1Polymorphisms of three gene-derived STS on pig chromosome 13q41 are associated with susceptibility to enterotoxigenic Escherichia coli F4ab/ac in pigs显示文摘Neonatal diarrhea caused by enterotoxigenic Escherichia coli(ETEC)F4 is a common and serious disease,resulting in significant economical loss in the pig industry.The locus encoding ETEC F4 receptor has been mapped to pig chromosome(SSC)13q41,and one of the most significantly linked markers is S0075.In this study,we selected three genes including SLC12A8,MYLK and KPNA1 from a chromosomal region flanking S0075 on SSC13 to develop pig specific sequence tagged sites(STS). Seven single nucleotide polymorphisms were identified in the three pig STS using DNA of four full-sib susceptible and resistant animals in a White Duroc×Erhualian intercross.All grandparents,parents and 755 offspring in the intercross were genotyped for three polymorphisms,including SLC12A8 g.159A>G,MYLK g.1673A>G and KPNA1 g.306A>G.Family-based transmission disequilibrium test(TDT) revealed that all polymorphisms and the corresponding haplotypes are significantly associated with ETEC F4ab/ac(especially F4ac)brush border adhesion phenotypes,indicating that these polymor- phism are in linkage disequlibrium with causal mutation(s)of the gene encoding ETEC F4ab/ac receptor. 2Energy-saving PPM schemes for WSNs显示文摘Energy conservation is a critical problem in recently-emerging wireless sensor networks (WSNs). Pulse position modulation (PPM), as an exploring-worthy modulation format for energy efficiency, is tailored for WSNs into two schemes, mono-mode PPM and multi-mode PPM, in this paper. Resorting to an idealized system model and a practical system model, which combine the power consumptions in transmission and reception modules of nodes with the idealized and realistic battery characteristics, the battery energy efficiencies of mono-mode PPM and multi-mode PPM are evaluated and compared. To minimize the battery energy consumption (BEC), these schemes are further optimized in terms of constellation size M for a link in path-loss channels. 3Protocatechuic aldehyde protects cardiomycoytes against ischemic injury via regulation of nuclear pyruvate kinase M2显示文摘Rescuing cells from stress damage emerges a potential therapeutic strategy to combat myocardial infarction.Protocatechuic aldehyde(PCA)is a major phenolic acid in Chinese herb Danshen(Salvia miltiorrhiza root).This study investigated whether PCA regulated nuclear pyruvate kinase isoform M2(PKM2)function to protect cardiomyocytes.In rats subjected to isoprenaline,PCA attenuated heart injury and protected cardiomyocytes from apoptosis.Through DARTS and CETSA assays,we identified that PCA bound and promoted PKM2 nuclear translocation in cardiomyocytes exposed to oxygen/glucose deprivation(OGD).In the nucleus,PCA increased the binding of PKM2 to β-Catenin via preserving PKM2 acetylation,and the complex,in cooperation with T-cell factor 4(TCF4),was required for transcriptional induction of genes encoding anti-apoptotic proteins,contributing to rescuing cardiomyocyte survival.In addition,PCA ameliorated mitochondrial dysfunction and prevented mitochondrial apoptosis dependent on PKM2.Consistently,PCA increased the binding of PKM2 to β-Catenin,improved heart contractive function,normalized heart structure and attenuated oxidative damage in mice subjected to artery ligation,but the protective effects were lost in Pkm2-deficient heart.Together,we showed that PCA regulated nuclear PKM2 function to rescue cardiomyocyte survival via β-Catenin/TCF4 signaling cascade,suggesting the potential of pharmacological intervention of PKM2 shuttle to protect the heart.Xunxun Wu Lian Liu Qiuling Zheng Haiping Hao Hui Ye Ping Li Hua Yang 2021Acta Pharmaceutica Sinica B2021,11,11:4

8Dihydrotanshinone I preconditions myocardium against ischemic injury via PKM2 glutathionylation sensitive to ROS显示文摘Ischemic preconditioning(IPC)is a potential intervention known to protect the heart against ischemia/reperfusion injury,but its role in the no-reflow phenomenon that follows reperfusion is unclear.Dihydrotanshinone I(DT)is a natural compound and this study illustrates its role in cardiac ischemic injury from the aspect of IPC.Pretreatment with DT induced modest ROS production and protected cardiomyocytes against oxygen and glucose deprivation(OGD),but the protection was prevented by a ROS scavenger.In addition,DT administration protected the heart against isoprenaline challenge.Mechanistically,PKM2 reacted to transient ROS via oxidization at Cys423/Cys424,leading to glutathionylation and nuclear translocation in dimer form.In the nucleus,PKM2 served as a co-factor to promote HIF-1a-dependent gene induction,contributing to adaptive responses.In mice subjected to permanent coronary ligation,cardiac-specific knockdown of Pkm2 blocked DT-mediated preconditioning protection,which was rescued by overexpression of wild-type Pkm2,rather than Cys423/424-mutated Pkm2.In conclusion,PKM2 is sensitive to oxidation,and subsequent glutathionylation promotes its nuclear translocation.Although IPC has been viewed as a protective means against reperfusion injury,our study reveals its potential role in protection of the heart from no-reflow ischemia.Xunxun Wu Lian Liu Qiuling Zheng Hui Ye Hua Yang Haiping Hao Ping Li 2023Acta Pharmaceutica Sinica B2023,13,1:1
9Deep reinforcement learning for optimal denial-of-service attacks scheduling显示文摘We consider an optimal denial-of-service(DoS)attack scheduling problem of N independent linear time-invariant processes,where sensors have limited computational capability.Sensors transmit measurements to the remote estimator via a communication channel that is exposed to DoS attackers.However,due to limited energy,an attacker can only attack a subset of sensors at each time step.To maximally degrade the estimation performance,a DoS attacker needs to determine which sensors to attack at each time step.In this context,a deep reinforcement learning(DRL)algorithm,which combines Q-learning with a deep neural network,is introduced to solve the Markov decision process(MDP).The DoS attack scheduling optimization problem is formulated as an MDP that is solved by the DRL algorithm.A numerical example is provided to illustrate the efficiency of the optimal DoS attack scheduling scheme using the DRL algorithm.Fangyuan HOU Jian SUN Qiuling YANG Zhonghua PANG 2022Science China(Information Sciences)2022,65,6:1

13Determining the in vitro Anti-Aging Effect of the Characteristic Components from Eucommia ulmoides显示文摘To evaluate the potential anti-aging ability of Eucommia ulmoides,four characteristic components(chlorogenic acid,geniposidic acid,aucubin,quercetin)were selected to assess their effects on H_(2)O_(2)-induced oxidative damage model of human umbilical vein endothelial cell(HUVEC).Oxidative damage indexes,inflammatory factors,cell cycle,cell apoptosis,cell senescence,and their related proteins were analyzed by methyl thiazolyl tetrazolium(MTT)assay,enzyme-linked immunosorbent assay(ELISA),propidium iodide(PI)staining,annexin V-FITC/PI double staining,SAβ-galactosidase staining,and western blotting(WB).The results showed that H_(2)O_(2)-induced cell growth inhibition rate decreased as supplementation with characteristic components when compared to H_(2)O_(2) group.Meanwhile,the contents of antioxidant indexes(reactive oxygen species,lactate dehydrogenase,molondialdehyde,superoxide dismutase,glutathione),inflammatory factors(nuclear factor kappa-B,intercellular cell adhesion molecule-1,vascular cell adhesion protein 1),and functional factors(NO,Endothelin-1)in characteristic components treated groups improved if comparison with H_(2)O_(2) group,suggesting the characteristic components of E.ulmoides could alleviate H_(2)O_(2)-induced oxidative damage.Moreover,cell cycle,cell apoptosis,cell senescence,and their related proteins under characteristic components treatment exhibited a better effect than under H_(2)O_(2) treatment,implying the characteristic components could participate in anti-aging via multiple pathways.These results manifested that the characteristic components of E.ulmoides posses the capacity of anti-aging,which provided a basis for investigating the anti-aging ability of E.ulmoides itself.Xuesong Wang Zhihong Wang Qiuling Yang Sheng Peng Mijun Peng 2022Journal of Renewable Materials2022,10,12:1

18A chromosome-level genome assembly for Dracaena cochinchinensis reveals the molecular basis of its longevity and formation of dragon’s blood显示文摘Dracaena,a remarkably long-lived and slowly maturing species of plant,is world famous for its ability to produce dragon’s blood,a precious traditional medicine used by different cultures since ancient times.However,there is no detailed and high-quality genome available for this species at present;thus,the molecular mechanisms that underlie its important traits are largely unknown.These factors seriously limit the protection and regeneration of this rare and endangered plant resource.Here,we sequenced and assembled the genome of Dracaena cochinchinensis at the chromosome level.The D.cochinchinensis genome covers 1.21 Gb with a scaffold N50 of 50.06 Mb and encodes 31619 predicted protein-coding genes.Analysis showed that D.cochinchinensis has undergone two whole-genome duplications and two bursts of long terminal repeat insertions.The expansion of two gene classes,cis-zeatin O-glucosyltransferase and small auxin upregulated RNA,were found to account for its longevity and slow growth.Two transcription factors(bHLH and MYB)were found to be core regulators of the flavonoid biosynthesis pathway,and reactive oxygen species were identified as the specific signaling molecules responsible for the injuryinduced formation of dragon’s blood.Our study provides high-quality genomic information relating to D.cochinchinensis and significant insight into the molecular mechanisms responsible for its longevity and formation of dragon’s blood.These findings will facilitate resource protection and sustainable utilization of Dracaena.Yanhong Xu Kaijian Zhang Zhonglian Zhang Yang Liu Feifei Lv Peiwen Sun Shixi Gao Qiuling Wang Cuicui Yu Jiemei Jiang Chuangjun Li Meifang Song Zhihui Gao Chun Sui Haitao Li Yue Jin Xinwei Guo Jianhe Wei 2022Plant Communications2022,3,6:0

20IGF2-NR4A2 Signaling Regulates Macrophage Subtypes to Attenuate Liver Cirrhosis显示文摘Background and Aims:Liver cirrhosis can lead to liver failure and eventually death.Macrophages are the main contributors to cirrhosis and have a bidirectional role in regulating matrix deposition and degradation.Macrophage-based cell therapy has been developed as an alternative to liver transplantation.However,there is insufficient evidence regarding its safety and efficacy.In this study,we aimed to explore the effect of combining insulin-like growth factor 2(IGF2)with bone marrow-derived macrophages(BMDMs)to treat mice with liver cirrhosis.Methods:We assessed liver inflammation,fibrosis regression,liver function,and liver regeneration in mice with CCl4-induced cirrhosis and treated with BMDM only or IGF2+BMDM.We performed in vitro experiments in which activated hepatic stellate cells(HSCs)were co-cultured with macrophages in the presence or absence of IGF2.The polarity of macrophages and the degree of inhibition of HSCs were examined.The effect of IGF2 on macrophages was also verified by the overexpression of IGF2.Results:Combining IGF2 with BMDM reduced liver inflammation and fibrosis and increased hepatocyte proliferation.Combining IGF2 with BMDM was more effective than using BMDM alone.In vitro experiments demonstrated that IGF2 inhibited HSCs activation by upregulating NR4A2 to promote the anti-inflammatory macrophages phenotype.IGF2 also increased the synthesis of matrix metalloproteinases(MMPs)by macrophages,which may explain why administering IGF2 combined with BMDM was more effective than administering BMDM only.Conclusions:Our study provides a theoretical basis for the future use of BMDM-based cell therapy to treat liver cirrhosis.Lichao Yao Xue Hu Mengqin Yuan Qiuling Zhang Pingji Liu Lian Yang Kai Dai Yingan Jiang 2023Journal of Clinical and Translational Hepatology2023,11,4:0
