| 1 | Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. | 2 | Development and validation of an endoscopic images-based deep learning model for detection with nasopharyngeal malignancies显示文摘Background:Due to the occult anatomic location of the nasopharynx and frequent presence of adenoid hyperpla-sia,the positive rate for malignancy identification during biopsy is low,thus leading to delayed or missed diagnosis for nasopharyngeal malignancies upon initial attempt.Here,we aimed to develop an artificial intelligence tool to detect nasopharyngeal malignancies under endoscopic examination based on deep learning.Methods:An endoscopic images-based nasopharyngeal malignancy detection model(eNPM-DM)consisting of a fully convolutional network based on the inception architecture was developed and fine-tuned using separate training and validation sets for both classification and segmentation.Briefly,a total of 28,966 qualified images were collected.Among these images,27,536 biopsy-proven images from 7951 individuals obtained from January 1st,2008,to December 31st,2016,were split into the training,validation and test sets at a ratio of 7:1:2 using simple randomiza-tion.Additionally,1430 images obtained from January 1st,2017,to March 31st,2017,were used as a prospective test set to compare the performance of the established model against oncologist evaluation.The dice similarity coef-ficient(DSC)was used to evaluate the efficiency of eNPM-DM in automatic segmentation of malignant area from the background of nasopharyngeal endoscopic images,by comparing automatic segmentation with manual segmenta-tion performed by the experts.Results:All images were histopathologically confirmed,and included 5713(19.7%)normal control,19,107(66.0%)nasopharyngeal carcinoma(NPC),335(1.2%)NPC and 3811(13.2%)benign diseases.The eNPM-DM attained an overall accuracy of 88.7%(95%confidence interval(CI)87.8%-89.5%)in detecting malignancies in the test set.In the prospective comparison phase,eNPM-DM outperformed the experts:the overall accuracy was 88.0%(95%CI 86.1%-89.6%)vs.80.5%(95%CI 77.0%-84.0%).The eNPM-DM required less time(40 s vs.110.0±5.8 min)and exhibited encouraging performance in automatic segmentation of nasopharyngeal malignant area from the background,with an average DSC of 0.78±0.24 and 0.75±0.26 in the test and prospective test sets,respectively.Conclusions:The eNPM-DM outperformed oncologist evaluation in diagnostic classification of nasopharyngeal mass into benign versus malignant,and realized automatic segmentation of malignant area from the background of nasopharyngeal endoscopic images. | Chaofeng Li Bingzhong Jing Liangru Ke Bin Li Weixiong Xia Caisheng He Chaonan Qian Chong Zhao Haiqiang Mai Mingyuan Chen Kajia Cao Haoyuan Mo Ling Guo Qiuyan Chen Linquan Tang Wenze Qiu Yahui Yu Hu Liang Xinjun Huang Guoying Liu Wangzhong Li Lin Wang Rui Sun Xiong Zou Shanshan Guo Peiyu Huang Donghua Luo Fang Qiu Yishan Wu Yijun Hua Kuiyuan Liu Shuhui Lv Jingjing Miao Yanqun Xiang Ying Sun Xiang Guo Xing Lv | 2018 | Cancer Communications2018,38,1: | 9 |
| 3 | Storage and Density of Soil Organic Carbon in Urban Topsoil of Hilly Cities:A Case Study of Chongqing Municipality of China显示文摘Rapid urbanization results in the conversion of natural soil to urban soil,and consequently,the storage and density of the soil carbon pools change.Taking Chongqing Municipality of China as a study case,this investigation attempts to better understand soil carbon pools in hilly cities.First,the vegetated areas in the study area were derived from QuickBird images.Then,topsoil data from 220 soil samples(0-20 cm) in the vegetated areas were collected and their soil organic carbon(SOC) densities were analyzed.Using the Kriging interpolation method,the spatial pattern of SOC was estimated.The results show that the SOC density exhibited high spatial variability in the urban topsoil of Chongqing.First,the SOC density in topsoil decreased according to slope in the order 2°-6° < 25°-90° < 0°-2° < 6°-15° < 15°-25°.Second,the newly developed areas during 2001-2010 had a lower SOC density than the areas built before 1988.Third,urban parks and gardens had a higher SOC density in topsoil,residential green land followed,and scattered street green land ranked last.For hilly cities,the variability of terrain affects the distribution of SOC.The Kriging results indicate that Kriging method combining slope with SOC density produced a high level of accuracy.The Kriging results show that the SOC density to the north of the Jialing River was higher than the south.The vegetated areas were estimated to amount to 73.5 km2 across the study area with an SOC storage of 0.192 Tg and an average density of 2.61 kg/m2. | LIU Yong WANG Cheng YUE Wenze HU Yanyan | 2013 | Chinese Geographical Science2013,23,1: | 6 |

| 10 | H_(2)S-mediated balance regulation of stomatal and non-stomatal factors responding to drought stress in Chinese cabbage显示文摘Increased evidence has shown that hydrogen sulfide(H_(2)S),a novel gasotransmitter,could enhance drought resistance in plants by inducing stomatal closure,with concurrent enhancement of photosynthetic efficiency,but little is known about the mechanism behind this contradictory phenomenon.This study examined the regulating mechanism of H_(2)S in response to drought stress fromstomatal and non-stomatal factors in Chinese cabbage.The results showed that exogenous H_(2)S could increase the accumulation of photosynthetic pigments and alleviate the damage caused by drought stress.It also regulated the expression in transcriptional level and the activity of ribulose 1,5-bisphosphate carboxylase/oxygenase(BrRuBisCO)under drought stress.The large subunit of BrRuBisCO was found to be modified by S-sulfhydration,which might be the reason for its increased enzyme activity.The fluxes of Cl^(−),K^(+),and H^(+)in the guard cells were detected by non-invasive micro-test techniques while under drought stress.The results indicated that H_(2)S signaling induced a transmembrane Cl^(−)and H^(+)efflux and inhibited K^(+)influx,and the Cl^(−)channel was the main responders for H_(2)S-regulated stomatal movement.In conclusion,H_(2)S signal not only activated the ion channel proteins located in the guard cell membrane to induce stomatal closure,but also regulated the transcriptional expression and the activity of RuBisCO,a non-stomatal factor to enhance the photosynthetic efficiency of leaves.There is therefore a beneficial balance between the regulation of H_(2)S signaling on stomatal factors and non-stomatal factors due to drought stress,which needs to be better understood to apply it practically to increase crop yields. | Wenze Zhang Lei Wang Liping Zhang Xiangqun Kong Jiao Zhang Xin Wang Yanxi Pei Zhuping Jin | 2023 | Horticulture Research2023,10,3: | 0 |

This paper presents a constitutive model for studying the anisotropy effects of pure titanium(CP-Ti) sheet on the springback behavior and forming properties during the microstamping process. Thin sheets of CP-Ti specimens with different orientations were examined using uniaxial tensile tests to assess the effects of anisotropy on their mechanical properties. Then an anisotropic constitutive model considering the off-axis elastic modulus was developed based on orthotropic elasticity and Hill's yield criterion. Numerical modeling and simulation of the microstamping process for fabricating multi-channel structures were performed. The effects of anisotropy on the springback of multi-channels were investigated and compared with experimental results;the effects of tool dimension parameters on the formability of microchannel structures were also analyzed. The results showed that the anisotropy of thin titanium sheets causes various degrees of forming loads and springback in the microstamping of microchannels at different orientations.
