| 1 | The preparation and characterization of folate-conjugated human serum albumin magnetic cisplatin nanoparticles显示文摘Objective:Nanoparticles are becoming an important method of targeted drug delivery.To evaluate the importance of folate-conjugated human serum albumin(HSA)magnetic nanoparticles(Folate-CDDP/HSA MNP),we prepared drug-loaded Folate-CDDP/HSA MNPs and characterized their features.Methods:First, folate was conjugated with HSA under the effect of a condensing agent,and the conjugating rate was evaluated by a colorimetric method using 2,4,6-trinitrobenzene sulfonic acid.Second,under N 2 gas,Fe 3 O 4 magnetic nanomaterials were prepared and characterized by using transmission electron microscopy(TEM),SEM-EDS and X-ray diffraction(XRD).Finally,Folate-CDDP/HSA MNP was prepared by using a solvent evaporation technique. TEM was used to observe particle morphology.The particle size and distribution of the prepared complexes were determined by a Laser particle size analyzer.Drug loading volume and drug release were investigated by a high performance liquid chromatography method(HPLC)in vitro.Results:We successfully prepared folate-conjugated HSA and its conjugating rate was 27.26μg/mg.Under TEM,Fe 3 O 4 magnetic nanoparticles were highly electron density and had an even size distribution in the range of 10-20 nm.It was confirmed by SEM-EDS and XRD that Fe 3 O 4 magnetic nanoparticles had been successfully prepared.Under TEM,drug-loaded magnetic nanoparticles were observed,which had a round shape,similar uniform size and smooth surface.Their average size was 79 nm which was determined by laser scattering,and they exhibited magnetic responsiveness.Encapsulation efficiency was 89.75%and effective drug loading was calculated to be 15.25%.The release results in vitro showed that the half release time(t1/ 2 )of cisplatin in cisplatin Solution and Folate-CDDP/HSA MNP was 65 min and 24 h respectively, which indicated that microspheres had an obvious effect of sustained-release.Conclusion:Folate-CDDP/HSA MNPs were prepared successfully.The preparation process and related characteristics data provided a foundation for further study,including the mechanism of the nanoparticles distribution in vivo and their intake by tumor cells. | Daozhen Chen Qiusha Tang Wenqun Xue Jingying Xiang Li Zhang Xinru Wang | 2010 | The Journal of Biomedical Research2010,24,1: | 13 |
| 2 | Arsenic trioxide alleviates acute graft-versus-host disease by modulating macrophage polarization显示文摘This study aimed to explore macrophage polarization in acute graft-versus-host disease after hematopoietic stem cell transplantation, and investigated if arsenic trioxide(ATO) could correct this imbalance. In the colon of GVHD mice, we found that the number of F4/80+iNOS+ cells as well as the expression intensity of TNF-α and IL-1β was greater in the GVHD group than in the BM group, whereas the number of F4/80+CD206+ cells and the expression intensity of IL-10 and TGF-β was greater in the BM group than in the GVHD group. We investigated the effect of ATO on GVHD mice, and found that ATO treatment clearly improved the survival of the mice and reduced the severity of GVHD. In addition, ATO reduced the number of F4/80+iNOS+ cells, and increased the number of F4/80+CD206+ cells in the colon of GVHD mice. Furthermore, ATO sharply decreased CD86 and CD80 expression, and increased CD163 and CD206 expression in macrophages induced from aGVHD patients. Therefore,ATO can modulate the M1 and M2 phenotype in GVHD mice or in macrophages from aGVHD patients. | 3 | Ultrafast dynamics of o-fluorophenol studied with femtosecond time-resolved photoelectron and photoion spectroscopy显示文摘The ultrafast dynamics of o-fluorophenol via the excited states has been studied by femtosecond time-resolved photoelectron imaging. The photoion and photoelectron spectra taken with a time delay between 267 nm pump laser and 800 nm probe laser provide a longer-lived S1 electronic state of about ns timescale. | 13 | Accurate Assessment and Tracking the Process of Liver-Specific Injury by the Residual Tissue Activity of Carboxylesterase 1 and Dipeptidyl Peptidase 4显示文摘Accurately assessing and tracking the progression of liver-specific injury remains a major challenge in the field of biomarker research.Here,we took a retrospective validation approach built on the mutuality between serum and tissue biomarkers to characterize the liver-specific damage of bile duct cells caused by a-naphthyl isothiocyanate(ANIT).We found that carboxylesterase 1(CES1),as an intrahepatic marker,and dipeptidyl peptidase 4(DPP-IV),as an extrahepatic marker,can reflect the different pathophysiologies of liver injury.Levels of CES1 and DPP-IV can be used to identify liver damage itself and the inflammatory state,respectively.While the levels of the conventional serological biomarkers alkaline phosphatase(ALP),alanine aminotransferase(ALT),and aspartate aminotransferase(AST)were all concomitantly elevated in serum and tissues after ANIT-induced injury,the levels of bile acids decreased in bile,increased in serum,and ascended in intrahepatic tissue.Although the level of γ-glutamyl transpeptidase(γ-GT)changed in an opposite direction,the duration was much shorter than that of CES1 and was quickly restored to normal levels.Therefore,among the abovementioned biomarkers,only CES1 made it possible to specifically determine whether the liver cells were destroyed or damaged without interference from inflammation.CES1 also enabled accurate assessment of the anti-cholestasis effects of ursodeoxycholic acid(UDCA;single component)and Qing Fei Pai Du Decoction(QFPDD;multicomponent).We found that both QFPDD and UDCA attenuated ANIT-induced liver damage.UDCA was more potent in promoting bile excretion but showed relatively weaker anti-injury and antiinflammatory effects than QFPDD,whereas QFPDD was more effective in blocking liver inflammation and repairing liver damage.Our data highlights the potential of the combined use of CES1(as an intrahepatic marker of liver damage)and DPP-IV(as an extrahepatic marker of inflammation)for the accurate evaluation and tracking of liver-specific injury—an application that allows for the differentiation of liver damage and inflammatory liver injury. | Qiusha Pan Peifang Song Zhenhua Ni Xingkai Qian Anqi Wang Liwei Zou Yong Liu Ping Wang Weidong Zhang Hong Ma Ling Yang | 2022 | Engineering2022,,12: | 0 |
