1Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. 2Metformin inhibits pancreatic cancer metastasis caused by SMAD4 deficiency and consequent HNF4G upregulation显示文摘Pancreatic ductal adenocarcinoma(PDAC)has poor prognosis due to limited therapeutic options.This study examines the roles of genome-wide association study identified PDAC-associated genes as therapeutic targets.We have identified HNF4G gene whose silencing most effectively repressed PDAC cell invasiveness.HNF4G overexpression is induced by the deficiency of transcriptional factor and tumor suppressor SMAD4.Increased HNF4G are correlated with SMAD4 deficiency in PDAC tumor samples and associated with metastasis and poor survival time in xenograft animal model and in patients with PDAC(log-rank P=0.036;HR=1.60,95%CI=1.03–2.47).We have found that Metformin suppresses HNF4G activity via AMPK-mediated phosphorylation-coupled ubiquitination degradation and inhibits in vitro invasion and in vivo metastasis of PDAC cells with SMAD4 deficiency.Furthermore,Metformin treatment significantly improve clinical outcomes and survival in patients with SMAD4-deficient PDAC(log-rank P=0.022;HR=0.31,95%CI=0.14–0.68)but not in patients with SMAD4-normal PDAC.Pathway analysis shows that HNF4G may act in PDAC through the cell-cell junction pathway.These results indicate that SMAD4 deficiency-induced overexpression of HNF4G plays a critical oncogenic role in PDAC progression and metastasis but may form a druggable target for Metformin treatment.Chengcheng Wang Taiping Zhang Quan Liao Menghua Dai Junchao Guo Xinyu Yang Wen Tan Dongxin Lin Chen Wu Yupei Zhao 2021Protein & Cell2021,12,2:5
3Hydrodynamics and bubble behaviour in a three-phase two-stage internal loop airlift reactor显示文摘Local hydrodynamics of a gas–liquid–solid system,such as bubble circulation regime,gas holdup,liquid velocity and axial profile of solid concentration,are studied in a two-stage internal loop airlift reactor.Empirical correlations for gas holdup and liquid velocity are proposed to ease the reactor design and scale-up.Different bubble circulation regimes were displayed in the first(lower) and second(upper) stages.Increasing superficial gas velocity and solid loading can promote regime transition of the second stage,and the gas holdup of the second stage is higher than that of the lower stage.In addition,the effects of solid loading on bubble behaviour are experimentally investigated for each stage.It is found that bubble size in the downcomer decreases with the presence of solid particles,and bubble size distribution widens under higher superficial gas velocity and lower solid loading.Dan Li Kai Guo Jingnan Li Yiping Huang Junchao Zhou Hui Liu Chunjiang Liu 2018Chinese Journal of Chemical Engineering2018,26,6:3

7Cyst fluid glycoproteins accurately distinguishing malignancies of pancreatic cystic neoplasm显示文摘Pancreatic cystic neoplasms(PCNs)are recognized as precursor lesions of pancreatic cancer,with a marked increase in prevalence.Early detection of malignant PCNs is crucial for improving prognosis;however,current diagnostic methods are insufficient for accurately identifying malignant PCNs.Here,we utilized mass spectrometry(MS)-based glycosite-and glycoform-specific glycoproteomics,combined with proteomics,to explore potential cyst fluid diagnostic biomarkers for PCN.The glycoproteomic and proteomic landscape of pancreatic cyst fluid samples from PCN patients was comprehensively investigated,and its characteristics during the malignant transformation of PCN were analyzed.Under the criteria of screening specific cyst fluid biomarkers for the diagnosis of PCN,a group of cyst fluid glycoprotein biomarkers was identified.Through parallel reaction monitoring(PRM)-based targeted glycoproteomic analysis,we validated these chosen glycoprotein biomarkers in a second cohort,ultimately confirming N-glycosylated PHKB(Asn-935,H5N2F0S0;Asn-935,H4N4F0S0;Asn-935,H5N4F0S0),CEACAM5(Asn-197,H5N4F0S0)and ATP6V0A4(Asn-367,H6N4F0S0)as promising diagnostic biomarkers for distinguishing malignant PCNs.These glycoprotein biomarkers exhibited robust performance,with an area under the curve ranging from 0.771 to 0.948.In conclusion,we successfully established and conducted MS-based glycoproteomic analysis to identify novel cyst fluid glycoprotein biomarkers for PCN.These findings hold significant clinical implications,providing valuable insights for PCN decision-making,and potentially offering therapeutic targets for PCN treatment.Ming Cui Ya Hu Zejian Zhang Tianqi Chen Menghua Dai Qiang Xu Junchao Guo Taiping Zhang Quan Liao Jun Yu Yupei Zhao 2023Signal Transduction and Targeted Therapy2023,8,11:0

11Modulating the Helicity of Sugar-Substituted Perylene Diimide Self-assemblies by Solvent Polarilities显示文摘Modulating the helicity of self-assemblies of conjugated molecules is important for their application in chiral electronics.In this communication,solvent effect has been investigated on how to modulate the helicity of supramolecular assemblies of sugar-substituted perylenediimide(PTCDI-BAG)by using UV-Vis,circular dichroism spectroscopy and transmission electron microscopy.Left-handed helical nanowires are obtained in the mixed solvent system of both N,N-dimethylformamide/ethanol(DMF/ethanol)and N,N-dimethylacetamide/ethanol(DMAC/ethanol).As changing the solvents into dimethyl sulfoxide/ethanol(DMSO/ethanol),ethylene glycol/ethanol(EG/ethanol),or N-methyl-2-pyrrolidone/ethanol(NMP/ethanol),only right-handed heical nanowires are observed.The chirality of supramolecular structures can be tuned by the controlling of the polarity of the solvents.A theoretical calculation is carried out to explain the chiral optical inversion phenomena of PTCDI-BAG aggregates in different solvents,which reveals that reversed CD spectra is due to the difference of clockwise and anticlockwise rotation of the dimer.Qiang Guo Junchao Wang Lingyun Zhu Zhixiang Wei 2015Chinese Journal of Chemistry2015,33,1:0

13Robot-assisted pancreaticoduodenectomy in situs inversus totalis patient with pancreatic cancer显示文摘Pancreatic cancer (PC) combined with situs inversus totalis (SIT) is rare, and the treatment strategy is obscure. We report the first patient with PC and SIT who underwent robot-assisted pancreaticoduodenectomy. A 57-year-old male patient presented to our hospital with epigastric pain, nausea, and weight loss over 1 month. Preoperative diagnostic modalities revealed a resectable pancreatic ductal adenocarcinoma at the head and neck junction of the pancreas. The patient also had a rare condition called SIT. Then, the patient underwent Da Vinci robot-assisted pancreaticoduodenectomy (RPD) in our center. After the operation, the patient recovered well without complications. Until now, the patient was followed up 5 months, and the quality of life was well without tumor recurrence or metastasis. 14Fucoidan-based dual-targeting mesoporous polydopamine for enhanced MRI-guided chemo-photothermal therapy of HCC via P-selectin-mediated drug delivery显示文摘The development of novel theranostic agents with outstanding diagnostic and therapeutic performances is still strongly desired in the treatment of hepatocellular carcinoma(HCC).Here,a fucoidan-modified mesoporous polydopamine nanoparticle dual-loaded with gadolinium iron and doxorubicin(FMPDA/Gd^(3+)/DOX)was prepared as an effective theranostic agent for magnetic resonance imaging(MRI)-guided chemo-photothermal therapy of HCC.It was found that FMPDA/Gd^(3+)/DOX had a high photothermal conversion efficiency of 33.4%and excellent T1-MRI performance with a longitudinal relaxivity(r1)value of 14.966 m M^(-1)·s^(-1).Moreover,the results suggested that FMPDA/Gd^(3+)/DOX could effectively accumulate into the tumor foci by dual-targeting the tumor-infiltrated platelets and HCC cells,which resulted from the specific interaction between fucoidan and overexpressed p-selectin receptors.The excellent tumor-homing ability and MRI-guided chemo-photothermal therapy therefore endowed FMPDA/Gd^(3+)/DOX with a strongest ability to inhibit tumor growth than the respective single treatment modality.Overall,our study demonstrated that FMPDA/Gd^(3+)/DOX could be applied as a potential nanoplatform for safe and effective cancer theranostics.Gaofeng Shu Lin Shen Jiayi Ding Junchao Yu Xiaoxiao Chen Xiaoju Guo Enqi Qiao Yaning Chen Chenying Lu Zhongwei Zhao Yongzhong Du Minjiang Chen Jiansong Ji 2022Asian Journal of Pharmaceutical Sciences2022,17,6:0

17Pancreatic cancer–Laparoscopic resection显示文摘Objective: To assess current role of laparoscopic resection for pancreatic cancer, so as to improve the surgical management of pancreatic cancer. Methods: A comprehensive review of articles from PubMed was carried out. Results: Cur- rently, the advantages of a complete laparoscopic pancreatoduodenectomy (LPD) are still outweighed by the morbidity associ- ated with the procedure. However, laparoscopic distal pancreatectomy (LDP) offers patients benefits in terms of postoperative recovery and the length of hospital stay with similar morbidity and mortality to open surgery. Hand-assisted laparoscopic sur- gery can help to overcome the limitation of a complete laparoscopic surgery while maintaining a minimally invasive approach. Conclusion: Current literature suggests that laparoscopic resection of pancreatic cancer is feasible and safe in experienced hands.
