3Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. 48-bromo-7-methoxychrysin inhibits properties of liver cancer stem cells via downregulation of β-catenin显示文摘AIM:To evaluate whether 8-bromo-7-methoxychrysin(BrMC),a synthetic analogue of chrysin,inhibits the properties of cancer stem cells derived from the human liver cancer MHCC97 cell line and to determine the potential mechanisms.METHODS:CD133+cells were sorted from the MHCC97 cell line by magnetic activated cell sorting,and amplified in stem cell-conditioned medium to obtain the enriched CD133+sphere forming cells(SFCs).The stem cell properties of CD133+SFCs were validated by the tumorsphere formation assay in vitro and the xenograft nude mouse model in vivo,and termed liver cancer stem cells(LCSCs).The effects of BrMC on LCSCs in vitro were evaluated by MTT assay,tumorsphere formation assay and transwell chamber assay.The effects of BrMC on LCSCs in vivo were determined using a primary and secondary xenograft model in Balb/c-nu mice.Expressions of the stem cell markers,epithelialmesenchymal transition(EMT)markers andβ-catenin protein were analyzed by western blotting or immunohistochemical analysis.RESULTS:CD133+SFCs exhibited stem-like cell properties of tumorsphere formation and tumorigenesis capacity in contrast to the parental MHCC97 cells.We found that BrMC preferentially inhibited proliferation and self-renewal of LCSCs(P<0.05).Furthermore,BrMC significantly suppressed EMT and invasion of LCSCs.Moreover,BrMC could efficaciously eliminate LCSCs in vivo.Interestingly,we showed that BrMC decreased the expression ofβ-catenin in LCSCs.Silencing ofβ-catenin by small interfering RNA could synergize the inhibition of self-renewal of LCSCs induced by BrMC,while Wnt3a treatment antagonized the inhibitory effects of BrMC.CONCLUSION:BrMC can inhibit the functions and characteristics of LCSCs derived from the liver cancer MHCC97 cell line through downregulation ofβ-catenin expression.Mei-Fang Quan Li-Hong Xiao Zhi-Hong Liu Hui Guo Kai-Qun Ren Fei Liu Jian-Guo Cao Xi-Yun Deng 2013World Journal of Gastroenterology2013,19,43:22

6Antioxidant properties of magnesium lithospermate B contribute to the cardioprotection against myocardial ischemia/reperfusion injury in vivo and in vitro显示文摘OBJECTIVE: To determine the cardioprotective effect of magnesium lithospermate B (MLB) on myocardial ischemia/reperfusion (MI/R) injury and to investigate the antioxidant potential in vivo and in vitro. METHODS: MI/R injury was induced by the occlusion of left anterior descending coronary artery for 30 min followed by reperfusion for 3 h in rats. After reperfusion, hearts were harvested to assess infarct size, histopathological damages, the levels of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), reduced glutathione (GSH) and malondialdehyde (MDA). Blood samples were col- lected to determine serum levels of creatine kinase-MB (CK-MB), cardiac troponin (cTnI) and lactate dehydrogenase (LDH). Furthermore, simulatedischemia/reperfusion (SI/R) injury in vitro was established by oxygen and glucose deprivation (OGD) for 2 h followed by 24-hour recovery period in cardiomyocytes. The activity of LDH in the cultured supernatant and the levels of intracellular reactive oxygen species (ROS), SOD and MDA in cardiomyocytes were also measured. Finally, cardiomyocytes apoptosis was determined with flow cytometry. RESULTS: MLB significantly limited infarct size, ameliorated histopathological damages and prevented leakage of CK-MB, cTnI and LDH. Additionally, SOD, CAT, GPx and GSH activities were notably increased by MLB, along with the MDA content decreased as compared with the model group in rats. In vitro study, MLB also decreased LDH activity in the cultured supernatant, increased SOD activity in cardiomyocytes, reduced intracellular ROS and MDA levels, and significantly suppressed cardiomyocytes apoptosis. CONCLUSION: MLB possessed remarkably cardioprotective effects on MI/R injury in vivo and in vitro. 7Impact of Heat Wave in 2005 on Mortality in Guangzhou, China显示文摘Objective To assess the impact of the heat wave in 2005 on mortality among the residents in Guangzhou and to identify susceptible subpopulations in Guangzhou, China. Methods The data of daily number of deaths and meteorological measures from 2003 to 2006 in Guangzhou were used in this study. Heat wave was defined as ≥7 consecutive days with daily maximum temperature above 35.0 ℃ and daily mean temperature above the 97th percentile during the study period. The excess deaths and rate ratio (RR) of mortality in the case period compared with the reference period in the same summer were calculated. Results During the study period, only one heat wave in 2005 was identified and the total number of excess deaths was 145 with an average of 12 deaths per day. The effect of the heat wave on non-accidental mortality (RR=1.23, 95% CI: 1.11-1.37) was found with statistically significant difference. Also, greater effects were observed for cardiovascular mortality (RR=1.34, 95% CI: 1.13-1.59) and respiratory mortality (RR=1.31, 95% CI: 1.02-1.69). Females, the elderly and people with lower socioeconomic status were at significantly higher risk of heat wave-associatedmortality. Conclusion The 2005 heat wave had a substantial impact on mortality among the residents in Guangzhou, particularly among some susceptible subpopulations. 8Generating dense and super-resolution ISAR image by combining bandwidth extrapolation and compressive sensing显示文摘Recent developing compressive sensing (CS) theory indicates that it is possible to obtain precise recovery of a sparse signal from very limited measurements,which provides a new way for data acquisition and signal processing as nature signals usually involve some degree of sparsity.In this paper,we present an algorithm for inversed synthetic aperture radar (ISAR) imaging with super resolution by combining CS and bandwidth extrapolation (BWE) technique.For ISAR imaging,the backscattering field of target is usually contributed by a few strong scattering centers,whose number is much less than that of image pixels.Thus,CS is intuitively suitable for constructing super resolution ISAR image.According to CS theory,the number of extracted dominating scatterers relies on the signal length,which indicates that if only limited data is available,it is difficult to generate dense ISAR image robustly by CS,and some signal components tend to lose.To soften this constraint,BWE is combined with CS imaging to increase the degree of freedom of signal while preserving its coherence.A refined CS-based formation for ISAR image-resolution enhancement is then developed.Both real and simulated data experiments are performed to evaluate the proposed approach,and an example of using this technique demonstrates the enhanced image resolution in application of maneuvering target imaging.QUAN YingHui ZHANG Lei GUO Rui XING MengDao BAO Zheng 2011Science China(Information Sciences)2011,54,10:15
9Effects of warming and clipping on plant and soil properties of an alpine meadow in the Qinghai-Tibetan Plateau, China显示文摘Climate warming and livestock grazing are known to have great influences on alpine ecosystems like those of the Qinghai-Tibetan Plateau(QTP) in China. However, it is lacking of studies on the effects of warming and grazing on plant and soil properties in these alpine ecosystems. In this study, we reported the related research from manipulative experiment in 2010–2012 in the QTP. The aim of this study was to investigate the individual and combined effects of warming and clipping on plant and soil properties in the alpine meadow ecosystem. Infrared radiators were used to simulate climate warming starting in July 2010, while clipping was performed once in October 2011 to simulate the local livestock grazing. The experiment was designed as a randomized block consisting of five replications and four treatments: control(CK), warming(W), clipping(C) and warming+clipping combination(WC). The plant and soil properties were investigated in the growing season of the alpine meadow in 2012. The results showed that W and WC treatments significantly decreased relative humidity at 20-cm height above ground as well as significantly increases air temperature at the same height, surface temperature, and soil temperature at the depth of 0–30 cm. However, the C treatment did not significantly decrease soil moisture and soil temperature at the depth of 0–60 cm. Relative to CK, vegetation height and species number increased significantly in W and WC treatment, respectively, while vegetation aboveground biomass decreased significantly in C treatment in the early growing season. However, vegetation cover, species diversity, belowground biomass and soil properties at the depth of 0–30 cm did not differ significantly in W, C and WC treatments. Soil moisture increased at the depth of 40–100 cm in W and WC treatments, while belowground biomass, soil activated carbon, organic carbon and total nitrogen increased in the 30–50 cm soil layer in W, C and WC treatments. Although the initial responses of plant and soil properties to experimental warming and clipping were slow and weak, the drought induced by the downward shift of soil moisture in the upper soil layers may induce plant belowground biomass to transfer to the deeper soil layers. This movement would modify the distributions of soil activated carbon, organic carbon and total nitrogen. 11Quality assessment of onboard GPS receiver and its combination with DORIS and SLR for Haiyang 2A precise orbit determination显示文摘The GPS,DORIS,and SLR instruments are installed on Haiyang 2A(HY2A)altimetry satellite for Precise Orbit Determination(POD).Among these instruments,the codeless GPS receiver is the state-of-art Chinese indigenous onboard receiver,and it is the first one successfully used for Low Earth Orbit(LEO)satellite.Firstly,the contribution assesses the performance of the receiver through an analysis of data integrity,numbers of all tracked and valid measurements as well as multipath errors.The receiver generally shows good performance and quality despite a few flaws.For example,L2 observations are often missing in low elevations,particularly during the ascent of GPS satellites,and the multipath errors of P1 show a slightly abnormal pattern.Secondly,the PCO(Phase Center Offset)and PCV(Phase Center Variation)of the antenna of the GPS receiver are determined in this contribution.A significant leap for Z-component of PCO up to-1.2 cm has been found on 10 October 2011.Thirdly,the obtained PCO and PCV maps are used for GPS only POD solutions.The post-fit residuals of ionosphere-free phase combinations reduce almost 50%,and the radial orbit differences with respect to CNES(Centre National d’Etudes Spatiales)Precise Orbit Ephemeris(POEs)improve about 13.9%.The orbits are validated using the SLR data,and the RMS of SLR Observed minus Computed(O-C)residuals reduces from 17.5 to 15.9 mm.These improvements are with respect to the orbits determined without PCO and PCV.Fourthly,six types of solutions are determined for HY2A satellite using different combinations of GPS,DORIS,and SLR data.Statistics of SLR O-C residuals and cross-comparison of orbits obtained in the contribution and the CNES POEs indicate that the radial accuracy of these orbits is at the 1.0 cm level for HY2A orbit solutions,which is much better than the scientific requirements of this mission.It is noticed that the GPS observations dominate the achievable accuracy of POD,and the combination of multiple types of observations can reduce orbit errors caused by data gaps and maintain more stable and continuous orbits.GUO Jing ZHAO QiLe GUO Xiang LIU XiangLin LIU JingNan ZHOU Quan 2015Science China Earth Sciences2015,58,1:13
12Efficient InGaN-based yellow-light-emitting diodes显示文摘Realization of efficient yellow-light-emitting diodes(LEDs) has always been a challenge in solid-state lighting.Great effort has been made, but only slight advancements have occurred in the past few decades. After comprehensive work on InGaN-based yellow LEDs on Si substrate, we successfully made a breakthrough and pushed the wall-plug efficiency of 565-nm-yellow LEDs to 24.3% at 20 A∕cm^2 and 33.7% at 3 A∕cm^2. 14SbHKT1;4, a member of the high‐affinity potassium transporter gene family from Sorghum bicolor, functions to maintain optimal Na^+/K^+ balance under Na^+ stress显示文摘In halophytic plants, the high‐affinity potassium transporter HKT gene family can selectively uptake Kt in the presence of toxic concentrations of Nat. This has so far not been well examined in glycophytic crops. Here, we report the characterization of SbHKT1;4, a member of the HKT gene family from Sorghum bicolor. Upon Nat stress, SbHKT1;4 expression was more strongly upregulated in salt‐tolerant sorghum accession, correlating with a better balanced Nat/ Kt ratio and enhanced plant growth. Heterogeneous expression analyses in mutants of Saccharomyces cerevisiae and Arabidopsis thaliana indicated that overexpressing SbHKT1;4 resulted in hypersensitivity to Nat stress, and such hypersensitivity could be alleviated with the supply of elevated levels of Kt, implicating that SbHKT1;4 may mediate Kt uptake in the presence of excessive Nat. Further electrophysiological evidence demonstrated that SbHKT1;4 could transport Nat and Kt when expressed in Xenopus laevis oocytes. 15Anti-diabetic effects and mechanisms of action of a Chinese herbal medicine preparation JQ-R in vitro and in diabetic KKAy mice显示文摘Refined-JQ(JQ-R) is a mixture of refined extracts from Coptis chinensis(Ranunculaceae),Astragalus membranaceus(Leguminosae) and Lonicera japonica(Caprifoliaceae), the three major herbs of Jin Qi-Jiang Tang tablet, a traditional Chinese medicine(TCM) formula. The mechanisms by which JQ-R regulates glucose metabolism and improves insulin sensitivity were studied in type 2 diabetic KK^(Ay)mice and insulin-resistant L6 myotubes. To investigate the mechanisms by which JQ-R improves insulin sensitivity, a model of insulin-resistant cells induced with palmitic acid(PA) was established in L6 myotubes. Glucose uptake and expression of factors involved in insulin signaling, stress, and inflammatory pathways were detected by immunoblotting. JQ-R showed beneficial effects on glucose homeostasis and insulin resistance in a euglycemic clamp experiment and decreased fasting insulin levels in diabetic KK^(Ay)mice. JQ-R also improved the plasma lipid profiles. JQ-R directly increased the activity of superoxide dismutase(SOD) and decreased malondialdehyde(MDA) as well as inducible nitric oxide synthase(iNOS) levels in insulin-resistant L6 cells, and elevated the insulin-stimulated glucose uptake with upregulated phosphorylation of AKT. The phosphorylation levels of nuclear factor kappa B(NF-κB p65), inhibitor of NF-κB(IκBα), c-Jun N-terminal kinase(JNK1/2) and extracellular-signal-regulated kinases(ERK1/2) were also changed after JQ-R treatment compared with the control group. Together these findings suggest that JQ-R improved glucose and lipid metabolism in diabetic KK^(Ay)mice. JQ-R directly enhanced insulin-stimulated glucose uptake in insulin-resistant myotubes with improved insulin signalling and inflammatory response and oxidative stress. 16Fibroblast-derived CXCL12/SDF-1α promotes CXCL6 secretion and co-operatively enhances metastatic potential through the PI3K/Akt/m TOR pathway in colon cancer显示文摘AIM To investigate the underlying mechanism by which CXCL12 and CXCL6 influences the metastatic potential of colon cancer and internal relation of colon cancer and stromal cells. METHODS Western blotting was used to detect the expression of CXCL12 and CXCL6 in colon cancer cells and stromal cells. The co-operative effects of CXCL12 and CXCL6 on proliferation and invasion of colon cancer cells and human umbilical vein endothelial cells(HUVECs) were determined by enzyme-linked immunosorbent assay,and proliferation and invasion assays. The angiogenesis of HUVECs through interaction with cancer cells and stromal cells was examined by angiogenesis assay. We eventually investigated activation of PI3K/Akt/m TOR signaling by CXCL12 involved in the metastatic process of colon cancer.RESULTS CXCL12 was expressed in DLD-1 cancer cells and fibroblasts. The secretion level of CXCL6 by colon cancer cells and HUVECs were significantly promoted by fibroblasts derived from CXCL12. CXCL6 and CXCL2 could significantly enhance HUVEC proliferation and migration(P < 0.01). CXCL6 and CXCL2 enhanced angiogenesis by HUVECs when cultured with fibroblast cells and colon cancer cells(P < 0.01). CXCL12 also enhanced the invasion of colon cancer cells. Stromal cell-derived CXCL12 promoted the secretion level of CXCL6 and co-operatively promoted metastasis of colon carcinoma through activation of the PI3K/Akt/m TOR pathway.CONCLUSION Fibroblast-derived CXCL12 enhanced the CXCL6 secretion of colon cancer cells,and both CXCL12 and CXCL6 co-operatively regulated the metastasis via the PI3K/Akt/m TOR signaling pathway. 17Formability,microstructure evolution and mechanical properties of wire arc additively manufactured AZ80M magnesium alloy using gas tungsten arc welding显示文摘Wire arc additive manufacturing(WAAM)technology has been used to fabricate the multi-layer single-pass deposited wall of AZ80M magnesium(Mg)alloy by gas tungsten arc welding.The formability,thermal cycles,microstructural evolution and mechanical properties of the WAAM AZ80M Mg alloy were investigated.The results show that there was significant difference in the temperature variation and the geometries between the original several layers and the subsequent deposited layers.Owing to the arc energy input,the interpass temperature rised rapidly and then stabilized at 150℃.As a result,the width of the deposited wall increased and then kept stable.There were obvious differences in the microstructure of the WAAM AZ80M Mg alloy among the top zone,intermediate zone and bottom zone of deposited wall.During the arc deposition process,theβphase of the WAAM AZ80M Mg alloy redissolved due to the cyclic heat accumulation,and then precipitated in the grain boundary.The cyclic heat accumulation also led to weakening of dendrite segregation.From the substrate to the top zone,the hardness of the deposited wall decreased gradually,and the intermediate zone which was the main body of deposited wall had relatively uniform hardness.The tensile properties of the WAAM AZ80M Mg alloy were different between the vertical direction and the horizontal direction.And the maximum ultimate tensile strength of the WAAM AZ80M Mg alloy was 308 MPa which was close to that of the as-extruded AZ80M Mg alloy.Yangyang Guo Gaofeng Quan Yinglong Jiang Lingbao Ren Lingling Fan Houhong Pan 2021Journal of Magnesium and Alloys2021,9,1:10
18Inactivated Sendai Virus Suppresses Murine Melanoma Growth by Inducing Host Immune Responses and Down-regulating β-catenin Expression显示文摘Objective This paper aims to investigate the anti-tumor mechanism of inactivated Sendai virus (Hemagglutinating virus of Japan envelope, HVJ-E) for murine melanoma (B16F10). Methods The murine dendritic cells (DCs) were treated with HVJ-E, and then the cytokines secreted from DCs and costimulation-related molecules on DCs were measured. Meanwhile, the expression of β-catenin in HVJ-E treated murine melanoma cells was detected. In addition, HVJ-E was intratumorally injected into the melanoma on C57BL/6 mice, and the immune cells, CTL response and tumor volume were analyzed. Results HVJ-E injected into B16F10 melanoma obviously inhibited the growth of the tumor and prolonged the survival time of the tumor-bearing mice. Profiles of cytokines secreted by dendritic cells (DCs) after HVJ-E stimulation showed that the number of cytokines released was significantly higher than that elicited by PBS (P<0.05). The co-stimulation-related molecules on DCs were comparable to those stimulated by LPS. Immunohistochemical examinations demonstrated the repression of β-catenin in B16F10 melanoma cells after HVJ-E treatment. Meanwhile, real-time reverse transcription PCR revealed that HVJ-E induced a remarkable infiltration of CD11c positive cells, chemokine ligand 10 (CXCL10) molecules, interleukin-2 (IL-2) molecule, CD4 + and CD8 + T cells into HVJ-E injected tumors. Furthermore, the mRNA expression level of β-catenin in the HVJ-E injected tumors was also down-regulated. In addition, B16F10-specific CTLs were induced significantly after HVJ-E was injected into the tumor-bearing mice. Conclusion This is the first report to show the effective inhibition of melanoma tumors by HVJ-E alone and the mechanism through which it induces antitumor immune responses and regulates important signal pathways for melanoma invasion. 19Arbuscular mycorrhizal fungi combined with exogenous calcium improves the growth of peanut(Arachis hypogaea L.)seedlings under continuous cropping显示文摘The growth and yield of peanut are negatively affected by continuous cropping.Arbuscular mycorrhizal fungi(AMF)and calcium ions(Ca^(2+))have been used to improve stress resistance in other plants,but little is known about their roles in peanut seedling growth under continuous cropping.This study investigated the possible roles of the AMF Glomus mosseae combined with exogenous Ca^(2+)in improving the physiological responses of peanut seedlings under continuous cropping.G.mosseae combined with exogenous Ca^(2+)can enhance plant biomass,Ca^(2+)level,and total chlorophyll content.Under exogenous Ca^(2+)application,the F_v/F_m in arbuscular mycorrhizal(AM)plant leaves was higher than that in the control plants when they were exposed to high irradiance levels.The peroxidase,superoxide dismutase,and catalase activities in AM plant leaves also reached their maximums,and accordingly,the malondialdehyde content was the lowest compared to other treatments.Additionally,root activity,and content of total phenolics and flavonoids were significantly increased in AM plant roots treated by Ca^(2+)compared to either G.mosseae inoculation or Ca^(2+)treatment alone.Transcription levels of AhCaM,AhCDPK,AhRAM1,and AhRAM2 were significantly improved in AM plant roots under exogenous Ca^(2+)treatment.This implied that exogenous Ca^(2+)might be involved in the regulation of G.mosseae colonization of peanut plants,and in turn,AM symbiosis might activate the Ca^(2+)signal transduction pathway.The combination of AMF and Ca^(2+)benefitted plant growth and development under continuous cropping,suggesting that it is a promising method to cope with the stress caused by continuous cropping.CUI Li GUO Feng ZHANG Jia-lei YANG Sha MENG Jing-jing GENG Yun WANG Quan LI Xin-guo WAN Shu-bo 2019Journal of Integrative Agriculture2019,18,2:9
20Advances of polymer binders for silicon-based anodes in high energy density lithium-ion batteries显示文摘Conventional lithium-ion batteries(LIBs)with graphite anodes are approaching their theoretical limitations in energy density.Replacing the conventional graphite anodes with high-capacity Si-based anodes represents one of the most promising strategies to greatly boost the energy density of LIBs.However,the inherent huge volume expansion of Si-based materials after lithiation and the resulting series of intractable problems,such as unstable solid electrolyte interphase layer,cracking of electrode,and especially the rapid capacity degradation of cells,severely restrict the practical application of Sibased anodes.Over the past decade,numerous reports have demonstrated that polymer binders play a critical role in alleviating the volume expansion and maintaining the integrity and stable cycling of Si-based anodes.In this review,the state-of-the-art designing of polymer binders for Si-based anodes have been systematically summarized based on their structures,including the linear,branched,crosslinked,and conjugated conductive polymer binders.Especially,the comprehensive designing of multifunctional polymer binders,by a combination of multiple structures,interactions,crosslinking chemistries,ionic or electronic conductivities,soft and hard segments,and so forth,would be promising to promote the practical application of Si-based anodes.Finally,a perspective on the rational design of practical polymer binders for the large-scale application of Si-based anodes is presented.Yu-Ming Zhao Feng-Shu Yue Shi-Cheng Li Yu Zhang Zhong-Rong Tian Quan Xu Sen Xin Yu-Guo Guo 2021InfoMat2021,3,5:8
