged Mice显示文摘Objective: To evaluate the effect of Heyan Kuntai Capsule(和颜坤泰胶囊, HYKT) on the ovarian function of aged mice and expressions of cohesion complexes in oocytes. Methods: Twenty-five 9-month-old female C57BL/6J mice were randomly divided into 5 groups by block randomization method(n=5 per group), including the control group(saline), 17β-estradiol group [E_2, 100 μg/(kg·d)], and low-, medium-, and highdose of HYKT groups [0.3, 0.9, 2.7 g/(kg·d), respectively]. All mice were treated by intragastric administration for 4 weeks. Hematoxylin and eosin staining and anti-VASA staining were used to detect the amounts of follicles. The apoptosis of follicles was measured by anti-gamma H_2A histone family member X(γH_2AX) staining and Td T-mediated d UTP Nick-End Labeling(TUNEL) assay. The density of cohesin subunits, REC8 meiotic recombination protein(REC8), structural maintenance of chromosome(SMC) 1β and SMC3 in oocytes were evaluated by immunofluorescent staining. Results: After administration of E_2 and high-dose of HYKT, the total number of follicles as well as the number of primordial and primary follicles were significantly increased(P<0.05). Anti-γH_2AX staining and TUNEL assay demonstrated that high-dose of HYKT and E_2 partly suppressed the apoptosis of follicles(P<0.05). Furthermore, it showed an increased trend in the levels of REC8 and SMC1β, after administration with E_2 and HYKT, and no obvious change in the level of SMC3. 7Improvement cues of lesion absorption using the adjuvant therapy of traditional Chinese medicine Qinbudan tablet for retreatment pulmonary tuberculosis with standard anti-tuberculosis regimen显示文摘Background:China is the second highest pulmonary tuberculosis(PTB)burden country worldwide.However,retreatment of PTB has often developed resistance to at least one of the four first-line anti-TB drugs.The cure rate(approximately 50.0–73.3%)and management of retreatment of PTB in China needs to be improved.Qinbudan decoction has been widely used to treat PTB in China since the 1960s.Previously clinical studies have shown that the Qinbudan tablet(QBDT)promoted sputum-culture negative conversion and lesion absorption.However,powerful evidence from a randomized controlled clinical trial is lacking.Therefore,the aim of this study was to compare the efficacy and safety of QBDT as an adjunct therapy for retreatment of PTB.Methods:We conducted a multicenter,randomized,double-blind,placebo-controlled clinical trial in China.People diagnosed with PTB were enrolled who received previous anti-TB treatment from April 2011 to March 2013.The treatment group received an anti-TB regimen and QBDT,and the control group was administered an anti-TB regimen plus placebo.Anti-TB treatment options included isoniazid,rifampicin,pyrazinamide,ethambutol,streptomycin for 2 months(2HRZES),followed by isoniazid,rifampicin,ethambutol for 6 months(6HRE),daily for 8 months.Primary outcome was sputum-culture conversion using the MGIT 960 liquid medium method.Secondary outcomes included lung lesion absorption and cavity closure.Adverse events and reactions were observed after treatment.A structured questionnaire was used to record demographic information and clinical symptoms of all subjects.Data analysis was performed by SPSS 25.0 software in the full analysis set(FAS)population.Results:One hundred eighty-one cases of retreatment PTB were randomly divided into two groups:the placebo group(88 cases)and the QBDT group(93 cases).A total of 166 patients completed the trial and 15 patients lost to follow-up.The culture conversion rate of the QBDT group and placebo group did not show a noticeable improvement by using the covariate sites to correct the rate differences(79.6%vs 69.3%;rate difference=0.10,95%confidence interval(CI):-0.02–0.23;F=2.48,P=0.12)after treatment.A significant 16.6%increase in lesion absorption was observed in the QBDT group when compared with the placebo group(67.7%vs 51.1%;rate difference=0.17,95%CI:0.02–0.31;χ2=5.56,P=0.02).The intervention and placebo group did not differ in terms of cavity closure(25.5%vs 21.1%;rate difference=0.04,95%CI:-0.21–0.12;χ2=0.27,P=0.60).Two patients who received chemotherapy and combined QBDT reported pruritus/nausea and vomiting.Conclusions:No significant improvement in culture conversion was observed for retreatment PTB with traditional Chinese medicine plus standard anti-TB regimen.However,QBDT as an adjunct therapy significantly promoted lesion absorption,thereby reducing lung injury due to Mycobacterium tuberculosis infection.Trial registration:This trial is registered at ClinicalTrials.gov,NCT02313610.Shao-Yan Zhang Ji-You Fu Xiao-Yan Guo Ding-Zhong Wu Tong Zhang Cui Li Lei Qiu Chang-Rong Shao He-Ping Xiao Nai-Hui Chu Qun-Yi Deng Xia Zhang Xiao-Feng Yan Zhao-Long Wang Zhi-Jie Zhang Xin Jiang Yue-Juan Zheng Pei-Yong Zheng Hui-Yong Zhang Zhen-Hui Lu 2020Infectious Diseases of Poverty2020,9,3:7
8Selective laser melting of near-α titanium alloy Ti-6Al-2Zr-1Mo-1V:Parameter optimization, heat treatment and mechanical performance显示文摘This paper presents a comprehensive study conducted to optimize the selective laser melting(SLM)parameters and subsequent heat-treatment temperatures for near-α high-temperature titanium alloy Ti-6 Al-2 Zr-1 Mo-1 V(TA15),which is widely used in the aerospace industry.Based on the surface morphology and relative density analysis,the optimized process parameters were:laser power from 230 W to 380 W,scan speed from 675 mm/s to 800 mm/s,scan spacing of 0.12 mm,and layer thickness of0.03 mm.The effects of the laser power and the layer thickness on the phase constitutions,microstructure features,as well as room-temperature and high-temperature(500℃) tensile properties,were then studied to obtain an in-depth understanding of SLM-built TA15.Six typical temperatures(650,750,850,950,1000 and 1100℃) covering three representative temperature ranges,i.e.,martensite partial decomposition temperature range,martensite complete decomposition temperature range and above βtransus temperature,were subsequently selected as heat-treatment temperatures.The heat treatmentmicrostructure-mechanical property relationships of SLM-built TA15 were elucidated in detail.These results provide valuable information on the development of SLM-built TA15 alloy for industrial applications,and these findings are also beneficial to additive manufacturing of other near-α Ti alloys with desirable high-temperature properties.Chao Cai Xu Wu Wan Liu Wei Zhu Hui Chen Jasper Chu Dong Qiu Chen-Nan Sun Jie Liu Qingsong Wei Yusheng Shi 2020Journal of Materials Science & Technology2020,54,22:7
9Resveratrol reduces brain injury after subarachnoid hemorrhage by inhibiting oxidative stress and endoplasmic reticulum stress显示文摘Previous studies have shown that resveratrol,a bioactive substance found in many plants,can reduce early brain injury after subarachnoid hemorrhage,but how it acts is still unclear.This study explored the mechanism using the experimental subarachnoid hemorrhage rat model established by injecting autologous blood into the cerebellomedullary cistern.Rat models were treated with an intraperitoneal injection of 60 mg/kg resveratrol 2,6,24 and 46 hours after injury.At 48 hours after injury,their neurological function was assessed using a modified Garcia score.Brain edema was measured by the wet-dry method.Neuronal apoptosis in the prefrontal cortex was detected by terminal deoxyribonucleotidyl transferase-mediated biotin-16-dUTP nick-end labeling assay.Levels of reactive oxygen species and malondialdehyde in the prefrontal cortex were determined by colorimetry.CHOP,glucose-regulated protein 78,nuclear factor-erythroid2-related factor 2 and heme oxygenase-1 mRNA expression levels in the prefrontal cortex were measured by reverse transcription polymerase chain reaction.Tumor necrosis factor-alpha content in the prefrontal cortex was detected by enzyme linked immunosorbent assay.Immunohistochemical staining was used to detect the number of positive cells of nuclear factor-erythroid 2-related factor 2,heme oxygenase 1,glucose-regulated protein 78,CHOP and glial fibrillary acidic protein.Western blot assay was utilized to analyze the expression levels of nuclear factor-erythroid 2-related factor 2,heme oxygenase 1,glucose-regulated protein 78 and CHOP protein expression levels in the prefrontal cortex.The results showed that resveratrol treatment markedly alleviated neurological deficits and brain edema in experimental subarachnoid hemorrhage rats,and reduced neuronal apoptosis in the prefrontal cortex.Resveratrol reduced the levels of reactive oxygen species and malondialdehyde,and increased the expression of nuclear factor-erythroid 2-related factor 2,heme oxygenase-1 mRNA and protein in the prefrontal cortex.Resveratrol decreased glucose-regulated protein 78,CHOP mRNA and protein expression and tumor necrosis factor-alpha level.It also activated astrocytes.The results suggest that resveratrol exerted neuroprotective effect on subarachnoid hemorrhage by reducing oxidative damage,endoplasmic reticulum stress and neuroinflammation.The study was approved by the Animals Ethics Committee of Shandong University,China on February 22,2016(approval No.LL-201602022).Yun-Kai Xie Xin Zhou Hong-Tao Yuan Jie Qiu Dan-Qing Xin Xi-Li Chu Da-Chuan Wang Zhen Wang 2019Neural Regeneration Research2019,14,10:6

11Diversity,bioactivities,and metabolic potentials of endophytic actinomycetes isolated from traditional medicinal plants in Sichuan,China显示文摘The present study was designed to determine the taxonomic diversity and metabolic activity of the actinomycetes community, including 13 traditional medicinal plants collected in Sichuan province, China, using multiple approaches such as morphological and molecular identification methods, bioactivity assays, and PCR screening for genes involved in antibiotics biosynthesis. 119 endophytic actinomycetes were recovered; 80 representative strains were chosen for 16 S r RNA gene partial sequence analyses, with 66 of them being affiliated to genus Streptomyces and the remaining 14 strains being rare actinomycetes. Antimicrobial tests showed that 12(15%) of the 80 endophytic actinomycetes displayed inhibitory effects against at least one indicator pathogens, which were all assigned to the genus Streptomyces. In addition, 87.5% and 58.8% of the isolates showed anticancer and anti-diabetic activities, respectively. Meanwhile, the anticancer activities of the isolates negatively correlated with their anti-diabetic activities. Based on the results of PCR screening, five genes, PKS-I, PKS-II, NRPS, ANSA, and oxy B, were detected in 55.0%, 58.8%, 90.0%, 18.8% and 8.8% of the 80 actinomycetes, respectively. 12Corrosion resistance and hydrophobicity of myristic acid modified Mg-AI LDH/Mg(OH)2 steam coating on magnesium alloy AZ31显示文摘A hydrophobic surface was successfully fabricated on the Mg-Al-layered double hydroxide(Mg-Al LDH)/Mg(OH)2-coated AZ31 magnesium alloy via an in-situ steam coating(SC)process and a subsequent surface modification with environment-friendly myristic acid(MA).The microstructure,composition and hydrophobicity of SC/MA composite coating were investigated by XRD,SEM,EDS,FTIR,and contact angle(CA)measurement.The corrosion behavior of the hybrid coating was evaluated by potentiodynamic polarization,EIS and hydrogen evolution test in 3.5 wt.%NaCl solution.The results showed that the LDH coating had nano-flake microstructure,which remained unchanged after modification with MA.The CA of the MA-modified coating surface reached up to 129°±3.5°,and the corrosion current density of SC/MA-2 coating decreased about three orders of the magnitude compared to that of the substrate.It is proven that the modified surface has an effective anti-corrosion effect on AZ31 alloy.The formation mechanism and the corrosion mechanism of the coating were also discussed.Zai-Meng Qiu Fen Zhang Jun-Tong Chu Yu-Chao Li Liang Song 2020Frontiers of Materials Science2020,14,1:5

16Effects of precipitation on soil organic carbon fractions in three subtropical forests in southern China显示文摘Aims The aim of this study was to investigate the effects of precipitation changes on soil organic carbon(SOC)fractions in subtropical forests where the precipitation pattern has been altered for decades.Methods We conducted field manipulations of precipitation,including ambient precipitation as a control(CK),double precipitation(DP)and no precipitation(NP),for 3 years in three forests with different stand ages(broadleaf forest[BF],mixed forest[MF]and pine forest[PF])in subtropical China.At the end of the experiment,soil samples were collected to assay SOC content,readily oxidizable organic carbon(ROC)and non-readily oxidizable organic carbon(NROC),as well as soil microbial biomass carbon(MBC),pH and total nitrogen content.Samples from the forest floors were also collected to analyze carbon(C)functional groups(i.e.alkyl C,aromatic C,O-alkyl C and carbonyl C).Furthermore,fine root biomass was measured periodically throughout the experiment.Important Findings Among the forests,ROC content did not exhibit any notable differences,while NROC content increased significantly with the stand age.This finding implied that the SOC accumulation observed in these forests resulted from the accumulation of NROC in the soil,a mechanism for SOC accumulation in the mature forests of southern China.Moreover,NP treatment led to significant reductions in both ROC and NROC content and therefore reduced the total SOC content in all of the studied forests.Such decreases may be due to the lower plant-derived C inputs(C quantity)and to the changes in SOC components(C quality)indicated by C functional groups analyses under NP treatment.DP treatment in all the forests also tended to decrease the SOC content,although the decreases were not statistically significant with the exception of SOC and ROC content in PF.This finding indicated that soils in MF and in BF may be more resistant to precipitation increases,possibly due to less water limitations under natural conditions in the two forests.Our results therefore highlight the different responses of SOC and its fractions to precipitation changes among the forests and suggest that further studies are needed to improve our understanding of SOC dynamics in such an important C sink region.Xiaomei Chen Deqiang Zhang Guohua Liang Qingyan Qiu Juxiu Liu Guoyi Zhou Shizhong Liu Guowei Chu Junhua Yan 2016Journal of Plant Ecology2016,9,1:4
17High‐sensitivity distributed dynamic strain sensing by combining Rayleigh and Brillouin scattering显示文摘The phase-sensitive optical time-domain reflectometry(φ-OTDR)is a good candidate for distributed dynamic strain sensing,due to its high sensitivity and fast measurement,which has already been widely used in intrusion monitoring,geophysical exploration,etc.For the frequency scanning basedφ-OTDR,the phase change manifests itself as a shift of the intensity distribution.The correlation between the reference and measured spectra is employed for relative strain demodulation,which has imposed the continuous measurement for the absolute strain demodulation.Fortunately,the Brillouin optical time domain analysis(BOTDA)allows for the absolute strain demodulation with only one measurement.In this work,the combination of theφ-OTDR and BOTDA has been proposed and demonstrated by using the same set of frequency-scanning optical pulses,and the frequency-agile technique is also introduced for fast measurements.A 9.9 Hz vibration with a strain range of 500 nεhas been measured under two different absolute strains(296.7μεand 554.8με)by integrating the Rayleigh and Brillouin information.The sub-micro strain vibration is demonstrated by theφ-OTDR signal with a high sensitivity of 6.8 nε,while the absolute strain is measured by the BOTDA signal with an accuracy of 5.4με.The proposed sensor allows for dynamic absolute strain measurements with a high sensitivity,thus opening a door for new possibilities which are yet to be explored.Benzhang Wang Dexin Ba Qi Chu Liqiang Qiu Dengwang Zhou Yongkang Dong 2020Opto-Electronic Advances2020,3,12:3
18Spacing graphene and Ni-Co layered double hydroxides with polypyrrole for high-performance supercapacitors显示文摘The urgent need of high-performance of energy storage devices triggers us to design newly class of materials.Generally,the materials feature with high conductivity,abundant pore s and excellent stability.Here,a sandwiched hybrid composite containing reduced graphene oxide,polypyrrole and Ni-Co layered double hydroxides(RGO/PPy/NiCo-LDH) was prepared in a facile way.The polypyrrole was incorporated in the two dimensional(2D) nanosheets,which not only serve as the spacer to increase the surface area,but also enhance the conductivity of the nanocomposite.The obtained architecture was employed as an advanced electrode in a supercapacitor.The electrode shows an ultrahigh specific capacitance(2534 F g^-1 at 1 A g^-1) and good cycling efficiency(78 % after 5000 cycles).Moreover,an asymmetric cell based RGO/PPy/NiCo-LDH composite demonstrates excellent electrochemical properties and good prospect of practical use.Jing Liang Cuili Xiang Yongjin Zou Xuebu Hu Hailiang Chu Shujun Qiu Fen Xu Lixian Sun 2020Journal of Materials Science & Technology2020,52,20:3
19The Colour Reaction of Mercury with 1-(6-Ethoxyl-2-benzothiazolyl)-3-(4-Nitrophenyl)-Triazene and its Application显示文摘A new triazene reagent 1-(6-ethoxyl-2-benzothiazolyl )-3-(4-nitrophenyl)-triazene(EtOBTNPT) has been synthesized. The colour reaction of EtOBTNPT with Hg(II) has beenstudied. At pH=10. 1-- 10. 7, in the presence of Triton X-100, the reagent with Hg(II) forms anorange-yellow complex with high sensitivity.
