1Reproductive management through integration of PGD and MPS-based noninvasive prenatal screening/diagnosis for a family with GJB2-associated hearing impairment显示文摘A couple with a proband child of GJB2(encoding the gap junction protein connexin 26)-associated hearing impairment and a previous pregnancy miscarriage sought for a reproductive solution to bear a healthy child. Our study aimed to develop a customized preconception-to-neonate care trajectory to fulfill this clinical demand by integrating preimplantation genetic diagnosis(PGD), noninvasive prenatal testing(NIPT), and noninvasive prenatal diagnosis(NIPD) into the strategy. Auditory and genetic diagnosis of the proband child was carried out to identify the disease causative mutations. The couple then received in-vitro-fertilization treatment, and eight embryos were obtained for day 5 biopsy. PGD was performed by short-tandem-repeat linkage analysis and Sanger sequencing of GJB2 gene. Transfer of a GJB2 c.235del C heterozygous embryo resulted in a singleton pregnancy. At the 13 th week of gestation, genomic DNA(g DNA) from the trio family and cell-free DNA(cf DNA) from maternal plasma were obtained for assessment of fetal chromosomal aneuploidy and GJB2 mutations. NIPT and NIPD showed the absence of chromosomal aneuploidy and GJB2-associated disease in the fetus, which was later confirmed by invasive procedures and postnatal genetic/auditory diagnosis. 5Genetic mapping of folate QTLs using a segregated population in maizeFA显示文摘As essential B vitamin for humans, folates accumulation in edible parts of crops, such as maize kernels, Is of great importance for human health. But its breeding is always limited by the prohibitive cost of folate profiling. The molecular breeding is a more executable and efficient way for folate fortification, but is limited by the molecular knowledge of folate regulation. Here we report the genetic mapping of folate quantitative trait loci (QTLs) using a segregated population crossed by two maize lines, one high in folate (GEMS31) and the other low in folate (DAN3130). Two folate QTLs on chromosome 5 were obtained by the combination of F2 whole-exome sequencing and F3 kernel-folate profiling. These two QTLs had been confirmed by bulk segregant analysis using F6 pooled DNA and F7 kernel-folate profiling, and were overlapped with QTLs identified by another segregated population. These two QTLs contributed 41.6% of phenotypic variation of 5-formyltetrahydrofolate, the most abundant storage form among folate derivatives in dry maize grains, in the GEMS31×DAN3130 population. 6Understanding auditory neuropathy spectrum disorder: a system- atic review in transgenic mouse models显示文摘Auditory neuropathy spectrum disorder is a unique group of hearing dysfunctions characterized by preserved outer hair cell function and abnormal neural conduction of the auditory pathway. However, the pathogenic mechanism underlying this disorder is not clear. We therefore performed a systematic review of genetic mouse models with different gene mutations to provide a valuable tool for better understanding of the process and the possible molecular mechanisms. Of the 18 articles retrieved, nine met the required criteria. All biochemical, histological, and electrophysiological results were recorded for each of the mouse models, as was the transgenic technology. 10Investigating the effect of cosolvents on P3HT/O-IDTBR film-forming kinetics and film morphology显示文摘The morphology of active layer in bulk heterojunction(BHJ) organic solar cells is decisive to the device performance. Previous works have shown that the solvent engineering is an effective method to optimize the morphology of active layer. However, screening the proper solvent is a tedious task, and we know very little about how to select a proper solvent for a particular system, especially for polymer/nonfullerene blend systems. Here, we combined the spectroscopic analysis in various solvent mixtures during film-forming process to reveal the relationship among the cosolvent characteristics, film-forming kinetics and film morphology. In this article, P3 HT/O-IDTBR blend was selected as model system due to being facile synthesized under a large-scale. Chlorobenzene(CB) was selected as main solvent, and the cosolvents were grouped into three categories according to its boiling point(bp) compared to CB.The cosolvents with lower bp, like chloroform(CF), can facilitate a faster film-forming process, reducing the domain size but sacrificing the crystallinity of both components. For the cosolvents with higher bp,like o-dichlorobenzene(DCB) and 1,2,4-trichlorobenzene(TCB), the self-organization process of P3 HT and O-IDTBR is separated and its duration was extended, constructing highly crystalline nanointerpenetrating network. However, the cosolvents with very high bp, such as chlornaphthalene(CN),would residue in film and keep P3 HT and O-IDTBR self-organizing for longer time, leading to larger phase separation. 11Immunogenicity and safety of a booster COVID-19 vaccination in patients with chronic liver disease:A multicenter study显示文摘Aim:Patients with chronic liver disease(CLD),especially cirrhosis,are at a high risk of severe illness or death from coronavirus disease-2019(COVID-19)and may have a suboptimal immune response to the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)vaccine.This study aimed to evaluate the safety and immunogenicity of the COVID-19 booster vaccination in patients with CLD.Methods:The study protocol was prospectively registered at ClinicalTrials.gov(No.NCT05204602)after approval by the Ethics Committee.Adult participants with CLD were enrolled in this multicenter prospective study.They completed two doses of the inactivated COVID-19 vaccine and received booster doses at least 6 months later.Adverse reactions were recorded within 14 days after the booster dose.Serum samples of the enrolled patients were collected before and after booster vaccination and tested for SARS-CoV-2 receptor-binding domain(RBD)immunoglobulin G and neutralizing anti-bodies.The chi-squared or Fisher's exact test was used to compare categorical data,and the Mann-Whitney U test was used to compare continuous variables.Two-sided p<0.05 were considered statistically significant.Results:In total,63 patients were enrolled from four hospitals in China,including 29 patients with cirrhosis.The median age of all patients was 55 years,and 61.9%(39/63)were male.The vaccines were well tolerated;most adverse reactions were mild and transient,and injection site pain(6.4%;4/63)and fatigue(3.2%,2/63)were the most frequent local and systemic adverse events.Following the booster vaccination,our results showed that in the whole cohort,the levels and positive rates of anti-RBD IgG and neutralizing antibodies were significantly higher than baseline levels(all p<0.05).Conclusions:The inactivated COVID-19 booster vaccine was safe and significantly increased antibody levels and positivity rates following standard vaccination regimens in patients with CLD,especially those with cirrhosis.Jitao Wang Jingwen Ai Huiling Xiang Yanliang Zhang Zhiyun Hou Qiran Zhang Jiaojian Lv Shubo Chen Chuan Liu Qianqian Li Jing Liang Faren Xie Shujun Jiang Nina Zhang Aiguo Zhang Xiaolin Lan Xuying Zhang Jinlong Li Dengxiang Liu Wenchuan Wang Wei Rao Zhang Qun Qiuju Tian Xiaolong Qi Wenhong Zhang 2023Portal Hypertension & Cirrhosis2023,2,3:0
12Boron:A key functional component for designing high-performance heterogeneous catalysts显示文摘Heterogeneous catalysis is a vivid branch of traditional catalysis field,with the advantage of high efficiency and being easily separated from reactants and products after reaction,and have received widespread attentions in large-scale industrial production,especially in the field of energy utilization.Boron has been found to be a key functional component for designing high-performance heterogeneous catalysts.In this review,we cover and categorize the past and recent progress in boron-containing materials and their applications in heterogeneous catalysis particularly in energy-related fields.The fundamental roles of boron components in the emerging heterogeneous catalysis of construction,regulation and stabilization of active phases/sites are highlighted,with the emphasis on how they regulating structural and electronic properties of host materials.We then categorize boron-containing catalysts into six kinds mainly including intermetallic boride catalysts,metal boride-derived catalysts,boron-doped catalysts,metal boride-decorated catalysts,boron-containing compounds as catalyst supports,and singleboron-site catalysts,as well as try to establish structure-catalytic activity relationship.The catalytic applications of these six boron-containing catalysts are discussed separately,focusing on the energy-related reactions such as hydrogen evolution reaction(HER),oxygen evolution reaction(OER),oxygen reduction reaction(ORR),carbon dioxide reduction reaction(CO_(2)RR)and nitrogen reduction reaction(NRR).Finally,the opportunities and challenges related to boron-containing compounds in the field of catalysis are prospected.Meihong Fan Xiao Liang Qiuju Li Lili Cui Xingquan He Xiaoxin Zou 2023Chinese Chemical Letters2023,34,1:0
13Dynamic m6A-ncRNAs association and their impact on cancer pathogenesis, immune regulation and therapeutic response显示文摘Several types of modifications have been proven to participate in the metabolism and processing of different RNA types,including non-coding RNAs(ncRNAs).N-6-methyladenosine(m6A)is a dynamic and reversible RNA modification that is closely involved in the ncRNA homeostasis,and serves as a crucial regulator for multiple cancer-associated signaling pathways.The ncRNAs usually regulate the epigenetic modification,mRNA transcription and other biological processes,displaying enormous roles in human cancers.In this review,we summarized the significant implications of m6A-ncRNA interaction in various types of cancers.In particular,the interplay between m6A and ncRNAs in cancer pathogenesis and therapeutic resistance are being widely recognized.We also discussed the relevance of m6A-ncRNA interaction in immune regulation,followed by the interference on cancer immunotherapeutic procedures.In addition,we briefly highlighted the computation tools that could identify the accurate features of m6A methylome among ncRNAs.In summary,this review would pave the way for a better understanding of the biological functions of m6A-ncRNA crosstalk in cancer research and treatment.Yuanliang Yan Jinwu Peng Qiuju Liang Xinxin Ren Yuan Cai Bi Peng Xi Chen Xiang Wang Qiaoli Yi Zhijie Xu 2023Genes & Diseases2023,10,1:0
