2Base-Editing-Mediated Artificial Evolution of OsALS1 In Planta to Develop Novel Herbicide-Tolerant Rice Germplasms显示文摘Recently developed CRISPR-mediated base editors,which enable the generation of num erous nucleotide changes in target genomic regions,have been widely adopted for gene correction and generation of crop germ plasms containing im portant gain-of-function genetic variations.How ever,to engineer target genes with unknown functional SNPs remains challenging.To address this issue,we present here abase-e diting-mediated gene evolution(BEMGE)m ethod,employing both Cas9n-based cytosine and adenine base editors as well as a single-guide RNA(sgRNA)library tiling the full-length coding region,for developing novel rice germ plasm swith mutations in any endogenous gene.To this end,OsALS1 was artificially evolved in rice cells using BEMGE through both Agrobacterium-mediated and particle-bom bardment-mediated transform ation.Four different types of amino acid substitutions in the evolved OsALS1,derived from two sites that have never been targeted by natural or human selection during rice dom estication,were identified,conferring varying levels of tolerance to the herbicide bispyribac-sodium.Furtherm ore,the P171F substitution identified in a strong OsALS1 allele was quickly introduced into the commercial rice cultivar Nangeng 46 through precise base editing w ith the corresponding base editor and sgRNA.Collectively,these data indicate great potential of BEMGE in creating important genetic variants of target genes for crop improvement.Yongjie Kuang Shaofang Li Bin Ren Fang Yan Carl Spetz Xiangju Li Xueping Zhou Huanbin Zhou 2020Molecular Plant2020,13,4:37

4Cas9-NG Greatly Expands the Targeting Scope of the Genome-Editing Toolkit by Recognizing NG and Other Atypical PAMs in Rice显示文摘CRISPR technologies enabling precise genome manipulation are valuable for gene function studies and molecular crop breeding. However, the requirement of a protospacer adjacent motif (PAM)y such as NGG and TTN, for Cas protein recognition restricts the selection of targetable genomic loci in practical applications of CRISPR technologies. Recently Cas9-NG, which recognizes a minimal NG PAM, was reported to expand the targeting space of genome editing in human cells, but it remains unclear whether this Cas9 variant can be used in plants. In this study, we evaluated the nuclease activity of Cas9-NG toward various NGN PAMs by targeting endogenous genes in transgenic rice. We found that Cas9-NG edits all NGG, NGA, NGT, and NGC sites with impaired activity, while the gene-edited plants were dominated by monoallelic mutations. Cas9-NG-engineered base editors were then developed and used to generate O s B Z R I gainof- function plants that can not be created by other available Cas9-engineered base editors. Moreover, we showed that a Cas9-NG-based transcriptional activator efficiently upregulated the expression of endogenous target genes in rice. In addition, we discovered that Cas9-NG recognizes NAC, NTG, NTT, and NCG apart from NG PAM. 5Improvement of the matrix and the interface quality of a Cu/Al composite by the MARB process显示文摘The matrix accumulative roll bonding technology (MARB) can improve the matrix performance of metal com- posite and strengthen the bonding quality of the interface. In this research, for the first time, the technology of MARB was proposed. A sound Cu/Al bonding composite was obtained using the MARB process and the bonding characteristic of the interface was studied using scanning electricity microscope (SEM) and energy-dispersive spectroscopy (EDS). The result indicated that accumulation cycles and diffusion annealing temperature were the most important factors for fabricating a Cu/Al composite material. 6High-efficiency and multiplex adenine base editing in plants using new TadA variants显示文摘Recently reported adenine base editors(ABEs)exhibit powerful potential for targeted gene correction as well as developing gain-of-function mutants and novel germplasms for both gene function studies and crop breeding.However,editing efficiency varies significantly among different target sites.Here,we investigated the activities of three evolved E.coli adenosine deaminase TadA variants(TadA8e,TadA8.17,and TadA8.20)side-by-side in transgenic rice.We found that TadA8e outperforms TadA8.17 and TadA8.20,and induces efficient A-to-G conversion at all tested sites in the rice genome,including those that were un-editable by ABE7.10 in our previous experiments.Furthermore,V82S/Q154R mutations were incorporated into TadA8e,resulting in a new variant that we named TadA9.Our data show that TadA9 is broadly compatible with CRISPR/SpCas9,CRISPR/SpCas9-NG,and CRISPR/SpRY,as well as CRISPR/ScCas9 nickase systems,achieving comparable or enhanced editing in a larger editing window at diverse PAM sites as compared with TadA8e.Finally,TadA9 was used to simultaneously install novel SNPs in four endogenous herbicide target genes in the commercial rice cultivar Nangeng 46 for potential field application in.weed control.Collectively,we successfully generated a series of novel ABEs that can efficiently edit adenosines in the rice genome.Our findings suggest that TadA9 and TadA8e have great potentials in the development of plant base editors and crop molecular breeding.Daqi Yan Bin Ren Lang Liu Fang Yan Shaofang Li Guirong Wang Wenxian Sun Xueping Zhou Huanbin Zhou 2021Molecular Plant2021,14,5:8
7Effect of RE on the ignition-proof, microstructure and properties of AZ91D magnesium alloy显示文摘The magnesium alloy is prone to burn during die-casting, which limits its applications severely, so the effect of adding rare earth (RE) on the ignition-proof of AZ91D Mg alloy is studied. The results indicate that the addition of mischmetal RE elements has a remarkable influence on the ignition-proof property of the magnesium alloy. It is found that the ignition temperature of the magne- sium alloy can be greatly raised by adding a proper amount of RE. When the amount is 0.1 wt%, the ignition temperature reaches 877°C which is 206°C higher than that of AZ91D without RE and the mechanical properties of the alloy are also improved. 8Two-dimensional simulation on the rolling process of semi-solid 60Si2Mn by finite element method显示文摘The effect of various process variables on the law of metal flow for semi-solid rolling 60Si2Mn was studied by finite element method. Semi-solid 60Si2Mn can be described as compressible rigid visco-plastic porous material saturated with liquid. In terms of thermo-mechanical coupling condition, the distributions of stress, velocity and temperature were studied using software MARC. 9Immersing Treatment of Steel Plate Surface in Steel-Aluminum Solid to Liquid Bonding显示文摘The interfacial status of the steel-aluminum solid to liquid bonding plates (their steel plate surfaces were or were not immersed in flux aqueous solution) were measured by using SEM (Scanning Electron Microscope) and X-ray diffraction . The results showed that the layer of flux (the minimum thickness was 15 μm on the steel plate surface) could protect the steel plate surface from oxidizing effectively at high temperature in solid to liquid bonding. The melt temperatUre of the flux should be lower than 580 ℃ so that it could be melted and removed completely. 13Effects of Long-term Fertilization on Phosphorus Accumulation and Migration in Purple Soil显示文摘Based on a long-term location test lasting for 20 years,the accumulation and migration of phosphorus in purple soil was studied.The results showed that P_2O_5 input in paddy-upland rotation was 120.0 kg/(hm^2· a)(a conventional level of phosphorus application),and P had a surplus of 53.9 kg/(hm^2· a).Phosphorus accumulating in soil increased available phosphorus(Olsen-P) content,and the positive correlation between P changing balance and Olsen-P change in wheat-rice rotation was extremely significant.The mixed application of organic fertilizer and chemical P fertilizer can speed up accumulation of available phosphorus in soil.Olsen-P content in all treatments with P fertilizer was higher in 0-60 cm soil than that of treatments without P fertilizer,and Phosphorus applied in surface soil moved to soil at a depth of 60 cm,increasing risk of P leakage and loss.The migration of phosphorus in soil is affected by P input,type of organic fertilizer and planting method.The quantities of phosphorus moving down increased with the increase of P input,and the application of pig manure can lead to migration of phosphorus in soil more easily compared with straw.The migration of phosphorus from soil to water mainly occurred in the first ten days after rice was transplanted,and paddy field drainage should not be conducted in the first 30 days after rice was transplanted.Li Xueping Zou Meiling Ren Jiayun 2015Meteorological and Environmental Research2015,6,8:1
14Electrochemical Detection of Superoxide Anion Released by Living Cells by Manganese(Ⅲ) Tetraphenyl Porphine as Superoxide Dismutase Mimic显示文摘Superoxide anion,one of the most active reactive oxygen species,is associated with the development of many diseases.So monitoring superoxide anion in living cells is of great significance for the pathological research of many diseases.In this work,a new non-enzymatic sensor tor the detection of superoxide anion(O^·-2)was developed,which was iabricated by the nanocomposites composed of manganese(Ⅲ)tetraphenyl porphine(MnTPP)as superoxide dismutase mimic and electrochemical reduced graphene oxide(ERGO)as electrode support material to modify the glassy carbon electrode(GCE).The electrochemical behavior of the fabricated electrode(MiiTPP/ERGO/GCE)was performed by electrochemical impedance spectroscopy(EIS)and cyclic voltammetry(CV),which revealed that MnTPP/ERGO/GCE possessed good catalytic ability to theelectrochemical reduction of O^·-2.The MnTPP/ERGO/GCE showed excellent electroanalysis perfonnance towards O^·-2 using the technique of diflerential pulse voltanimetry(DPV)with a linear relationship in the range of 0.2—110.0μmol/L,a sensitivity of 445 μA· L·mmol^-1·cm^- 2 and a detection limit of 0.039 μmol/L(S/N=3).The real-time monitoring of O^·-2 from MCF-7 breast cancer cells stimulated by zymosan was realized in this work,which indicates that the MnTPP/ERGO/GCE hold potential application for electrochemical quantification of superoxide anions in biological applications.CUI Min REN Jujie WEN Xiaofang LI Na XING Yifei ZHANG Cong HAN Yuanyuan JI Xueping 2020Chemical Research in Chinese Universities2020,36,5:1
15Superplastic deformation of commercial OOCr22Ni5Mo3N0.17 duplex stainless steel显示文摘The superplastic behavior of a commercial duplex stainless steel has been studied by means of isothermal hot tensile testat temperatures of 850-1050℃ for the initial strain rates ranging from 3×l0-4 s-1 to 5X10-2 s-1. At 960℃, the best superplastic de-formation that caused the maximum elongation greater than 840% was obtained for an initial strain rate of 1.2×10-3 s-1. At 850℃, thebest elongation 500% was achieved for an initial strain rate of 2.5×10-3 s-1. During the deformation in higher temperature region,coarse γ grains formed during the prior treatments were broken into spherical particles, resulting in a homogeneous dispersion of γparticles within the δ-ferrite matrix. However, at lower temperatures between 800 and 950℃, the σ phase was formed through theeutectoid decomposition of δ→γ+σ, resulting finally in the stable equiaxed micro-duplex structures with δ/γ and γ/σ, respectively.The precipitation of the σ phase played an important role in improving the superplasticity at 850℃. The strain-rate sensitivity coeffi-cient, m-values, were also determined by the strain rate change tests. 19Comprehensive and deep profiling of the plasma proteome with protein corona on zeolite NaY显示文摘Proteomic characterization of plasma is critical for the development of novel pharmacodynamic biomarkers.However,the vast dynamic range renders the profiling of proteomes extremely challenging.Here,we synthesized zeolite NaY and developed a simple and rapid method to achieve comprehensive and deep profiling of the plasma proteome using the plasma protein corona formed on zeolite NaY.Specifically,zeolite NaY and plasma were co-incubated to form plasma protein corona on zeolite NaY(NaY-PPC),followed by conventional protein identification using liquid chromatography-tandem mass spectrometry.NaY was able to significantly enhance the detection of low-abundance plasma proteins,minimizing the“masking”effect caused by high-abundance proteins.The relative abundance of middleand low-abundance proteins increased substantially from 2.54%to 54.41%,and the top 20 highabundance proteins decreased from 83.63%to 25.77%.Notably,our method can quantify approximately 4000 plasma proteins with sensitivity up to pg/mL,compared to only about 600 proteins identified from untreated plasma samples.A pilot study based on plasma samples from 30 lung adenocarcinoma patients and 15 healthy subjects demonstrated that our method could successfully distinguish between healthy and disease states.In summary,this work provides an advantageous tool for the exploration of plasma proteomics and its translational applications.Congcong Ma Yanwei Li Jie Li Lei Song Liangyu Chen Na Zhao Xueping Li Ning Chen Lixia Long Jin Zhao Xin Hou Li Ren Xubo Yuan 2023Journal of Pharmaceutical Analysis2023,13,5:0
20Hormones and carbohydrates synergistically regulate the formation of swollen roots in a Chinese cabbage translocation line显示文摘The genus Brassica contains a rich diversity of species and morphological types,including leaf,root,and oil crops,all of which show substantial phenotypic variation.Both Chinese cabbage and cabbage are typical leaf-type crops with normal roots.We created translocation lines based on interspecific crosses between Chinese cabbage and cabbage and identified qdh225,which exhibited a swollen-root phenotype.The swollen root of qdh225 contained a large number of granular substances,and the formation of its irregular morphological tissue was caused by a thickening of the phloem.Transcriptomic and metabolomic data suggested that differential expression of genes encoding nine types of enzymes involved in starch and sucrose metabolism caused changes in starch synthesis and degradation in the swollen root.These genes jointly regulated sucrose and starch levels,leading to significant enrichment of starch and soluble proteins in the swollen root and a reduction in the content of soluble sugars such as d-glucose and trehalose 6-phosphate.A significant increase in auxin(IAA)and abscisic acid(ABA)contents and a decrease in gibberellin(GA)content in the swollen root likely promoted the differential expression of genes associated with hormone signal transduction,thereby regulating the development of the swollen root.Taken together,our data suggest that accumulation of IAA and ABA and reduction in GA promote swollen root formation by regulating hormone-mediated signaling,leading to a thickening of phloem,root enlargement,and substantial accumulation of starch and soluble proteins.The latter provide materials,energy,and nutrient sources for the development of swollen roots.Xiaojing Ren Wei Ma Shuxin Xuan Dandan Li Yanhua Wang Yuanchao Xu Daling Feng Jianjun Zhao Xueping Chen Shuangxia Luo Shuxing Shen Aixia Gu 2023Horticulture Research2023,10,8:0
