1α-Synuclein aggregation and transmission in Parkinson’s disease: a link to mitochondria and lysosome显示文摘The presence of intraneuronal Lewy bodies(LBs) and Lewy neurites(LNs) in the substantia nigra(SN) composed of aggregatedα-synuclein(α-syn) has been recognized as a hallmark of pathological changes in Parkinson’s disease(PD). Numerous studies have shown that aggregated α-syn is necessary for neurotoxicity. Meanwhile, the mitochondrial and lysosomal dysfunctions are associated with α-syn pathogenicity. The hypothesis that α-syn transmission in the human brain contributes to the instigation and progression of PD has provided insights into PD pathology. 2Poly-PR in C9ORF72-Related Amyotrophic Lateral Sclerosis/Frontotemporal Dementia Causes Neurotoxicity by Clathrin-Dependent Endocytosis显示文摘GGGGCC repeat expansions in the C9 ORF72 gene are the most common cause of amyotrophic lateral sclerosis and frontotemporal dementia(c9 ALS/FTD). It has been reported that hexanucleotide repeat expansions in C9 ORF72 produce five dipeptide repeat(DPR) proteins by an unconventional repeat-associated non-ATG(RAN)translation. Within the five DPR proteins, poly-PR and poly-GR that contain arginine are more toxic than the other DPRs(poly-GA, poly-GP, and poly-PA). Here, we demonstrated that poly-PR peptides transferred into cells by endocytosis in a clathrin-dependent manner, leading to endoplasmic reticulum stress and cell death. In SH-SY5 Y cells and primary cortical neurons, poly-PR activated JUN amino-terminal kinase(JNK) and increased the levels of p53 and Bax. The uptake of poly-PR peptides by cells was significantly inhibited by knockdown of clathrin or by chlorpromazine, an inhibitor that blocks clathrin-mediated endocytosis. Inhibition of clathrin-dependent endocytosis by chlorpromazine significantly blocked the transfer of poly-PR peptides into cells, and attenuated poly-PRinduced JNK activation and cell death. 3Effect of a compound starter cultures inoculation on bacterial profile and biogenic amine accumulation in Chinese Sichuan sausages显示文摘Effects of a starter culture composed of Lactobacillus sakei,Pediococcus pentosaceus,Staphylococcus xylosus and S.carnosus at the ratios(m/m)of 1:1:1:1 on bacterial profile and biogenic amine accumulation in Chinese Sichuan sausages were demonstrated.In starter culture-inoculated sausages,Lactobacillus spp.,Pediococcus spp.and Staphylococcus spp.were predominant bacterial genus and their relative abundances ranged from 70.14%to 25.98%,57.66%to 14.08%,and 15.71%to 13.40%during fermentation,respectively.Accordingly,Lactobacillus spp.and Weissella spp.were predominant bacterial genus and their relative abundances ranged from 63.14%to 24.70%and 49.40%to 11.96%during spontaneous fermentation,respectively.Furthermore,the abundances of undesirable microorganisms such as Yersinia spp.,Enterobacter spp.,Acietobacter spp.and Psychrobacter spp.were lower than that of the control.The levels of histamine,putrescine,tyramine and cadaverine in Chinese Sichuan sausages with starter cultures inoculation were significantly lower(P<0.05)than that of the control,and were decreased by 83.09%,69.38%,51.87%and 57.20%,respectively,at the end of the ripening.These results revealed that the starter cultures inoculation was a better alternative for microbial quality improvement and biogenic amine reduction of Chinese Sichuan sausages with good sensory attributes.Hongyang Ren Yuanpeng Deng Xinhui Wang 2022Food Science and Human Wellness2022,11,2:2

8Aristolochic acids exposure was not the main cause of liver tumorigenesis in adulthood显示文摘Aristolochic acids(AAs) have long been considered as a potent carcinogen due to its nephrotoxicity. Aristolochic acid I(AAI) reacts with DNA to form covalent aristolactam(AL)-DNA adducts,leading to subsequent A to T transversion mutation, commonly referred as AA mutational signature. Previous research inferred that AAs were widely implicated in liver cancer throughout Asia. In this study, we explored whether AAs exposure was the main cause of liver cancer in the context of HBV infection in China's Mainland. Totally 1256 liver cancer samples were randomly retrieved from 3 medical centers and a refined bioanalytical method was used to detect AAI-DNA adducts. 5.10% of these samples could be identified as AAI positive exposure. Whole genome sequencing suggested 8.41% of 107 liver cancer patients exhibited the dominant AA mutational signature, indicating a relatively low overall AAI exposure rate. In animal models, long-term administration of AAI barely increased liver tumorigenesis in adult mice, opposite from its tumor-inducing role when subjected to infant mice. Furthermore, AAI induced dose-dependent accumulation of AA-DNA adduct in target organs in adult mice, with the most detected in kidney instead of liver. 9Experimental Investigation on Pouch Lithium-ion Battery Thermal Management with Mini-Channels Cooling Plate Based on Heat Generation Characteristic显示文摘An electrochemical thermal coupling model of lithium battery was established to study the heat generation characteristic in this study.The simulation results showed that the heat generation density of the battery increased with the discharge rate.With the discharge process,the heat generation density of the battery increased continuously.With 2.5C discharge rate,the heat generation density at the end of discharge was 1.82 times of that at the beginning of discharge.The heat generation density at different areas of the battery was not uniform and 46%of the total ohmic heat was generated near the electrode tabs.A cooling plate with variable mini-channels was designed to improve the temperature non-uniformity caused by the heat generation characteristic.A cooling plate with uniform mini-channels was designed for compared experiment.The experiments were conducted with deionized water and refrigerant R141b and carried out with 1.5C,2C and 2.5C discharge rates.Experimental results showed that the cooling plate with variable mini-channels had a better cooling performance in both single-phase and two-phase cooling conditions.REN Honglei JIA Li DANG Chao YANG Chengliang JIA Hongyang LIU Junjie 2022Journal of Thermal Science2022,31,3:1
10An RNA-RNA crosstalk network involving HMGB1 and RICTOR facilitates hepatocellular carcinoma tumorigenesis by promoting glutamine metabolism and impedes immunotherapy by PD-L1+ exosomes activity显示文摘Hepatocellular carcinoma(HCC)is the global leading cause of cancer-related deaths due to the deficiency of targets for precision therapy.A new modality of epigenetic regulation has emerged involving RNA-RNA crosstalk networks where two or more competing endogenous RNAs(ceRNAs)bind to the same microRNAs.However,the contribution of such mechanisms in HCC has not been well studied.Herein,potential HMGB1-driven RNA-RNA crosstalk networks were evaluated at different HCC stages,identifying the mT0RC2 component RICTOR as a potential HMGB1 ceRNA in HBV^(+)early stage HCC.Indeed,elevated HMGB1 mRNA was found to promote the expressio n of RICTOR mRNA through competitively bin ding with the miR-200 family,especially miR-429.Functio nal assays emplo ying overexpressi on or in terference strategies dem on strated that the HMGB1 and RICTOR 3zuntra nslated regions(UTR)epigenetically promoted the malignant proliferation,self-renewal,and tumorigenesis in HCC cells.Intriguingly,in terference agai nst HMGB1 and RICTOR in HCC cells promoted a stron ger an ti-PD-L1 immuno therapy resp on se,which appeared to associate with the production of PD-L1^(+)exosomes.Mechanistically,the HMGB1-driven RNA-RNA crosstalk network facilitated HCC cell glutamine metabolism via dual mechanisms,activating a positive feedback loop involving mT0RC2-AKT-C-MYC to upregulate glutamine synthetase(GS)expression,and inducing mTORCI signaling to derepress SIRT4 on glutamate dehydrogenase(GDH).Meanwhile,this crosstalk network could impede the efficacy of immunotherapy through mTORCI-P70S6K dependent PD-L1 production and PD-L1^(+)exosomes activity.In conclusion,our study highlights the non-coding regulatory role of HMGB1 with implicatio ns for RNA-based therapeutic targeting together with a predictio n of an ti-PD-L1 immuno therapy in HCC.Yanping Wei Xuewu Tang Yibin Ren Yun Yang Fengliang Song Jingbo Fu Shuowu Liu Miao Yu Jing Chen Suyang Wang Kecheng Zhang Yexiong Tan Zhipeng Han Lixjn Wei Baohua Zhang Zhangjun Cheng Liang Li Hongyang Wang 2022Signal Transduction and Targeted Therapy2022,7,1:0
11Dynamic compression behavior of TiZrNbV refractory high-entropy alloys upon ultrahigh strain rate loading显示文摘In this study,the dynamic compressive response behavior of a body-centered cubic(BCC)single-phase TiZrNbV refractory high-entropy alloy(RHEA)was investigated under impact at speeds of 313-1584 m s^(-1)using two-stage,gas-gun-driven,high-speed plate-impact experiments;recovery sample analysis;and theoretical calculations.The strain rate and pressure were approximately 10^(7) s^(−1) and 5.07-29.37 GPa,respectively.The results showed that the TiZrNbV RHEA had a Hugoniot elastic limit of 4.12-5.86 GPa and a spall strength of 1.84-2.03 GPa.The initial yield strength of the alloy showed a strong strain-rate dependence and could be described by the modified Zerilli-Armstrong model,while the phonon-damping effect was the main reason for its high strain-rate sensitivity.Microstructural analysis showed that the dynamic deformation of the TiZrNbV RHEA was controlled by the dislocation slip,dislocation proliferation,intersection of the deformation bands,and grain refinement.The analysis also showed that the intergranular,transgranular,and mixed-type cracks dominated the spall failure of the material.The dynamic Hall-Petch effect and pinning from the lattice distortion led to high dynamic yield strength.The critical strain rate for the phonon drag effect was positively related to the relative atomic mass and local strain field of the metals.Within the experimental loading range,the RHEA showed good structural stability,and simultaneously,the theoretical calculation method for the equation of state based on a cold-energy mixture could accurately predict its shock-response behavior.The valence-electron concentration(VEC)had a direct effect on the shock-compression properties of the HEAs;higher VEC implied more difficulty in compressing the HEAs.The findings of this study provide insights into understanding the mechanical response characteristics of RHEAs under extreme conditions such as high-speed impact and ultrahigh strain-rate loading.Kerong Ren Hongyang Liu Rong Ma Sen Chen Siyuan Zhang Ruixin Wang Rong Chen Yu Tang Shun Li Fangyun Lu 2023Journal of Materials Science & Technology2023,,30:0

13LncRNA ZFAS1 regulates cardiomyocyte differentiation of human embryonic stem cells显示文摘Background:Cardiomyocytes derived from human embryonic stem cells(hESCs)are regulated by complex and stringent gene networks during differentiation.Long non-coding RNAs(lncRNAs)exert critical epigenetic regulatory functions in multiple differentiation processes.However,the involvement of lncRNAs in the differentiation of hESCs into cardiomyocytes has not yet been fully elucidated.Here,we identified the key roles of ZFAS1(lncRNA zinc finger antisense 1)in the differentiation of cardiomyocytes from hESCs.Methods:A model of cardiomyocyte differentiation from stem cells was established using the monolayer differentiation method,and the number of beating hESCs-derived cardiomyocytes was calculated.Gene expression was analyzed by quantitative real-time PCR(qRTPCR).Immunofluorescence assays were performed to assess the expression of cardiac troponin T(cTnT)andα-actinin protein in cardiomyocytes.Results:qRT-PCR showed that ZFAS1 expression in the mesoderm was significantly higher than that in embryonic stem cells,cardiac progenitor cells,and cardiomyocytes.Knockdown of ZFAS1 inhibited cardiomyocyte differentiation from hESCs,which was characterized by reduced expression of the cardiac-specific markers cTnT,α-actinin,myosin heavy chain 6(MYH6),and myosin heavy chain 7(MYH7).In contrast,ZFAS1 overexpression remarkably increased the percentage of spontaneously beating cardiomyocytes.In terms of the mechanism,we found that ZFAS1 is an antisense lncRNA at the 5′end of the protein-coding gene ZNFX1.Knockdown of ZFAS1 could increase the mRNA expression level of ZNFX1.Furthermore,qRT-PCR demonstrated that the silencing of ZNFX1 led to an increase in cardiac-specific markers that predicted the promotion of cardiomyocyte differentiation.Conclusion:Altogether,these data suggest that lncRNA-ZFAS1 is required for cardiac differentiation by functionally inhibiting the expression of ZNFX1,which may provide a reference for the treatment of heart disease to a certain extent.YANG CAO YINING LIU YANG YU XIAOFEI GUO XIUXIU WANG WENYA MA HANJING LI ZHONGYU REN XINLU GAO SIJIA LI HAOYU JI HONGYANG CHEN HONG YAN YANAN TIAN XIN WANG BENZHI CAI 2023BIOCELL2023,47,6:0
